Biphasic Release Characteristics of Dual Drug-loaded Alginate Beads

  • The dual drug-loaded alginate beads simultaneously containing drug in inner and outer layers were prepared by dropping plain (single-layered) alginate beads into $CaCl_2$ solution. The release characteristics were evaluated in simulated gastric fluid for 2 h followed by intestinal fluids thereafter for 12 h. The surface morphology and cross section of dual drug-loaded alginate beads was also investigated using scanning electron microscope (SEM). The poorlv water-soluble ibuprofen was chosen as a model drug. The surface of single-layered and dual drug-loaded alginate beads showed very crude and roughness, showing aggregated particles, surface cracks and rough crystals. The thickness of dual drug-loaded alginate beads surrounded by outer layer was ranged from about 57 to 329mcm. The distinct chasm between inner and outer layers was also observed. In case of single-layered alginate bead, the drug was not released in gastric fluid but was largely released in intestinal fluid. However, the release rate decreased as the reinforcing $Eudragit^{\circledR}$ polymer contents increased. When the plasticizers were added into polymer, the release rate largely decreased. The release rate of dual drug-loaded alginate beads was stable in gastric fluid for 2 h but largely increased when switched in intestinal fluid. The drug linearly released for 4 h followed by another linear release thereafter, showing a distinct biphasic release characteristics. There was a difference in the release profiles between single-layered and dual drug-loaded alginate beads due to their structural shape. However, this biphasic release profiles were modified by varying formulation compositions of inner and outer layer of alginate beads. The release rate of dual drug-loaded alginate beads slightly decreased when the outer layer was reinforced with $Eudragit^{\circledR}$ RS1OO polymers. In case of dual drug-loaded alginate beads with polymer-reinforced outer layer only, the initial amount of druc released was low but the initial release rate (slope) was higher due to more swellable inner cores when compared to polymer-reinforced inner cores. The current dual drug-loaded alginate beads may be used to deliver the drugs in a time dependent manner.

  • One of the characteristics features of Chinese Medicine is recognized as the practical use of combinations of crude drugs in the forms of various prescriptions according to the symptoms of illness. Such crude drugs are mostly dried plant roots, rhizomes, leaves, fruits and seeds, sometimes dried animla organ preparations even fossils or minerals. In the oldest book of Chinese Medicine, Sang Han Ron, 113 prescriptions were described in which about 80 crude drugs were included, and in other old literature, Shin Nong Bon Tcho Keun, 363 crude drugs were recorded. Such drugs, in particular, those included in Sang Han Ron are generally low toxic, and the prescriptions are acting mild with less side effects, but obviously not so easy task to determine the effect of the indivisual drug from the total effects of various prescriptions which are generally consisted of many kinds of crude drugs. About 200 years ago, in the middle of Edo era, a Japanese medical doctor, Yoshimasu Todo wrote a series of books named "Yakucho", in which he described the essential various prescriptions in which the certain drugs plays the main role. This book is very suggestive to evaluate the effects of some Chinese drugs. The actual effects of Chinese medicine and drugs have widely been recognized by the East Asian peoples during their long age experiences, but still it has not been studied sufficiently by modern scientific methods to give generally acceptable evidences. During past some years, my collaborators and I have been engaged to study some well-known Chinese drugs chemically to find out the active principles in determining their chemical structures under the collaboration of pharmacologists, Prof. K. Takagi and his coworkers. In the present lecture, I would like to present two topics from our recent investigations: The studies on the principles of Paeony and Ginseng roots.

한약재중의 잔류농약 분석 - GC에 의한 18종 잔류농약의 분석 - (Determination of residual pesticides in crude drugs - Gas chromatographic Analysis of 18 pesticides -)

  • This study was carried out to determine the 11 organochlorine, 7 organophosphorus residual pesticides in 251 crude drugs. These residual pesticides in herbal drugs were extracted with acetonitrile and the extracts were cleaned up via LC-florisil solid phase extraction column. The prepared samples were assayed for pesticide residues using GC-ECD, NPD with capillary column and identified by GC-MSD. Recoveries were $63.9{\sim}111.5%$ in the organochlorine pesticides and $69.8{\sim}92.4%$ in the organophosphorus pesticides, and detection limits were $0.001{\sim}0.65\;ppm$ in the organochlorine pesticides and $0.0009{\sim}0.0074\;ppm$ in the organophosphorus pesticides. Pesticide residues were detected in 9 cases.

  • To get basic data for the utilization of S. herbacea L. as a raw material in food and Chinese herbs, chemical compositions of its leaves, stem and root were investigated. Leaves had the highest level of moisture and the lowest bevel of total sugar. The crude protein and crude lipid contents of the stem were similar to those of the root. Crude ash and salt contents (dry basis) in leaves were considerably higher than those of the stem and root. Total amino acid contents of leaves, stem and root were 1,270 mg/100 g, 1,525 mg/100 g, and 1,569 mg/100 g, respectively. Although the amino acid compositions of loaves, stein, and root were different, their major amino acids were glutamic acid, leuicine, isoleucine and aspartic acid. The rich minerals in leaves, stem and root were Na, K and Ca.

  • 'Si Ho'(柴胡) is one of the chinese crude drugs used mainly to cure inflammation, fever, cough, hepatitis, jaundice, etc. Though the botanical origin of the crude drug has been considered to be Bupleurum species of Umbelliferae, there has been no pharmacognostical study. To clarify the botanical origin of 'Si Ho', the morphological and anatomical characteristics of the roots of Bupleurum species growing in Korea, such as Bupleurum euphorbioides Nakai, Bupleurum falcatum L., Bupleurum leveillei Bois., Bupleurum longiradiatum Turcz. and Bupleurum scorzoneraefolium Wild. were studied. As a result, it is authentified that 'Si Ho' was proved to be the root of Bupleurum falcatum and B. scorzoneraefolium and 'Jug Si Ho' was the root of B. longiradiatum.

한국산 속단의 생약학적 연구 (Pharmacognostical Studies on the 'SogDan' from Korea)

  • 'SogDan(續斷)' is one of the crude drugs used mainly as a tonic and analgesic. The botanical origin of the crude drug has never been studied pharmacognostically. To clarify the botanical origin of SogDan from Korea, the anatomical characteristics of Phlomis umbrosa Turcz. and Phlomis koraiensis Nakai were studied. As a result, it was clarified that SogDan from Korea was the root of Phlomis umbrosa.

Antinociceptive and sedative effects of the bark of Cerbera odollam Gaertn.

  • The crude methanolic extract of the bark of Cerbera odollam Gaertn. was evaluated for its possible antinociceptive and neuropharmacological activities in animal models. At the dose of 250 and 500 mg/kg body weight, the extract showed a significant antinociceptive effect in acetic acid induced writhing in mice comparable to that produced by aspirin, used as standard drug (P<0.001). The extract significantly reduced the time of onset of sleep (P<0.01) and potentiated the pentobarbital induced sleeping time in mice at the dose of 400 mg/kg of body weight significantly (P<0.001). It also decreased the open field score in open field test significantly at the dose of 400 mg/kg of body weight (P < 0.05). The obtained results tend to suggest the probable antinociceptive and neuropharmacological activities of the crude extract.

  • 'NuRo (漏蘆)' is one of the crude drugs which has been used for removal of fever and skin disease. With regard to the botanical origin of 'NuRo', it has been considered to be Echinops species of Compositae, but there has no pharmacognostical confirmation on it. To clarify the botanical origin of 'NuRo', the anatomical characteristics of the root of Echinops latifolius and E. setifer were studied. As a result, it was clarified that 'NuRo' from Korea was the root of Echinops setifer.

후박(厚朴)의 생약학적 연구 (Pharmacognostical Studies on the ‘Hoo Bak’)

  • 'Hoo Bak' is one of the Chinese crude drugs used mainly to cure a headache, apoplexy and dyspepsia. With regard to the botanical origin of 'Hoo Bak', it has been considered to be Machilus thunbergii of Lauraceae in Korea. But there has no pharmacognostical confirmation on it. To clarify the botanical origin of 'Hoo Bak', the anatomical characteristics of the bark of Machilus thunbergii, Magnolia officinalis and Magnolia obovata were studied. As a result, it was clarified that 'Hoo Bak' from Korea was the bark of Machilus thunbergii of Lauraceae.

  • Hepatoprotective activities of 26 traditional Korean herbal prescriptions described in Donghibogam were evaluated in vivo and in vitro screening system. Twenty one prescriptions appeared to be active. Six prescriptions among them showed highly significant hepatoprotective activities; Shihogyegy-Tang(柴胡桂枝湯), Soshiho-Tang(小柴胡湯), Shiho plus Younggolmoryo-Tang(柴胡加龍骨牡蠣湯), Yinjinho-Tang(茵蔯蒿湯), Yinsam-Tang(人蔘湯) and Shashim-Tang(瀉心湯). Shihogyegy-Tang was most active. They reduced the release of transaminases from dissociated hepatocytes by thioacetamide, and serum transaminases and alkaline phosphatase activities of rat with carbontetrachloride-induced hepatitis. They inhibited significantly lipid peroxidation and cellular fatty change of liver by carbontetrachloride.
