Developmental Abnormalities in Zebrafish Angiogenesis with Chronic Exposure to Crude Oil and Dispersant

  • Oil spills have occurred throughout the years of industrialization and represent a global challenge as they affect vast areas of the ocean. The toxicity of crude oil to aquatic organisms has been extensively investigated, but the potential impacts of crude oil on vertebrate development remain largely unknown. Here, we investigated the effects of dispersants used in treating a recent oil spill, as well as that of crude oil, on vertebrates by using the zebrafish (Danio rerio) model species, which has been widely used in empirical studies of both early embryonic development and adult physiology. Chronic exposure to crude oil resulted in marked developmental abnormalities, including pericardial edema, abnormal trunk vessel development, retardation of axonal branching, and abnormal jaw development. Embryonic development was affected more severely by exposure to the oil-dispersant combination than to the oil alone. Thus, the zebrafish in vivo model system suggests that dispersant treatment can have detrimental developmental effects on vertebrates and its potential impact on marine life, as well as humans, should be carefully considered in clean-up efforts at the site of an oil spill.

Excretory-secretory antigen is better than crude antigen for the serodiagnosis of clonorchiasis by ELISA

  • Although stool examination is the standard diagnostic method of clonorchiasis, serodiagnosis by ELISA using crude antigen is now widely used because of its convenience. However, ELISA diagnosis still suffers from cross-reactions, and therefore there is a need to improve the present conventional ELISA. The present study was undertaken to evaluate the diagnostic value of ELISA using excretory-secretory antigen (ESA) instead of crude antigen (CA) of Clonorchis sinensis. The diagnostic sensitivity of ELISA using excretory-secretory antigen was 92.5%, which was higher than that of ELISA using crude Clonorchis sinensis antigen (88.2%). In addition, the specificity of excretory-secretory antigen was found 93.1% while that of crude antigen was 87.8%. In summary, Clonorchis sinensis ESA was found to be a better serodiagnostic antigen than CA for ELISA.

Component Analysis of Acorns of Quercus mongolica and Quercus Variabilis

  • To compare seed components of plus trees, seed powder ground after seedcoat removal was analyzed for two oak species, i. e., Quercus monglica (white oak) and Quercus variabilis (red oak), which are typical oak trees in Korea but have different fruiting characteristics. Thus we aimed at analyzing and comparing many ingredients including minerals, sugars, etc. Two species were similar to each other in the content of water, crude ash, crude protein and carbohydrates, but crude lipid content in Q. variabilis was 2.5 times higher than that in Q. mongolica. Crude proteins of Clone 124 was 1.5 times higher than that of Clone 75 in Q. mongolica. Crude lipid content showed the highest value in Clone 0511 of Q. variabilis, and more phosphate and iron was found in Q. monglica than in Q. variabilis. Glucose showed 85.4% and 88.3% on average of the total monosacchrides in two species, and galactose and arabinose were also found. In the content of phosphate, iron, and crude lipid, differences were found between two species and among clones of two species.

Physicochemical Properties of Acorn Crude Starch and Acorn Refined Starch (도토리 조전분 및 정제전분의 이화학적 특성)

  • The physicochemical properties of acorn starch were investigated by comparing acorn crude starch and refined starch, Proximate composition analysis revealed that the contents of crude protein, fat and ash were higher in acorn crude starch than in refined starch. Water binding capacity and swelling power of crude starch were a little higher than refined starch. Gelatinization temperature determined by optical transmittance and by Amylograph were $68^{\circ}C$ and $73^{\circ}C$, respectively. The viscosity of crude starch had the amylose content of 28.8%, and the molecular weight of amylose was 20, 425 and glucose unit per segment of amylopectin was 21.

The Nutritional Composition of Bamboo Shoots and the Effects of its Fiber on Intestinal Microorganisms (죽순의 영양성분 및 죽순의 식이섬유가 장내미생물에 미치는 영향)

  • This study evaluated the composition two popular species of edible bamboo shoots in Korea (Phyllostachyspubescens and Sinoarundinarianigra) and the effect of their abundant dietary fiber on intestinal microorganisms in healthy young women. The ranges of total moisture, crude protein, crude lipid, crude ash, and dietary fiber content were 87.190.8, 2.943.5, 0.150.39, 0.411.05, and 4.206.15% (wet weight basis), respectively. Moisture and crude ash content increased after heat treatment; however, crude protein, crude lipid, and dietary fiber content were reduced after heating. The major minerals found in bamboo shoots were potassium, phosphorous, sulfur, magnesium, and calcium. In addition, glucose and fructose were abundant free sugars, while asparagine and tyrosine were the most abundant free amino acids. Approximately 70% of the total free fatty acids found in bamboo shoots were linoleic acid and linolenic acid. The ascorbic acid content was 6.60~17.56 mg/100 g (wet weight basis), and one phenolic compound, p-hydroxy benzoic acid, was 0.10.2% (wet weight basis) and detected by HPLC analysis. The intake of bamboo shoots for seven days significantly increased viable cell counts of Lactobacillus spp. and reduced viable cell counts of Bacteriodes spp. in feces (p<0.05). In our data, bamboo shoots may be useful in the food industry as high dietary fiber ingredients.

Effects of Seeding Dates on Yield and Feed Value of Italian Ryegrass in Paddy Field Cultivation

  • This study was carried out to investigate the influence of the seeding dates on the yield and feed value in cultivating Italian rye grass on paddy soil. The field experiment was designed in a randomized block design of 3 repetitions with seeding dates of Oct. 19 (T1), Oct. 26 (T2), Nov. 2 (T3), Nov. 9 (T4), and Nov. 16 (T5) treatments. Plant length, fresh yield, dry matter yield and TDN yield were increased with early seeding dates (p<0.05). Crude protein, Crude fat and crude ash were increased with early seeding dates (p<0.05). On the other hand, NDF, ADF and Crude fiber were decreased with early seeding dates (p<0.05). Total mineral contents were higher in the order of T4 > T3 > T5 > T1 > T2 (p<0.05). The total composition amino acid content was decreased significantly (p<0.05) as the seeding dates delayed. Total free sugar content was highest in T1, whereas the lowest in T5 treatment (p<0.05). Nutrition yields (crude protein, crude fat, amino acid and free sugar) were increased significantly with early seeding dates (p<0.05). The above result indicated that it is favorable to seed soon after rice harvest to increase dry matter yield and nutrition yield of Italian ryegrass in the midlands.

  • This research was conducted to determine contents of saponins and alkloids for plants distributed in the sourthern region of Korea. The extracts from each plants were purified by solvent fractionation, column chromatography, TLC and analyzed GC and HPLC. As a result, contents crude gingseng saponins and saikosaponins were the highest in Oenothera odorata and Metaplexis japonica, respectively. and contents crude alkaloids was the highest in Metaplexis japonica among the all plants xamined. HPLC was conducted to detect of saponins. As a result, ginseng saponin-like substances was detected in the extracts of Euphobia splendens, Taraxacum mongolicum and Metaplexis japonica, gingsen-like substances and its of saikosaponin c-like substances was detected in the extracts of Camellia japonica and Aleurites fordii. GC was conducted to detect of alkaloid. As a result, nicotine-like substances was detected in only the extracts of Fatsia japonica.

Study on Corrosion Law of Large Crude Oil Storage Tank Floor and Risk-Based Inspection and Maintenance Technology

  • In this paper, the author's team has carried out a comparative experimental study on the corrosion characteristics of Q235 steel commonly used in large-scale storage tanks under the specific bottom water environment found with Russian and Daqing crude oil. It was found that there is a certain degree of uniform or local corrosion on the tank floor depending on the kind of bottom water. The bottom water corrosion of Daqing crude oil is a uniform corrosion caused by carbon dioxide. While the Russian crude oil bottom water corrosion is clearly local corrosion caused by co-corrosion of carbon dioxide and hydrogen sulfide, here the corrosion rate is obviously higher than that caused by Daqing crude oil. There are two modes of storage tank inspection and maintenance that have been currently adopted by Chinese refining and chemical enterprises: a regular inspection mode and a API581-2016 risk-based detection mode. These modes have been effectively combined to form an intelligent tank inspection and maintenance mode, software tools to support this intelligent inspection and maintenance management have been developed.

  • The bioremediation potential of crude oil by Polyporus sp. S133 pre-grown in wood meal was investigated in two separate experiment trials; liquid medium and soil. The effect of three nutrients (glucose, polypeptone, and wood meal), oxygen flow, and some absorbent on the efficiency of the process was also evaluated. Degradation of crude oil in soil was significantly increased with an addition of oxygen flow and some absorbent (kapok and pulp). The highest degradation rate of crude oil was 93% in the soil with an addition of 10% kapok. The present study clearly demonstrates that, if suitably developed, Polyporus sp. S133 could be used to remediate soil contaminated with crude oil.

Acute and Subacute Oral Toxicity Evaluation of Crude Antifungal Compounds Produced by Lactobacillus plantarum HD1 in Rats

  • The aim of this study was to investigate the acute and subacute oral toxicity of crude antifungal compounds produced by Lactobacillus plantarum HD1 in Sprague-Dawley rats. In the acute toxicity study, the crude antifungal compounds (0.625, 1.25, 2.5, and 5.0 g/kg) did not produce mortality, significant changes in general behavior, or changes in the gross appearance of the organs. In the subacute toxicity study, the crude antifungal compounds were administered orally to rats at doses of 0, 0.5, 1.0, and 2.0 g/kg daily for 28 days. There were no test article-related deaths, abnormal clinical signs, or body weight changes. The study also showed no significant differences between the control and treated groups in hematological and serum biochemical parameters, histopathological examination, or any other findings. These results suggest that acute or subacute oral administration of crude antifungal compounds from L. plantarum HD1 is not toxic in rats.
