Occurrence of Clubroot in Cruciferous Vegetable Crops and Races of the Pathogen in Korea

  • Cruciferous vegetable crops grown in several locations in Korea were surveyed from 1996 to 2000. Clubroot severely occurred up to a maximum of 100% in Chinese cabbage fields in 15 out of 42 locations, and in cabbage fields in 5 out of 13 locations surveyed. The disease also severely occurred up to a maximum of 40% in radish fields in 6 out of 35 locations, and up to a maximum of 40% and 100% in turnip and brown mustard fields in one each out of the few locations surveyed, respectively. The disease occurred less than l% in one kale field in one out of two locations surveyed. A total of 268 isolates of Plasmodiophora brassicae was obtained from six cruciferous vegetable crops. The isolates were classified into 13 races based on their pathogenicity to the differential varieties of cabbage and rutabaga. There were 13 races found in isolates from Chinese cabbage, while 6 races each were found in isolates from cabbage and radish. There were five and three races found in turnip and brown mustard isolates, respectively. One isolate from kale was identified as race 8. Race 8 was the most frequently isolated from five cruciferous vegetable crops, except brown mustard. Races 3 and 14 were isolated only from Chinese cabbage.

병 저항성 기반 십자화과 작물의 검은무늬병 관리 (Disease Resistance-Based Management of Alternaria Black Spot in Cruciferous Crops)

  • 검은무늬병은 세계적으로 재배되는 십자화과 작물을 황폐화시키고 있으며, 브로콜리, 배추, 케일, 무, 유채 등의 생산에 경제적인 손실을 가져왔다. 십자화과 작물의 검은무늬병은 A. brassicae, A. brassicicola, A. raphani 등을 포함하는 다수의 Alternaria 종에 의하여 발생하며 이러한 병원균들은 감염된 종자나 매개충에 의하여 전반된다. 본 총설에서는 검은무늬병에 대한 십자화과 작물의 병 저항성 형질을 발굴하기 위한 그 동안의 노력을 보여주었다. 병 저항성을 위한 유전 자원이 십자화과 작물의 근연 야생종으로부터 조사되었으며, 십자화과 작물의 상이한 품종들이 다른 접종 조건에서 선별되었다. 검은무늬병 저항성 작물의 개발을 위하여 다양한 식물 방어 관련 유전자들을 이용한 십자화과 작물의 형질 전환이 이루어지기도 하였다. β-Amino-n-butyric acid나 멜라토닌과 같은 화학 물질을 전처리하여 활성화된 식물 면역이 십자화과 작물의 검은무늬병을 줄이기 위하여 제안되었다. 검은무늬병 저항성 형질은 다른 서식지에서 유래한 모델 식물 Arabidopsis 생태형들에서도 평가되었다. 야생형 Arabidopsis와는 상이한 병 저항성을 보이는 식물 면역 관련 돌연변이체들은 십자화과 작물에서 검은무늬병을 관리하는 데에 가치있는 정보를 제공하였다. 특히 이러한 돌연변이체에서 변동된 산화-환원 조절과 항산화 반응에 대하여 본 총설에서 논하였다.

Assessing persistence of cruciferous crops in the field

  • We assessed the persistence of eight major cruciferous crops-leaf mustard, oilseed rape, cabbage, broccoli, cauliflower, Chinese cabbage, turnip, and radish-growing in the field. In the first part of our experiment, we tested the viability of seeds that had been buried at two different soil depths for up to 16 months. We then broadcast seeds over the soil surface and left them undisturbed to investigate the survivorship of the resultant plants over two years. Seed viability was significantly affected by plant taxa and burial depth, but not substantially affected by the duration of burial. Although seeds of leaf mustard had the greatest viability among all crops examined here, the viability rates were significantly lower at 2 cm depth than at 15 cm. Seeds of leaf mustard, oilseed rape, broccoli, turnip, and Chinese cabbage remained viable throughout the 16-month period. A study of plant demography revealed that only leaf mustard and oilseed rape succeeded in producing seeds and overwintering in the undisturbed field. However, neither of those species competed well with other plants long-term and their overall growth and survival rates declined during the evaluation period. In addition, insect herbivory severely decreased the growth of all of these crops. Our results suggest that populations of leaf mustard and oilseed rape do not tend to persist in the field for more than a few years without disturbance and external seed inputs.

십자화과 채소작물에서의 후사리움 시들음병 발생과 그 원인균의 병원성 분화 (Occurrence of Fusarium Wilt on Cruciferous Vegetable Crops and Pathogenic Differentiation of the Causal Fungus)

  • 1996년부터 1998년까지 한국내 십자화과 채소작물의 포장에서 시들음병 발생을 조사한 결과, 배추와 무에서는 발생율이 최대 40%까지 심하게 발생하였으며, 양배추에서는 일부 포장에서만 약간 발생하였다. 시들음증상을 보이는 십자화과 채소작물의 병든 뿌리로부터 총 123균주의 Fusarium균이 분리하였으며, 공시균주들의 형태적 및 배양적 특성을 조사한 결과, 모두 Fusarium of oxysporum으로 동정되었다 공시균주들의 배추 및 양배추에 대한 병원성은 공시품종에 따라 다소 차이는 있었으나 균주들간에 뚜렷하게 구분되지 않았다. 무에 대한 병원성에 있어서는 대체로 무분리 균주들이 배추 및 양배추분리 균주들보다 강한 것으로 나타났다 공시균주들의 십자화과 이외의 채소작물에 대한 병원성검정 결과, 고추 등 8개 작물에 대해 모두 병원성이 없었다 이 연구결과, 배추감염 균주들의 병원형은 양배추감염 균주들의 병원형과 동일하고, 무감염 균주들의 병원형과는 다른 것으로 나타났다.

Virulence Differentiation of Eight Turnip mosaic virus Isolates Infecting Cruciferous Crops

  • Turnip mosaic virus (TuMV) is an infectious viral pathogen on the cruciferous crops, predominantly Chinese cabbage (Brassica campestris subsp. pekinensis) and radish (Raphanus sativus). On the basis of the symptom development in selective differential hosts from indicator host species, Chinese cabbage and Korean radish inbred lines, the representative eight isolates of TuMV were divided into two major groups/or six types. Group I includes Th 1, Ca-ad7, and Cj-ca2-1 isolates, while group II includes the other isolates (rg-pfl, r 9-10, Rhcql-2, Stock and Mustard). According to the molecular phylogenetic analysis, these isolates, however, divided into two groups and two independent isolates. Phylogenetic analysis indicated that four isolates (Tu 1, r9-10, Stock and Rh-cql-2) formed a distinct phylogenetic group, and the other two isolates (Ca-ad7 and Cj-ca2-1) also formed another group. Mustard and rg-pfl isolates did not seem to have any relationship with these two groups. Taken together, these results indicated that virulence differentiation on host plants, molecular phylogenetic analysis of the nucleotide and the deduced amino acid of TuMV coat proteins did not show any relationship. The multi-resistant lines, Wonyae 20026 and BP058 in Chinese cabbage represent valuable genetic materials that can be used for crucifer breeding programs on TuMV resistance, but not in Korean radish.

Occurrence of Sclerotinia Rot in Cruciferous Crops Caused by Sclerotinia spp.

  • Cruciferous crops grown in greenhouses and fields in Korea were surveyed from 1995 to 2000. Sclerotinia rot most severely occurred up to 30% in cabbage. Incidence of the disease was as high as 20% at its maximum in Chinese cabbage and rape and 10% in radish, but as low as less than 1 or 2% in broccoli and kale. Symptoms of Sclerotinia rot commonly developed on loaves and stems of the crucifers, but rarely on rachises of broccoli. A total of 112 isolates of sclerotinia species was obtained from the diseased crucifers. Out of the isolates, 103 isolates were identified as S. sclerotiorum, and the rest as 5. minor based on their morphological and cultural characteristics. S. sclerotiorum was isolated from all the crucifers, while S. minor was isolated from Chinese cabbage, broccoli, and kale. Six isolates of S. sclerotiorum and three isolates of S. minor were tested for their pathogenicity to the crucifers by artificial inoculation. All the isolates of the two Sclerotinia spp. induced rot symptoms on the plants of the crucifers tested, which were similar to those observed in the fields. The pathogenicity tests revealed that there was no significant difference in the susceptibility of the crucifers to the isolates of S. sclerotiorum. However, in case of S. minor, radish was relatively less susceptible to the pathogen.

십자화과(十字花科) 채소(菜蔬) 검은무늬병균(病菌)(Alternaria spp.)의 종자전염(種子傳染)에 관(關)한 연구(硏究) (Studies on Seed Transmission of Alternaria spp. in Three Cruciferous Vegetable Crops)

  • 종묘상(種苗商)에서 시판중(市販中)인 무우 종자(種子) 38개(個) 시료(試料), 배추 종자(種子) 20개(個) 시료(試料), 양배추종자(種子) 3개(個) 시료(試料) 등(等) 모두 61개(個) 시료(試料)의 십자화두(十子花枓) 채소종자(菜蔬種子)를 수집(蒐集)하여 Alternaria spp.를 검출(檢出)하고 종자(種子)에서의 감염상태(感染狀態) 및 병원성(病原性)을 조사(調査) 결과(結果)는 다음과 같다. 1. 배추 종자(種子) 시료(試料)에서 Alternaria spp.를 검출(檢出)한 결과(結果) 20개(個) 시료중(試料中) A. raphani는 7개(個) 시료(試料)에서 평균(平均) 3.8%, A. brassicicola는 6개(個)의 시료(試料)에서 평균(平均) 1.8%, A. brassicae는 1개(個) 시료(試料)에서 0.5%가 검출(檢出)되었다. 2. 무우 38개(個) 시료(試料)의 종자(種子) 중(中)에서는 A. raphani 균(菌)이 11개(個) 시료(試料)에서 7.2%, A. brassicae는 2개(個) 시료(試料)에서 1.0%가 검출(檢出)되었다. 3. 양배추종자(種子)는 공시(供試)한 3개(個) 시료(試料)의 종자(種子)에서 A. brassicicola가 21.8% 검출(檢出)되었다. 4. 종자(種子)의 감염(感染)된 Alternaria spp.는 대부분 종피(種皮)에 존재(存在)하여 발아(發芽)를 저해(沮害)하거나 유묘(幼苗) 감염(感染)을 일으켰으며 A. brassicicola는 무, 배추, 양배추종자(種子)의 배(胚)에서 A. raphani는 무우종자(種子)의 배(胚)에서 검출(檢出)되었다. 5. 종자(種子)에서 분리(分離)된 A. brassicae, A. brassicicola, A. raphani를 배추, 무우, 양배추에 접종(接種)한 결과 유엽(幼葉)보다는 노숙엽(老熟葉)에서 흑색(黑色)의 작은 반점(斑點)이 나타나고 점차(漸次) 회색(灰色) 및 담갈색(淡褐色)의 둥근 병반(病斑)을 형성(形成)하고 병반주위(病斑周圍)가 황변(黃變)되면서 엽고(葉枯)를 일으키고 부정형(不定形)으로 확대(擴大)되어 고사(枯死)하였다.

Genetic Diversity and Phylogenetic Relationships between Chinese Cabbages [B. campestris (syn. rapa) L.] and Cabbages (B. oleracea L.) in Korea

  • Members of the genus Brassica, which are known as oil crops or cruciferous vegetables, are widely cultivated in Canada, Australia, Asian and Europe. Because Brassica species have high yields, are well adapted to their environments, and are self-incompatible, the germplasm is abundant. Previous studies have reported abundant genetic diversity even within Brassica subspecies. In Korea, fresh cabbage leaves are eaten with roast meat, and to meet the current popular demand, new varieties are being increasingly bred. To determine the genetic diversity and relationships among the cabbage vegetables in Korea, we evaluated the genetic variation of 18 accessions based on 5S and 18S ribosomal RNA (rRNA) gene sequences. We detected many variable nucleotide sites, especially in the 5S rRNA gene sequences. Because the length of the 18S rRNA gene might influence the dissimilarity rate statistics, we used both the 5S and 18S sequences to analyze the phylogenetic relationships. S7 (B. oleracea) showed the most distant phylogenetic relationship with the other Brassica species. Interestingly, B2 (B. oleracea), B15, and B18 (B. campestris) have three different types of leaf profiles, and were divided into one group, and the other Brassica species formed another group. Statistical analysis of interspecies and intraspecies genetic distances revealed that B. campestris L. showed higher genetic diversity than B. oleracea L. This work provides additional data that facilitates the evaluation of the genetic variation and relationships among Brassica species. The results could be used in functional plant breeding programs to improve Brassica crops.

Current Status of Plasmodiophora brassicae Researches in Korea

  • Clubroot disease is caused by the soil-born obligate plant pathogen Plasmodiophora brassicae. This pathogen can infect all cruciferous vegetables and oil crops, including Brassica rapa, B. oleracea, B. napus, and other Brassica species. Clubroot disease is now considered to be a major problem in Chinese cabbage production in China, Korea, and Japan. We collected several hundreds of P. brassicae infected galls from Korea, and isolated the single spore from the collection. For establishment of novel isolation, and mass-propagation methods for singe spore isolates of P. brassicae pathogen, we developed new filtration method using both cellulose nitrate filter and syringe filter. Accurate detection of P. brassicae pathogen in the field was done by using real-time PCR in the potential infested soil. When we tested the different pathogenicity on commercial Chinese cabbage varieties, P. brassicae from collected galls showed various morphological patterns about clubroot symptom on roots. To date, 8 CR loci have been identified in the B. rapa genome using the quantitative trait loci (QTL) mapping approach, with different resistant sources and isolates. We are trying to develop the molecular marker systems for detect all 8 CR resistant genes. Especially for the study on the interaction between pathogens and CR loci which are not well understood until now, genome wide association studies are doing using the sequenced inbred lines of Chinese cabbage to detect the novel CR genes.

고랭지 여름 무에서 시들음병 발생 현황과 병원균의 병원학적 특성 연구 (Status of Fusarium Wilt Incidence on Summer Radish and Etiological Characteristics of the Causal Fungus in Korea)

  • 2018년과 2019년에 강원도 여름 재배 무 포장에서 시들음병 발생이 조사되었다. 시들음병은 7월 상순 발생했고, 7월 하순과 8월 뜨거운 여름에 급속히 확산되었으며, 심한 경우, 일부 포장에서는 80%까지 발생하였다. 유묘기에 병든 무는 생육이 불량하고 뿌리 내부조직이 갈변되었다. 생육중기 동안 시간이 지나면서 병든 잎은 황화되고 뿌리표면은 흰색에서 흑색으로 변색되고, 도관조직은 갈변하였다. 병든 식물체로부터 23개 Fusarium균이 분리되었고, EF-1α와 IGS 염기서열 분석에 의해 Fusarium oxysporum f. sp. raphani로 동정되었다. 무와 다른 작물에 대한 인공접종에 의해 병원성이 검정되었다. 11개 품종 간에 차이는 있었으나 모든 균주들은 무에서 병원성이었다. 그러나 로켓, 방울양배추, 스토크, 순무를 포함하는 배추과 작물을 제외하고는 병원성이 없었다. 병원성 검정결과는 무 포장에서 시들음병을 방제하기 위해서는 배추과 채소 이외의 작물로 돌려짓기하는 것이 필요하다는 것을 나타낸다.
