Antigenicity of DW-116, a New Quinolone Antibiotic, in Guinea Pigs rind Mice (기니픽과 마우스에서 신규 퀴놀론 항균제 DW-116의 항원성 시험)

  • Antigenic potential of DW-116, a newly synthesized fluoroquinolone, was examined by conduc-ting active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA) and passive hemagglutination (PHA) tests. In ASA test, mild to moderate signs of anaphylactic responses were observed in the groups sensitized with low (2 mg/body) and high (10 mg/body) doses of DW-116 alone and the group sensitized with DW-116 plus adjuvant. Some moderate to severe anaphylactic reactions were observed in the group sensitized with a DW-116-bovine serum albumin (BSA) conjugate plus adjuvant when challenged with a DW-116-guinea pig senHn albumin (GSA) conjugate. However these reactions were considered to be a cross-reaction between BSA and GSA since similar reactions were induced when challenged by GSA alone. In heterologous PCA test using mice and rats, positive responses were not detected in any of the experimental groups. In PHA test, positive responses were observed in the groups sensitized with low and high doses of DW-116 alone and the group sensitized with DW-116 plus adjuvant. However, these responses were not considered to be drug-specific because some positive responses were also seen in the negative control group. From these results, it was concluded that DW-116 is not likely to have specific antigenic potential in clinical use.

Parallel Synthesis of Unsymmetrical trans-Stilbenes

  • New unsymmetrical trans-stilbenes have been prepared by the sequential coupling reactions of bromobenzenesulfonate with formylarylboronic acids, benzylphosphonates and arylmagnesium bromides and characterized. The nickel-catalyzed reactions of stilbenesulfonates with aryl Grignard reagents produced the corresponding stilbenes via the nucleophilic aromatic substitution of the neopentyloxysulfonyl group by aryl nucleophiles. The great chemoselectivity of the alkyloxysulfonyl group allows the stepwise construction of unsymmetrical trans-stilbenes possessing terphenyl moieties. This procedure appears to be a promising and conceptually straightforward route for the parallel synthesis of various unsymmetrical stilbenes as well as other highly conjugated hydrocarbons.

  • The Mechanical strength and the electrical insulation properties of the Epoxy Resin vary with the formation of chemical reactions between Epoxy and curing agent. Therefore, to study their reactions is important for understanding the properties of the cured Epoxy. In this paper, to get the cured Epoxy having good electrical insulation property, it is studied the effects of the surrounding temperature during the cure process on the property of the cured Epoxy. In the experiment, Epoxy is cured with the surrounding temperature varied between -5[$^{\circ}C$] to 50[$^{\circ}C$]. As the results, it follows that the higher the temperature is, the faster the cross-link process is.

Dual Substituent Effects on Anilinolysis of Bis(aryl) Chlorothiophosphates

  • The reactions of bis(Y-aryl) chlorothiophosphates (1) with substituted anilines and deuterated anilines are investigated kinetically in acetonitrile at $55.0^{\circ}C$. The Hammett plots for substituent Y variations in the substrates show biphasic concave upwards with a break point at Y = H. The cross-interaction constants (${\rho}_{XY}$) are positive for both electron-donating and electron-withdrawing Y substituents. The kinetic results of 1 are compared with those of Y-aryl phenyl chlorothiophosphates (2). The cross-interaction between Y and Y, due to additional substituent Y, is significant enough to result in the change of the sign of ${\rho}_{XY}$ from negative with 2 to positive with 1. The effect of the cross-interaction between Y and Y on the rate changes from negative role with electron-donating Y substituents to positive role with electron-withdrawing Y substituents, resulting in biphasic concave upward free energy correlation with Y. A stepwise mechanism with a rate-limiting leaving group departure from the intermediate involving a predominant frontside attack hydrogen bonded, four-center-type transition state is proposed based on the positive sign of ${\rho}_{XY}$ and primary normal deuterium kinetic isotope effects.

  • A computer program COVAFIT has been developed and applied to the evaluation of experimental cross sections for MeV energy incident particles. The program utilizes weighted least-square linear regression method with high-order polynomials derived in this study. Meeting the growing demand for the treatment of covariances in nuclear data, it deals with the variance and covariance data provided along with experimental cross sections and yields those for the evaluated ones. The evaluated results on two sets of neutron total cross section of oxygen and three sets of proton cross section for $C^{11}$ production reactions confirm the methodology formulated in and the applicability of the program.

  • The average fission neutron cross sections were determined for the following reactions, $^{93}Nb(n,\alpha)^{90}Y$, $^{90}Zr(n,p)^{90}Y$,$^{93}Nb(n,\alpha)^{90m}Y$and$^{90}Zr(n,p)^{90m}Y$. The cation exchange column was used for the quantitative separation of the product nuclides using $\alpha-$hydroxyisobutyric acid as the eluent. The absolute activites of $^{90m}Y$ and $^{90}Y$were determined by the gamma ray spectrometry and a calibrated $2\pi$gas flow counter, respectively. The cross sections of $^{93}Nb(n,\alpha)^{90}Y$, $^{90}Zr(n,p)^{90}Y$,$^{93}Nb(n,\alpha)^{90m}Y$and $^{90}Zr(n,p)^{90m}Y$ reactions were found to be$0.14\pm0.01mb$, $0.83\pm0.02mb$, $0.018\pm0.02mb$ and $0.33\pm0.02mb$, respectively. The possible use of $^{90m}Y$ instead of $^{90}Y$ was discussed as a better means for the determination of niobium.

  • During the industrial preparation and the storage of foods, the side chain of some protein-bound amino acids can react chemically each other or with other molecules present in the food. The following reactions have been described : destruction of amino acids, racemization, protein-protein interactions, reactions of proteins with reducing sugars, oxidizing agents, or polyphenols. Apart from total destruction, the main reacitons are the forming of Maillard reactions products(e.g. fructoselysine) and the crosslinking with other amino acids in the same or in another protein molecule(e.g. lysinoalanine). The most often involved amino acid is lysine because of its free functional ${\varepsilon}-amino$ acid group. Generally derivatives of amino acids or crosslinks in polypeptides influence the bioavailability and the overall digestibility of the protein. This work reviews the technological, analytical, nutritional, and physiological problems related to the formation of fructoselysine and lysinolalnine in human foods, and evaluates the possible health risk for humans. A summary of the available information is of help in considering whether or not the presence of fructoselysine/lysinoalanine in foods represents a danger to man. The reduction in protein quality through these reactions is not a problem for the general population, but it is extremely important in infant foods, since infants are often nourished with a limited number of food product(e.g. formular foods) which are sensitive to the Mailard reaction.

  • Contrast media are substances that are used to enhance the contrast of structures or fluids within the body in medical imaging. Those are commonly used to enhance the visibility of blood vessels and the gastrointestinal tract. The aim of this study is to evaluate patients who got injected iopamidol, developed as one of contrast medium regarding changes in cardiovascular system and especially, adverse drug reactions. This study were performed on randomly selected a total of 51 patients (range of 15~85 years old) who got injected contrast medium, Iopamidol at a P hospital in Gyeonggi province from September 22nd, 2008 to January 24th, 2009. Data were collected by questionnaire though person to person. We analyzed the data by SPSS (Version 12. for windows). Methods of analysis were frequency and cross analysis. In results, 19.6% (n=10) were founded for adverse drug reactions of contrast medium. 25.0% (n=4) of 10 patients who had adverse drug reactions of contrast medium had took abdomen contrast test. 2 patients of those took contrast test for chest. Also, 3.9% (n=2) patients felt vomiting and nausea after injection of contrast medium. In conclusion, 19.6% patients experienced adverse drug reaction of contrast medium. Therefore, when taking radiation test, we should make efforts to minimize adverse drug reactions to achieve suitable and effective treatment.

  • The cross-coupling reaction of organo tin reagents with a variety of organic halides, catalyzed by palladium, provides a novel method for generating a carbon-carbon bond. We use this method for antibacterial agents, and synthesis of new quinolone derivatives which have carbon-carbon bond at C-7 position of general quinolone moieties. Aryl tin, quinolone moieties, and palladium catalyst were refluxed in DMF to afford new quinolone derivatives. This palladium catalyzed coupling reactions have capacity for further synthetic elaboration.

  • The cause of Huntington's disease (HD) is a pathological expansion of the polyglutamine domain within the N-terminal region of huntingtin. Neuronal aggregates composed of mutant huntingtin within certain neuronal populations are a characteristic hallmark of HD. Because tissue transglutaminase (tTG) cross-links proteins into aggregates and polypeptide-bound glutamines are primary determining factors for tTG-catalyzed reactions, it has been hypothesized that tTG may contribute to the formation of aggregates.
