오제스키병 바이러스 검출을 위한 Polymerase Chain Reaction (Polymerase Chain Reaction for the Detection of Aujeszky's Disease Virus)

  • Polymerase chain reaction (PCR) was evaluated for the early detection of Aujeszky's disease virus (ADV) DNA from virus-infected cell cultures. For the purposes, the Korean ADV NYJ1-87 was propagated in swine kidney (SK) cells and subjected to the amplification of DNA (217 bp) by PCR using sense and antisense primers specific to gp50 gene of the ADV. In detection of cell-associated viral DNA, reliable PCR conditions were determined as 30 cycles of reaction consisting 1 minute each of denaturation at $94^{\circ}C$, annealing at $55^{\circ}C$ and polymerization at $72^{\circ}C$. The PCR encountered best results with reagent mixtures of $50{\mu}l$ containing $200{\mu}M$ dNTPs, $0.2{\mu}M$ each sense and antisense primers, 1 mM $MgCl_2$ and 10% (v/v) template DNA in the final concentrations. ADV-specific DNAs were detected as early as 6, 6, and 9 hours post-infection, respectively, from lysates of the SK cells infected with ADV of $10^3$, $10^2$ and $10^1\;TCID_{50}/ml$ by this condition. In culture supernatant, the DNAs were detected from ADV of as low infectivity as $10^ {-3}\;TCID_{50}/ml$ by the reduced reagent concentrations and 30 cycles of 1 minute each of denaturation at $94^{\circ}C$ and annealing at $55^{\circ}C$, and 2 minutes of polymerization at $72^{\circ}C$. The lowest amount of detectable ADV DNA was 1 fg. In conclusion, the PCR condition established in the present study was recognized as a feasible alternative to time-consuming procedures in isolation and characterization of the virus.

  • Simple sequence repeats (SSR)는 진핵생물체에 널리 산재되어 있으며, 큰 다형현상을 나타내고, polymerase chain reaction (PCR)으로 쉽게 분석된다. 이 연구의 목적은 서로 다른 Chlamydomonas reinhardtii계통간의 다형현상과 Chlamydomonas의 SSR 좌위에서의 유전양상을 결정하는데 있었다. C. reinhardtii의 genomic DNA library를 만들어 $^{32}$P로 라벨링한 (AC)$_{11}$ probe를 이용하여 (CA/GT)n 반복서열을 가지는 clone을 선택하기위해 screen하였다. 선택된 clone을 sequencing하여 (CA/GT)n sequence에 인접한 PCR primer set를 제조하였다. PCR은 여러 C. reinhardtii 계통의 SSR 좌위를 증폭하기 위하여 사용하였다. 그 좌위는 및몇 C. reinhardtii 계통에서 다형현상을 보였다. 그러나 그 좌위에서 C. reinhardtii의 6계통중 4계통만 DNA가 PCR 증폭을 하였고 2계통은 증폭을 하지 않았다. C. reinhardtii와 C. smithii의 교배로 생긴 4배체에서 2:2의 분리비를 보여주었는데, 이는 단순한 멘델의 유전양상을 나타낸다. 이러한 결과로 미루어 SSR 다형현상은 Chlamydomonas의 개체 식별, 개체군 연구, 연쇄 분석, 그리고 유전자 지도 작성을 하는데 유용할 것이다.다.

자외선 조사가 Ovalbumin의 분자적 성질에 미치는 영향 (Effect of Ultraviolet Irradiation on Molecular Properties of Ovalbumin)

  • 식품 allergen 저감화 수단으로 자외선 조사의 타당성을 검토하고 자외선조사가 식품 단백질의 분자적 성질에 미치는 영향을 조사하고자 ovalbumin 용액에 자외선을 조사한 후 단백질의 분자량 분포와 2차구조 및 3차구조의 변화를 조사하였다. SDS-PAGE와 Gel permeation chromatography 결과 ovalbumin은 자외선 조사에 의하여 단백질이 분해되고 조사시간이 증가할수록 펩티드가 중합하는 형태를 나타냈다. Circular dichroism 연구는 자외선 조사에 의하여 ${\alpha}$-helix 구조가 감소하고 조사시간이 증가할 경우 compact denatured ovalbumin의 구조를 나타내는 2차구조의 변화를 나타냈다. 자외선 조사된 ovalbumin의 형광스펙트럼은 조사시간이 증가할수록 단백질의 maximum emission intensity가 감소하는 3차구조의 변화를 나타냈다. 결과적으로 자외선 조사는 ovalbumin의 분자적 성질을 변화시키며 allergen의 항원성을 변화시키는 수단으로 이용가능성을 시사한다.

한국의 전통 엮음직물에 관한 고찰 (Study on Traditional Korean Intertwinement of Textile)

  • Different method of textile fabrication had existed since the ancient times, but the most dominant form was the 'weaving' method which embodies the cross fabrication of vertical and horizontal threads using special weaving machines (looms). In addition, fabrication methods like knitting, felt, and lace were commonly practiced while intertwinement methods like twisting, braiding, and knotting techniques were frequently used for weaving strings and braid type textiles. In the past, people did not pay attention to strings and braids because they were classified as non-textiles, but, in this paper, we have expanded the scope of definition of textile to include strings and braids, and seek to conduct in-depth research on various different intertwinement of textiles. The adaptation of intertwinement method in arts and crafts enables limitless creative works reflecting the ever-changing taste and style of the creator. This creativity is further broadened by the fact that intertwinement methods encompass no set patterns and the subject and shape of its creation is as broad as its artistic pursuits. We can infer the knowledge of our ancestors, their aesthetic taste, and the pattern of life from these creations. In addition, these creations can provide comfort and improve the quality of life of modem people who are deeply deprived of sentimental and emotional solitude in the technological civilization of the 21stcentury. The main purpose of this paper is to examine traditional Korean arts and crafts made using the intertwinement methodology through the context of strings, braids, straw crafts and knotting method. Then it will conduct through research on the different materials, forms, characteristics, usage and other key notions based on the currently preserved records and relics to contribute to the research on traditional textiles and provide the foundations to the development of the Korean culture and cultural products, reflecting the true sole and aesthetics of traditional Korean textile crafts.

A Study on CRM Practices for Public sector Insurance Companies

  • Organizations pursue a CRM strategy for the purpose of increasing business performance and value. However, firms face a multitude of organizational challenges associated with this endeavor. To reduce their risk of failure, it is suggested that firms undertake a deep analysis of organizational readiness prior to committing to a CRM initiative. Insurance sector is no exception to this fact. There is an increased need to concentrate on the various challenges thrown open by the public insurance firms in implementing CRM. Many insurance firms have invested into customer driven CRM but research indicates varying outcomes (Schmith 2004). While it is clear that there are significant issues involved in the CRM implementation and success and environment faced by the public sector. It is clear that business should have an easier time in applying CRM systems is the strategic value for public sector. With customers demanding more service and accessibility from administrators, public sector CRM software technologies have to offer best solutions for achieving process and cost objectives (Souder 2001). With results which go far beyond improved service delivery and include sustained cost reductions, increased customer knowledge and better employee morale, CRM software implementation and post product environments offer great upside value. Although there are material differences in public sector use of CRM strategy, they share at least one glaring similarity - they have much to gain from proven CRM software technology. As business methods cross over in the public sector, many government bodies are investigating how they can adopt and adapt various CRM models (Bleyer 2003). There is a need to understand the similarities and differences in public sector CRM to foster shared knowledge, business processes and planning functions to integrate disparate technologies and software platforms and then, of course, the organizational culture to support knowledge sharing (Peters 1997). For the public sector, there are clearly identified CRM processes which have resulted in increased profits and improved efficiency. These have focused on sales, marketing and customer service activities, which often operate along fundamentally different lines in various public sector insurance companies. Thus the present research paper makes an attempt to explore how public sector CRM methods can be adopted and subsequently adapted.

Rice genotype, parental lineage and physiological tolerance to soil salinity shapes the community structure of rice seed bacterial endophytes

  • Rice seeds are a home to endophytic bacterial communities which serve as a source of the plant's endophytes. As rice undergo physiological and adaptive modifications through cross breeding in the process of attaining salinity tolerance, this may also lead to changes in the endophytic bacterial community especially those residing in the seeds. This study explores the community structure of seed bacterial endophytes as influenced by rice parental lineage, genotype and physiological adaptation to salinity stress. Endophytic bacterial diversity was studied through culture dependent technique, cloning and Terminal-Restriction Fragment Length Polymorphism (T-RFLP) analysis. Results revealed considerably diverse communities of bacterial endophytes in the interior of rice seeds. The richness of ribotypes ranges from 5-14 T-RFs corresponding to major groups of bacterial endophytes in the seeds. Endophytic bacterial diversity of the salt-sensitive IR29 is significantly more diverse compared to those of salt-tolerant cultivars. Proteobacteria followed by Actinobacteria and Firmicutes dominated the overall endophytic bacterial communities of the indica rice seeds based on 16S rDNA analysis of clones and isolates. Community profiles show common ribotypes found in all cultivars of the indica subspecies representing potential core microbiota belonging to Curtobacterium, Flavobacterium, Enterobacter, Xanthomonas, Herbaspirillum, Microbacterium and Stenotrophomonas. Multivariate analysis showed that the bacterial endophytic community and diversity of rice seeds are mainly influenced by their host's genotype, physiological adaptation to salt stress and parental lineage.

지방공무원의 직무스트레스가 구강건강 삶의 질에 미치는 영향 (The effects of occupational stress on oral health impact profile (OHIP) in local government workers)

  • Objectives : This study looked at the relationship between occupational stress and the Oral Health Impact Profile (OHIP), to evaluate the effect of occupational stress-related factors. Methods : Data was obtained from a cross-sectional survey of 260 local officials in Gangwondo. The research comprised three questions relating to subjective oral symptoms, an occupational stress measurement tool and an oral health impact factor which was composed of questions. The data was analysed using t-test, one-way ANOVA, Pearson correlation and path analysis in Amos. Results : Occupational stress had a positive correlation to drinking frequency, Oral symptoms had a negative correlation. And drinking frequency, smoking amount and occupational stress had a positive correlation to oral symptoms. It denoted that drinking frequency, occupational stress and oral symptoms had a negative correlation to OHIP. The path model had an excellent goodness of fit (p=0.07, namely p>0.05). Five 'goodness-of-fit indices' of the model were all above 0.9: GFI=0.987, AGFI=0.952, NFI=0.902, IFI=0.939, CFI=0.934), and its RMSEA was 0.045. Occupational stress and oral symptoms had a firsthand impact on OHIP. In addition, it affected OHIP through the parameters of oral symptoms. Occupational stress exercised a firsthand influence on drinking frequency, drinking frequency exercised a firsthand influence on smoking amount. Smoking amount had a firsthand impact on oral symptoms. Conclusions: Oral health education programs for the development of an improved oral hygiene environment through reduction in drinking and smoking also need to focus on relieving stress by improving workplace culture. In addition, due to good communication is required to reduce occupational stress caused by interpersonal conflict.

황색종 연초의 약배양 및 종간교배에 의한 반수체 배가계통의 특성비교 (Comparison of Lines from Anther and Maternally-derived Dihaploids in Flue-cured Tobacco(Nicotiana tabacum L.))

  • The present study was conducted to compare the relative efficiency of two different haploid breeding methods in tobacco varietal development. A single F t hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L., Bright Yellow4(BY4) and NC 95, was used to develop the 30 dihaploid lines by anther culture(F1-ADH) and maternally-derived doubled haploid utilizing Nicotiana africana(F1-MDH), respectively. As compared with mid-parent, ADH lines showed increasing in number of leaves, delaying in days to flower and narrowing in leaf width. However, no significant differences in the other characters investigated were recognized. MDH lines also showed narrow leaf width, while no significant differences in the other characters were observed. The variations of the characters investigated were generally greater in ADH than MDH lines. MDH lines had higher plant height and shorter days to flower than ADH lines, while other characters did not show remarkable differences. The degree of heritability for each of the characters observed between ADH and MDH was almost the same. The characters showing high heritability value were plant height, leaf number, days to flower, and yield, while those showing relatively low value were leaf length, leaf width, and total alkaloid content. Predicted gains from selection for increased yield were calculated for both populations(F1-ADH, F1-MDH) and correlated responses associated with selection for yield were estimated. Plant height, leaf width, days to flower, percent reducing sugar and disease resistance would be expected to improve with selection for yield much faster in the MDH population than in the ADH.

Helicobacter pylori Infection Impacts on Functional Dyspepsia in Thailand

  • Background: Helicobacter pylori (H. pylori) is a well known major cause of gastric cancer and even when asymptomatic infected patients are at elevated risk. Functional dyspepsia (FD) is also one of the most common gastrointestinal diseases, which greatly impacts the quality of life. H. pylori infection and psychosocial stress are frequently associated with FD but limited studies have confirmed the relationships, especially in Southeast Asian countries. Here we aimed to investigate the prevalence and impact of H. pylori infection, anxiety and depression on Thai FD patients. Materials and Methods: This cross-sectional study was conducted in a tertiary care center in Thailand, during February 2013-January 2014. All FD patients were diagnosed and categorized by Rome III criteria into epigastric pain syndrome (EPS) and postprandial distress syndrome (PDS) groups. The Hospital Anxiety and Depression Scale was used to evaluate psychological status. The presence of H. pylori was defined as positive with H. pylori culture, positive rapid urease test or positive histology. Results: Three hundred FD patients were included, 174 (58%) female. Overall mean age was 54.8+15.1 years. There were 192 (64%) patients with PDS and 108 (36%) with EPS. H. pylori infection was demonstrated in 70 (23.3%) patients. Anxiety and depression were documented in 69 (23%) and 22 (7.3%), respectively. H. pylori infection, anxiety and depression were significantly higher in PDS than EPS patients (27.1% vs 16.7%; p=0.04; OR=1.86; 95%CI=1.01-3.53 and 29.7% vs 11.1%; p=0.0002; OR=3.4; 95%CI=1.7-7.1 and 10.4% vs 1.9%; p=0.006; OR=6.2; 95%CI=1.4-38.9, respectively). Conclusions: H. pylori infection, anxiety and depression were commonly found in Thai FD patients and more prevalent in PDS than EPS. H. pylori eradication might be the key to success for the treatment of Thai FD patients and prevent the development of gastric cancer.

Removal and Inactivation of Human Immunodeficiency Virus(HIV-1) by Cold Ethanol Fractionation and Pasteurization during the Manufacturing of Albumin and Immunoglobulins from Human Plasma

  • Viral safety is a prerequisite for manufacturing clinical albumin and immunoglobulins from human plasma pools. This study was designed to evaluate the efficacy of cold ethanol fractionation and pasteurization (60$\^{C}$ heat treatment for 10h) for the removal/inactivation of human immunodeficiency virus type 1 (HIV-1) during the manufacturing of albumin and immunoglobulins. Samples from the relevant stages of the production process were spiked with HIV-1, and the amount of virus in each fraction was quantified by the 50% tissue culture infectious dose(TCID(sub)50). Both fraction IV fractionation and pasteurization steps during albumin processing were robust and effective in inactivating HIV-1, titers of which were reduced from an initial 8.5 log(sub)10 TCID(sub)50 to undetectable levels. The log reduction factors achieved were $\geq$ 4.5 and $\geq$ 6.5, respectively. In addition, fraction III fractionation and pasteurization during immunoglobulins processing were robust and effective in eliminating HIV-1. HIV-1 titers were reduced from an initial 7.3 log(sub)10 TCID(sub)50 to undetectable levels. The log reduction factors achieved in this case were $\geq$ 4.9 and $\geq$ 5.3, respectively. These results indicate that the process investigated for the production of albumin and immunoglobulins have sufficient HIV-1 reducing capacity to achieve a high margin of safety.
