항균성분의 지속력에 대한 인체적용 시험 평가 (Clinical Evaluation of Residual Effectiveness of Antibacterial Agents)

  • 항균비누에 흔히 사용되는 항균성분인 PCMX와 IPMP에 대하여 ASTM E2752-10 (Standard Guide for Evaluation of Residual Effectiveness of Anti- bacterial Personal Cleansing Products)에 기술된 cup scrub method에 따라 항균 지속력을 평가하였다. 건강한 남녀 총 80명의 피험자를 대상으로 한 실험에서 5% PCMX와 0.1% IPMP를 함유한 액체비누는 대조군에 비해서 E. coli와 S. aureus의 수를 통계학적으로 유의하게 감소시켰으며 27시간 동안 항균효과가 지속되는 것을 확인하였다. 항균성분의 지속력에 대한 인체적용 시험은 국내에서는 처음 시행된 것으로 결과와 함께 보고하는 바이다.

  • 미백 물질 탐색 방법으로, MC1R 발현 인자인 Mitf (microrhthalmia transcription factor)를 이용한 protein chip을 적용하였다. MC1R promoter와 Mitf 결합의 저해 인자로써, DNA 상의 결합 부위인 E-box (CATGTG)와 유사한 서열을 가진 oligomer를 사용하였고, E-box 내외부의 서열 변화에 따른 저해율 또한 측정하였다. 그 결과 DNA-Mitf 결합 저해율에 있어서, E-box 서열 내 변화를 준 oligonucleotide 경쟁자는, E-box 이외의 서열 변화를 준 경쟁자보다 낮은 수치를 보였다.

Antimelanogenic and antioxidant effects of trimethoxybenzene derivatives: methyl 3,4,5-trimethoxybenzoate, ethyl 3,4,5-trimethoxybenzoate, methyl 3,4,5-trimethoxycinnamate, and ethyl 3,4,5-trimethoxycinnamate

  • In this study, derivatives of trimethoxybenzene were investigated as inhibitors of melanogenesis. We examined the effects of methyl 3,4,5-trimethoxybenzoate (MTB), ethyl 3,4,5-trimethoxybenzoate (ETB), methyl 3,4,5-trimethoxycinnamate (MTC), and ethyl 3,4,5-trimethoxycinnamate (ETC). First, the inhibitory effects of these agents on melanin production were evaluated using α-melanocyte-stimulating hormone (α-MSH)-stimulated B16F10 melanoma cells. We found that all derivatives decreased α-MSH-induced melanin production in B16F10 melanoma cells; ETC showed a strong inhibitory effect at half of the concentration of the other derivatives. As tyrosinase is considered a key enzyme of melanogenesis, we also examined whether the derivatives inhibited tyrosinase activity. MTC and ETC reduced mushroom tyrosinase activity and expression levels of α-MSH-induced B16F10 cellular tyrosinase protein. Inhibitory effects of all derivatives on α-MSH-induced B16F10 cellular tyrosinase activity were shown in a dose-dependent manner. Additionally, the derivatives were exposed to diphenylpicrylhydrazyl free radical to examine their antioxidant characteristics. All derivatives showed considerable antioxidant activity, which was 2-fold higher than that of arbutin. In conclusion, the trimethoxybenzene derivatives, including MTB, ETB, MTC, and ETC exerted anti-melanogenic and antioxidant effects on α-MSH-stimulated melanogenesis, demonstrating their potential for use as novel hypopigmenting agents and antioxidants.

기포를 이용한 차별화된 화장품 제형 개발 (Development of Novel Cosmetic Formulations using Foams)

  • 본 총설에서는 기포의 화장품 이용에 대해서 논의하고자 하였다. 기포의 생성은 기포형성제와 기포 생성 장치 등을 이용하며 생성된 기포는 다양한 특성을 나타낸다. 기포는 기체의 용매에 대한 분산 형태로 발생된다. 기포의 안정성과 유변학적인 성질 등을 평가하는 것은 기포를 제형으로 이용하기 위한 출발점이다. 기포의 의약품과 화장품에 대한 이용은 점차 증가 추세에 있다. 의약품의 경우 기포 제형은 직장(rectal), 질(vaginal) 및 피부(dermal)용으로 대별될 수 있으며 화장품 영역의 경우 기포 제형은 모발 및 메이크업 영역에서는 헤어 무스, 파운데이션 등에서 이용되고 있으며 기초화장품에서는 자외선 차단 화장품에 이용되고 있다. 최근 유효성분의 안정화 및 유효성분의 피부 전달 측면에서 많은 연구와 특허가 출원되고 있어 향후 기능성화장품의 새로운 제형으로 활용이 기대된다.

화장품 소재로서의 꽃 10 종 에탄올추출물 생리활성 특성연구 (Biological Activities of Cosmetic Material from Ten Kinds of Flower Ethanol Extracts)

  • Background: We investigated the antioxidant, anti-wrinkles, whitening, and moisturizing properties and amounts of phenolic compounds of ethanol extracts from flowers of 10 resource plants from Namwon and Mt. Jiri., Korea. Methods and Results: We measured antioxidant efficacy based on the total polyphenol, and total flavonoid content, and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. We evaluated the inhibitory effect on melanin synthesis and tyrosinase activity for the whitening effect. Furthermore, we analyzed the elastase and matrix metalloproteinase-1 (MMP-1) inhibition activity for anti-wrinkle capacity. To evaluate the moisturizing effect, we examined hyaluronan synthase (HAS) mRNA expression. In addition, the 19 phenolic compounds were detected using high performace liquid chromatography (HPLC). Among the 10 flowers, the antioxidant effect was high in the order of Rosa multiflora, Nelumbo nucifera, and Elsholtzia splendens. Whitening effect was high in the order of N. nucifera, R. multiflora, and Dendranthema zawadskii. As for the anti-wrinkle property, N. nucifera was the most effective followed by R. multiflora. Taraxacum coreanum was the best for moisturizing effect, followed by D. zawadskii, and E. splendens. Seven phenolic compounds were detected in the extracts of the 10 flowers. Conclusions: Overall, the extracts of five flowers extracts showed strong potential as antioxidant, whitening, anti-wrinkle, and moisturizing functional cosmetic agents.

광감작성 시험에서의 동물대체 시험법 (Invirto alternatives to photosensitization Test)

  • 동물시험을 최소화 하고, 동물시험에 사용되는 노력과 비용을 줄이기 위해 동일한 동물에서 광독성과 광알러지성을 함께 보는 새로운 방법을 제안하고, in vitro alternatives로서 photostability와 spectrophotometric carbonyl assay의 유용성을 검토하였다. Modified photosensitization model에서는 Ichikawa 방법과는 달리 동물수, 휴식 기간, 첩포 횟수를 단순히하고 폐쇄 첩포를 추가하였다. 방법의 유용성을 보기 위해 Benzocaine, 6-methyl coumarine을 Ichikawa 방법 및 본 시험 방법으로 결과를 비교한 결과 macroscopic, histologic examination에서 비슷한 결과를 얻었다. Photostability 시험에서는 pbotoreactive agent 들이 모두 20%이상의 흡광도 감소를 보여 pbotosensitization potential을 측정하기 위한 전 단계 실험으로서의 가능성을 보여주었고, spectrophotometric carbonyl assay에서는 salmonella를 이용한 광독성 실험에서는 나타나지 않았던 저농도의 Amthracene, Hematoporphyrin에서 조차도 phtotoxic prtential을 보여주어 in vitro alternatives로서의 가능성을 보여주었다.

Repairing Damaged Hair Using Pentapeptides of Various Amino Acid Sequences with Crosslinking Reaction

  • The aim of this study is to investigate the effect of various pentapeptides on hair repair depending on the characteristics of comprising amino acids using crosslinking agents in hair. Total ten peptides were synthesized with two kinds of amino acids respectively, of which were previously categorized according to R group of the amino acids contributing to the characteristic of each peptide: STTSS (Ser-Thr-Thr-Ser-Ser), LIILL (Leu-Ile-Ile-Leu-Leu), CMMCC (Cys-Met-Met-Cys-Cys), DEEDD (Asp-Glu-Glu-Asp-Asp), RKKRR (Arg-Lys-Lys-Arg-Arg), TAMRA-STTSS, TAMRA-LIILL, TAMRA-CMMCC, TAMRA-DEEDD, and TAMRA-RKKRR. Pentapeptide alone, or pentapeptides with crosslinking agents such as polymeric carbodiimide (PCI) and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) were treated to chemically damaged hair. Hair diameter and break strength (N = 40/case) were measured to calculate tensile strength of hair for computing hair repair ratio, and fluorescence yields (N = 20/case) were collected for hair treated with TAMRA-peptides. The tensile strength of hair treated with pentapeptides alone, or pentapeptides with cross-linking agents is consistent with the fluorescence yield from the microscope images of the cross-sectioned hair in vision and in numerical values. Pentapeptides consisting of hydrophobic amino acids (LIILL), amino acids with sulfur (CMMCC), and basic amino acids (RKKRR) increased the tensile strength in perm-damaged hair. Pentapeptides with no extra carboxyl/amine groups in R group of amino acids resulted in no significant differences in hair strength and fluorescence yield among hairs treated with alone and with crosslinkers. Pentapeptides with extra carboxyl groups or amine groups enabled further strengthening of hair due to increased bonds within the hair after carbodiimide coupling reaction. The hair repairs of pentapeptides with various amino acid sequences were studied using crosslinking. Depending on the physical characteristics of comprising amino acids, the restoration of damaged hair was observed with tensile strength of hair and fluorescence signals upon cross-sectioned hair in parallel to possibly understand the binding tendency of each pentapeptide within the hair.

감초 성분의 항균활성, 정량 및 방부력에 관한 연구 (Antimicrobial Activity, Quantification and Bactericidal Activities of Licorice Active Ingredients)

  • 본 연구에서는 감초성분인 glycyrrhizin, liquiritin, liquiritigenin, isoliquiritigenin의 항균활성을 측정하고, 원산지별 감초 추출물에서 항균활성 성분인 isoliquiritigenin과 liquiritigenin을 정량하여 비교평가하였다. 또한 challenge test를 통하여 가장 뛰어난 항균활성을 나타낸 isoliquiritigenin과 합성 방부제인 메틸 파라벤과의 방부력을 비교하였다. 감초 성분의 항균활성을 disc diffusion assay를 통해 측정한 결과, Liquiritigenin과 isoliquiritigenin이 B. subtilis, P. acnes, E. coli, P. aeruginosa에 대해 저해활성을 보였다. 특히 감초성분 중 큰 항균활성을 나타낸 isoliquiritigenin과 합성 방부제로 사용되는 메틸 파라벤과 프로필 파라벤의 최소저해농도를 비교한 결과 isoliquiritigenin이 강력한 항균활성을 나타내는 것을 확인하였다. HPLC를 통해 원산지별 감초 추출물 isoliquiritigenin과 liquiritigenin을 정량한 결과, 이들의 함량은 한국산 감초가 중국산 감초와 우즈베키스탄산 감초보다 높은 것을 확인하였다. 이를 통해 한국산 감초가 우수한 항균력을 나타내는데 isoliquiritigenin, liquiritigenin 함량이 영향을 미치는 것으로 판단된다. Isoliquiritigenin의 B. subtilis, P. acnes, E. coli, P. aeruginosa에 대한 challenge test를 통해 메틸 파라벤 보다 isoliquiritigenin이 뛰어난 방부력을 나타냄을 확인하였다. 이상의 본 연구 결과로부터 감초 성분의 항균활성을 확인하였고, 원산지별 감초(한국, 중국, 우즈베키스탄) 중 한국 감초가 우수한 항균력을 나타내는데 isoliquiritigenin과 liquiritigenin 함량이 영향을 미치는 것으로 사료된다. 또한 감초 성분 중 가장 우수한 항균활성을 나타낸 isoliquiritigenin은 파라벤류를 대체 할 수 있는 천연 보존제로써 이용 가능성이 있다고 판단된다.

Role of Micronutrients in Skin Health and Function

  • Skin is the first line of defense for protecting our bodies against external perturbations, including ultraviolet (UV) irradiation, mechanical/chemical stress, and bacterial infection. Nutrition is one of many factors required for the maintenance of overall skin health. An impaired nutritional status alters the structural integrity and biological function of skin, resulting in an abnormal skin barrier. In particular, the importance of micronutrients (such as certain vitamins and minerals) for skin health has been highlighted in cell culture, animal, and clinical studies. These micronutrients are employed not only as active compounds in therapeutic agents for treating certain skin diseases, but also as ingredients in cosmetic products. Here, the author describes the barrier function of the skin and the general nutritional requirements for skin health. The goal of this review is to discuss the potential roles and current knowledge of selected micronutrients in skin health and function.

Application of Perfluoropolymethlisopropyl Ether to Long Wearing Lipstick

  • It has been used that fluorinated compounds could be applied to Make-up products. It is that fluorinated compounds are hydrophobicity, lipophobicity and homophobicity. These fluorinated compounds are reported to form a highly protective and effective film against most aggressive chemical and physical agents. In this study we maed a success to formulate the new type of lipstic using perfluoropolymethylisopropylether with low molecular weight which is long wearing and comfortable to use. This PPIE was adsorbed more than 90% in this experiment and formed the well-balanced gel networks even if the selected PPIE is 100% volatile at room temperature. On the basis of the adsorption of PPIE, we developed the new type lipstick, which was highly-lip feeling and had long wearing and non coloration. And these characteristics were verified by actual consumer test.
