Influence of Horse and Rider on Stress during Horse-riding Lesson Program

  • The present study aims to confirm the influence of a horse-riding lesson program (HRLP) on the stress level of horses and riders by respectively analyzing their salivary cortisol concentration. Twenty-four healthy horses and 23 riders participated in this study. The horses were randomly classified into two groups for the horse riding lesson program: Class 1 (for the beginner lesson) and Class 2 (for the intermediate lesson). The Class 1 group consisted of 12 horses and 12 riders, while the Class 2 group consisted of 12 horses and 11 riders. Salivettes cotton wool swabs were used for saliva collection and the saliva analyses were conducted using a two-way analysis of variance for repeated measures with SAS version 8. As for the results, the average salivary cortisol concentration of all horses before HRLP significantly increased compared to the baseline (p<0.001) while it decreased after the HRLP. The results of the salivary cortisol concentration of the riders were similar to the horses' results. However, there was no difference during the HRLP between Class 1 and Class 2 in the horse or rider groups. The results suggest that the HRLP did not influence the stress level of the horses or riders. Thus, this study provides the necessary information and guidelines for future studies on stress in horses during riding and gives insight into better horse welfare and management options.

Students' Salivary Cortisol level and Emotional intensity vary by teacher's teaching style in Secondary School Science Class

  • This study was conducted to examine my hypothesis that how teacher's teaching style influences emotional and physiological states of students in the secondary school science classroom. Sixty healthy secondary school students were participated in this study and divided into two groups: manipulation and non-manipulation. Each group underwent different styles of teaching on the scientific hypothesis-generating of com starch experiment. Before and after the class, the strength of emotion was measured using adjective emoticon check lists and they extracted their saliva sample for salivary hormone analysis. Here are the results of this study. First, the intensity of positive emotions in the manipulation group was significantly stronger than the one in the non-manipulation group, whereas the intensity of negative emotions in the non-manipulation group was significantly stronger than the one in the manipulation group. Second, the cortisol level, an indicator of stress, was decreased in the manipulation group whereas it was increased in non-manipulation group. Third, the quality of scientific hypotheses which is generated by students during the class had no connection with types of instructions. Fourth, this study found significantly negative correlation between students' emotional intensity of interest and concentration changes of salivary cortisol. Therefore, the different teaching styles have influence upon students' attitude and interest in science.

생활 스트레스에 대한 심박변이와 혈청 코티졸과의 관계 (Relationship between Heart Rate Variability (HRV) and Serum Cortisol Level to Life Stress)

  • 일상생활에서 경험하는 다양한 사건들은 모두 스트레스인자로 작용하여 이에 대한 대처가 부적절하면 인체의 항상성이 파괴되어 정신 및 신체질환의 발생에 영향을 주며 인체의 면역반응에 부정적인 영향을 주어 질병에 대한 대처능력을 떨어뜨려 다양한 만성질환에 대한 감수성을 높일 수 있다. 스트레스에서 내분비학적 및 생화학적 반응을 이용하는 근거는 신체 대부분의 시스템이 스트레스 중 변화하고 이런 변화를 나타내는 생물학적 지표가 스트레스와 관련된다는 근거에 의해 신체적 반응에 대한 생화학적 반응으로서 스트레스 호르몬인 코티졸 측정과 생리적인 스트레스반응으로서 여러 가지의 생리신호 중에 혈압(blood pressure)과 심박변이도(heart rate variability, HRV)를 측정하여 스트레스 반응 수준을 측정하여 상관관계를 분석하였다. 전체 94명의 피실험 대상자의 혈청 코티졸의 농도는 평균 11.22 ng/mL 이었고 최저 3.6ng/mL에서 최고 23.3 ng/mL의 분포를 나타냈다. 남녀 집단을 나누어 분석하면 남자가 13.71 ng/mL 으로 여자 10.47 ng/mL 보다 유의하게 높았다(p<0.05). 생리신호 중 혈압을 남녀 집단으로 구분하여 분석한 결과 수축기혈압 평균이 남자가 133 mmHg으로 여자 117 mmHg보다 통계적으로 유의하게 높았다(p<0.05).확장기 혈압의 평균은 남자가 81mmHg로 여자의 76.4 mmHg 보다 높았으나 유의하지 않았다. 심박변이도 중 교감신경 활동을 보여주는 LF/HF ratio 비도 남자가 3.09 로 여자 1.77 보다 유의하게 높게 나타났다(p<0.05).

사육방식의 차이가 거세한우의 등심내 콜티졸 수준 및 지방산조성에 미치는 영향 (Effect of different rearing systems on cortisol level and fatty acid composition in M-Longissimus of Korean native steers)

  • 본 연구는 사육방식의 차이가 한우 등심내 콜티졸과 지방산함량에 미치는 영향을 구명하고자 실시하였다. 무항생제 및 관행축산으로 생산된 한우육 1등급 등심 부위를 3종류의 브랜드별로 각각 3회씩 총 18점을 구매하여 시료로 공시하였고, 분석한 결과는 다음과 같다. 한우육 등심내 콜티졸 함량은 무항생제 처리구에서 관행사육 처리구 대비 유의적 (p=0.0176)으로 낮게 나타났으며, 포화지방산 및 불포화지방산 함량에서는 유의적 (p >0.05)인 차이를 나타내지는 않았으나, 관행사육 처리구에서는 포화지방산이, 무항생제 처리구에서는 불포화지방산이 각각 높은 경향으로 분석되었다. 또한, 불포화지방산 중 n-3과 n-9 지방산을 제외한 n-6 지방산에서 유의적 (p <0.01)인 차이를 보였다. 또한, 한우 등심내 콜티졸 농도와 n-6 지방산 농도간의 상관관계를 분석한 결과 유의적 (p=0.0014)으로 부 (-)의 상관관계가 성립되었다.

일부 치과위생사의 스트레스가 타액 코티졸 농도에 미치는 영향 (The Effects of Stress on Salivary Cortisol Level of Some of the Dental Hygienists)

  • 연구는 설문지를 통하여 치과위생사의 인구사회학적 특성, 근무관련 특성, 건강행태와 생활스트레스, 사회심리적 스트레스, 직무스트레스, 자아존중감을 조사하고 이들이 타액 내 코티졸 농도에 미치는 영향을 알아보고자 시행하였다. 연구대상은 2010년 2월 현재 대구 경북 치과 위생사 협회에 등록된 219명의 치과위생사로 하였으며, 타액 채취는 오후 5시를 기준으로 전후 30분까지 하였으며, 타액채취와 설문지를 동시에 할 수 있는 날짜를 선정, 2010년 3월 12일부터 3월 26일까지 실시하였다. 생활스트레스에서는 현재직장 근속년수(p<0.05), 자가 건강상태, 규칙적인 식사(p<0.01)에 유의한 차이가 있었고, 사회심리적 스트레스는 연령(p<0.05), 일일 근무시간(p<0.01), 자가건강상태, 수면시간, 규칙적인 식사, 취미여가활동(p<0.01)에 유의한 차이가 있었다. 직무스트레스는 학력(p<0.05), 직무경력, 직급, 같은부서의 인력 적절성, 현재직장 근속년수(p<0.05)가 유의한 차이가 있었다. 자아존중감은 자가건강상태(p<0.01), 수면시간(p<0.05), 취미여가활동(p<0.01)에 유의한 차이가 있었다. 타액 코티졸 농도에서는 정규시간 외 근무일(p<0.05), 자가건강상태(p<0.05)가 유의한 차이가 있었고 직무스트레스와 타액 코티졸 농도에는 약한 음의 상관관계를 보였다(p<0.01). 건강행태가 생활스트레스에 대한 경로계수는 -36.807(p<0.05)로, 건강행태가 사회심리적 스트레스 -5.209(p<0.001)로, 건강행태가 자아존중감 0.561(p<0.05)에 유의한 영향을 미쳤다. 생활스트레스가 사회심리적 스트레스 0.012(p<0.05)로, 자아존중감이 사회심리적 스트레스 -2.437(p<0.001)로, 사회심리적 스트레스가 직무스트레스 0.523(p<0.001)로 유의하게 영향을 미쳤다. 직무스트레스가 타액 코티졸농도 -0.060(p<0.05)로 유의하게 영향을 미쳤다. 이 연구에서는 직무스트레스가 타액 코티졸 농도에 유의하게 영향을 미치나 음의 상관성을 보여 만성적 직무스트레스에 의한 코티졸 농도의 고갈을 고려해 볼 수도 있겠다.

자침(刺鍼)이 마취(痲醉)에 미치는 작용기전(作用機轉) 연구(硏究) (제(第) 2 보(報)) (A Study on the Effect of Acupuncture on Anesthesia and the Mode of Action (The Second Report))

  • This study was carried to identify whether acupuncture at several meridian points can affect the human anesthesia or not through the analysis of serum intermediates, such as, melatonin, serotonin, TSH, cortisol in human serum. We investigated the effect of accupuncture on the change of serum intermediates which have frequently been made a subject of discussion owing to its hyponic and sedative properties in practice. The result obtained were as follow ; 1. Serum melatonin levels was hardly affected at 40 minutes after general acupuncture on Hab-Gog, Nae-Gwan, and the other meridian points as compared with that($5.64{\pm}1.02pg/ml$) of resting control group. But on day 1 after acupuncture, these tend to increase significantly ($7.95{\pm}2.05pg/ml$ to $8.21{\pm}1.57pg/ml$, p <0.05). On the other hand, control group under sleep showed the serum level of $7.39{\pm}1.03pg/ml$. Additional acupuncture at In-Dang induced the increased melatonin level, especially at 40minutes after acupuncture(p <0.05). 2. Serotonin level in resting control serum recorded $51.14{\pm}8.17{\mu}g/ml$. Acupuncture at determined meridian point intend to increase average level of serotonin, but not significant. However observation on day 1 after acupuncture at Hab-Gwan meridian point and under sleeping showed the significantly increased serotonin level, $74.05{\pm}35.83{\mu}g/ml$, $70.44{\pm}13.08{\mu}g/ml$, respectively. 3. TSH level in intact human serum recorded $0.85{\pm}0.24{\mu}IU/ml$. and the serum exposed to acupuncture showed the irregular pattern of TSH level in a mean range of $0.76{\pm}0.44{\mu}IU/ml$ to $1.06{\pm}0.38{\mu}IU/ml$, regardless of meridian point or time after acupuncture. 4. The values of serum cortisol in control group were $11.76{\pm}2.06{\mu}g/dl$ in resting, $7.51{\pm}2.85{\mu}/dl$ under sleep, respectively. The level of serum cortisol in 40minutes after acupuncture was markedly (p <0.05) reduced to the serum levels of $5.65{\pm}2.58{\mu}g/dl$ (Hab-Gog, Nae-Gwan), $7.58{\pm}3.21{\mu}g/dl$ (Gyo-Gam, Sin-Mun besides Hab-Gog, Nae-Gwan), $6.43{\pm}3.54{\mu}g/dl$ (In-Dang besides to Hab-Gog, Nae-Gwan), respectively, as compared with control, intending to increase a little on day 1 after acupuncture. From the above results, the analysis of serum intermediates suggest that acupunctuation at meridian points applied to at this study act upon the phase of light anaesthesia or hypnosis, at the same time affect pituitary-adrenal axis rather than hypothalamus-pituitary axis in the secretary system of hormone and also don't affect dorsal-raphe nucleus according to the observation of change transition in serum intermediates, such as TSH, serotonin, and cortisol.

동물 복지 융합형 디지털 콘텐츠 제작을 위한 야외효력시험에서 이유 자돈의 혈중 Cortisol 분석 (Analysis of Plasma Cortisol from Nursery Pigs in Outdoor Efficacy Test for Digital Content - Based Approach in Animal Welfare Convergence Types)

  • The plasma cortisol of nurserypigs was examined using an outdoor efficacy testwith a digital content-based approach in animal welfare convergence types. Nine nurserypigs,without discriminating between female and male, were classified into 2 groups of 3 pigs each: control and group 1 (effect+nature), control and group 2(effect+nature+music). The control group was the same for group 1 and 2 to compare the effects using a t-test. There was no significant difference in plasma cortisol levels between the control group and group 1 until 4 h after stress induction. However, significant differences were subsequently found between the control group and group 1 from 8 h to 72 h (p<0.05). Further, plasma cortisol was not affected in group 2 at 0 h through 8 h and 72 h. At 12 h through 48 h, group 2 showed a reduction in plasma cortisol level compared to the control group(p<0.05). These results indicated that after stress induction, applying effect plus nature or effect plus nature plus music can effectively decrease plasma cortisol levels in nursery pigs within8 h through 72 h and may serve as a better model for digital content-based approach in animal welfare convergence types.

무항생제 사육방식으로 생산된 한우육내 코티졸과 지방산 함량 (Cortisol and Fatty Acid Contents in Hanwoo Meat Produced by Antibiotics-free Rearing System)

  • 본 시험은 무항생제 사육방식이 한우육내 코티졸과 지방산 함량에 미치는 영향을 구명하고자 실시하였다. 무항생제 및 관행 축산으로 생산된 한우육 1등급 등심(M. longissimus)부위를 3가지의 브랜드별로 각각 3회씩 구매하여 시료로 공시하였고, 분석한 결과는 다음과 같다. 한우육 등심내 코티졸 함량은 무항생제 처리구에서 관행사육 처리구 대비 유의적(p=0.0176)으로 낮게 나타났으며, 포화지방산 및 불포화지방산 함량에서는 유의적(p>0.05)인 차이를 나타내지는 않았으나, 관행사육 처리구에서는 포화지방산이, 무항생제 처리구에서는 불포화지방산이 각각 높은 경향으로 분석되었다. 또한, 불포화지방산 중 n-3과 n-9 지방산을 제외한 n-6 지방산(linoleic acid, ${\gamma}$-linolenic acid)에서 유의적(p<0.01)인 차이를 보였다. 또한, 한우육내 코티졸 농도와 n-6 지방산 농도간의 상관관계를 분석한 결과, 특히 linoleic acid와 유의적(p=0.0140)으로 부(-)의 상관관계가 성립되었다. 결과적으로, 무항생제 인증으로 사육된 한우육은 관행적으로 사육된 한우육에서 보다 상대적으로 코티졸 함량이 낮았으며, 건강에 유익한 불포화지방산 함량이 다소 높은 경향으로 분석됨으로써 육질을 향상시키는 것으로 나타났다.

Measurement of Cortisol in Human Serum by Isotope Dilution Liquid Chromatography/Mass Spectrometry and a Comparison through a Proficiency Testing

  • Liquid chromatography/mass spectrometry using isotope dilution method has been established as a primary method for the measurement of cortisol in human serum. Verification of this method was accomplished by the participation in Consultative Committee for Amount of Substance-Metrology in Chemistry (CCQM) pilot study. Two levels of cortisol certified reference materials were prepared and certified by the established method. They were used as sample materials for the proficiency testing. The expanded uncertainty in the measurement of cortisol in human serum was approximately 1.2% at 95% confidence level. The results of the proficiency testing showed a good precision among the participants, but some bias to the certified values. This means that commercial field laboratories should keep traceability chain to SI unit through available reference measurement procedures and/or available reference materials.

직장온·멜라토닌·코티졸 분석을 통한 노년기 여성의 의복 착용 쾌적성 평가 (Assessment of the Clothing Wear Comfort for Elderly Women based on Rectal Temperature, Melatonin and Cortisol Analyses)

  • This study examines the wearing comfort of elderly women through a physiological analysis based on rectal temperature and biochemical analysis with salivary melatonin and cortisol. This study was conducted on 7 elderly women aged 65 or over. Two kinds of clothing ensemble (control and prototype) were used as experimental clothing. The control clothing was a general clothing ensemble and the prototype clothing lowered clothing pressure by adding an extra gap. The experimental schedule included daily living activities with randomly assigned experimental clothing. Rectal temperature was constantly measured every 5 minutes during the experiment and saliva samples for melatonin and cortisol were collected twice per day before and after sleep. The rectal temperature was lower for the prototype than the control throughout the experiment, and its circadian rhythm was prompt and clear in prototype. In addition, melatonin was secreted more but cortisol was secreted less when the subjects wore the prototype clothing. With these results, we assumed that regular circadian rhythm and low level of stress might be caused by wearing prototype clothing that lowered clothing pressure. The results demonstrate the necessity to develop clothing that considers body changes in elderly women.
