Vertical Profiles of Alkaline Phosphatase Activity in Dam Reservoirs and its Relation with Microbial Parameters (댐 저수지에서 alkaline phosphatase 활성의 수직변화와 미생물 요인들과의 상관관계)

  • The alkaline phosphatase activity (APA) of two dam reservoirs and inflowing streams were measured monthly in 2000. During summer months in 2001, the vertical profiles of APA and related parameters were also examined in one of the reservoirs. The APA was relatively high during the summer season in the epilimnion while it was almost invariable in the hypolimnion. A small increase in APA was observed at just above the bottom. The APA fluctuation was independent of the concentration of soluble reactive phosphorus. It was assumed that APA is not indicative of the phosphorus availability status. An examination of size-fractionated samples suggested that APA in reservoirs was attached to particles larger than $0.4{\mu}m$, whereas in streams it existed in a dissolved form. There was a positive significant correlation between chlorophyll a concentration and APA in the photic zone. In the aphotic zone, APA correlated positively with the colony count of heterotrophic bacteria, but not with microscopic total bacterial counts.

Comparative Analysis between Multilevel Sectioning with Conventional Haematoxylin and Eosin Staining and Immunohistochemistry for Detecting Nodal Micrometastases with Stage I and II Colorectal Cancers

  • Management of patients with stage II colorectal carcinomas remains challenging as 20 - 30% of them will develop recurrence. It is postulated that these patients may harbour nodal micrometastases which are imperceptible by routine histopathological evaluation. The aims of our study were to evaluate (1) the feasibility of multilevel sectioning method utilizing haematoxylin and eosin stain and immunohistochemistry technique with cytokeratin AE1/AE3, in detecting micrometastases in histologically-negative lymph nodes, and (2) correlation between nodal micrometastases with clinicopathological parameters. Sixty two stage I and II cases with a total of 635 lymph nodes were reviewed. Five-level haematoxylin and eosin staining and one-level cytokeratin AE1/AE3 immunostaining were performed on all lymph nodes retrieved. The findings were correlated with clinicopathological parameters. Two (3.2%) lymph nodes in two patients (one in each) were found to harbour micrometastases detected by both methods. With cytokeratin AE1/AE3, we successfully identified four (6.5%) patients with isolated tumour cells, but none through the multilevel sectioning method. Nodal micrometastases detected by both multilevel sectioning and immunohistochemistry methods were not associated with larger tumour size, higher depth of invasion, poorer tumour grade, disease recurrence or distant metastasis. We conclude that there is no difference between the two methods in detecting nodal micrometastases. Therefore it is opined that multilevel sectioning is a feasible and yet inexpensive method that may be incorporated into routine practice to detect nodal micrometastases in centres with limited resources.

CCNA1 Promoter Methylation: a Potential Marker for Grading Papanicolaou Smear Cervical Squamous Intraepithelial Lesions

  • Background: From our previous study, we established that cyclin A1 (CCNA1) promoter methylation is strongly correlated with multistep progression of HPV-associated cervical cancer, suggesting potential use as a diagnostic maker of disease. Objectives: The purpose of the present study was to assess the prevalence of CCNA1 promoter methylation in residual cervical cells isolated from liquid-based cytology that underwent hrHPV DNA screening for cervical cancer, and then to evaluate this marker for diagnostic accuracy using parameters like sensitivity, specificity, predictive values and likelihood ratio. Methods: In this retrospective study, histopathology was used as the gold standard method with specimens separated into the following groups: negative (n=31), low-grade squamous intraepithelial lesions (LSIL, n=34) and high-grade squamous intraepithelial lesions or worse (HSIL+, n=32). The hrHPV was detected by Hybrid Capture 2 (HC2) and CCNA1 promoter methylation was examined by CCNA1 duplex methylation specific PCR. Results: The results showed the frequencies of CCNA1 promoter methylation were 0%, 5.88% and 83.33%, while the percentages of hrHPV were 66.67%, 82.35% and 100% in the negative, LSIL and HSIL+ groups, respectively. Although hrHPV infection showed high frequency in all three groups, it could not differentiate between the different groups and grades of precancerous lesions. In contrast, CCNA1 promoter methylation clearly distinguished between negative/LSIL and HSIL+, with high levels of all statistic parameters. Conclusion: CCNA1 promoter methylation is a potential marker for distinguishing between histologic negative/LSIL and HSIL+using cervical cytology samples.

A Design of Inverted-Triangle UWB Monopole Antenna with Band Rejection Slot (대역 저지 슬롯이 추가된 역삼각형 모노폴 UWB 안테나의 설계)

  • In this paper, an inverted-triangle patch UWB antenna with an uneven ground planes and an inverted T-slot for 5 GHz WLAN band rejection is presented. The operating bandwidth of the proposed antenna fed with the CPW line is expanded from 3.1 GHz to 10.6 GHz for about -10 dB return loss using three angular parameters correlated to the main patch and the ground plane. The fabricated antenna on Taconic RF-60A substrate with 6.16 relative dielectric constant and 0.64 mm thickness has a main antenna patch size of 36 mm${\times}$19.5 mm. The measured results show return losses of about -10 dB and nearly omni-directional radiation patterns. The proposed UWB antenna has advantages of easily adjustable impedance characteristics by the three angular parameters and easily accomplishable band rejection characteristics by the inverted T-slot.

Clinicopathological Significance of Osteopontin in Cholangiocarcinoma Cases

  • Cholangiocarcinoma (CCA) is generally a rare primary liver tumor of the bile duct with extremely poor clinical outcomes due to late diagnosis. Osteopontin (OPN) is the most abundant expressed gene in intrahepatic CCA and its involvement in tumor aggressiveness suggests it could be a useful prognostic biomarker. However, the prognostic significance of OPN expression in CCA is still controversial. We therefore immunohistochemically studied OPN expression in 354 resected CCAs and correlated the results with patient clinicopathological parameters. OPN expression was separately scored according to the percentage of cancer cells or degree of stromal tissue staining and classified as low (score 0-1) and high (score 2-3). OPN expression in CCA cells was found in 177 out of 354 patients (56.5%), whereas stroma was positive in 185 out of 354 patients (52.3%). Univariate analysis with several of the aforementioned parameters revealed that stromal but not cancer cell OPN expression was significantly associated with tumor size, tumor direct invasion into normal liver parenchyma, regional lymph node metastasis and higher staging. The combination of cancer cell and stromal OPN expression demonstrated a positive trend for linkage with lymph node metastasis. Multivariate analysis identified gender, the presence of lymphatic permeation and lymph node metastasis, but not OPN expression, as independent prognostic factors. This study confirms the presence of stromal OPN expression in tumor aggressiveness but not survival in CCA patients.

Genetic Relationship between Ultrasonic and Carcass Measurements for Meat Qualities in Korean Steers

  • Real time ultrasonic measurements for 13th rib fat thickness (LBF), longissimus muscle area (LEMA) and marbling score (LMS) of live animal at pre-harvest and subsequent carcass measurements for fat thickness (BF), longissimus muscle area (EMA), marbling score (MS) as well as body weight of live animal, carcass weight (CW), dressing percentage (DP), and total merit index (TMI) on 755 Korean beef steers were analyzed to estimate genetic parameters. Data were analyzed using multivariate animal models with an EM-REML algorithm. Models included fixed effects for year-season of birth, location of birth, test station, age of dam, linear and quadratic covariates for age or body weight at slaughter and random animal and residual effects. The heritability estimates for LEMA, LBF and LMS on RTU scans were 0.17, 0.41 and 0.55 in the age-adjusted model (Model 1) and 0.20, 0.52 and 0.55 in the weight-adjusted model (Model 2), respectively. The Heritability estimates for subsequent traits on carcass measures were 0.20, 0.38 and 0.54 in Model 1 and 0.23, 0.46 and 0.55 in Model 2, respectively. Genetic correlation estimate between LEMA and EMA was 0.81 and 0.79 in Model 1 and Model 2, respectively. Genetic correlation estimate between LBF and BF were high as 0.97 in Model 1 and 0.98 in Model 2. Real time ultrasonic marbling score were highly genetically correlated to carcass MS of 0.89 in Model 1 and 0.92 in Model 2. These results indicate that RTU scans would be alterative to carcass measurement for genetic evaluation of meat quality in a designed progeny-testing program in Korean beef cattle.

Evaluation of KiSS1 as a Prognostic Biomarker in North Indian Breast Cancer Cases

  • Background: Breast cancer is the commonest female cancer worldwide and its propensity to metastasize negatively impacts on therapeutic outcome. Several clinicopathological parameters with prognostic/predictive significance have been associated with metastatic suppressor expression levels. The role of metastatic suppressor gene (MSG) KiSS1 in breast cancer remains unclear. Our goal was to investigate the possible clinical significance of KiSS1 breast cancer. Materials and Methods: The study was conducted on 87 histologically proven cases of breast cancer and background normal tiisue. Quantitative reverse transcriptase polymerase chain reaction (qRT PCR) and immunohistochemistry (IHC) were used to investigate KiSS1 at gene and protein levels, respectively, for correlation with several patient characteristics including age, family history, hormonal receptor status, stage, tumor size, nodal involvement and metastatic manifestation and finally with median overall survival (OS). Results: Our study revealed (i) KiSS1 levels were generally elevated in breast cancer vs normal tissue (P < 0.05). (ii) however, a statistically significant lower expression of KiSS1 was observed in metastatic vs non metastatic cases (P = 0.04). (iii) KiSS1 levels strongly correlated with T,N,M category, histological grade and advanced stage (p<0.001) but not other studied parameters. (iv) Lastly, a significant correlation between expression of KiSS1 and median OS was found (P = 0.04). Conclusions: Conclusively, less elevated KiSS1 expression is a negative prognostic factor for OS, advancing tumor stage, axillary lymph node status, metastatic propensity and advancing grade of the breast cancer patient. Patients with negative KiSS1 expression may require a more intensive therapeutic strategy.

Genetic Parameters and Responses in Growth and Body Composition Traits of Pigs Measured under Group Housing and Ad libitum Feeding from Lines Selected for Growth Rate on a Fixed Ration

  • The main objective of this study is to examine genetic changes in growth rate and carcass composition traits in group housed, ad libitum fed pigs, from lines of Large White divergently selected over four years for high and low post-weaning daily gain on a fixed but restricted ration. Genetic parameters for production and carcass traits were also estimated by using average information-restricted maximum likelihood applied to a multivariate individual animal model. All analyses were carried out on 1,728 records of group housed ad libitum fed pigs, and include a full pedigree of 5,324 animals. Estimates of heritability (standard errors in parentheses) were 0.11 (0.04) for lifetime daily liveweight gain (LDG), 0.13 (0.04) for daily carcass weight gain (CDG) and 0.28 (0.06) for carcass backfat (CFT). Genetic correlations between LDG and CDG were highly positive and between LDG and CFT negative, suggesting that selection for lifetime daily gain under commercial conditions of group housing with ad libitum feeding would result in favourable improvement in carcass traits. CFT showed negative genetic correlations with CDG. Correlated genetic responses evaluated as estimated breeding values (EBVs) were obtained from a multivariate animal model-best linear unbiased prediction analysis. After four years of divergent selection for 6 week post-weaning growth rate on restricted feeding, pigs performance tested on ad libitum feeding in groups exhibited changes in EBVs of 6.77 and -9.93 (g/d) for LDG, 4.25 and -7.08 (g/d) for CDG, and -1.42 and 1.55 (mm) for CFT, in the high and low lines, respectively. It is concluded that selection for growth rate on restricted feeding would significantly improve genetic performance and carcass composition of their descendants when group housed and ad libitum fed as is a common commercial practice.

Postoperative Flat Back : Contribution of Posterior Accessed Lumbar Interbody Fusion and Spinopelvic Parameters

  • Objective : Posterior accessed lumbar interbody fusion (PALIF) has a clear objective to restore disc height and spinal alignment but surgeons may occasionally face the converse situation and lose lumbar lordosis. We analyzed retrospective data for factors contributing to a postoperative flat back. Methods : A total of 105 patients who underwent PALIF for spondylolisthesis and stenosis were enrolled. The patients were divided according to surgical type [posterior lumbar inter body fusion (PLIF) vs. unilateral transforaminal lumbar interbody fusion (TLIF)], number of levels (single vs. multiple), and diagnosis (spondylolisthesis vs. stenosis). We measured perioperative index level lordosis, lumbar lordosis, pelvic tilt, sacral slope, pelvic incidence, and disc height in standing lateral radiographs. The change and variance in each parameter and comparative group were analyzed with the paired and Student t-test (p<0.05), correlation coefficient, and regression analysis. Results : A significant perioperative reduction was observed in index-level lordosis following TLIF at the single level and in patients with spondylolisthesis (p=0.002, p=0.005). Pelvic tilt and sacral slope were significantly restored following PLIF multilevel surgery (p=0.009, p=0.003). Sacral slope variance was highly sensitive to perioperative variance of index level lordosis in high sacral sloped pelvis. Perioperative variance of index level lordosis was positively correlated with disc height variance ($R^2=0.286$, p=0.0005). Conclusion : Unilateral TLIF has the potential to cause postoperative flat back. PLIF is more reliable than unilateral TLIF to restore spinopelvic parameters following multilevel surgery and spondylolisthesis. A high sacral sloped pelvis is more vulnerable to PALIF in terms of a postoperative flat back.

Perfusion-Weighted MRI Parameters for Prediction of Early Progressive Infarction in Middle Cerebral Artery Occlusion

  • Objective : Early progressive infarction (EPI) is frequently observed and related to poor functional outcome in patients with middle cerebral artery (MCA) infarction caused by MCA occlusion. We evaluated the perfusion parameters of magnetic resonance imaging (MRI) as a predictor of EPI. Methods : We retrospectively analyzed patients with acute MCA territory infarction caused by MCA occlusion. EPI was defined as a National Institutes of Health Stroke Scale increment ${\geq}2$ points during 24 hours despite receiving standard treatment. Regional parameter ratios, such as cerebral blood flow and volume (rCBV) ratio (ipsilateral value/contralateral value) on perfusion MRI were analyzed to investigate the association with EPI. Results : Sixty-four patients were enrolled in total. EPI was present in 18 (28%) subjects and all EPI occurred within 3 days after hospitalization. Diabetes mellitus, rCBV ratio and regional time to peak (rTTP) ratio showed statically significant differences in both groups. Multi-variate analysis indicated that history of diabetes mellitus [odds ratio (OR), 6.13; 95% confidence interval (CI), 1.55-24.24] and a low rCBV ratio (rCBV, <0.85; OR, 6.57; 95% CI, 1.4-30.27) was significantly correlated with EPI. Conclusion : The incidence of EPI is considerable in patients with acute MCA territory infarction caused by MCA occlusion. We suggest that rCBV ratio is a useful neuro-imaging parameter to predict EPI.
