Phytotoxicity and Translocation of Residual Diquat Dibromide from Sandy Loam and Loam Soil to Following Crops Cultivating in the Soils

  • BACKGROUND: Diquat dibromide is a fast-acting nonselective herbicide and plant growth regulator. In this study, in order to understand the possibility of unintentional pesticide contamination in the following crops, the phytotoxicity and transition of diquat dibromide residue in soil into the following crops such as pepper, radish, lettuce and corn have been assessed through phytotoxicity trial and residual evaluation in the unintentional contamination of the higher residual diquat dibromide. METHODS AND RESULTS: The pepper, radish, lettuce and corn were cultivated in the sandy soil and loam soil where the 35 mg/kg and 90 mg/kg diquat dibromide were applied, respectively. Mild growth inhibition symptoms were observed in radish, lettuce and corn crops at the 90 mg/kg- diquat dibromide treatment on the 30 day of cultivation. Diquat dibromide was analyzed using liquid chromatography QTRAP (LC-MS/MS). The recovery rates of diquat dibromide from soil and crop were determined within range from 89.1 to 116.4% with relative standard deviation less than 14.7%. Diquat dibromide residues in soil were found to be 23.90-30.22 and 69.59-82.57 mg/kg from the 35 mg/kg and 90 mg/kg of diquat dibromide-treated soil, respectively after 30 days of crop cultivation. This result implicates that diquat dibromide did not convert to metabolites and remained mostly in the soil, even though it was partially decomposed during crop cultivation. In addition, the diquat dibromide in pepper and radish that were grown for 47 days, and lettuce and corn that were cultivated for 30 days were detected to be 0.01 mg/kg or less in the sandy loam and loam soil where the 90 mg/kg diquat dibromide was applied. CONCLUSION(S): Diquat dibromide did not cause severe phytotoxicity in the following crops as well as it did not uptake and distribute to the following crops, even though it was considered to be residual in the soil.

Utilization of Robinia pseudoacacia as Sawdust Medium for Cultivation of Edible and Medicinal Mushrooms

  • This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.

  • A novel integrated method for increasing dissolved oxygen concentration in culture media has been developed. It involves adding hydrogen peroxide to the medium, which is then decomposed to oxygen and water by catalase and adding vegetable oil to the medium as antifoam agent and oxygen vector. A new apparatus for automated addition of hydrogen peroxide to the bioreactor to keep the dissolved oxygen concentration constant over the range $10-100%\;{\pm}\;5%$ was tested. A significant increase (over threefold) of cultivation time was obtained while the dissolved oxygen concentration remained stable ($30%\;{\pm}\;5%$). Therefore, use of corn oil mixed with Ca-stearate as oxygen vector and antifoam and hydrogen peroxide as oxygen source to control excessive foam that was generated by microorganism biosurfactant, GB16-BS produced at Bacillus sp. GB16 cultivation was appropriate for stable cultivation.

  • On the basis of a brief introduction of the mechanized dry farming in Heilongjiang Province, the author states the developing process from the combination of single technics of farm machinery and agronomy to the technical assembly of high yield cultivation technics and its mathematical expression. According to the main temperature accumulated zones, 5 typical comprehensive technical assembly models for the mechanized cultivation technics and their agricultural machinery systems have been listed. They are, the Heihe " 261" wheat and soybean model : the Yi'an big ridge double row film mulching corn model : the Yongchang high yield mechanized soybean and other grain crops six year rotation model for Keshan state farms. The author conclude that the application of mechanized high yield cultivation technical assembly is the key point to transform the Heilongjiang province from big agriculture to strong agriculture, we have to take " high yield , high quality , high efficiency , s stain -ability and earning foreign currency" as the general target and carry out the corresponding policy and measures for the further development of agricuture.

  • 잣버섯에 적합한 액체종균용 주배지 재료는 대두분, 배양 기간은 12일, 통기량은 0.9 vvm이 적합하였다. 또한, 잣버섯 재배용 톱밥종균용 배지는 미송과 옥분을 부피비로 95 : 5로 혼합하는 것이 적합하였다. 액체종균 및 톱밥종균을 사용하였을 경우 잣버섯 생산성을 비교한 결과 액체종균으로 재배한 경우 재배일수 43일로 톱밥종균으로 재배할 때보다 재배기간 보다 2일 단축되었고, 유효경수 11.3개, 수량 111.9 g로 우수하였다.

Efficient Production of ε-Poly-L-Lysine by Streptomyces ahygroscopicus Using One-Stage pH Control Fed-Batch Fermentation Coupled with Nutrient Feeding

  • ε-Poly-L-lysine (ε-PL) is a homopolymer of L-lysine molecules connected between the epsilon amino and alpha carboxyl groups. This polymer is currently used as a natural preservative in food. Insufficient biomass is a major problem in ε-PL fermentation. Here, to improve cell growth and ε-PL productivity, various nitrogen-rich nutrients were supplemented into flask cultures after 16 h cultivation, marking the onset of ε-PL biosynthesis. Yeast extract, soybean powder, corn powder, and beef extract significantly improved cell growth. In terms of ε-PL productivity, yeast extract at 0.5% (w/v) gave the maximum yield (2.24 g/l), 115.4% higher than the control (1.04 g/l), followed by soybean powder (1.86 g/l) at 1% (w/v) and corn powder (1.72 g/l) at 1% (w/v). However, supplementation with beef extract inhibited ε-PL production. The optimal time for supplementation for all nutrients examined was at 16 h cultivation. The kinetics of yeast-extract-supplemented cultures showed enhanced cell growth and production duration. Thus, the most commonly used two-stage pH control fed-batch fermentation method was modified by omitting the pH 5.0-controlled period, and coupling the procedure with nutrient feeding in the pH 3.9-controlled phase. Using this process, by continuously feeding 0.5 g/h of yeast extract, soybean powder, or corn powder into cultures in a 30 L fermenter, the final ε-PL titer reached 28.2 g/l, 23.7 g/l, and 21.4 g/l, respectively, 91.8%, 61.2%, and 45.6% higher than that of the control (14.7 g/l). This describes a promising option for the mass production of ε-PL.

옥수수 연작지에서 Fall panicum의 발생과 연작 피해 방지를 위한 대상작의 도입효과 (Introducing Strip Cropping for Decreasing the Damage of the Continous Corn Cultivation)

  • 옥수수연작 피해방지를 위해 레드클로버 등을 대상작으로 도입하여 그 효과를 기대하였다. 특히 옥수수 연작지에 발생하는 난지형 잡초인 Fall panicum(FP)를 대상으로 옥수수/호밀연작 정도와 토양 습윤 정도에 따른 발생량을 조사하였다. 옥수수 재배와 동시에 대상 경작한 작물은 레드클로버(RC), 알팔파(A), 목초혼파(G/ R)이었다. 2년 동안 대상작물 재배 후 마지막 3년차에 옥수수 재배로 다시 환원하였는데 옥수수 연작지와 옥수수 단작에서의 얻어진 평균 생초수량의 범위는 각각 119.4${\~}$169,6 및 64.9${\~}$83.3톤/ha 이었으나 옥수수만의 수량에 있어서는 각각 45.6${\~}$52.8 및 64.9${\~}$83.3톤/ha로 서로 다른 결과를 얻었다. 이 같은 수량차이의 대부분은 옥수수 연작지에서의 FP의 발생량에 기인한 것으로 파악되었는데 실제 옥수수 연작지에서의 FP 발생량의 범위는 평균 45.6${\~}$63.4에 달하여 연작지에서의 호밀 28.2${\~}$41.6을 초과하였다. 대상작으로 도입한 레드클로버, 알팔파 및 목초 혼파의 3년간 건물 생산량은 각각, 10.2, 10.6 및 10.6톤/ha로. 연작구 평균 옥수수 수량의 대략 4분의 1정도인 것으로 나타났는데 대상작 도입 후 옥수수 생초 수량은 연작구 및 레드클로버, 알팔파 및 목초 혼파 대상작 도입구에서 각각 56, 82, 88 및 79톤/ha 로 대상작 도입구에서 크게 증가하였고 알팔파 도입 시 가장 크게 증가하는 것으로 나타났다.

중국 동북3성에서의 옥수수 수확량과 위성기반의 식생 지수 및 농업기후요소와의 상관성 연구 (Correlation between the Maize Yield and Satellite-based Vegetation Index and Agricultural Climate Factors in the Three Provinces of Northeast China)

  • 본 연구에서는 지난 20년간(1996~2015) 중국 동북 3성에서의 옥수수 수확량과 위성기반 식생지수인 NDVI (Normalized Difference Vegetation Index) 그리고 여러 기후요소들간의 월별 상관성을 분석하고자 하였다. 중국 동북 3성의 옥수수 재배지역에서 옥수수 수확량은 작황시기의 NDVI와 통계적으로 유의한 양의 상관관계를 보였고, 8월과 9월의 최고기온 및 여름철 $30^{\circ}C$ 이상의 고온 발생빈도와 음을 상관관계를 가졌다. 옥수수 수확량과 강수량간의 상관관계는 7월에 요녕성에서만 유의한 양의 계수를 나타내었고 길림성과 흑룡강성에서는 상관성이 나타나지 않았다. 본 연구를 통해 중국 동북 3성의 옥수수 수확량을 추정하기 위해서는 NDVI와 최고기온 자료를 예측인자로 사용하는 것이 적합할 것으로 생각된다.

간척지에서 SCB 액비를 이용한 여름철 사료작물 재배에 관한 연구 (Study on Summer Forage Crop Cultivation Using SCB (Slurry Composting-Biofilteration) Liquid Fertilizer on Reclaimed Land)

  • 본 연구는 간척지에서 여름철 대표 사료작물인 옥수수와 수수${\times}$수수 교잡종의 생산성 및 화학비료와 가축분뇨의 시용시 생산성을 비교하기 위하여 2008년은 화옹간척지에서 2009년은 석문간척지에서 시험을 실시하였다. 간척지에서 사료작물의 생산성은 옥수수가 수수${\times}$수수 교잡종보다 높았으며 사료가치도 옥수수가 수수${\times}$수수 교잡종보다 높았다. 화학비료와 가축분뇨의 시용시 생산성은 옥수수의 경우 화학비료에서 높은 수량을 보였으며 수수${\times}$수수 교잡종의 경우 화옹간척지에서 시험한 결과 SCB 액비구에서 높았고 석문간척지에서는 돈분액비구에서 높은 수량을 보였다. 사료가치는 화학비료구와 가축분뇨 시용구에서 비슷한 결과를 나타냈다. 반면 사료작물의 생산성과 토양염류도와의 관계성은 보이지 않았다. 이상의 시험결과로 보아 간척지에서 여름철 대표사료작물인 옥수수와 수수${\times}$수수 교잡종의 재배가 가능하며 가축분뇨를 시용하여 재배가 가능하다고 판단되어지며 돈분액비와 SCB 액비의 경우 새로운 비료로서 대체 가능성이 있으나 추가적인 연구가 필요할 것으로 판단되어진다.

Growth characteristics comparison per planting density on the waxy corn early-planting culture for the paddy field in the southern province

  • The study was conducted to analyze the growth characteristics comparison per planting density on the waxy corn early-planting culture for the paddy field in the southern province of south Korea. The cultivation period of early-planting culture for the paddy farming of the waxy corn are sown on February 15, 2016years, transplanting March 15 and harvest June 20. And it grew 126 days. The weather change according to the cultivation period of unheated plastic house early-planting culture, it was average temperature $14.6^{\circ}C$ and humidity 62.5%. And the temperature was 5.6 degrees Celsius warmer compared with the outside temperature and the humidity was 0.7 percent higher tendency. At the growth per planting density of waxy corn, culm length was average 224cm, the more it is high density culture the more was high trend. Stem diameter and ear length the more it is high density culture the more was lowed trend. The node number of $60{\times}20Cm$ was 12 nodes, fruit seting 5.7 nodes, tasseling number 94 days and silking number 96 days. In the ear characteristics per planting density, the size of ear length, seed setting length, ear width and ear weight the more planting density is high the more lowed that trend. The commodity percentage of planting density $60{\times}35Cm$ was the highest among other treatment as 69.1%. But, marketable yield was the highest planting density of $60{\times}20Cm$ as 4,543 ears/10a, and appeared in order $60{\times}25Cm$ 95%> $60{\times}30Cm$ 93%> $60{\times}35Cm$ 92%. The planting density on the waxy corn early-planting culture for the paddy farming in the southern province, the planting density analyzed to be effective planting of over 25% than normal season culture.
