A Novel Method for Production of Concentrated Purity Maltose Using Swollen Extruded Starch (Extrusion시킨 팽윤 전분을 기질로 한 새로운 Maltose 생산법)

  • A novel method for production of concentrated purity maltose using swollen extruded corn starch was investigated. Degree of gelatinization of extruded starch suitable for maltose formation was found to be around 70%. The optimal amiunt of enzyme was 400 unit fungal $\alpha $-amylase per g of starch, and the reaction time was 12 hours. At extruded starch concentration of 300 g/l(w/v), maltose concentration and content were reached up to 220 g/l(w/v) and 77%(w/w), respectively. The maltose forming reaction was also successfully proceeded at high starch concentration of 700 g/l(w/v), however, the conversion yield and content were decreased. By the addition of extruded starch by fed-batch wise, the maltose concentration, purity, and conversion yield could be improved up to 465 g/l(w/v), 70%(w/w), and 0.63, respectively. The investigated maltose production process seems to have many potential advantages over the conventional process utilizing liquefied starch, and the feasibility for industrial application needs to be evaluated.

Response Surface Methodological Approach for Optimization of Enzymatic Synthesis of Sorbitan Methacrylate

  • Sorbitan methacrylate was synthesized from sorbitan dehydrated from D-sorbitol using an immobilized lipase. To optimize the enzymatic synthesis of sorbitan methacrylate, response surface methodology was applied to determine the effects of five-level-four-factors and their reciprocal interactions on sorbitan methacrylate biosynthesis. A total of 30 individual experiments were performed, which were designed to study reaction temperature, reaction time, enzyme amount and substrate molar ratio. A statistical model predicted that the highest conversion yield of sorbitan methacrylate was 100%, at the following optimized reaction conditions: a reaction temperature of 43.06 $^{\circ}C$, a reaction time of 164.25 mins., an enzyme amount of 7.47%, and a substrate molar ratio of 3.98:1. Using these optimal factor values under experimental conditions in four independent replicates, the average conversion yield reached 98.7%${\pm}$1.2% and was well within the value predicted by the model.

  • Effect of $H_2$ spillover rate as functon of calcination temperature on reaction kinetics was evaluated. Reaction kinetics including yield, conversion and selectivity of 1-butene isomerization over $Pt/HxMoO_3/SiO_2$ were measured as reaction temperature was increased. While conversion of 1-butane was decreased, yield of iso-butene was increased. Two kinds of reaction mechanism were proposed from the change of selectivity as function of temperature.

Whole Cell Bioconversion of Ricinoleic Acid to 12-Ketooleic Acid by Recombinant Corynebacterium glutamicum-Based Biocatalyst

  • The biocatalytic efficiency of recombinant Corynebacterium glutamicum ATCC 13032 expressing the secondary alcohol dehydrogenase of Micrococcus luteus NCTC2665 was studied. Recombinant C. glutamicum converts ricinoleic acid to a product, identified by gas chromatography/mass spectrometry as 12-ketooleic acid (12-oxo-cis-9-octadecenoic acid). The effects of pH, reaction temperature, and non-ionic detergent on recombinant C. glutamiucm whole cell bioconversion were examined. The determined optimal conditions for production of 12-ketooleic acid are pH 8.0, 35℃, and 0.05 g/l Tween80. Under these conditions, recombinant C. glutamicum produces 3.3 mM 12-ketooleic acid, with a 72% (mol/mol) maximum conversion yield, and 1.1 g/l/h volumetric productivity in 2 h; and 3.9 mM 12-ketooleic acid, with a 74% (mol/mol) maximum conversion yield, and 0.69 g/l/h maximum volumetric productivity in 4 h of fermentation. This study constitutes the first report of significant production of 12-ketooleic acid using a recombinant Corynebacterium glutamicum-based biocatalyst.

  • Microbiological conversion of sterols to 17-ketosteroids has been recognized as a source for commercial preparation of steroidal drugs. In order to develop bacterial strains and process with Brevibacterium lipolyticum IAM 1398 capable of converting cholesterol to 1,4-Androstadiene-3,17-dione (ADD) at about 27% yield, we studied on strain improvement, fermentation condition and whole cell immobilization. By using UV and/or NTG as mutagens, a mutant to convert cholesterol to ADD with higher yield than 60% was selected. Better production of ADD was manifested in the case of maltose used as a supplemental carbon source, and yeast extract or soytone as a nitrogen source. Addition of tween 80 (0.05%) as a surfactant beneficial for increasing the productivity. The optimal initial pH of the medium was 6.5 and optimal culture temperature was $30^{\circ}C$. Whole cell immobilization by using carrageenan, agar, alginate and acrylamide was carried out and the activity of conversion was tested. In the case of carrageenan and agar, immobilized cells were active for at least two cycles of fermentation.

  • In this study, the conversion of glucosamine, which is a major monomer in chitin/chitosan of crustacean shell, using solid acid catalyst was performed to obtain chemical intermediates such as levulinic acid and 5-hydroxymethyl furfural (5-HMF). The conversion reaction was optimized with four reaction factors of selection of ionic resin catalyst, reaction temperature, catalyst amount, and reaction time. As an optimized result, the highest levulinic acid yield was achieved approximately 36.86% under the determined conditions (Amberlyst 15 as a solid-acid catalyst, $180^{\circ}C$, 5% catalyst amount and 60 min). On the other hand, 5-HMF yield was found to be 0.91% at the condition.

  • In order to investigate effective solar energy conversion system, the light-induced electron transfer reactions have been examined across single-lamellar liposomes incorporated organic photosensitizers such as anthracene and naphthalene derivatives. We have observed photosensitized reduction of methyl viologen (1,1'-dimethyl-4,4'-$bipyridinium^{2+}$) dissolved in the exterior aqueous phase of the pigmented phospholipid liposomes when EDTA, as electron donor, is dissolved in the enclosed aqueous phase of the liposomes. The anthroyl stearic acid incorporated in the hydrophobic bilayer of liposomes leads to much less quantum yield for the photosensitized reduction of $MV^{2+}$ than the anthracene carboxylate incorporated in the outer hydrophilic layer. However, ${\beta}$-carotene with anthroyl stearic acid incorporated into the bilayer enhances the quantum yield significantly (${\Phi}{\simeq}0.2-0.3$), preventing the reverse reaction of electron transfer ($MV^+_\ {\rightarrow}MV^{2+}$) so that it might be useful for solar energy conversion into chemical energy. A naphthalene derivative, octadecyl naphthylamine sulfonic acid incorporated into the outer layer of liposomes results in less efficiency of $MV^{2+}$ reduction than anthroyl stearic acid. These results have been also tested with respect to lipid components of liposomes.

Conversion Characteristics of Liquid Fuels from Sawdust by Acetone-Solvolysis (아세톤-용매분해반응에 의한 톱밥으로부터 액체 연료물질의 전환 특성 연구)

  • Sawdust, produced as an wood by-product, is usable biomass as liquid fuels if decomposed to monomer unit, because the chemical structure are similar to high octane materials found in gasoline. In this study, parameters of thermochemical degradation by acetone-solvolysis reaction of sawdust such as the effect of reaction temperature, reaction time and type of solvent on conversion yield and degradation products were investigated. The liquid products by acetone-solvolysis from sawdust produced various kind of ketone, phenol and furan compounds. The optimum sawdust conversion was observed to be 88.7% at $350^{\circ}C$, 40min. Combustion heating value of liquid products from thermochemical conversion processes was as high as 7,824 cal/g. The energy yield and mass yield in acetone-solvolysis of sawdust was 60.8% and 36.4 g-oil/100g-sawdust after 40 min of reaction at $350^{\circ}C$, respectively. The major components of the acetone-solvolysis products, that could be used as liquid fuel, were 4-methyl-3-pentene-2-one, 1,3,5-trimethylbezene, 2,6-dimethyl-2,5-heptadiene-4-one, 3-methyl-2-cyclopenten-1-one as ketone compounds.

Motion Linearity-based Frame Rate Up Conversion Method (선형 움직임 기반 프레임률 향상 기법)

  • A frame rate up-conversion scheme is needed when moving pictures with a low frame rate is played on appliances with a high frame rate. Frame rate up-conversion methods interpolate the frame with two consecutive frames of the original source. This can be divided into the frame repetition method and motion estimation-based the frame interpolation one. Frame repetition has very low complexity, but it can yield jerky artifacts. The interpolation method based on a motion estimation and compensation can be divided into pixel or block interpolation methods. In the case of pixel interpolation, the interpolated frame was classified into four areas, which were interpolated using different methods. The block interpolation method has relatively low complexity, but it can yield blocking artifacts. The proposed method is the frame rate up-conversion method based on a block motion estimation and compensation using the linearity of motion. This method uses two previous frames and one next frame for motion estimation and compensation. The simulation results show that the proposed algorithm effectively enhances the objective quality, particularly in a high resolution image. In addition, the proposed method has similar or higher subjective quality than other conventional approaches.

Operation Characteristics of Coal Syngas Production and DME Conversion Facilities (석탄 합성가스 제조 및 화학원료(DME) 전환설비의 운전 특성)

  • In this study, the syngas producing facility that consists of pulverized coal feeding/gasification and hot gas clean-up system was tested for Indonesian subbituminous coal. And the DME conversion facilities have been developed and tested for converting syngas to DME by reactions with catalysts. So, the entrained-bed slagging type pi lot scale coal gasifier was operated normally in the temperature range of $1,400{\sim}1,450^{\circ}C,\;7{\sim}8kg/cm^2$ pressure. And Roto middle coal produced syngas that has a composition of $36{\sim}38%$ CO, $14{\sim}16%\;H_2,\;and\;5{\sim}8%\;CO_2$. Particulates in syngas were 99.8% removed by metal filters. $H_2S$ composition in syngas was also desulfurized by the Fe chelate system to yield less than 0.1 ppm level. When the clean syngas $70{\sim}100 Nm^3/h$ was provided to DME conversion rector, normally operated in the temperature range of $230{\sim}250^{\circ}C$ and $60kg/cm^2$ pressure, 4.5% DME was yielded.
