Fungal development is largely affected by many environmental factors. In a model filamentous fungus Aspergillus nidulans, asexual development is promoted by exposure of light, presence of salt and non-fermentable sugars. In other hand, sexual development is largely induced by absence of light, fermentable sugars and hypoxic condition. Also, some important genes including veA and nsdD play positive roles in activating sexual development. Here, we reported that the effect of high concentration of $CO_2$ on developmental decision in A. nidulans. When wild-type $veA^+$ strain was cultured in normal condition, sexual and asexual development occurred in balanced manner. However, high concentration of $CO_2$ (~5%) strongly activated sexual development and inhibited asexual development. Furthermore, this $CO_2$ effect was controlled by the veA or nsdD gene. High $CO_2$ culture of $veA^-$ or $nsdD^-$ mutant didn't activate sexual development, suggesting that the activation of sexual development induced by high $CO_2$ cannot overcome the genetic requirement of sexual development such as veA or nsdD. Since 5% $CO_2$ is an important condition for human pathogenic fungi for surviving and adapting in human body, this developmental pattern of A. nidulans affected by $CO_2$ concentration may provide interesting clues for comparative study with human fungal pathogens including Aspergillus fumigatus.
Endogenous 84 amino acid parathyroid hormone(PTH) is synthesized as a pre-pro hormone by the chief cells of the parathyroid glands. Physiological actions of PTH include regulation of bone metabolism, renal tubular reabsorption of calcium and phosphate, and intestinal calcium absorption. In addition, PTH stimulates new bone formation by extraordinary stimulation of osteoblastic activity and decreasing calcium excretion by the kidney. In this study, we constructed and tested retrovirus vectors designed to express the human parathyroid hormone(hPTH) gene under the control of the tetracycline-inducible promoters. To increase the hPTH gene expression at turn-on state, woodchuck hepatitis virus posttranscriptional regulatory element(WPRE) sequence was also introduced into retrovirus vector at downstream region of either the hPTH gene or the sequence encoding reverse tetracycline-controlled transactivator(rtTA). Transformed primary culture cells(porcine fetal fibroblast, PFF, chicken embryonic fibroblast, CEF) were cultured in the medium supplemented with or without doxycycline(tetracycline derivative) for 48 hours, and induction efficiency was measured by comparing the hPTH gene expression level using two step RT-PCR and ELISA Higher hPTH expression($3{\tims}10^4\;pg/ml,\;5.3{\times}10^4\;pg/ml$) and tighter expression control(up to 8 fold) were observed from the vector in which the WPRE sequence was placed at downstream of the hPTH gene. The resulting tetracycline inducible vector system may be helpful in solving serious physiological disturbance problems which have been a major obstacle in successful production of transgenic animals.
Effect of NO$_3$:NH$_4$ratio and pH of nutrient solution on nitrate content of leaves was investigated for leaf lettuce(Lactuca sativa L.) grown by a deep flow technique. Ratios(in me.L$^{-1}$ ) of NO$_3$:NH$_4$tested were 12:1, 10:3 and 8:5. The treatment of 8:5 NO$_3$:NH$_4$ratio had two solutions, one with uncontrolled pH and the other with automatically controlled pH. Solution pH continuously increased in 12:1 NO$_3$:NH$_4$treatment. Solution pH decreased gradually more as NH$_4$ratio increased. Treatment of 8:5 NO$_3$:NH$_4$with automatic pH control satisfactorily maintained the solution pH in the range of pH 5.5-6.0. Nitrate content in leaves was the greatest in treatment of 12:1 NO$_3$:NH$_4$ and the least in treatment of 8:5 NO$_3$:NH$_4$with automatic pH control. Fresh weight decreased in treatments of 10:3 and 8:5 NO$_3$:NH$_4$, whereas it increased in treatments of 12:1 and 8:5 NO$_3$:NH$_4$with pH control. It was concluded that the growth and leaf nitrate content were the greatest in high NH$_4$treatment with automatic pH control.
In an attempt to improve the productivity of ${\beta}-galactosidase$ from Bacillus sp. A1, which was isolated from soil and has remarkably higher transgalactosylation activity than lactose hydrolysis activity, a chemical mutation procedure using N-methyl-N'-nitro-N-nitrosoguanidine followed by selection was conducted. The final selection, designated as Bacillus sp. A4442, turned out to show a substantially increased enzyme productivity. Catabolite repression by glucose and lactose requirement as an inducer for the enzyme biosynthesis, which were shown in the parent strain, was markedly diminished; instead it was found out that galactose acts as another inducer. Because pH of medium, one of the most important factors for cell growth as well as enzyme production, is closely related with the sugar concentration during culture, it was kept in the optimum range of $6.5{\sim}7.5$; for this the initial glucose concentration was adjusted to be 0.5% which was thereafter maintained by the controlled pumping-in of lactose using the pH-stat technique. By doing so, we were able to increase the productivity of ${\beta}-galactosidase$ with high transgalactosylation activity up to $44\;unit/m{\ell}-broth$.

A new Phalaenopsis cultivar 'SM 333' was bred by Sangmiwon Orchid, Korea, which produces young plants through tissue culture techniques. The new cultivar 'SM 333', showing the phenotype of multiflora with pink color and small, multibranching-type characteristics, was derived from crossing between Phalaenopsis 'Odoriko' and 'Be Tris'. An elite individual number '02-03-33' later termed 'SM 333' was selected among about 300 individual progenies, based on an intensive selection process covering vegetative and flowering distinctiveness over more than 2 years. In year 2004-2005, the 1st and 2nd characteristic analyses were carried out through performance and uniformity tests. 'SM 333' shows flower color that is bright clean pink (RHS # RP69D) and flower shape that is formal type with 5.0 and 5.8 cm in flower height and width, respectively. 'SM 333' is regarded as raceme flower type suitable for the small casual flower market. The leaves of 'SM 333' grow horizontally and about 20.8 cm in length and 6.5 cm in width. This cultivar also possesses no genetic variation, and is amenable to fast in vitro propagation and easy growth due to its vigorous growth habit. This 'SM333' was registered (Reg. # 2916) with Korea Seed & Variety Service (KSVS) on 1st December, 2009, and the plant breeder's right is currently controlled by Sangmiwon Orchid Company, Korea.

Perhaps "Subject" is nothing more than the name of distance of object which is "too hot" to come close, and of this circulating movement. It's because of this object that the real subject stands against generalization and the subject can't be restored to any place in symbolic order-even though it is empty. The part which is told from Lacan's structural theory, that is to say, an importance to Lacan is that his Subject theory is not suggested or denied as a manual structure. On the contrary, it is a study of the relationship between the settled symbol that included in "real subject which is a unconscious one" and the symbolic subject hold- that is a metaphysical subject in general meaning. In Lacan's enlarged concept of subject beyond symbolic reality, it is noticeable that it gives justifiability to the union of a medium of different nature in artistic expression. We can recognize that the unconscious world is a living space which enables it to be a "condition of human being", not something dark under the surface of water through Magritte's(Rene Magritte, 1898~1967) surrealistic works. In other words, Magritte's art secures a core dimension of human nature through a mysterious gap of conscious and settled space. Magritte's drawings often evokes strange and unsettling feelings in people who view his paintings. This is because routine objects are found in "unsuitable" places from which we usually find them in our everyday lives. "Reality" in Magritte's paintings makes it aware that it is a strained field of concealment and disclosure basically between truths, and we can learn that his behavior to overturn to paint in-visible things is finally an effort to restore the "real subject" to the viewer's reality. In other words, such reversion arouses a nostalgic desire for the objects existing in their original appearance as they are - natural condition that our gaze had not been distorted yet by anamorphic stains. - and the state when we are conscious of them normally. Such desire offers an opportunity for us to get out of mental depression rather than operates to us as an abnormal crack. It's a successive process of effort to search for lost subject and Paradise Lost facing up to reality of subject human that is to be a subject of world and life are ousted from their place by structure and authority of culture.
In the 17th and 18th centuries, when the Ryukyu kingdom had been invaded and controlled by Japan, some official texts about 'history' were documented. In that texts, there are some myths about the kings-Syunten(舜天), Eiso(英祖), Satto(察度), Syouhasi(尙巴志), Syouen(尙圓)-who founded dynasties of Ryukyu kingdom. This study attempts to delineate the features and meaning of those founder myths as a polotial discourse for kingship. The results of this study are as follows. 1. The founder myths are discourses to describe the genealogy of the Ryukyu kingdom as the result of Mandate of Heaven(T'ien Ming, 天命). 2. In ancient okinawa, the sun was a symbol of ruler. The king of Ryukyu kingdom was thought as 'son of the sun', 'Tedako(てだこ, 太陽子) in the time of Syou dynasty(尙王朝). But in the founder myths, the only one Tedako is Eiso. 3. In the myths, the 'historical' founders are described as the king for agriculture. They were said to make the farming instruments with iron and rule the spring water for farming. The iron and water for fariming was thought to represent the power of king and the kingship. 4. The writers of myths aimed to be separated from the mythical kingship of past Ryukyu(古琉球). So, they threw the idea of Tedako(=mythical kingship) into the prison of mythical ages by rewriting myth of King Eiso. Instead of it, they built up the new image of agriculture-king who rules and controlls the iron and water for good farming. The meaing of this change, from 'shiny sun' to 'cultural king', is that the abstract and general power of kingship was turned into a concrete and restricted power of kingship. On the bases of it, there was the confucian political ideas from China, T'ien Ming thought(天命論). Under the influence of not only China but also Japan, the writers of myths attempt to find a new way for their kingdom by reconstructing kingship with ruminating over the kingship of past. The myth of founders in the 17th and 18th Centuries shows the aspects of kingship of Ryukyu kingdom and tells us that those myths are the political discourses identifying kingship and Ryukyu kingdom.
Life table experiments were performed in order to examine the effects of food quality on Moina macrocopa fed with four kinds of algal foods, Botryococcus sp., Scenedesmus subspicatus, Selenastrum capricornutum, and Chlorella sp., at $20^{\circ}C$ . The temperature effects on M. macrocopa were also assessed, feeding Chlorella at $17^{\circ}C$,$20^{\circ}C$,$25^{\circ}C$, and a combination of $28^{\circ}C$ (light) and $25^{\circ}C$ (dark). The cartilaginous Botryococcs cells were Inappropriate food for Moina. Among the foods tested, Chlorella was the food of the best quality in all accounts of life history traits. Moina grown on Chlorella showed higher net reproductive rate ($R_0$), longer mean and maximum longevities, earlier mean age at maturity, longer mean carapace length at maturity, larger mean clutch size, and shorter mean time interval between clutch productions than those grown on Selenastrum and Scenedesmus. An optimal temperature for Moina was $20^{\circ}C$ . When Moina were grown on Chlorella at $20^{\circ}C$, they showed the highest r, the highest $R_0$, the shortest T, the longest mean longevity, the earliest mean age at maturity, the longest mean carapace length at maturity, and the largest mean clutch size. The results of life table experiments showed that the individual and population growth patterns were much more affected by low temperature $17^{\circ}C$, than by high temperature ($\geq 20^{\circ}C$). In the optimal condition, the r value was very high, 5.1 in $d^{-1}$. In conclusion, the food quality and the temperature are the most important factors to govern the size and continuity of Moina population, by which the individual growth rates and reproductivity of members in the population can be controlled to survive in their environment such as small and temporary water bodies in nature.
This experiment was conducted to find out the possibility of use of reused rockwool and comparison of growth, productivity and quality of tomatoes according to the use of rockwool and coir medium. The experiment was conducted in an automatic controlled greenhouse at Andong National University, College of Life Science, located in Andong, Gyeongsangbuk-do.. As a result of the experiment, there was no difference in the number of leaves, plant height, and leaf area between treatments, and the crown diameter was slightly higher in rockwool medium, also there was no difference between reused rockwool and coir medium. Fruit productivity showed different responses depending on the cultivation environment, but there was no significant difference between rockwool, reused rockwool and coir medium. In addition, the quality of fruit was observed to be different according to the concentration of EC in the medium. Therefore, in tomato hydroponic cultivation, there was no difference in the type of medium in growth, productivity, fruit quality and the environmental and water management had a great effect, and it is expected that the reuse of rockwool will have a positive effect on the economic point of view.
Background: The purpose of the study was to explore whether the effect of Huanglian-jie-du granule depends on the baseline heart rate variability in patients with Hwa-byung. Methods: We used the supplementary data from a randomized, double-blind, placebo-controlled trial of Huanglian-jie-du granule for Hwa-byung patients. Study drugs were taken orally three times per day for one week. Heart rate variability was measured three times; before the treatment, after the treatment, and month follow-up period. Spearman's rho test was done to explore the role of the baseline heart rate variability to the effect of Huanglian-jie-du granule. Results: Baseline low frequency power of heart rate variability was negatively correlated with the reduction of the insomnia severity index in experimental group (r=-0.493 p=0.02). Also, Huanglian-jie-de granule significantly decreased the low frequency power of heart rate variability in patients with Hwa-byung compared to placebo (mean difference 266 [95% CI: 29~503], p=0.028). In Hwa-byung symptoms, stuffy, pushing up in the chest, and flush of anger were correlated with low frequency power of heart rate variability (p=0.010, p=0.000, and p=0.016, respectively). Conclusions: The effect of the Huanglian-jie-de granule for insomnia seems to be lower when the baseline low frequency power of heart rate variability is higher in Hwa-byung patients. Also, Huanglian-jie-de granule is likely to decrease the sympathetic activity in patients with Hwa-byung. Measurement of heart rate variability may be the useful to understand the state of Hwa-byung patients.
