  • 수소 생산량은 기질로부터 생산될 수 있는 수소의 분자수와 균주간의 특성에 의해서 좌우된다. 광합성 세균에 의한 수소 가스 생산은 유기물질 및 물을 전자 공여체로 하여 광합성에 의해 생산되는 것으로 알려져 있다. 이론적으로 수소생산량은 glucose 한 분자로부터 12분자의 수소가스가 생산된다.

  • This study was conducted to develop the simple and convenient irrigation control model which can maintain the appropriate rates of irrigation and drainage of nutrient solution according to the environmental conditions and growth stages in soilless culture of cucumber. In order to obtain fundamental data for development of the model, investigation of the actual state of soilless culture practices was carried out. Most irrigation systems of soilless culture were controlled by the time clock. Evapotranspiration of cucumber in soilless culture was investigated and correlations with environmental conditions were analyzed, and its prediction model was developed. A irrigation control model based on the time clock control and there were considered seasons, weather conditions, and growth stages was developed. Applicability of the model was tested by simulation. Drainage rates of irrigation system controlled by conventional time clock, integrated solar radiation, and the developed model were 61%, 20%, and 32%, respectively in cucumber perlite culture.

연골세포와 중간엽줄기세포의 3차원 Co-culture를 통한 연골화 향상 (Enhanced Chondrogenesis by Three-dimensional Co-culture of Chondrocytes and Mesenchymal Stem Cells)

  • Two-dimensional cultivation is typically used for cell growth, but the method reduces the characteristics of chondrocytes and stem cells, and limits culture area. Therefore, development of three-dimensional culture method is needed to mimic in vivo environment, improve quality of cells and scale-up efficiently. Improving proliferation and chondrogenesis is available by co-culture of chondrocytes and mesenchymal stem cells (MSCs) that leads to interaction between two kinds of cells. However, the co-culture has problems that permeability of sphere diminishes as aggregate size increased and ratio of two kinds of cells composing each spheres is different. In this work, co-cultivation method using controlled sphere composed of chondrocytes and MSCs was established and enhanced chondrogenesis. Periosteum-derived progenitor cells (PDPCs) that are appropriate for cell therapy source of articular cartilage were used as MSCs. Controlled spheres were formed in the hanging-drop plates and shifted for being induced chondrogenesis in 35-mm non-adhesive culture dishes at a rotation rate of 60 rpm. After inducing chondrogenesis, gene expressions related with chondrogenesis were found to be improved and it was apparent that the utilization of controlled spheres promoted chondrogenesis. As a result, available numbers of cells per unit area were increased and chondrogenic differentiation ability was improved compared to typical two-dimensional culture. This approach shows the potential in cartilage regeneration as it can provide sufficient numbers of chondrocytes.

배양 조건에 따른 Pseudomonas sp.CH-414의 Phospholipid 생산능의 변화 (Phospholipid Production by Pseudomonas sp.CH-414 under Various Culture Conditions)

  • Pseudomonas sp. CH-414를 이용하여 통기속도 빛 pH 변화에 대한 균체량 벚 인지질 생산 변화에 대한 실험을 수행하였다. pH를 7.0에서 8.0으로 변 화시켰을 때는 인지질의 생산량뿐만 아니라 균체량 이 동시에 급격하게 감소하였으나, pH를 7.0에서 6 6.0으로 낮추어 주였을 경우 pH 7.0과 비교하여 인 지질이 약 20% 정도 증가하였으며 균체량의 변화는 없었다. 통기속도가 증가할수록 균체량은 증가하였으나 인지질의 생산은 변화가 심하였으며 3vvm에서는 인지질 농도의 급격한 감소를 보였다. 이상의 결과를 토대로 배양 30시간까지는 pH 7.0, 3vvm으로 배양한 후 그 이후부터는 pH 6.0, 1 vvm 으로 변화를 주어 배양하였을 때 균체량은 약 18.5g d dry cell weight/$$\ell$ 이었고 인지 질의 양은 약 $0.83g/\ell$(45mg/g cell)이였다.

  • Emulsan is an extracellular emulsifying agent produced by the hydrocarbon-degrading Acinetobacter species RAG-1. In this study emulsan production of Acinetobacter calcpaceticus RAG-1 was investigated under various culture modes such as batch, fed-batch, membrane cell recycle, and continuous culture. The productions of emulsan under both ethanol-sufficient fed-batch and membrane cell recycle cultures were all 15.0U/ml, which was 53% increase in emulsan activity compared to that of pH controlled batch culture. Emulsan production was found to be strongly dependent on the residual ethanol concentration. In continuous culture the emulsan productivity increased with dilution rate.

  • Cephalosporium 배양시 균체의 형태학적 측면을 고려한 cephalosporin C 생합성에 대한 모델식을 제안하였다. 제한기질로 glucose와 methionine을 고려한 double-substrate 모델을 설정하였는데 여기서 glucose는 균체의 증식 정도를, methionine은 균체의 증식속도를 제어하게 된다. Cephalosporin C의 생산은 균체의 형태학적 분화와 밀접한 관계가 있다. 한편 cephalosporin C의 생산성을 증대시키기 위해 모델식을 유가식 배양에 응용하였다.

  • 광합성세균에 의한 광의존 수소생산에 있어서 상시 가변적인 태양광 이용조건에 대응되는 자동조절 반연속배양장치를 구성하였다. 이는 pulse 발생장치를 설비한 가스메터에 의해 일정의 수소생산량이 측정되면, 수소생산에 소비된 만큼의 기질이 timer와 연결된 정량펌프에 의해 공급될 수 있는 시스템이다. 이 배양장치를 이용하여, 옥외조건에서 Rhodopseudomanas sphaeroides B6의 agar gel 고정화균체에 의한 수소생산 실험을 시도하였다. 일조와 기후의 격심한 변동에도 불구하고 배양이 잘 조절되어 수소생산성이 유지되었다. 이는 광합성세균의 수소생산을 위한 옥외 반연속배양의 자동조절 가능성을 나타내는 것이다.

  • It is reported that mechanical stimulation takes a role in improving cell growth in skeletal system. And various research groups have showed that developed bioreactor to stimulate cell-seeded and threedimensional scaffold. In this study, we designed a custom-made bioreactor capable of applying controlled compression to cell-seeded agarose gel. This device consisted of a circulation system and compression system. In circular system, culture chamber was sealed for prohibiting contamination and media solution was circulated by pump. In compression system, mechanical stimuli were controlled by LabVIEW software and mechanical transfer system. Cell-encapsulated agarose gels were cultured for up to 7 days. There were significant differences between the number of cells grown in dynamic cell culture and in static cell culture from 3 days to 7 days.

습관성 유산에 대한 최신 연구 경향 및 실험적 방법 연구 (A study on the latest trend of researches concerning recurrent pregnancy loss, and controlled experiment on animals about antiphospholipid antibody)

  • Objectives : Recurrent pregnancy loss occurs in approximately 0.5-3% of women. There are many studies concerning immunological factor recently. Therefore, aim of this study is to examine the latest trend of researches concerning recurrent pregnancy loss, and controlled experiment on animals about antiphospholipid antibody. Method : We referred Pubmed site by using searching word of 'recurrent pregnancy loss' (Limits : 2000.1-2004.3, animal) Results and conclusions : 1. We searched 29 papers. Immunological factor : 18 ( about antiphospholipid antibody : 10 ), Chromosomal abnormality : 6, The others : 5 2. In five papers about controlled experiment on animals, (1) Materials : 8-12 weeks old mice / 11.5-day old-rat embryos / New Zealand rabbit (2) Inductions : inject intraperitoneally with human IgG containing antiphospholipid antibodies / culture in a solution of 1 ml medium which contained IgG purified from sera of women / inject intradermally with cardiolipin (3) Treatments : inject intraperitoneally with complement component before / culture in a solution of 1 ml medium which contained IgG purified from sera of women with SLE and RPL or from healthy women / inject intradermally with TFX, 0.9% NaCl (4) Measurements : weight fetuses and placentas, calculate frequency of fetal resorption / after culture, examine the embryos / examine platelet counts, APTT and numbers of live and dead newborns, resorbed fetuses, body mass, newborn viability and survival rates.
