Eicosapentaenoic Acid (EPA) Biosynthetic Gene Cluster of Shewanella oneidensis MR-1: Cloning, Heterologous Expression, and Effects of Temperature and Glucose on the Production of EPA in Escherichia coli

  • The putative EPA synthesis gene cluster was mined from the entire genome sequence of Shewanella oneidensis MR-1. The gene cluster encodes a PKS-like pathway that consists of six open reading frames (ORFs): ORFSO1602 (multi-domain beta-ketoacyl synthase, KS-MAT-4ACPs-KR), ORFSO1600 (acyl transferase, AT), ORFSO1599 (multi-domain beta-ketoacyl synthase, KS-CLF-DH-DH), ORFSO1597 (enoyl reductase, ER), ORFSO1604 (phosphopentetheine transferase, PPT), and ORFSO1603 (transcriptional regulator). In order to prove involvement of the PKS-like machinery in EPA synthesis, a 20.195-kb DNA fragment containing the genes was amplified from S. oneidensis MR-1 by the long-PCR method. Its identity was confirmed by the methods of restriction enzyme site mapping and nested PCR of internal genes orfSO1597 and orfSO1604. The DNA fragment was cloned into Escherichia coli using cosmid vector SuperCos1 to form pCosEPA. Synthesis of EPA was observed in four E. coli clones harboring pCosEPA, of which the maximum yield was 0.689% of the total fatty acids in a clone designated 9704-23. The production yield of EPA in the E. coli clone was affected by cultivation temperature, showing maximum yield at $20^{\circ}C$ and no production at $30^{\circ}C$ or higher. In addition, production yield was inversely proportional to glucose concentration of the cultivation medium. From the above results, it was concluded that the PKS-like modules catalyze the synthesis of EPA. The synthetic process appears to be subject to regulatory mechanisms triggered by various environmental factors. This most likely occurs via the control of gene expression, protein stability, or enzyme activity.

  • Arteria radialis is a branch of the brachial artery extending down the forearm around the wrist where it closes to skin surface. In the oriental medicine, the skin above arteria radialis has an important role because oriental medicine practitioners put their finger tips on the area, and diagnose patient's health conditions by feeling the pulsation of the arterial contraction. The finger tip diagnostic method relies on subjective decision of the practitioner; and there is a need to develop an objective diagnostic modality. The pulsation of the arterial contraction appears not only a movement on the site but also as temperature fluctuation due to pulsatile feeding of warmer blood. The goal of this study is to demonstrate a feasibility of using an infrared camera quantitatively to detect the temperature fluctuation on the skin. Clinical important three different areas, called chon, gwan, chuk, near a wrist where the arteria radialis reaches close to skin surface are marked with small pieces of surgical tape. A high-speed and high-resolution infrared camera with a 3 cm of field of view measures these areas for 10 second at 200 frames per second with a 320*240 pixel size. The pulsatile temperature fluctuation is calculated after passing a band pass filter to remove any stationary temperature over 10 second. The temperature fluctuation of a healthy male volunteer is measured at a room temperature as a control, and is compared with another measurement performed after 20 minutes staying in a room at a 40 degree Celsius. This comparison is repeated for three times, and indicates that the fluctuation increases after staying 20 minutes in the warm room. This increase becomes smaller when the person stays in the warm room with an acupuncture treatment that decreases body temperature. So that an objective diagnostics on the site may become feasible.

  • The steel plate reinforced concrete structure(SC structure) is suggested for the reasons of the saving of construction period, the saving of manpower and the advantage of quality control. The objective of this study is to evaluate basic structural behavior of SC structure under pure shear load, and shear with axial load condition and to suggest the method of in-plane pure shear loading. From the test results, structural behaviors of SC structure under pure shear load and shear with axial load were investigated the combination of validity of pure shear loading method by using 4 hinge frames was verified.

Characterization of Hibiscus Latent Fort Pierce Virus-Derived siRNAs in Infected Hibiscus rosa-sinensis in China

  • Although limited progress have been made about pathogen system of Hibiscus rosa-sinensis and Hibiscus latent Fort Pierce virus (HLFPV), interaction between plant host and pathogen remain largely unknown, which led to deficiency of effective measures to control disease of hibiscus plants caused by HLFPV. In this study, infection of HLFPV in Hibiscus rosa-sinensis was firstly confirmed for the first time by traditional electron microscopy, modern reverse transcription polymerase chain reaction and RNA-seq methods in China (HLFPV-Ch). Sequence properties analyzing suggested that the full-length sequences (6,465 nt) of HLFPV-Ch had a high sequence identity and a similar genomic structure with other tobamoviruses. It includes a 5'-terminal untranslated region (UTR), followed by four open reading frames encoding for a 128.5-kDa replicase, a 186.5-kDa polymerase, a 31-kDa movement protein, 17.6-kDa coat protein, and the last a 3'-terminal UTR. Furthermore, HLFPV-Ch-derived virus-derived siRNAs (vsiRNAs) ant its putative target genes, reported also for the first time, were identified and characterized from disease Hibiscus rosa-sinensis through sRNA-seq and Patmatch server to investigate the interaction in this pathogen systems. HLFPV-Ch-derived vsiRNAs demonstrated several general and specific characteristics. Gene Ontology classification revealed predicted target genes by vsiRNAs are involved in abroad range of cellular component, molecular function and biological processes. Taken together, for first time, our results certified the HLFPV infection in China and provide an insight into interaction between HLFPV and Hibiscus rosa-sinensis.

Unified equivalent frame method for post-tensioned flat plate slab structures

  • The post-tensioned (PT) flat plate slab system is commonly used in practice, and this simple and fast construction method is also considered to be a very efficient method because it can provide excellent deflection and crack control performance under a service load condition and consequently can be advantageous when applying to long-span structures. However, a detailed design guideline for evaluating the lateral behavior of the PT flat plate slab system is not available in current design codes. Thus, typical design methods used for conventional reinforced concrete (RC) flat plate slab structures have inevitably been adopted in practice for the lateral load design of PT flat plate structures. In the authors' previous studies, the unified equivalent frame method (UEFM) was proposed, which considers the combined effect of gravity and lateral loads for the lateral behavior analysis of RC flat plate slab structures. The aim of this study is to extend the concept of the UEFM to the lateral analysis of PT flat plate slab structures. In addition, the stiffness reduction factors of torsional members on interior and exterior equivalent frames were newly introduced considering the effect of post-tensioning. Test results of various PT flat plate slab-column connection specimens were collected from literature, and compared to the analysis results estimated by the extended UEFM.

The Evaluation of the Swing and Aerial Motion of Kovacs on Horizontal Bar (국내 철봉 선수들의 Kovacs의 흔들기 및 체공 동작에 대한 평가)

  • Objective: The aim of this study was to investigate the swing and aerial motion of Kovacs, and evaluate the skill level of Kovacs by Korean adult players on horizontal bar. Method: The subjects for this study were 6 male top athletes participated in the 46th National Gymnastics against Cities and Provinces. After the motions of Kovacs were filmed by digital highspeed camcorder setting in 90 frames/s, kinematical data were calculated through DLT method. The variables were computed in the lapse time, the joint angle, the position·velocity of body COG, the inferred tension force of bar, and body COG path were simulated according to skill level of Kovacs. Results: Firstly, it was revealed that the lapse time was 1.19±0.03 s in the swing phase, and 0.83±0.03 s in the aerial phase. Secondly, it was revealed that the shoulder·hip joint motions of S1 and S2 were better than the other subjects in the swing phase, and the knee joint motions of S1 and S2 were better than the other subjects in the aerial phase. Thirdly, it was revealed that the horizontal·vertical velocity of body COG were -1.40±0.03 m/s, 3.80±0.07 m/s respectively, and the vertical positions of S1 and S2 were higher a little than the other subjects. Lastly, the skill level of Kovacs of this subjects was evaluated into 3 steps; excellent, advanced, normal. They need to train the swing motion including a giant circle, and body motions in the air. Conclusion: It would be suggested that Korean domestic players should improve to increase the vertical velocity at release instant and train to control the limbs elaborately in the air.

Packet Loss Concealment Algorithm Using Pitch Harmonic Motion Estimation and Adaptive Signal Scale Estimation (피치 하모닉 움직임 예측과 적응적 신호 크기 예측을 이용한 패킷 손실 은닉 알고리즘)

  • In this paper, we propose a packet loss concealment (PLC) algorithm using pitch harmonic motion prediction and adaptive signal amplitude prediction and. The spectral motion prediction method divides the spectral motion of the previous usable frame into predetermined sub-bands to predict and restore the motion of the lost signal. In the proposed algorithm, the speech signal is classified into voiced and unvoiced sounds. In the case of voiced sounds, it is further divided into pitch harmonics using the pitch frequency to predict and restore the pitch harmonic motion of the lost frame, and for the unvoiced sound, the lost frame is restored using the spectral motion prediction method. When the continuous loss of speech frames occurs, a method of adjusting the gain using the least mean square (LMS) predictor is proposed. The performance of the proposed algorithm was evaluated through the objective evaluation method, PESQ (Perceptual Evaluation of Speech Quality) and was showed MOS 0.1 improvement over the conventional method.

Transcriptional profiles of rock bream iridovirus (RBIV) using microarray approaches

  • Rock bream iridovirus (RBIV) causes high mortality and economic losses in the rock bream (Oplegnathus fasciatus) aquaculture industry in Korea. Viral open reading frames (ORFs) expression profiling at different RBIV infection stages was investigated using microarray approaches. Rock bream were exposed to the virus and held for 7 days at 23 ℃ before the water temperature was reduced to 17 ℃. Herein, 28% mortality was observed from 24 to 35 days post infection (dpi), after which no mortality was observed until 70 dpi (end of the experiment). A total of 27 ORFs were significantly up- or down-regulated after RBIV infection. In RBIV-infected rock bream, four viral genes were expressed after 2 dpi. Most RBIV ORFs (26 genes, 96.2%) were significantly elevated between 7 and 20 dpi. Among them, 12 ORF (44.4%) transcripts reached their peak expression intensity at 15 dpi, and 14 ORFs (51.8%) were at peak expression intensity at 20 dpi. Expression levels began to decrease after 25 dpi, and 92.6% of ORFs (25 genes) were expressed below 1-fold at 70 dpi. From the microarray data, in addition to the viral infection, viral gene expression profiles were categorized into three infection stages, namely, early (2 dpi), middle (7 to 20 dpi), and recovery (25 and 70 dpi). RBIV ORFs 009R, 023R, 032L, 049L, and 056L were remarkably expressed during RBIV infection. Furthermore, six ORFs (001L, 013R, 052L, 053L, 058L, and 061L) were significantly expressed only at 20 dpi. To verify the cDNA microarray data, we performed quantitative real-time PCR, and the results were similar to that of the microarray. Our results provide novel observations on broader RBIV gene expression at different stages of infection and the development of control strategies against RBIV infection.

Construction of a Novel Shuttle Vector for Tetragenococcus species based on a Cryptic Plasmid from Tetragenococcus halophilus

  • A cryptic plasmid (pTH32) was characterized from Tetragenococcus halophilus 32, an isolate from jeotgal, Korean traditional fermented seafood. pTH32 is 3,198 bp in size with G+C content of 35.84%, and contains 4 open reading frames (ORFs). orf1 and orf2 are 456 bp and 273 bp in size, respectively, and their translation products showed 65.16% and 69.35% similarities with RepB family plasmid replication initiators, respectively, suggesting the rolling-circle replication (RCR) mode of pTH32. orf3 and orf4 encodes putative hypothetical protein of 186 and 76 amino acids, respectively. A novel Tetragenococcus-Escherichia coli shuttle vector, pMJ32E (7.3 kb, Emr), was constructed by ligation of pTH32 with pBluescript II KS(+) and an erythromycin resistance gene (ErmC). pMJ32E successfully replicated in Enterococcus faecalis 29212 and T. halophilus 31 but not in other LAB species. A pepA gene, encoding aminopeptidase A (PepA) from T. halophilus CY54, was successfully expressed in T. halophilus 31 using pMJ32E. The transformant (TF) showed higher PepA activity (49.8 U/mg protein) than T. halophilus 31 cell (control). When T. halophilus 31 TF was subculturd in MRS broth without antibiotic at 48 h intervals, 53.8% of cells retained pMJ32E after 96 h, and only 2.4% of cells retained pMJ32E after 14 days, supporting the RCR mode of pTH32. pMJ32E could be useful for the genetic engineering of Tetragenococcus and Enterococcus species.

Exploring the Combined Use of LiDAR and Augmented Reality for Enhanced Vertical and Horizontal Measurements of Structural Frames (골조 수직, 수평 측정작업 시 LiDAR 및 AR 기술 적용방안 제시)

  • This study is centered on the combined use of LiDAR(Light Detection and Ranging) and AR(Augmented Reality) technologies during vertical and horizontal frame measurements in construction projects. The intention is to enhance the quality control procedure, elevate accuracy, and curtail manual labor along with time expenditure. Present methods for accuracy inspection in frame construction often grapple with reliability concerns due to subjective interpretation and the scope for human error. This research recommends the application of LiDAR and AR technologies to counter these issues and augment the efficiency of the inspection process, along with facilitating the dissemination of results. The suggested technique involves the collection of 3D point cloud data of the frame utilizing LiDAR and leveraging this data for checks on construction accuracy. Furthermore, the inspection outcomes are fed into a BIM (Building Information Modeling) model, and the results are visualized via AR. Upon juxtaposing this methodology with the current approach, it is evident that it offers benefits in terms of objective inspection, speed, precise result sharing, and potential enhancements to the overall quality and productivity of construction projects.
