자소음자(紫蘇飮子)가 GUINEA PIG의 기관지(氣管支) 평활근(平滑筋)에 미치는 영향(影響) (Effects of Jasoeumja Extract On the Contraction of Isolated Guinea pig Trachea Smooth Muscle)

  • This study was carried out to investigate the effect of Jasoeumja extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate its mechanism. The results were obtained as follows: 1. The isolated trachea smooth muscle was suspended in the Organ bath with oxygenated Kreb's Henseleit bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with Isometric transducer (Nacro F-60). The resting tension was approximately 0.5g. 2. The trachea smooth muscle of the isolated guinea pig was significantly relaxed by the administration of Jasoeumja extract. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine $10^{-4}M$ was significantly inhibited by Jasoeumja extract. 4. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine $10^{-4}M$ was considerably inhibited by Jasoeumja extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to 5-hydroxytryptamine $10^{-4}M$ was considerably inhibited by Jasoeumja extract.

  • This study was carried out to investigate the effect of Heapyoyangjintang and Haepyoejintang extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate its mechanism. The results were obtained as follows : 1. The isolated trachea smooth muscle was suspended in the organ bath with oxygenated kreb's Henseleit bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with Isometric transducer(nacro F-60). The resting tension was approximately 0.5g. 2. The trachea smooth muscle of the isolated guinea pig was significantly relaxed by the administration of Haepyoyangjintang and Haepyoejintang extract. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine$10^4M$ Was significantly inhibited by Heapyoyangjintang and Haepyoejintang extract. 4. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine$10^4M$ was considerably inhibited by Haepyoyangjintang and Haepyoejintang extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to 5-hydorxytrtrypptamine $10^4M$ was considerably inhibited by Haepyoyangjintang and haepyoejintang extract.

  • The following results were obtained from the observation on the change of plasma cortisol concentration in the experiment of intravenous administration of Haengsoyeum Water Extract in the rabbit. And the effects of Haengsoyeum extract on the contractile force of the isolated guinea pig trachea smooth muscle. 1. In intravenous administration the plasma cortisol concentration increased significantly about 1 hours after with a does of 0.2 ml/kg. 2. In intravenous administration the plasma cortisol concentration increased significantly about from 1 to 3 hours after with a does of 0.4 ml/kg. 3. The contractile response of the trachea smooth muscle of isolated guinea pig to histamine $10^{-4}\;M$ was significantly inhibited by Haengsoyeum extract. 4. The contractile response of the trachea smooth muscle of isolated guinea pig to acetylcholine $10^{-4}\;M$ was considerably inhibited by Haengsoyeum extract. 5. The contractile response of the trachea smooth muscle of isolated guinea pig to 5-hydroxytryptamine $10^{-4}\;M$ was inhibited by Haengsoyeum extract.

Cholecystokinin-pancreozymin의 식도절편 수축반응에 미치는 phenoxybenzamine의 영향 (Effect of Phenoxybenzamine on the Contraction of the Isolated Esophagus Strip Induced by CCK-PZ)

  • In this study, the effects of phenoxybenzamine and related drugs on the action of CCK-PZ and caerulein were examined in isolated gall bladder of guinea pig and higher esophagus strip of fowl. The strips were placed in a bath containing Locke-Ringer solution maintained at $38^{\circ}C$. Oxygen was continuously bubbled through the solution. The contractile response was measured isometrically by a force displacement transducer connected to polygraph. In isolated gall bladder preparation caerulein produced contractile response of CCK-PZ type, but the relative potency on a weight basis was 30 times stronger than CCK-PZ. The response of caerulein or CCK-PZ was not blocked by cholinergic blocking agent and both alpha and beta adrenergic blockades, however, the response of caerulein or CCK-PZ was exceptionally blocked by phenoxybenzamine. In isolated esophagus strip CCK-PZ with high concentration produced marked contraction which was not modified by atropine and other blocking agents, whereas the response was blocked by phenoxybenzamine. These results lead to the conclusion that phenoxybenzamine inherently inhibits the contractile response of CCK-PZ and caerulein on esophagus and other smooth muscle.

가토의 근위와 원위대장 평활근의 운동성 비교 (Comparision of Proximal and Distal Large Intestinal Motility in Rabbit)

  • To validate the comparision of proximal and distal large intestinal motility, the amplitude and frequency of spontaneous motility, the effect of acetylcholine, the effect of atropine on the response of acetylcholine, the effect of histamine and the effect of pyrilamine and cimetidine on the response of histamine were investigated in rabbit. The results were summarized as follows: 1. The amplitude of spontaneous motility was more powerful on the proximal large intestine than that of the distal large intestine, but the frequency of spontaneous motility was similar on the both proximal and distal large intestine in rabbit. 2. Acetylcholine caused the contraction of proximal and distal large intestine, and the contractile response were increased between the concentration of acetylcholne $10^{-9}$ and $5{\times}10^{-6}M$ and $10^{-7}$ and $10^{-4}M$ on the proximal and distal large intestine, respectively, with dose-dependent manner in rabbit. 3. The contractile response induced by acetylcholine was completely blocked by the post-treatment with cholinergic receptor blocker, atropine $10^{-6}M$. 4. Histamine caused the contraction of proximal and distal large intestine and the contractile response were increased between the concentration of histamine $10^{-9}$ and $5{\times}10^{-5}M$ and $10^{-5}$ and $10^{-3}M$ on the proximal and distal large intestine, respectively, with dose-depend ent manner in rabbit. 5. The contractile response induced by histamine was completely blocked by the pretreatment with $H_1$-receptor blocker, pyrilamine $10^{-6}M$, but not blocked by the pretreatment with $H_2$-receptor blocker, cimetidine $10^{-6}M$.

  • These studies were carried out to investigate the effect of Insameumja water extract on the inhibitory contractile action of acetylcholine in guinea pig. The results of these studies were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in guinea pig was significantly inhibited by Insameumja. 2. Dose-response of acetylcholine pretreated Insameumja in guinea pig was not significantly changed. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in guinea pig was decreased. 4. Inhibitory contractile action of acetylcholine pretreated indomethacin was significantly changed by Insameumja. 5. Inhibitory contractile action of acetylcholine pretreated methylene blue was not significantly changed by Insameumja.

  • These studies were carried out to investigate the effects of Ohotang water extract on the inhibitory contractile action of acetylcholine in control rat. The results of these were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in control rat was significantly inhibited by Ohotang. 2. Dose-response of acetylcholine pretreated Ohotang in control rat was significantly inhibited. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in control rat was significantly inhibited by Ohotang. 4. Contractile force of acetylcholine pretreated indomethacin from trachea smooth muscle in control rat was not significantly changed. 5. 개 적출 신동맥에 있어서 histamine receptor의 생리적 특성 (Physiological characteristics of histamine receptor of the isolated renal artery in dog)

  • To elucidate the physiological characteristics of histamine receptor of the isolated renal artery in dog, effects of various receptor blockers and $Ca^{{+}{+}}$ channel blockers on the contractile responses of histamine were investigated in the isolated renal artery of dog. The results were summarized as follows: 1. Histamine caused the contraction in the isolated renal artery of dog, and the contractile responses increased between the concentration of $5{\times}10^{-6}M$ and $10^{-2}M$ in dose-dependent manner. 2. The contractile response induced by histamine ($10^{-3}M$) was not blocked by pretreatment with atropine ($10^{-6}M$), phentolamine ($10^{-6}M$), propranolol ($10^{-6}M$) or cimetidine ($10^{-6}M$), but was completely blocked by pretreatment with pyrilamine ($10^{-6}M$). 3. The contractility induced by histamine ($10^{-3}M$) was decreased in the $Ca^{{+}{+}}$ free medium. 4. The contractile response induced by histamine ($10^{-3}M$) was markedly inhibited by pretreatment with papaverine ($5{\times}10^{-5}M$) or verapamil ($5{\times}10^{-5}M$). 5. After the pre-contraction induced by norepinephrine ($10^{-7}M$) or prostaglandin $F2{\alpha}$ ($5{\times}10^{-6}M$), the contractile response induced by the additional treatment of histamine ($10^{-3}M$) was markedly increased compared with that of histamine ($10^{-3}M$) alone.
