For large and rapid screening of high-yielding mutants of lovastatin produced by filamentous fungal cells of Aspergillus terreus, one of the most important stage is to test as large amounts of mutated strains as possible. For this purpose, we intended to develop a miniaturized cultivation method using $7m{\ell}$ culture tube instead of traditional $250m{\ell}$ flask (working volume $50m{\ell}$). For obtaining large amounts of conidiospores to be used as inoculums for miniaturized cultures, 4 components i.e., glucose, sucrose, yeast extract and $KH_2PO_4$ were intensively investigated, which had been observed to show positive effect on enhancement of spore production through Plackett-Burman design experimet. When optimum concentrations of these components that were determined through application of response surface method (RSM) based on central composite design (CCD) were used, maximum spore numbers amounting to $1.9\times10^{10}$ spores/plate were obtained, resulting in approximately 190 fold increase as compared to the commonly used PDA sporulation medium. Using the miniaturized cultures, intensive strain development programs were carried out for screening of lovastatin high-yielding as well as highly reproducible mutants. It was observed that, for maximum production of lovastatin, the producers should be activated through 'PaB' adaptation process during the early solid culture stage. In addition, they should be proliferated in condensed filamentous forms in miniaturized growth cultures, so that optimum amounts of highly active cells could be transferred to the production culture-tube as reproducible inoculums. Under these highly controlled fermentation conditions, compact-pelleted morphology of optimum size (less than 1 mm in diameter) was successfully induced in the miniaturized production cultures, which proved essential for maximal utilization of the producers' physiology leading to significantly enhanced production of lovastatin. As a result of continuous screening in the miniaturized cultures, lovastatin production levels of the 81% of the daughter cells derived from the high-yielding producers turned out to be in the range of 80%$\sim$120% of the lovastatin production level of the parallel flask cultures. These results demonstrate that the miniaturized cultivation method developed in this study is efficient high throughput system for large and rapid screening of highly stable and productive strains.
One of the objectives of this work is to obtain information relevant to the industrial production of alcohol from sugar. The fermentation of alcohol by a strain of saccharomycess cerevisiae ATCC 24858 was studied In a continuous single-stage process with recycle of the cells via tangential flow microfiltration membranes. The experimental results reported in this study pertain to continuous cultures with total cell-recycle by varying the dilution rate (D=0.3, 0.5, and 0.7 $hr^{-1}$) and glucose concentration (50, 100, 150, and 200g/l sugar solution). Productivity using a repeated cell recycle system was found extremely high, 1.e., over 10 to 29 times higher than that of a smile batch system. When a sugar concentration of 200g/1 at dilution rate, 0.7 hr-1 was used, 83.9g/l ethanol was formed with an ethanol yield of 0.42(82% of theoretical) based on sugars utilized.
We evaluated a more practical and cost-effective immobilization carriers for ethanol production using the yeast Saccharomyces cerevisiae. Three candidate materials-rice hull, rice straw, and sawdust-were tested for their cell-adsorption capacity and operational durability. Derivatizations of rice hull, rice straw, and sawdust with the optimal concentration of 0.5 M of 2-(diethylamino)ethyl chloride hydrochloride (DEAE HCl) resulted in > 95% adsorption of the initial yeast cells at 2 hr for DEAE-rice hull and DEAE-sawdust and in only approximately 80% adsorption for DEAE-rice straw. In addition, DEAE-sawdust was found to be a more practical carrier for immobilizing yeast cells in terms of operational durability in shaking flask cultures with two different speeds of 60 and 150 rpm. Furthermore, the biosorption isotherms of DEAE-rice hull, -rice straw, and -sawdust for yeast cells revealed that the $Q_{max}$ of DEAE-sawdust (82.6 mg/g) was greater than that of DEAE-rice hull and DEAE-rice straw. During the 404-hr of continuous column reactor operation using yeast cells immobilized on DEAE-sawdust, no serious detachment of the yeast cells from the DEAE-sawdust was recorded. Ethanol yield of approximately 3.04 g/L was produced steadily, and glucose was completely converted to ethanol at a yield of 0.375 g-ethanol/g-glucose (73.4% of the theoretical value). Thus, sawdust is a promising practical immobilization carrier for ethanol production, with significance in the production of bioethanol as a biofuel.

The level of sunk cost and risk-taking theory have been offered as one explanation for the escalation of commitment behavior. This Study attempted to replicate Keil's study in Korea. Keil examined the level of sunk cost associated with the risk propensity and risk perception of decision-makers, and these factors are assessed for cross-cultural robustness using matching laboratory experiments carried out in three countries. The level of sunk cost and the risk perception of decision-makers contributed significantly to their continuous willingness to their project. Moreover, the risk propensity of decision-makers was inversely related to risk perception, and this inverse relationship was significantly more weak in Korea than in Singapore. These results show that the sunk-cost effect exists across cultures, and that the risk-taking behaviors are partially mediated by cultural factors.
When reserpine-producing cell strains of Rauwolfia serpentina were transferred from the dark to the light irradiation, the production of reserpine was extremely enhanced whereas the cell growth was suppressed. In an incubation period of 20 days, the most effective culture condition for reserpine production was the combination of 8 days of dark culture and following 12 days of light culture. The time courses of both cell growth and reserpine production were measured in vitro in order to clarify the effect of wave length range of light on the biosynthesis of reserpine. Although the growth of cultured cells which had been incubated under continuous red, yellow, and green lights, respectively, was similar to that of the cultured cells subcultured in the dark. The cells cultured under red light irradiation produced less reserpine than dark-grown cultures. Both blue and near-ultraviolet light inhibited the growth of cultured cells. The production of reserpine was strikingly enhanced by blue light, but was strongly inhibited by near-ultraviolet light.

This study was to establish the use of parthenogenetic mouse ES (P-mES02) cells as a reproducible differentiation system for mouse cardiomyocytes. To induce differentiation, P-mES02 cells were dispersed by dissociation and the formation of ES cell aggregates in differentiation medium. After 7 days in differentiation culture, the embryoid bodies (EBs) were plated onto gelatin-coated dish. Cultures were observed daily using an inverted light microscope to determine the day of contraction onset and total duration of continuous contractile activity for each contracting focus. (omitted)

The goal of the current research was to assess the influence of the growth rate of Nocardia amarae on its overall metal binding capacity. Batch sorption isotherms for cadmium (Cd), copper (Cu), and nickel (Ni) showed that Nocardia cells harvested from chemostat cultures at a dilution rate of $0.33d^-1$ had a significantly higher metal sorption capacity than cells grown at 0.5 and $1d^-1$. The cell surface area estimated using a dye technique indicated that pure N. amarae cells grown at a lower growth rate had a significantly more specific surface area than cells harvested from a higher growth rate operation. Accordingly, this difference in the specific surface area seemed to indicate that the higher metal sorption capacity of the slowly growing Nocardia cells was due to their higher specific surface area.
The stabilities of plasmid vectors in Zymomonas mobilis were tested in batch and continuous cultures. It was found that the growth of the host Zymomonas strain was greatly affected by the size of plasmids as well as the composition of nutrient media: the host cells grew taster when harboring plasmids of smaller sizes and in n non-selective medium. All the Zymomonas plasmid vectors containing antibiotics selective markers and Zymomonas replication origins could be maintained in a stable manner over 30 generations without being integrated into host chromosomes.
In order to investigate the characteristics for RNA accumulation, continuous cultures of S. cerevisiae MTY62 under the carbon-limitation or carbon-phosphate-limitation were performed. In the carbon-limited chemostat, the highest RNA content of 144 mg-RNA/g-dry cell was observed at the dilution rate of $0.30\;hr^{-1}$. In the carbon and phosphate-limited chemostat, the culture condition with the dilution rate of $0.25\;hr^{-1}$ gave the maximal RNA content of 209 mg-RNA/g-dry cell. The accumulation rate of RNA $(mg-RNA/L\;{\cdot}\;hr)$ was decreased at higher dilution rate in the carbon and phosphate-limited chemostat.
In this study, significant factors influencing on the quality and stability of fermented sausage, such as materials, processing conditions, and microbiological characteristics as well as topography during ripening, were documented. Since most fermented sausages are not heated during manufacture or before consumption, a strict control of the growth of pathogens and the selection of favourable conditions that encourage the specific growth and development of desirable microflora are particularly important. With respect to microbiological safety, hurdles, i.e., preservations(nitrite), redox potential, competitive flora, acidity(pH), and water activity($a_{w}$) are matters of importance to prevent proliferation of bacterial pathogens. Today, for ensuring the safety and quality of the final product, the application of starter cultures in combination with the proper processing is subsequently used in practice. For improving the efficiency of microbiological utility in the production of fermented sausages, the understanding of their topography is essential. The documented different points must be taken into account when HACCP systems set up for the manufacture of fermented sausages. There are continuous researches concerning desirable improvements to sausage fermentation with health enhancing properties.
