벼에 대한 원소상 요오드 증기$(I_2)$의 직접오염경로 해석 실험 (An Experimental Study on the Direct Contamination Pathway of Elemental iodine Vapor $(I_2)$ for Rice Plants)

  • 피폭상자 내에서 벼를 생육단계별로 80 분 간 $I_2$ 증기에 피폭시키고 작물체 직접오염 관련 인자를 조사하였다. $I_2$ 증기의 경엽부 침적속도$(m\;s^{-1})$는 피폭시기에 따라 $1.4{\times}10^{-5}-1.3{\times}10^{-4}$의 범위로 생육 전반기 피폭시 비교적 낮았다. 이삭 침적속도는 $2.5{\times}10^{-5}-6.7{\times}10^{-5}$의 범위였다. 전체 작물체에 대한 침적속도는 $1.4{\times}10^{-5}-1.8{\times}10^{-4}$의 범위로 8월 18일(출수 시작후 7일) 피폭시 가장 높았다. 침적속도를 생육밀도 기준으로 나타내면 시기에 따른 변이 정도가 대체로 감소하였다. 기온, 일광 및 습도에 대한 이렇다할 침적속도의 경향성은 관찰되지 않았다. 요오드의 현미 전류계수는 $3.3{\times}10^{-5}-4.7{\times}10^{-4}$의 범위로 8월 23일 피폭에서 가장 높았다. 출수전 경엽부에 침적한 요오드도 상당량 현미로 전류하는 것으로 나타났다.

위해성 평가 기법에 따른 토양 불소 기준안 연구 (Derivation of Soil Fluorine Standards Based on a Human Health Risk Assessment Method)

  • This study established risk-based fluoride soil contamination standards according to the Korean Soil Contaminant Risk Assessment Guidelines (SRAG). Ten exposure scenarios were evaluated, broadly categorized into Scenario 1, which used the default parameters from the current SRAG, and Scenario 2, which used the latest exposure factors and bio-concentration factors. Fluoride soil standards corresponding to a total hazard index (HI) of 1.0 were determined for each scenario. For children in agricultural areas, the derived risk-based soil fluoride standard was 70 mg/kg for Scenario 1 and 27 mg/kg for Scenario 2. In industrial areas, the risk-based fluoride soil standard was 2200 mg/kg in Scenario 1 and 2300 mg/kg in Scenario 2. This study clearly demonstrated that the crop ingestion exposure pathway exerted predominent influence on the estimated human health risk standards. Additionally, using the Added Risk Approach and considering soil background concentrations, the total fluoride soil standards for residential areas ranged from 232 mg/kg to 444 mg/kg, while the standards for industrial areas ranged from 2405 mg/kg to 2674 mg/kg.

해양 환경의 미세 플라스틱과 인간의 건강에 미치는 영향 (Microplastics in the Marine Environment and Their Impacts on Human Health)

  • 미세 플라스틱은 입자의 크기가 5 mm 이하인 플라스틱 조각을 말하며 미세 플라스틱의 오염은 해양 생태계와 인간의 건강과 관련되어 전 세계적인 관심사이다. 광범위하게 오염되어 있는 관계로, 미세 플라스틱은 물고기, 담치, 굴, 조개, 가리비와 같이 다양한 동물에 섭취되어 체내에 축적된다. 게다가, 섭취된 미세 플라스틱은 소장, 간, 신장 그리고 심지어 뇌에서도 높은 농도로 관찰된다. 해산물은 인간에게 있어 주요 단백질 공급원 중 하나이므로, 해산물의 소비는 인간이 미세 플라스틱에 노출되는 경로가 될 수 있다. 많은 근거들은 설치류에서 미세 플라스틱의 반복적인 경구 투여가 생식, 심장, 소화기, 내분비 그리고 심지어 신경계에서 병리적, 기능적 변화를 유도함을 가리킨다. 더욱이, 임신기와 수유기 동안 모체가 미세 플라스틱에 노출되면 새끼에서 대사의 항상성에 변화가 일어난다. 해산물은 세계적으로 3억 1천만 명 이상의 사람들에게 20% 이상의 단백질 공급원이라는 사실을 고려할 때, 미세 플라스틱은 인간의 몸에 축적되어 생리적 기능에 장애를 유발할 수 있다고 가정하는 것이 타당하다. 본 리뷰에서 우리는 해양에서 미세 플라스틱 오염의 현재 실태와 해양 동물 및 설치류에서 미세 플라스틱의 축적과 독성, 그리고 인간에게의 노출과 인간 건강에 미치는 잠재적인 영향에 대해 요약하였다.

내열성 효소를 이용한 전분으로부터 6-인산과당의 제조 (Production of Fructose 6-Phoschate from Starch Using Thermostable Enzymes)

  • 인산당은 모든 유기체에서 발견되며 무척 다양한 유용성을 지니고 있다. 특히 glucose 1-phosphate (G1P), glucose 6-phosphate (G6P), fructose 6-phosphate (F6P) 등은 해당과 정, 당합성과정, 5탄당 인산화과정 및 캘빈회로와 같은 탄수화물 대사와 에너지 생산 대사의 주요한 핵심 중간물질이다. 특히 해당과정에서 F6P는 G6P의 이성질화반응에 의하여 생성된다. F6P의 대량생산은 전분을 이용하는 것이 가능한데, 우선 전분에 인산화효소를 가하여 G1P를 얻고, 이 G1P를 자리옮김효소 (phosphoglucomutase, GM)와 이성질화효소 (phosphoglucoisomerase, GI)를 순차적으로 적용하여 G6P와 F6P를 생산하게 된다. 효소반응의 경우 전분의 용해도 증가, 반응속도의 향상 및 미생물의 오염방지 등을 위하여 중온성 효소보다는 고온성 효소 혹은 내열성 효소가 선호된다. 본 연구는 세 가지 내열성 효소를 이용하여 전분으로부터 두 단계반응으로 F6P를 생산하는 것에 관한 것이다. 실험에 사용된 효소는 대장균에서 발현된 재조합 효소로서, 효소의 생산은 유가식 배양을 이용하였다. 1.2% 가용성 전분 200 L를 이용하여 1,253 g의 순수한 G1P를 생산하였으며 이를 이용하여 최종적으로 30% 수율로 F6P를 생산할 수 있었다. 최대수율을 얻기 위하여 반응표면분석법을 이용하여 GM : GI = 1 : 1.23, 63.5$^{\circ}C$, pH 6.85의 조건이 도출되었으며, 이 조건하에서 실험을 통하여 20 g/L의 전분을 이용하여 30% 수율로 F6P가 생성됨을 확인할 수 있었다.

해양바이오물질이 PCB의 독성작용에 미치는 영향 (Effects of Chitosan on the Toxicity of Environmental Pollutants)

  • 산업의 발달과 함께 환경오염에 대한 국민적인 관심도는 날로 증가하고 있다. PCB는 우리 주변에 널리 퍼져 있고 먹이사슬을 통해 체내에도 축적되어 인체의 위해성이 우려되는 대표적인 환경오염물질이다. PCB의 노출은 성장기의 두뇌에서 가장 큰 신경독성을 나타내며 영아 및 유아는 상대적으로 높게 노출되어 위험집단으로 분류된다. 본 연구는 PCB의 신경독성에 구조-활성관계가 미치는 영향을 분석하고 PCB에 의한 독성을 최소화 할 수 있는 방안으로서 해양활성물질의 사용가능성을 이해하고자 하였다. PCB노출에 따른 신경세포의 신호전달 체계변화를 분석하기 위하여 Protein Kinase C (PKC)의 변화를 측정하였다. PKC의 전체적인 활성을 [$^3H$]PDBu로 분석한 결과 ortho-position(PCB-105, -123)을 가지고 있는 PCB가 non-ortho (pCB-77, -81) 구조보다 신경에 미치는 영향은 더 높았다. Westem blot 결과 PKC isofonn 중에는 PKC-beta II 및 epsilon의 경우 ortho-position PCB에서 더 높은 활성을 보였다. 이러한 PKC의 변화는 성장기 신경세포에서 신호전달기작의 변화에 많은 영향을 미치므로 이를 예방하거나 차단 할 수 있는 물질을 발견하고자 다양한 키토산을 처리하였다. 그 결과 1백만 달톤 이상의 고분자 키토산의 경우 PCB에 의한 신호전달 기작 변화를 억제할 수 있음을 보였다. 본 연구는 환경오염 등에 의한 독성예방에 키토산의 활용가능성을 제시하였다.

Polycyclic Aromatic Hydrocarbons (PAHs) and their Bioaccessibility in Meat: a Tool for Assessing Human Cancer Risk

  • Polycyclic aromatic hydrocarbons (PAHs) are primarily formed as a result of thermal treatment of food, especially barbecuing or grilling. Contamination by PAHs is due to generation by direct pyrolysis of food nutrients and deposition from smoke produced through incomplete combustion of thermal agents. PAHs are ubiquitous compounds, well-known to be carcinogenic, which can reach the food in different ways. As an important human exposure pathway of contaminants, dietary intake of PAHs is of increasing concern for assessing cancer risk in the human body. In addition, the risks associated with consumption of barbecued meat may increase if consumers use cooking practices that enhance the concentrations of contaminants and their bioaccessibility. Since total PAHs always overestimate the actual amount that is available for absorption by the body, bioaccessibility of PAHs is to be preferred. Bioaccessibility of PAHs in food is the fraction of PAHs mobilized from food matrices during gastrointestinal digestion. An in vitro human digestion model was chosen for assessing the bioaccessibility of PAHs in food as it offers a simple, rapid, low cost alternative to human and animal studies; providing insights which may not be achievable in in vivo studies. Thus, this review aimed not only to provide an overview of general aspects of PAHs such as the formation, carcinogenicity, sources, occurrence, and factors affecting PAH concentrations, but also to enhance understanding of bioaccessibility assessment using an in vitro digestion model.

Aflatoxin과 그 생성(生成)에 관련되는 주요인(主要因) (Aflatoxin: Factors Affecting Aflatoxin Production)

  • Aflatoxins are toxic and carcinogenic secondary metabolites which are produced by trains of A. flavus and A. parasiticus during their growth on foods and feedstuffs. Aflatoxins are a group of closely related heterocyclic compounds of which $B_1$, $B_2$, and $G_2$ are the major members. Aflatoxins are synthesized via a polyketide pathway in which the general steps are acetate, an-thraquinones, xanthone and aflatoxins. Aflatoxin formation is favored by high moisture or high $a_w$(0.95${\sim}$0.99). The limiting $a_w$ for aflatoxin production on agricultural commodities is 0.83. Optimum temperature for aflatoxin production by the molds is $25{\sim}30^{\circ}C$ and the incubation time for the maximum production of the toxin is 7${\sim}$15 days. The limiting temperatures for aflatoxin production are ${\leq}7.5^{\circ}C\;and\;\geq40^{\circ}C$. Cycling temperatures may or may not stimulate aflatoxin production depending on the amplitude of cycling, substrate and strains of molds. Aflatoxin pro-ducing molds are aerobic organisms and thus have a requirement for oxygen. A decreasing $O_2$ concentration and/or increasing concentrations of $CO_2$ or $N_2$ depress the mold growth and aflatoxin formation. A. flavus grows competitively or associatively in the presence of other microorganisms and occasionally loses the competition with other microorganisms. Some lactic acid bacteria have been shown to reduce growth and aflatoxin production by A. parasiticus. Carbon source is the most important nutritional factors affecting aflatoxin formation by the molds. Sucrose, fructose and glucose are the most favorable carbon sources. Food substrates of plant derived products which have high carbohydrate content such as agricultural commodities and their products are most vulnerable to contamination by aflatoxins.

광주지역 황사시 미세먼지 유입경로별 중금속 오염도 평가 (Estimation of Heavy Metal Contamination by PM10 Inflow Pathways while Asian Dust in Gwangju)

  • The purpose of this study is to investigate the relationship of fine dust PM10 and heavy metals in PM10 in Asian dust flowing into Gwangju from 2013 to 2018. The migration pathways of Asian dust was analyzed by backward trajectory analysis using HYSPLIT (Hybrid Single Particle Lagrangian Integrated Trajectory) model, and the change of heavy metal concentration and heavy metal content per 1 ㎍/㎥ of fine dust PM10 in Gwangju area were analyzed. Also, the characteristics of the heavy metals were analyzed using the correlation between the heavy metals in PM10. As a result of analyzing Asian dust entering the Gwangju region for 6 years, the average concentration of PM10 measured in Asian dust was 148 ㎍/㎥, which was about 4.5 times higher than in non-Asian dust, 33 ㎍/㎥. A total of 13 Asian dust flowed into the Gwangju during 6 years, and high concentration of PM10 and heavy metals in that were analyzed in the C path flowing through the Gobi/Loess Plateau-Korean Peninsula. As a result of the correlation analysis, in case of Asian dust, there was a high correlation between soil components in heavy metals, so Asian dust seems to have a large external inflow. On the other hand, in case of non-Asian dust, the correlation between find dust PM10 and artificial heavy metal components was high, indicating that the influence of industrial activities in Gwangju area was high.

대형조선소 천장크레인 운전원의 용접흄 노출 실태 (Exposure status of welding fumes for operators of overhead traveling crane in a shipyard)

  • Objectives: Operators of overhead traveling crane in a ship assembly factory perform work to transmit large vessel blocks to an appropriate working process. Hazardous matters such as metal dusts, carbon monoxide, carbon dioxide, ozone, loud noise and fine particles are generated by variable working activities in the factory. The operators could be exposed to the hazardous matters during the work. In particular, welding fumes comprised of ultra fine particles and heavy metals is extremely hazardous for humans when exposing a pulmonary through respiratory pathway. Occupational lung diseases related to welding fumes are increasingly on an upward tendency. Therefore, the objective of this study is to assess properly unknown occupational exposure to the welding fumes among the operators. Methods: This study intended to clearly determine an equivalence check whether or not chemical constituents and composition of the dusts, which existed in the driver's cab, matched up with generally known welding fumes. Furthermore, computational fluid dynamics program(CFD) was used to identify a ventilation assessment in respect of a contamination distribution of welding fumes in the air. The operators were investigated to assess personal exposure levels of welding fumes and respirable particulate. Results: The dust in an operation room were the same constituents and composition as welding fumes. Welding fumes, which caused by the welding in a floor of the factory, arose with an ascending air current up to a roof and then stayed for a long time. They were considered to be exposed to the welding fumes in the operation room. The personal exposure levels of welding fumes and respirable particulate were 0.159(n=8, range=0.073-0.410) $mg/m^3$ and 0.138(n=8, range=0.087-0.178) $mg/m^3$, respectively. They were lower than a threshold limit value level($5mg/m^3$) of welding fumes. Conclusions: These findings indicate that an occupational exposure to welding fumes can exist among the operators. Consequently, we need to be keeping the operators under a constant assessment in the operator process of overhead traveling crane.

Hexachlorobenzene Dechlorination Ability of Microbes from Canal and Estuary Sediments

  • This study aimed to investigate the hexachlorobenzene (HCB) dechlorinating ability of sediment microbes collected from a natural canal receiving secondary effluents from an industrial estate and nearby factories. Nine sites along the stream and one in the estuary in the Gulf of Thailand into which the canal spills were specified and sampling for sediment and water. Preliminary analysis of the sediments showed that the first four sites nearest to the discharging location were contaminated by HCB within the range of 0.18 to 1.25 ppm. Apart from that, 1,3,5-trichlorobenzene which has never been commercially produced or used in any manufacturing processes except for the transformation from higher chlorinated benzene was also identified in the range of 0.16 to 0.24 ppm. This suggested a possibility of sporadically HCB contamination in this stream. Of more important, people in the community along this canal earn their living by coastal fishery; hence, posing a risk of spreading HCB and its less chlorinated congeners via food chain from caught marine creatures to human. As a result, there is an urgent need to understand the behavior of HCB dechlorination in this stream sediment which can lead to a clean-up action in the future. Serum bottles with sediment slurries (sediment to water ratio of 1:1 (v/v) and filtered to remove particles larger than 0.7 mm) from each site were inoculated with 2 mg/l of HCB, kept anaerobically in the dark at room temperature without any nourishment, and analyzed for HCB and its less-chlorinated congeners every 6 days. Total chemical oxygen demand, suspended solids, and volatile suspended solids were in the range of 21,492-73,584, 158,100-518,100 and 6,000-32,700 mg/l, respectively. It was found that all sediment slurries began to dechlorinate HCB in 12 to 30 days and the HCB was completely removed within 42 to 60 days or so. On the other hand, there was no HCB dechlorination occurred in the controlled set which was sterilized by autoclaving prior to the addition of HCB. This implies that the HCB transformation was solely due to microorganisms' activities. HCB was dechlorinated principally via pentachlolobenzene to 1,2,3,5-tetrachlorobenzene and terminated at 1,3,5-trichlorobenzene which is the major pathway as reported by many researchers. Dichlorobenzene has not been detected in any samples within the dechlorination period of 60 days. The results indicate that the microbial matrix in the sediment of this stream has an outstanding capability to dechlorinate HCB. Existing substrates and nutrients which mainly sorbed onto the solid phase and the typical temperature in Thailand were sufficient and suitable to promote the activities of these HCB-dechlorinating microbes.
