Characterization of Styrene Catabolic Genes of Pseudomonas putida SN1 and Construction of a Recombinant Escherichia coli Containing Styrene Monooxygenase Gene for the Production of (S)-Styrene Oxide

  • Some Pseudomonas species can grow on styrene as a sole carbon and energy source. From the new isolate Pseudomonas putida SN1, the genes for styrene catabolism were cloned and sequenced. They were composed of four structural genes for styrene monooxygenase (styA and styB), styrene oxide isomerase (styC), and phenylacetaldehyde dehydrogenase (styD), along with two genes for the regulatory system (styS and styR). All the genes showed high DNA sequence (91% to 99%) and amino acid sequence (94% to 100%) similarities with the corresponding genes of the previously reported styrene-degrading Pseudomonas strains. A recombinant Escherichia coli to contain the styrene monooxygenase from the SN1 was constructed under the control of the T7 promoter for the production of enantiopure (S)-styrene oxide, which is an important chiral building block in organic synthesis. The recombinant E. coli could convert styrene into an enantiopure (S)-styrene oxide (ee >99%) when induced by IPTG The maximum activity was observed as 140 U/g cell, when induced with 1 mM IPTG at $15^{\circ}C$.

Prediction of primary lining loads for soft ground tunnels based on case studies (토사터널의 일차 라이닝에 작용하는 하중 예측 사례 연구)

  • Prediction of lining loads is one of the key issues to be addressed in the design of a tunnel. The validity of the existing design methods is reviewed by comparing the loads calculated using the methods with the field measurements obtained from several tunnels in Edmonton, carada. However, the existing methods are determined not to be fully satisfactory for the prediction of primary lining loads. To account for the stress reduction occurring prior to lining installation, the stress reduction factor is used coupled with an analytical solution for calculation of lining loads. Typical values of dimensionless load factors nD/H for tunnels in Edmonton are obtained from parametric analyses and presented in a table. The loads calculated using the proposed method are compared with field measurements collected from tunnels in Edmonton to verify the method. The method can be used for other tunnels if the tunnels are built in stiff or dense soils, where good ground control is accomplished during the tunnel construction.

Improvement of RTL system for location monitoring of multi-laborers in the underground space (지하공간에서 노무자의 위치 인식을 위한 RTL 시스템 개선)

  • Real time location (RTL) system which can control the safety of laborers in the underground space is developed in this study. This innovative system can monitor the location and history of movement of multiple laborers in real time. To accommodate constrains of the underground space, the system is portable and composed with low-battery mode. Since conventional method to detect the location of laborer with radio signal strength indicator (RSSI) contains high errors, new measurement system using accelerometers and correction method are suggested in this study. Field test is performed to validate the developed system. The error for moving laborer was 0.4 m and for non-moving laborers was 0.2 m. Thus, it is found that the new RTL system can be used to monitor the location of laborers in the underground construction space.

  • Recent, the construction of the multi-purpose double-deck tunnel is required to solve the flood protection and congested area at urban city. The multi-purpose double-deck tunnel is desperately needed for the introduction of efficient utilization of underground space in addition to the main feature of road capabilities. A basic review was performed for site selection to consider the control capability and features of road tunnel at the same time, and the processable flow in accordance with tunnels cross section of double deck tunnel. Site Selection Criteria for multi-purpose use of the double-deck tunnel has been proposed through the site selection criteria by use of the tunnels review. Also the estimation processable flow was performed to review the versatility of double-deck tunnel due to design of tunnel cross-section. Site Selection of double-deck tunnel from this study can be seen the need for a complex consideration through a variety of analyzes.

  • Several attempts to construct multi-purpose tunnel for both road and waterway have been made. The multi-purpose tunnel is mainly used as a road tunnel, however it is transferred to waterway to control flood during rainy season. The planning of the multi-purpose tunnel is recognized as cost-effective way of infrastructure construction. In case of the multi-purpose tunnel constructed beneath groundwater table, seasonal fluctuation of groundwater table and repeated flow in the tunnel may cause long-term deterioration of the tunnel system. In this study, the behavior of multi-purpose tunnel in view of groundwater table or flow in the tunnel is investigated using model test and numerical modeling method. The results have shown that rising of groundwater table caused buoyant force to the tunnel and the fluctuation of rainwater in the tunnel generated loosening of surrounding ground. It is recommended to evaluate the effect of the long-term water pressure variation in the design of a multi-purpose tunnel.

Efficient and Precise Construction of Markerless Manipulations in the Bacillus subtilis Genome

  • We have developed an efficient and precise method for genome manipulations in Bacillus subtilis that allows rapid alteration of a gene sequence or multiple gene sequences without altering the chromosome in any other way. In our approach, the Escherichia coli toxin gene mazF, which was used as a counter-selectable marker, was placed under the control of a xylose-inducible expression system and associated with an antibiotic resistance gene to create a "mazF-cassette". A polymerase chain reaction (PCR)-generated fragment, consisting of two homology regions joined to the mazF-cassette, was integrated into the chromosome at the target locus by homologous recombination, using positive selection for antibiotic resistance. Then, the excision of the mazF-cassette from the chromosome by a single-crossover event between two short directly repeated (DR) sequences, included in the design of the PCR products, was achieved by counter-selection of mazF. We used this method efficiently and precisely to deliver a point mutation, to inactivate a specific gene, to delete a large genomic region, and to generate the in-frame deletion with minimal polar effects in the same background.

Study on a Scheme to Increase Flood Reduction Effect of Washlands (강변저류지군의 홍수저감효과 개선방안에 대한 연구)

  • Due to climate change and difficulty in construction of large dam, it is on the increase to use small hydraulic facilities such as washland for the flood control purpose. Since there can be many potential locations for washland in a basin, it is important for determination of optimal location to calculate flood reduction effect of washlands substantially and exactly. In this study, a new scheme to increase flood reduction effect of washlands is suggested. Suggested scheme uses sensitivity analysis of flood reduction effect depending on washland characteristics such as storage volume, spillway elevation and spillway width. The characteristics of the washland is determined by results of sensitivity analysis to maximize flood reduction effect at basin outlet and downstream of a washland. The methodology is applied to Anseong river basin to show its applicability and applied result is compared with those of another studies.

SS and COD Runoff from a Rice Field Watershed during Storm Events in the Growing and Non-growing Seasons (강우시 영농기와 비영농기의 광역논에서의 부유물질 (SS)과 COD의 유출특성)

  • The objective of this research was to investigate runoff characteristics of suspended solid (SS) and chemical oxygen demand (COD) from a paddy field watershed during storm events in the growing and non-growing seasons. Average of event mean concentration (EMC) of pollutants were 56.9 mg/L for SS and 23.9 mg/L for COD in the non-growing season and 50.3 mg/L for SS and 11.9 mg/L for COD in the growing season. The average EMC of SS in the study area was much lower than that in the uplands irrespective of cultivation, suggesting that paddy fields control soil erosion. This may be because flooding and wet soil in the growing season, and rice straw residue and stubble on the topsoil in the non-growing season reduce soil erosion. The changing tillage practice from fall tillage to spring tillage avoids soil erosion due to shortening of the tilled fallow period. However, the average EMC of COD in the non-growing season was about twice as much that in the growing season likely due to the runoff of organics like rice straw residues. The relationship between SS and COD loads and stormwater runoff volume was expressed by power function. The exponent for SS was higher than that for COD, suggesting that SS load increased with stormflow runoff more than COD load did. The mean SS and COD loads per storm during the non-growing season were much lower than those in the growing season, and therefore non-point source pollution in the growing season should be managed well.

Development of Self-propelled Explosive Subsoiler (2) - Construction of Prototype and Performance Evaluation - (자주식 심토환경 개선기 개발(2) - 본체 제작 및 성능 평가 -)

  • This study was carried out to develop a self-propelled type explosive subsoiler for improving the root zone soil conditions in orchard and other forest fields. Prototype was designed to be able to inject air and other soil improving material such as lime into soil at the same time, and thus improve the air permeability and drainage of orchard soils to promote the root growth of tree for high quality fruit production. Soil penetration device of explosive subsoiler is composed of air hammer, penetration rob and air injection nozzle. To support the soil penetration device of explosive subsoiler to penetrate vertically, modified Scott-Russel mechanism was used. Timing control device for simultaneous injection of soil improving material with air was attached to the out side wall of air cylinder and as the cylinder move, the soil improving material was injected into soil at the same time. Turning radius of prototype was 2.2-2.3 m with good mobility in sloped land. It took approximately 1 minute for lime injection system to reach the optimum pressure of 9.9 kg/$cm^2$, average 10-20 seconds were required to rupture soil with the depth of 50 cm and 2-3 seconds were required for explosion, so all in all about 1 minute and 20 seconds were required for one cycle of explosion. Maximum soil rupture depth and diameter were 50 cm and 3-4 m respectively depending on the soil type and soil moisture content. For final design of explosive subsoiler inclination angle of lime hopper was increased from 60 degree to 70 degree and the shape of hopper was changed from rectangular cone to circular cone to solve the clogging problem of lime at out let. Agitating system operated by compressed air was attached to the metering device of the prototype, thus more than 90 cc of lime was discharged per cycle from metering device without clogging problems.

Construction of Improved Tetracycline-Inducible Expression System for the Effective Regulation of Transgene Expression (외래 유전자의 효율적인 발현 조절을 위한 개선된 Tetracycline-Inducible Expression System의 구축)

  • In this study we tried to construct a more efficient tetracycline-inducible gene expression system by replacing previous key elements with more advance ones. At the beginning, we substituted PGK (phophoglycerate kinase) promoter for CMV (cytomegalovirus) promoter to control "$rtTA2^sM2$" which has been known for high induction efficiency in response to tetracycline. With this modification, expression of the EGFP marker gene under the induction condition was significantly increased. Next, we replaced "TRE" fragment with a modified version named "TRE-tighf" which has been reported to have higher affinity and specificity to the transactivator by minor base change of the "TRE" DNA fragment sequence. Use of "TRE-tighf" instead of "TRE" resulted in more than 10 fold increment in terms of induction efficiency and significant decrement of background expression in non-inducible condition. By combining PGK promoter and "TRE-tight" fragment, we could upgrade previous tetracycline-inducible system to show more stringent turn on/off gene switch ability and stronger expression of the gene of our interest. Use of this newly developed system must be very helpful to the studies of gene expression, especially to the transgenic animal study in which non-controllable constitutive expression of the transgene has been one of the urgent problems to be solved.
