We have developed an efficient system of assessing the ability of a gene silencing cassette to silence transcripts from recalcitrant or poorly studied plant species by using a model plant as a host for the gene of interest. Tobacco plants transgenic for Lachrymatory Factor Synthase (LFS) enzyme activity from onion were first produced by introducing a CaMV 35S-onion-lfs gene construct. These plants were then subjected to a second transformation with an RNAi construct directed against the lfs gene sequence. LFS enzyme activity assay showed that the transgenic plants, containing both the lfs gene and the RNAi construct, had significantly reduced LFS activity. This observation was supported by Western analysis for the LFS protein and further validated by quantitative RT-PCR analysis that demonstrated a significant reduction in the lfs transcript level in the dual transformants. In this work, we have demonstrated that the RNAi construct is a suitable candidate for the development of a non-lachrymatory onion. Our model plant RNAi system has wide-reaching applications for assessment and targeting of plant secondary pathway genes, from poorly studied or recalcitrant plant species, that are important in the pharmacological, food and process industries.

The aim of this paper is to make embodiment and structure of the mission concept, named mission construct, in the National R&D Programs as an effective tool which manages the life-cycle processes and examines their validity and duplication problems. While it is generally accepted the premise that the mission is the fundamental reason for being in planning and implementing a public program, its role is being overlooked because of its vagueness and abstractness. To derive the mission construct of a program level, existing literature on corporate/organization missions and their components is examined. The study identifies and proposes the following four dimensions and their attributes: raison d'etre, program definition, program identification and program goals.

In order to gain insight into the mechanism of the regulation of cytochrome P450IAl by arylhydrocarbon, the 5'-flanking region of a trout CYP450IAl 5'flanking DNA was cloned into pCAT-basic vector and it was transfected into Hepa-1 cells. 3MC treatment to hepa Ⅰ cells transfected with fish CYP450IAl-CAT construct results in mRNA increased by 2.81 fold when it was compared with that of control This increase of mRNA was decreased by concomitantly treated flavonoids such as morin. The levels of CAT mRNA that was treated with morin was 29.2-58.0% of 3MC stimulated CAT mRNA. Further investigation to find out if there are DRE, XRE or negative regulatory cis element in CYP450IA1 gene was undertaken. Results of the deletion study of 5'flanking DNA of trout P450IA indicate the existance of the negative(-1600 ～ -1300). CAT mRNA was about two-fold higher in deleted trout CYP450IAl-CAT construct transfected cells compared to the wi Id type trout CYP450IAl-CAT construct transfected cells. This paper presents a method of constructing the universal multiple processing element unit(UMPEU) by De Bruijn Graph. The second method is as following. First, we propose transformation operators in order to construct the De Bruijn UMPEU using properties of graph. Second, we construct the transformation table of De Bruijn graph using above transformation operators. Finally we construct the De Bruijn graph using transformation table. The proposed UMPEU be able to construct the De Bruijn graph for any prime number and integer value of finite fields. Also the UMPEU is applied to fault-tolerant computing system, pipeline class. parallel processing network, switching function and its circuits.

The purpose of this study was to examine the psychometric structure of Sasang typology with four personality measures and examine the construct validity of Sasang Personality Questionnaire (SPQ). We measured biopsychological traits of 104 university students using SPQ, Temperament and Character Inventory (TCI), NEO-Personality Inventory (NEO-PI) and Myers-Briggs Type Indicator (MBTI). The construct validity of SPQ was analyzed with TCI, NEO-PI and MBTI using Pearson's correlation. The temperament features of each Sasang groups were compared with Analysis of Variance and Profile Analysis. The SPQ showed significant correlation with TCI Novelty-Seeking (r=.398), TCI Harm-Avoidance (r=-.333), TCI Reward-Dependence (r=.465), TCI Self-Directedness (r=0.340), NEO-PI Extraversion (r=0.489) and MBTI Extraversion-Introversion (r=-0.509). The SPQ, TCI and NEO-PI revealed significant differences between Sasang constitutional groups. We examined the temperament construct of the Sasang typology and the validity of SPQ with well established western biopsychosocial instruments. This study would contribute to the objective studies on the biopsychosocial mechanism of Sasang medicine.

In the self-directed learning, learner can construct learning course. But it is very difficult for learner to construct learning course with understanding the various learning contents's characteristics. This research proposed the method to support to learner the information of learning contents type to fit the learner as calculate the learner's learning preference when learner construct the learning course. The calculating method of learning preference used preference vector value of topic map. To apply this method, we tested 20 learning sampling group and presented that this method help to learner to construct learning course as getting the high average degree of learning satisfaction.

This study tried to find out how to overcome multicollinearity problems in the structural equation model by creating a hierarchical construct model about the repurchase intention of social commerce. This study selected, as independent variables, price, quality, service, and social influence, based on literature review about social commerce, and then, as detailed variables of independent variables, selected system quality, information quality, transaction safety, order fulfillment and after-sales service, communication, subjective norms, and reputation. As results of empirical analysis about hierarchical construct model, all the independent variables were accepted having a significant impact on repurchase intention of social commerce. Next, this study analyzed the competition model that eight independent variables of price, system quality, information quality, transaction safety, order fulfillment and after-sales service, communication, subjective norm, and reputation directly influence the repurchase intention of social commerce. As results of empirical analysis, system quality, information quality, transaction safety, communication appeared to be insignificant. This study showed that hierarchical construct model is useful to overcome the multicollinearity problem in structural equational model and to increase explanatory power.

Double-stranded RNA (dsRNA) induces gene silencing in a sequence-specific manner by a process known as RNA interference (RNAi). The RNA-induced silencing complex (RISC) is a multi-subunit ribonucleoprotein complex that plays a key role in RNAi. VIG (Vasa intronic gene) has been identified as a component of Drosophila RISC; however, the role VIG plays in regulating RNAi is poorly understood. Here, we examined the spatial and temporal expression patterns of VIG-1, the C. elegans ortholog of Drosophila VIG, using a vig-1::gfp fusion construct. This construct contains the 908-bp region immediately upstream of vig-1 gene translation initiation site. Analysis by confocal microscopy demonstrated GFP-VIG-1 expression in a number of tissues including the pharynx, body wall muscle, hypodermis, intestine, reproductive system, and nervous system at the larval and adult stages. Furthermore, western blot analysis showed that VIG-1 is present in each developmental stage examined. To investigate regulatory sequences for vig-1 gene expression, we generated constructs containing deletions in the upstream region. It was determined that the GFP expression pattern of a deletion construct (${\Delta}-908$ to -597) was generally similar to that of the non-deletion construct. In contrast, removal of a larger segment (${\Delta}-908$ to -191) resulted in the loss of GFP expression in most cell types. Collectively, these results indicate that the 406-bp upstream region (-596 to -191) contains essential regulatory sequences required for VIG-1 expression.

This study investigated elementary students' (grade 4~6) learning progressions for water cycling drawn from iterative assessments using ordered multiple-choice (OMC) items. An assessment system, which consisted of construct map, item design, outcome space, and measurement model, was employed in this study to examine children's learning progressions. At the first stage of the assessment system, a construct map was designed on which children's conceptual understandings from naive to most sophisticated were represented. At the item design stage, 8 OMC items were drawn from the construct map. Each item option of the OMC items was scored from 0 to 3 according to its level of understanding at the stage of outcome space. As a measurement model, Rasch model, a branch of item response theory, was applied to interpreting the outcomes of the OMC items. This cycle of assessment system was furtherly implemented iteratively in order to elaborate on the first version of water cycling learning progression. In conclusion, children's understanding of water cycling could be described in two aspects: water distribution and water movement. We identified children's conjectural developmental pathways about water cycling existed from superficial and naive accounts to more complex and abstract accounts.

Objectives : To investigate reliability and validity of Korean translation of Western Ontario and McMaster Universities(WOMAC) osteoarthritis index. Methods : The reliablity, construct validity of the Korean WOMAC Index in the patient of knee osteoarthritis was investigated. Test-retest reliability was quantified with pearson's correlation coefficient and intraclass correlation coefficient. Internal consistency was quantified with Cronbach's ${\alpha}$. and construct validity with pearson's correlation coefficient by correlating of the Visual Analog Scale(VAS). Results : Test-retest reliability of Korean WOMAC Index for pain was 0.76 to 0.95, stiffness was 0.89 to 0.94, and physical function was 0.71 to 0.95. Intraclass correlation coefficient for pain was 0.76 to 0.94, stiffness was 0.54 to 0.89, and physical function was 0.70 to 0.95. Internal consistency were 0.94 and 0.94 for the first and second time, respectively. Construct validity for pain was 0.79, for stiffness was 0.66, and physical function was 0.67. Conclusions : The Korean translation of Western Ontario and McMaster Universities(WOMAC) osteoarthritis index is reliable, valid assessment tool in knee osteoarthritis.
