최근 급속도로 당뇨병 환자가 증가하면서 인슐린 시장이 크게 성장하고 있다. 또한 최근 식물체를 이용하여 의약용 단백질 생산이 경제적인 측면과 안정성 측면에서 매우 효과적임이 보고되고 있어 이를 이용한 분자농업이 주목을 받고 있다. 본 연구에서는 인슐린 단백질을 식물체에서 생산하기 위한 유전자 발현 construct를 설계하기 위한 실험으로서 식물발현용 preprominiinsulin construct를 제조하기 위한 단계적 실험을 수행하였다. 우선 proinsulin이 무세포 식물 전사/번역시스템에서 성공적으로 발현됨을 확인하였다. Prominiinsulin construct를 제조하여 대장균에서 발현시키는데 성공하였으며, 이를 트립신으로 절단한 후 인간 항인슐린 항체를 이용한 western 분석을 통하여 효과적으로 A-펩타이드와 B-펩타이드가 형성되며 이후 적절하게 접힘이 일어나고 hexamer로 조립됨을 확인하였다. 이후 식물체에서 재조합 인슐린 유전자가 발현되는지를 확인하기 위하여 RFP 결합 construct를 제조하여 담배의 현탁배양세포인 BY-2 세포에 형질전환시켜 RFP 결합 preprominiinsulin이 성공적으로 발현됨을 확인 하였다. 이러한 성공적인 연구 결과를 토대로 향후 이 construct는 RFP 단백질을 제거하여 35S 프로모터에 직접 유도되는 [N 말단 ${\rightarrow}$ tobacco E2 시그널 펩타이드 ${\rightarrow}$ B-펩타이드(1-29 AA) ${\rightarrow}$ AAK ${\rightarrow}$ A-펩타이드(1-21 AA) ${\rightarrow}$ RR ${\rightarrow}$ His6 ${\rightarrow}$ KDEL ${\rightarrow}$ C 말단] construct를 제조하여 담배 식물체에 형질전환시켜 분자농업을 통해 인간 인슐린 단백질을 생산하는데 활용하고자 한다.
D. melanogaster를 대상으로 두 종류의 P element construct, 즉 Pc[(ry+)B]와 p[(ry+)$\Delta$SX9]을 사용한 형질전환 실험을 통하여 자율적 및 비자율적인 P element construct간의 vector로써의 효율성을 분석했다. 자율적인 P element 내에 rosy 유전자를 포함하고 있는 Pc[(ry+)B] construct를 진정 M 계통형인 ry506 돌연변이 계통에 주입시켰다. 또한 비자율적인 P element 내에 rosy 유전자를 포함하고 있는 p[(ry+)$\Delta$SX9] construct와 전이효소의 공급원으로써 p-$\Delta$2-3hs$\pi$ helper plasmid를 역시 같은 계통의 ry506 돌연변이 계통에 주입시켰다. Dechorination 방법에 의해 총 1143 embryo를 대상으로 microinjection 한 결과, 모두 35 계통의 형질전환된 정상형의 눈을 가진 계통을 얻었다. P element construct를 주입시킨 전체 1143 embryo 중에서 약 20% 정도가 성체로 부화되었으며, 부화된 개체 가운데 약 40% 정도가 눈 색깔이 거의 정상형과 같은 G0 transformant 로 나타났다. 그러나 생식력을 가진 G0 transformant의 약 40% 만이 G1 transformant로 나타남으로써 모두 35 계통의 transformant를 얻었다. 따라서 G0 단계에서 나타난 형질전환 현상에는 반드시 rosy transformant의 염색체 삽입에 의해서 만이 아니라 세포질내에서도 construct내의 rosy 유전자의 발현이 가능한 것으로 분석되었다. 또한 형질전환율에 있어서는 Pc[(ry+)B]와p[(ry+)$\Delta$SX9] construct 간에 유의한 차가 없는 것으로 나타났다. 이상의 결과로 볼 때, 실험 목적에 따라 자율적인 P element 또는 helper DNA를 이용한 비자율적인 P element를 효과적인 vector 로 선택 이용함으로써 특정 유전자를 곤충집단내로 침투 및 고정시킬 수 있다고 판단된다.
The purpose of this study is to devise the concept of properties of the light on learning progressions for 3-6 grade students in the elementary school. For the purpose, this study creates a construct map of the properties of the light. Ordered multiple-choice items were developed in view of the construct map and presented to 200, 3~6 grade students in the elementary school. A partial credit model of Rasch model was used to analyze the results of those items, and the learning progressions was devised from the analysis results. The study can be summarized as follows: the construct modeling approach was used to devise the properties of light on learning progressions. As a result, the concept was selected for the core standards of construct map. Based on the construct map, the multiple-choice items were developed for students' conceptual understanding. The items were analyzed using the partial credit model, and the analysis findings showed that they were appropriate to assess students about their level of understanding of the properties of light. Finally, learning progressions were devised with the use of item analysis results.
This study aims to develop the constructs for evaluating team presentations performed by undergraduate international students in Korea. To achieve the goal, this study reviews related research and sets the criteria to analyze the problems of team presentations performed in Korean by undergraduate international students. Through this analysis we got to know which problems they were making. They were failing to reorganize the resources they had referenced and to organize the contents into one. And errors or mistakes of grammar and spelling were found often in their PPT files, and the editing styles of the PPTs were not consistent. Moreover, their performances were not unlike their PPT files. One made a good presentation but the other students just recited the script without interaction with the audience. Moreover, they could not operate the Q&A part and lead the discussion well, and components for wrapping up the presentation also were omitted frequently. After the analysis, this study conducted a survey with 23 Korean language educators asking about the validity of the construct and description of the survey. Based on the collected feedback, the construct list has been modified primarily, and carried out a second survey with an expert group. Based on their opinion, the final construct list was fine-tuned.
The purpose of this paper is to examine what is meant by a ralid measure of health. Guyatt, Kirshner and Jaeschke propose that health tests should be designed so as to have one of several kinds of validity: 'longitudinal construct validity' for those which are used for longitudinal research designs, and 'cross-sectional construct validity' for those which are used for cross-sectional designs. Williams and Naylor argue that this approach to test classification and validation confuses what a test purports to measure with the purpose for which it is used, and that some tests have multiple uses. A review of the meanings of validity in the psychologica test literature shows that both sets of authors use the term validity in an idiosyncratic way. It is central to use a scale to measure a person's level of a construct in mathematics education research. This article explains a practical process through which a researcher rapidly can develop an instrument to measure the construct. The process includes research questioning, reviewing the literature, framing a background theory, treating the data, and reviewing the instrument. The statistical treatment of data includes normality analysis, item-total correlation analysis, reliability analysis, and factor analysis. The purpose of this research is to construct real-time image acquisition system for a line-scan inspection system as a computer vision system. To construct the line-scan inspection system, many problem arise. However, problems of image acquisitions using linear CCD are practically studied in this paper. These problems are concerned to proper lighting systems, relations with handling system, camera systems, and impacts due to the noise of environment. These problems are examined separately, and proper configurations for each case are suggested to construct the line-scan image acquisition system.
Despite the noteworthy increase in the number of FL anxiety studies, inconsistencies associated with the effects of FL anxiety on language learner performance have been reported in literature. Such conflicting results seem to be attributable in part to unstable conceptualization of the FL anxiety construct and its measure. This paper purported to address the emerging call for a theoretical clarification of the construct at hand as a preface to a clear picture of language anxiety on a conceptual ground. This paper not only covers aspects of general anxiety from psychological perspectives, but examines how FL anxiety and its associated concepts have been conceptualized in the literature. Inconsistent results that pertain to FL learning were also delineated. Given the drawbacks found in the exiting theories of FL anxiety, several points were taken into account for a refinement of the conceptual framework. This attempt will hopefully shed new light on the construct per se and prove conducive to the development of the field of English education.
