Critical Design Status of the G-CLEF Flexure Control Camera

  • The GMT-Consortium Large Earth Finder (G-CLEF) is the very first light instrument of the Giant Magellan Telescope (GMT) to be commissioned in 2022. The instrument is a fiber feed, optical band echelle spectrograph that is capable of extremely precise radial velocity measurement. Korea Astronomy and Space Science Institute (KASI) has been involved in the development of the G-CLEF as one member of the international consortium consisted of five astronomical institutes including Smithsonian Astrophysical Observatory (SAO), Observatories of the Carnegie Institution of Washington (OCIW). It is scheduled to have KASI side Critical Design Review in December 2017. In this presentation we will report the recent progress on the critical design activities for the G-CLEF Flexure Control Camera (FCC).

  • Phenotypic analysis of gene-specific knockout (KO) mice has revolutionized our understanding of in vivo gene functions. As the use of mouse embryonic stem (ES) cells is inevitable for conventional gene targeting, the generation of knockout mice remains a very time-consuming and expensive process. To accelerate the large-scale production and phenotype analyses of KO mice, international efforts have organized global consortia such as the International Knockout Mouse Consortium (IKMC) and International Mouse Phenotype Consortium (IMPC), and they are persistently expanding the KO mouse catalogue that is publicly available for the researches studying specific genes of interests in vivo. However, new technologies, adopting zinc-finger nucleases (ZFNs) or Transcription Activator-Like Effector (TALE) Nucleases (TALENs) to edit the mouse genome, are now emerging as valuable and effective shortcuts alternative for the conventional gene targeting using ES cells. Here, we introduce the recent achievement of IKMC, and evaluate the significance of ZFN/TALEN technology in mouse genetics.

A new paradigm for cancer therapeutics development

  • The number of cancer patients has increased due to longer life spans and treatment has become a universal problem. Since molecular-targeted therapies were introduced as a new developmental strategy, certain targets have been examined hundreds of times, with developers overlapping their research efforts. We need to focus our energy and resources on novel drug candidate identification and optimization, in order to enhance the entry of early-stage drug candidates into the therapeutics pipeline. This presents a major opportunity for Korea to jump the decades-old development gap between our programs and those that are more advanced in other countries. Although this country does not have a specific center for validation and development of cancer therapeutics, we do have cutting-edge scientists performing research in many institutions. In this paper, I will review cancer drug development in Korea and suggest future directions, while urging colleagues to utilize their networking expertise so we can move toward a new paradigm of novel therapeutics development. An example of such efforts has begun with the Drug Development Consortium, which was described in the KSBMB chapter. This consortium was launched in 2010 by biochemists, chemists, cell and molecular biologists and pharmacologists. It is clear that effective cancer therapeutics will be developed more efficiently when we all strive for the same goal.

효율적인 GML 문서 저장을 위한 저장 스키마의 설계 및 성능평가 (Design and Performance Analysis of Storage Schema for Efficient Storing of GML Documents)

  • GML은 OGC(OpenGIS Consortium)에서 공간지리정보의 저장 및 전송을 위한 인코딩 표준으로 제안한 마크업 언어이다. 일반적인 공간 네트워크 데이터베이스에서 GML 지원을 위한 연구는 GML 문서의 파싱, GML 문서의 저장, 그리고 GML 문서의 질의어로 분류된다. GML 문서 저장에 관한 연구는 효율적인 GML 문서 검색을 위해 필수적인 연구이다. 그러나 GML 문서의 저장 스키마에 관한 연구는 거의 전무한 형편이다. 또한 기존 XML 문서 저장 스키마는 공간지리정보 저장에 적합하지 않다. 따라서 본 논문에서는 공간지리정보를 포함한 GML 문서를 효율적으로 저장하기 위한 저장 스키마를 제안한다. 아울러 제안하는 저장 스키마의 성능평가를 실시한다.

해외 주요 도서관 컨소시엄의 운영현황 비교·분석 - 도서관 컨소시엄 전문가와의 면담을 중심으로 - (Library Consortia Characteristics Comparative Analysis around the World)

  • 본 연구에서는 해외의 주요 도서관 컨소시엄의 운영현황과 특성에 대하여 선진국의 도서관 컨소시엄 전문가를 직접 면담하고 자료를 수집하고 비교 분석하였다. 조사결과, 해외 도서관 컨소시엄들은 구성 목적, 조직, 재원, 기능 등에 있어서 각각의 독특한 특성을 가지고 있었으며, 컨소시엄별 전문가들의 인식에도 차이가 있는 것으로 나타났다. 전문가들은 공통적으로 도서관 컨소시엄의 가치를 정보가격 협상력으로 인식하고 있었으며 정보가격 인상의 대안으로 OA(Open Access)를 지목하였다. 본 연구에서는 해외 도서관 컨소시엄 운영에 관한 이해를 돕고자 하였다.

β-Secretase (BACE1) Purification by Refolding Method and Complex with Hispidin

  • Alzheimer's disease (AD) is a devastating neurodegenerative disease that represents the most common form of dementia among the elderly population. The deposition of aggregated ${\beta}$-amyloid ($A{\beta}$) senile plaques in the human brain is a classic observation in the neuropathology of AD, yet an understanding of the mechanism of their formation remains elusive. $A{\beta}$ is formed through endoproteolysis of the amyloid precursor protein (APP) by ${\beta}$-secretase (BACE1, ${\beta}$-site APP-cleaving enzyme) and ${\gamma}$-secretase. In this study, BACE1 protein was successfully over-expressed, purified, and refolded and utilized in a binding study with hispidin. We developed a simpler refolding method using a urea gradient and size-exclusion gel filtration to purify an active BACE1 protein variant, in larger quantities than that reported previously, and measured the binding affinity of hispidin to the BACE1 protein variant through isothermal titration calorimetry.

Dissemination of Advanced Mouse Resources and Technologies at RIKEN BioResource Center

  • RIKEN BioResource Center (BRC) has collected, preserved, conducted quality control of, and distributed mouse resources since 2002 as the core facility of the National BioResource Project by the Ministry of Education, Culture, Sports, Science and Technology (MEXT), Japan. Our mouse resources include over 5,000 strains such as humanized disease models, fluorescent reporters, and knockout mice. We have developed novel mouse strains such as tissue-specific Cre-drivers and optogenetic strains that are in high demand by the research community. We have removed all our specified pathogens from the deposited mice and used our quality control tests to examine their genetic modifications and backgrounds. RIKEN BRC is a founding member of the Federation of International Mouse Resources and the Asian Mouse Mutagenesis and Resource Association, and provides mouse resources to the one-stop International Mouse Strain Resource database. RIKEN BRC also participates in the International Gene Trap Consortium, having registered 713 gene-trap clones and their sequences in a public library, and is an advisory member of the CREATE (Coordination of resources for conditional expression of mutated mouse alleles) consortium which represents major European and international mouse database holders for the integration and dissemination of Cre-driver strains. RIKEN BRC provides training courses in the use of advanced technologies for the quality control and cryopreservation of mouse strains to promote the effective use of mouse resources worldwide.

  • 일반적으로 비용이 많이 들고 복잡한 처리과정을 거치는 디지털 항공사진측량은 독립형 워크스테이션 시스템을 기반으로 사용되어 왔다. 본 연구에서는 Open Geospatial Consortium에서 제안한 국제 GIS 표준인 Web Processing Service을 통해 항공사진측량 처리 과정에 대한 새로운 접근 방안을 제시하고자 한다. 실험을 위해 항공사진측량 업무를 기본단위로 정의하는 WPS 프로파일링 과정을 거쳐 여러 개의 제네릭 프로세스로 정의하였고, 정의된 프로세스는 WPS 표준에 따라 각각 서버 S/W와 클라이언트 S/W 모듈을 구현하였다. 본 연구를 통해 WPS의 기본 단위 항공사진측량 프로세스를 다수의 사용자가 웹을 사용하여 공유, 재활용 측면의 접근을 기대할 수 있었다.

GML 문서에서 연관규칙 생성 시스템 구현 (Implementation of Association Rules Creation System from GML Documents)

  • 지리 정보에 대한 관심이 증가되면서 이러한 연구와 활용 분야도 다양해지고 있다. OGC(Open GIS Consortium)에서는 XML(Extensible Markup Language)을 GIS 분야에 도입한 GML(Geography Markup Language)을 개발하였으며 여러 활용 분야에서 GML을 사용하고 계속적으로 연구되고 있다. 본 논문에서는 기존의 XML 문서를 기반으로 연구되었던 데이터 마이닝 방법 중 하나인 연관규칙 기법(Apriori)을 GML 문서들에 사용하여 의미 있는 규칙을 찾아내려 한다. 규칙을 찾는 방법에는 2가지가 있을 수 있다. 하나는 GML 문서에서 내용만을 뽑아내어 그에 따른 규칙을 찾아내는 방법이고, 다른 하나는 GML 문서에서 사용된 태그와 속성을 기반으로 규칙을 찾아내는 방법이다. 본 논문에서는 2가지 방법을 통해 규칙을 찾는 것에 대하여 기술하고 2가지 방법을 적용한 시스템을 보일 것이다.

Analysis of differences in human leukocyte antigen between the two Wellcome Trust Case Control Consortium control datasets

  • The Wellcome Trust Case Control Consortium (WTCCC) study was a large genome-wide association study that aimed to identify common variants associated with seven diseases. That study combined two control datasets (58C and UK Blood Services) as shared controls. Prior to using the combined controls, the WTCCC performed analyses to show that the genomic content of the control datasets was not significantly different. Recently, the analysis of human leukocyte antigen (HLA) genes has become prevalent due to the development of HLA imputation technology. In this project, we extended the between-control homogeneity analysis of the WTCCC to HLA. We imputed HLA information in the WTCCC control dataset and showed that the HLA content was not significantly different between the two control datasets, suggesting that the combined controls can be used as controls for HLA fine-mapping analysis based on HLA imputation.
