Preparation and Characteristics of Calcium Lactate from Black Snail

  • Calcium lactate (CL) prepared from powdered black snail (PBS) or its ashed powder (ABS), was investigated for ideal manufacturing conditions to optimize color, solubility and sensory quality. Based on the amount of PBS and 100 mL lactic acid (LA), the yields of PBS-CL were 300% and 15 g in 10% LA and 260% and 20 g in 20% LA. Yields of ABS-CL based on the amount ABS and 100 mL LA were 400% and 60 g in 10% LA and 329% and 66 g in 20% LA. Both of the yields were decreased with an increase of the LA concentration on the basis of PBS and ABS amounts, but proportionally increased with the increment in the LA concentration on the basis of LA volume. Optimal preparation times of the dehydrated PBS-CL and ABS-CL were, respectively, 4 hr and 5 hr at 10$0^{\circ}C$, 3 hr and 4 hr at 12$0^{\circ}C$, and 1 hr and 2 hr at 15$0^{\circ}C$, which showed shorter time in preparing the dehydrated ABS-CL. PBS-CL and ABS-CL were confirmed to be Ca($CH_3$CHOH$CO_2$)$_2$ by the analysis results of IR and $^1$H-NMR. Calcium contents of the anhydrous PBS-CL and ABS-CL were individually 15.4% (w/w) and 17.3% (w/w) representing 84.2% and 94.5% or each theoretical value. Colors or PBS-CL and ABS-CL were light yellow and light-greenish white each. Solubilities of PBS- CL and ABS-CL in distilled water at pH 3~8 were 5.43 and 6.11 g/100 mL, respectively, which demonstrated higher mean solubilities rather than the 4.74 g/100 mL of standard CL. Solubilities of PBS-CL (3.14~5.03 g/100 mL) and ABS-CL (4.69~6.05 g/100 mL) against soup soy sauce, 3% brine, Soju (Korean distilled liquor), thick soy sauce, grape juice and orange juice were higher than those of standard CL (2.94~5.84 g/100 mL). ABS-CL was believed to have a wide use range due to its low sourness while different applications of PBS-CL in food are expected due to its mild astringent taste and strong savory taste despite its strong bitter taste as estimated by sensory evaluation.

천연 색소 추출물을 첨가한 수박 음료의 제조 (Manufacturing of Watermelon Beverage Added with Natural Color Extracts)

  • 음료 제조를 위한 수박 원료 희석 농도, 당, 구연산, 비타민 C 첨가농도, 살균 방법, 천연 색소 추출 방법, 풍미증진 재료의 첨가량에 따른 색과 맛에 대하여 조사하였다. 주스 제조를 위한 조건을 실험한 결과 수박의 맛과 색이 남아있는 수박즙의 최적 희석 농도는 40%이었고, 음료로서 적정 당도는 $12^{\circ}Brix$였으며, 산미와 항산화 효과를 위한 구연산과 비타민 C첨가 농도는 0.5+0.3 g/L 또는 1.0+0.3 g/L이 적절하였다. 가열살균과 시간을 비교한 결과 수박 음료를 $70^{\circ}C$이상의 온도에서 살균 시 수박 고유의 적색도가 감소되었으며, 관능 검사 결과 $60^{\circ}C$ 10-30분 또는 $70^{\circ}C$에서 10 분간 살균 시 양호하였다 또 $60^{\circ}C$이상으로 살균 시 $30^{\circ}C$에서 한 달 동안 저장 후 처리시간에 관계없이 부패된 것은 발견되지 않았다. 천연 색소 추출 방법에 있어서는 열수 추출에 비해 알콜 추출은 전반적으로 효과가 낮았으며, 첨가 재료에 있어서도 알콜 추출보다는 열수 추출에 유리한 재료인 복분자와 오미자가 효과적이었고 복분자 20 g/L와 오미자 30g/L를 열수추출한 것을 7:3으로 배합하여 음료를 제조했을 때 가장 기호도가 높았다. 추후 다른 재료를 탐색 할 때도 수용성 가용분이 많은 재료가 적합하다고 생각된다. 본 실험에서는 주스 제조 시 침전의 문제를 해결하기 위해 희석과 예열 처리로 응고되는 침전물을 미리 여과하는 방법을 사용했으나 이 보다 더 좋은 개선 방법을 찾기 위한 연구가 앞으로 더 필요하리라 생각된다.

녹엽단백질 급여가 흰쥐 혈청 콜레스테롤 및 유리 아미노산 농도에 미치는 영향 (Leaf Protein Affect the Concentration on Serum Cholesterol and Free Amino Acids in Rats)

  • 이탈리안 라이그라스로부터 갈변을 방지시켜 white LP를 제조하여 4주령의 Wistar 계 수컷 흰쥐에 14일 급여, 혈청지질농도 및 혈중 유리 아미노산 패턴에 대하여 단백질원으로 대두단백질 및 카제인 급여와 비교하였으며 혈중 유리 아미노산이 혈청 지질농도에 대하여 어떠한 관련이 있는지를 검토하였다. 갈변을 방지하여 조제한 LP 의 질소 함량 및 소화율이 거의 대두단백질 수준까지 높여 단백질의 품질에 대한 영향을 확인하였다. 혈청 총 콜레스테롤 및 LDL-콜레스테롤은 카제인군에 비하여 이탈리안 라이그라스 white LP 군이 유의적으로 낮고, 대두 단백질군과 같은 수준이었다. 이때 혈중유리아미노산을 비교한 결과, LP 군과 대두단백질군과는 거의 닮은 패턴을 나타내고 카제인군에 대해서는 glycine 만 높고 다른 아미노산은 차가 없든지 유의적으로 낮았다. 그리고, 카제인 수준을 15.0% 로 낮추어 대두 단백질과 비교하였다. 그 결과, 혈청 콜레스테롤 농도는 카제인군이 유의적으로 높고 혈중 glycine 농도가 낮고, methionine 농도의 높은 폐턴은 일치하였다. 이상의 결과로부터 혈중 유리아미노산 패턴은 급여 단백질의 종류에 의해 어느정도 규제되어지며 아미노산 패턴은 혈청지질과 어떠한 관련이 있는 것으로 인정되었다. 더욱이, LP 와 대두단백질의 혈중 유리 아미노산 패턴은 매우 유사한 것이 관찰되어 LP 의 혈청 콜레스테롤 강하작용의 메카니즘은 대두 단백질의 메카니즘과 유사한 것으로 생각되어진다.

Gallic acid가 Lipopolysaccharide로 활성화된 마우스 대식세포의 인터루킨 생성에 미치는 영향 (Inhibitory Effect of Gallic acid on Production of Interleukins in Mouse Macrophage Stimulated by Lipopolysaccharide)

  • Objectives: Gallic acid (GA) is the major component of tannin which could be easily founded in various natural materials such as green tea, red tea, grape juice, and Corni Fructus. The purpose of this study is to investigate the effect of Gallic acid (GA) on production of interleukin (IL) in mouse macrophage Raw 264.7 cells stimulated by lipopolysaccharide (LPS). Methods: Productions of interleukins were measured by High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on $xMAP^{(R)}$ (multi-analyte profiling beads) technology. Firstly, cell culture supernatant was obtained after treatment with LPS and GA for 24 hour. Then, it was incubated with the antibody-conjugated beads for 30 minutes. And detection antibody was added and incubated for 30 minutes. And Strepavidin-conjugated Phycoerythrin (SAPE) was added. After incubation for 30 minutes, the level of SAPE fluorescence was analyzed on Bio-plex Suspension Array System and concentration of interleukin was determined. Results: The results of the experiment are as follows. 1. GA significantly inhibited the production of IL-3, IL-10, IL-12p40, and IL-17 in LPS-induced mouse macrophage RAW 264.7 cells at the concentration of 25, 50, 100, 200 uM (p<0.05). 2. GA significantly inhibited the production of IL-6 in LPS-induced mouse macrophage RAW 264.7 cells at the concentration of 50, 100, 200 uM (p<0.05). 3. GA diminished the production of some cytokine such as IL-4, IL-5, and IL-13 in LPS-induced mouse macrophage RAW 264.7 cells. 4. GA did not show the inhibitory effect on the production of IL-$1{\alpha}$ and IL-9 in LPS-induced mouse macrophage RAW 264.7 cells. Conclusions: These results suggest that GA has anti-inflammatory activity related with its inhibitory effects on the production of interleukins such as IL-3, IL-10, IL-12p40, IL-17, and IL-6 in LPS-induced macrophages.

Pharmacoat Coating in an Aqueous System : The Dissolution Behavior and Reduction in Coating Time

  • It is sometimes said lately that the pH of the human gastric juice is significantly different among individuals. Thus, the dissolution behavior of coated solid dosage forms should preferably be independent of the pH of the test solution. With these points as a background, the effect of pH on the dissolution velocity of coated tablets was studied to compare that of Pharmacoat with other gastric soluble film coating materials. Three viscosity types of Pharmacoat have been available(3, 6 and 15cP) until now. the 6cP type has been considered to be the most suitable for a tablet coating amongst the three types. The 3 cP type with a low degree of polymerization, is capable of providing high concentration, but the film strength is so inferior that sometimes cracking of the film may occur. On the other hand, in the case of the 15cP type, high polymer concentration cannot be achieved because of the high dgree of polymerization, and thus it is uneconomical for coating. Now, there is a strong demand to reduce the coating time even when HPMC is used in the 6cP type in order to reduce the coating cost. In order to improve this problem, we have concentrated our attention on reducing the viscosity value of HPMC to an allowable lower limit from 6cP. As a result of this study, it was found that the reduction of the viscosity value to around 4.5cP enabled the use of a higher solution concentration and an incidental shorter coating time without giving any substantial adverse effects on the properties of coated preparations. These experiment results are presented in the later part of this presentation. Based on this study, we have added the viscosity type of 4.5cP as one of the Pharmacoat products as Pharmacoat-645.

저온-저염 절임기술이 절임배추의 품질 특성에 미치는 영향 (Effects of the Low Temperature and Low Salt Solution on the Quality Characteristics of Salted Chinese Cabbage)

  • The principal objective of this study was to evaluate the influence of temperature and salt concentration on the physicochemical properties of salted Chinese cabbage. Salted Chinese cabbage samples were prepared with various concentrations of salt(4, 5, 6 and 7%), and were stored for 10 days at three temperatures(8, 14, and $25^{\circ}C$). The salting ratio of Chinese cabbage evidenced a decreasing trend regardless of the salting temperature and salt concentration, and their decrement appeared relatively high as the salting temperature increased. The period required to achieve a critical salting ratio(85%) decreased with increases in the salt concentration at $25^{\circ}C$, and a similar trend was observed at lower temperatures(8 and $14^{\circ}C$). The salinity of all samples evidenced an increasing trend during the salting period, and at $25^{\circ}C$, in particular, a continuous increment was observed. At salt concentrations of 4%, the critical salinity(2.2%) was not achieved regardless of the temperature and salting period. The pH of salted Chinese cabbage achieved critical pH in 3 days at a salting temperature of $25^{\circ}C$, but the critical pH 5.5 of samples at 8 and $14^{\circ}C$ appeared after a long period of approximately 4 to 10 days. The average hardness values of salted Chinese cabbage at a salting ratio of 85% were approximately 1.49 MPa, 1.87 MPa, and 1.97 MPa, respectively, at three temperatures($25^{\circ}C$, $14^{\circ}C$, and $8^{\circ}C$). The initial reducing sugar content of cabbage juice was 11.8 mg/$m{\ell}$, and this value decreased substantially to 3 to 5 mg/$m{\ell}$ on day 1.

수성이상계에서 K.fragilis의 에탄올 추출발효 특성에 관한 연구 (Extractive Ethanol Fermentation Characteristics of K.fragilis in an Aqueous Two Phase System)

  • 여러 가지 조성의 PEG 20000jDx로 구성한 수성 이상계를 사용한 에탄올 추출발효실험을 통하여 K. fragilis에 의한 돼지감자 주스의 발효 특성을 초기 당농도 및 생성 알코올 농도의 함수로 규명하여 다음의 결과를 얻었다. 균체의 비증식속도 빛 에탄올 비생성속도는 PEG 및 Dx 농도가 낮을수록 증가하였다. 비증식속도는 조성에 무관하게 초기 당농도 $80g/\ell$에서 최대치를 보였고, 에탄올 비생성속도는 $120g/\ell$최대치를 보였다. 생성 알코올 농도에 의한 저해 영향은 비증식속도에 대하여 지수함수형 을, 에탄올 비생성속도에 대하여 쌍곡선함수형을 나타냈다. Dx 농도가 2.5~5% 범위에셔 PEG 농도 가 낮을수록 에탄올 저해 영향은 감소하였다. 총괄 에탄올 생산성은 초기 당농도가 증가할수록 높았으 며 생산성이 최대치에 도랄하는 소요시간도 당농도가 증가함에 따라 증가하였다. 시간별 에탄올 생산 속도는 초기 당농도 $160g/\ell$ 에서 가장 높은 값을 보 였다. 총괄 에탄올 생산성은 대조실험에 비하여 수 성이상계 추출발효를 통하여 1.2배 이상 증가하였고 초기 당농도가 높을수록 이상계 추출발효가 더욱 유리한 것으로 나타났다.

Comparative antidiabetic activity of different fractions of methanolic extract of Zingiber officinale Roscoe in streptozotocin induced NIDDM rats

  • Earlier we have reported the antidiabetic activity of fresh juice of rhizomes of Zingiber officinale (Z. officinale) and its correlation with 5-HT receptor antagonism. Since 6-gingerol the marker compound of Z. officinale is reported to posses 5-HT anatgonistic activity, the present investigation, was undertaken to find out the concentration of 6-gingerol present in methanolic extract of Z. officinale and its different fractions (petroleum ether, toluene and chloroform). We also evaluated these fractions for antidiabetic activity in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in non insulin dependent diabetes mellitus (NIDDM) rats were found to be significantly (P < 0.05) higher than control rats and these were significantly decreased by treatment with methanolic extract of Z. officinale and its fractions. The results of oral glucose tolerance test (OGTT) showed that methanolic extract and its fractions significantly (P < 0.05) decreased both STZ-induced increase in $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM groups. Treatment with petroleum ether fraction produced a greater reduction in elevated glucose and $AUC_{glucose}$ levels as compared to treatment with other fractions. Treatment with methanolic extract of Z. officinale and its fractions also produced significant reduction in the elevated lipid, serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) levels in NIDDM rats. The effect of petroleum ether fraction on elevated lipid, SGOT and SGPT levels was significantly greater as compared to treatment with other fractions. The concentration of 6-gingerol was found to be maximum in petroleum ether fraction (11.430%) and minimum in chloroform fraction (0.973%). The methanolic extract and toluene fraction was found to contain 3.080% and 2.191 %, 6-gingerol respectively. In conclusion, our data suggest that methonolic extract and its fractions possess significant antidiabetic activity in NIDDM rats. The extent of activity appears to be dependent on the concentration of 6-gingerol present in the extract or its fractions.

미선나무 잎 추출물을 이용한 음료 개발 및 이화학적 특성 (Beverage Development using Abeliophyllum distichum Leaf Extract and Its Physicochemical Properties)

  • A beverage was developed using the Abeliophyllum distichum leaf (AL). The beverage was prepared by adding it to apple juice by concentration, and physicochemical quality, antioxidant activities, and sensory evaluation were measured. Soluble solid and reducing sugar content of the control were 12.57 °Brix and 11.40%, respectively, and there was no difference from the group with addition of the AL extract. However, pH was slightly increased upon addition of AL extract. Lightness and yellowness increased when AL extract was added. Verbascoside content was not detected in the control, but it increased as the concentration of AL extract increased. The contents of ascorbic acid and flavonoids were 5.38 and 20.42 mg%, respectively, and there was no significant difference between the groups. However, the content of polyphenols increased as the concentration of the AL extract increased. DPPH radical and metal ion scavenging activity were increased by addition of the AL extract, but there was no difference in the ABTS radical scavenging activity. As a result of the sensory evaluation, there was no difference from the control even wihen the AL extract was added; thus, it was considered that there was no problem with the degree of acceptability when added within about 300 ppm.

염건조기의 인공소화시 Nitrite, Thiocyanate 및 Ascorbic acid가 N-nitrosamine의 생성에 미치는 영향 (Effect of Nitrite, Thiocyanate and Ascorbic acid on N-nitrosamine Formation in Salted and Dried Yellow Corvenia under Simulated Gastric Digestion)

  • 우리나라 고유의 염건품인 굴비를 섭취하였을 때 체내에서 생성되는 NA를 예측하기 위해서 인공타액과 위액을 혼합하여 37。C에서 2시간동안 배양한 굴비의 인공소화물로부터 NA를 분석하였고, 동시에 nitrite, thiocyanate 및 ascorbic acid가 공존할 경우 NA생성에 미치는 영향을 실험하였다. 굴비시료에 인공타액과 위액을 가하여 소화시킨 인공소화물에서는 N-nitrosodimethylamine(NDMA)이 전혀 검출되지 않았으나, nitrite농도를 증가시킬수록 이에 비례하여 NDMA의 생성은 증가하였다. 굴비시료에 thiocyanate를 농도별(0∼6.4 mM)로 첨가하여 인공소화 시킨 결과 nitrite첨가량에 비례하여 NDMA 생성이 증가되었으나, 고농도의 nitrite(100 mM)를 함유한 시료에 thiocyanate를 8mM이상 첨가한 경우에는 뚜렷한 촉매효과를 나타내지 않았다. 반면에 10g의 굴비시료에 ascorbic acid를 농도별로 첨가한 인공 소화물에서 NDMA생성억제를 실험한 결과 nitrite농도가 0.5 및 1 mM인 경우에는 NDMA의 생성을 억제시키지 못하였으나 nitrite와 ascorbic acid의 농도가 높아짐에 따라 비례적으로 NDMA 생성억제도 뛰어났다.
