Contamination of Green Vegetable Juice by E. coli O157:H7 during Storage (E. coli O157:H7에 의한 녹즙 저장 환경에서의 미생물학적 오염도 조사)

  • The market for green vegetable juice (GVJ) is growing owing to the increasing demand for healthy food; however data on the safety and quality of GVJ are poorly reported. The objective of this study was to investigate the change in microbial community in GVJ during storage and its contamination by E. coli O157:H7. The microbial community was analyzed via culturable and non-culturable methods at 5, 10, and $25^{\circ}C$ for different storage times. In the non-culturable method, denaturing gradient gel electrophoresis (DGGE) was used. The initial bacterial concentration was $2.92{\times}10^5CFU/mL$, which exceeded the limit prescribed by the Korean Food Hygiene law. The results of the DGGE analysis indicated that the microbial community during storage was diverse and the spoilage lactic acid bacteria were prevalent at a later stage. Other bacteria such as Rahnella, Citrobacter, Pseudomonas, and Cyanobacteria were identified. Thus, the results strongly emphasize the need to pay attention to GVJ production safety, especially with respect to temperature control, in order to prevent the growth of foodborne pathogens such as E. coli O157:H7 and other spoilage bacteria.

  • Tuna cooking juice concentrate (TCJC) is by-produced during the canning processing of skipjack tuna Katsuwonus pelamis and it is well known that TCJC contains various nutritional components. Therefore, the immuno-stimulating activity of TCJC was investigated using macrophage RAW 264.7 cell line and the spleen cell isolated from BALB/c mice. The TCJC increased the production of IL-6, TNF-${\alpha}$, and IL-$1{\beta}$ in a dose-dependent manner compared to the control in RAW 264.7 cells without any toxicity. In particular, the production of TNF-${\alpha}$ was increased over 300-fold. The production of both Th1 cytokine (such as IFN-${\gamma}$, TNF-${\alpha}$, IL-2, and IL-12) and Th2 cytokine (IL-4, IL-6, IL-10) was also increased by TCJC treatment in splenocytes. Moreover, the TCJC increased the splenocyte proliferation in a concentration-dependent manner compared to control. These results indicate that TCJC may enhance immune function by promoting various cytokine production.

A STUDY ON THE ENAMEL EROSION CAUSED BY ORANGE JUICES (오렌지쥬스의 치아 법랑질 침식효과에 대한 연구)

  • Acidic drink has been known as a cause of tooth erosion. The purpose of this study is to verify the acid levels of orange juices in market and evaluate the erosion effects on the enamel of deciduous teeth by orange juices in relatively short period of time. With four kinds of orange juice selected from market, pH, buffering capacity and concentration of Ca and P ion were measured. And forty segments of normal enamel of deciduous incisors were divided into four groups and level of erosion was measured by surface hardness test before and after soaking at 50ml of orange juice for 10 minutes. The results of this study showed that the average pH of orange juices was 4.0 and this pH value was so acidic as to cause the tooth erosion. There were differences in small quantity for each group of teeth, however, the result was statistically so significant that orange juices can cause enamel erosion in relatively short period of time.

Inhibition Effects of Pulp on Seed Germination of American Ginseng (과육이 서양삼 종자 발아에 미치는 영향)

  • The germination inhibitory effects of American ginseng (Panax quinquefolium Linne) pulp were discussed. The germination inhibitory effects of pulp juice were decreased in a concentration dependent manner. When the pulp juice was diluted 0 (original juice), 1, 2, 5, 10, 20, 50 and 100 times, the radicle lengths of the assay plant, Chinese cabbage (Brassica chinensis Linne), showed 0, 0.32, 0.72, 3.13, 4.83, 16.07, 16.73 and 23.50 mm, respectively (CK=25.98 mm). The pulp evidently inhibited the embryo growth in natural fruit. The longer was the duration that the pulp stayed around the seed, the longer was the time course needed for embryo getting free from the inhibitory effects of pulp. When the depulping was performed on the day 0, 15, 30 and 60 after harvest, the time courses needed for embryo extricating the residual inhibitory effects from pulp were 30, 75, 135 and 135 days, respectively. Moreover, if the pulp stayed around the seed with time, that would make the seed rotten ratio increase. When the pulp stayed around the seed for 0, 15, 30, 60 and 270 days, the seed rotten ratios were 5.47, 5.71, 19.05, 27.14 and 33.33%, respectively. Therefore, we concluded that the pulp could be included in the inhibitory components which made American ginseng seed get into dormancy.

A Criteria on Nitrate Concentration in Soil Solution and Leaf Petiole Juice for Fertigation of Cucumber (Cucumis sativus L.) under Greenhouse Cultivation (시설 오이의 관비재배를 위한 토양용액과 엽병즙액중 질산태 농도 기준 설정)

  • To develope a technique for efficiently managing fertilizer for cucumber, a quick test method to quantify nitrate content in soil solution and leaf petiole juice using a simple instrument was investigated. Among the nitrate analyzing instruments such as compact ion meter, nitrate ion meter, and test strip with reflectometer, the paper test-strip used in conjunction with a hand-held reflectometer was most closely correlated with ion chromatography method in nitrate content, and then it would be suggested with a tool that a farmer can use rapidly, conveniently and accurately for nitrate analysis in a field. Nitrate content in soil solution collected by porous cup was very variable on the lapsed time after drip irrigation and the sampling positions such as soil depth and the distance from dripper. As a result, a significant correlation between nitrate contents of soil solutions and 2M KCl soil extract was not found. However, nitrate content in soil solution extracted with a volume basis (soil:water=1:2) showed the highly significant correlation with that in 2M KCl extract. Nitrate contents of cucumber leaf petiole juices was greatly different between upper and lower leaves. Eleven to sixteen positioned-leaf would be a proper sampling position to determine nitrate content in leaf petiole for evaluating nutrient state by plant tissue analysis. From the secondary regression equations between nitrate contents of soil and petiole juice and the yield of cucumber, nitrate levels for real time diagnosis were estimated as $400mg\;l^{-1}$ soil solution by porous cup. $300mg\;l^{-1}$ in a soil volume extraction, and $1400mg\;l^{-1}$ in petiole juice from spring to summer season. In addition, the maximum yield of cucumber fruit in pot test was obtained in nitrate $1500mg\;l^{-1}$ level of petiole juice, which was similar to nitrate $1400mg\;l^{-1}$ in greenhouse trial.

Dietary Copper Intakes and Nutritional Status of Copper in Serum among Elementary Schoolchildren in Chungnam Province in Korea: Comparison between Remote Rural and Urban Areas (일부 학령기 아동의 구리 섭취량 및 구리 영양 상태에 관한 연구: 충남 벽지농촌과 도시간의 비교)

  • This study is performed to compare the dietary intakes and food sources of copper (Cu) using the database of Cu content developed in this study between the elementary schoolchildren in remote rural areas (RA, n=58, $9.9{\pm}1.7$ yrs) and those in urban area (UA, n=60, $9.4{\pm}1.8$ yrs), and to analyze the relationship between serum Cu concentration and serum antioxidant status in the RA. The results obtained in this study were as followings: 1) Dietary intakes of calorie, calcium and iron in the RA were in the 3/5-4/5 of the Korean RDA while the UA were similar to or more than the Korean RDA, 7th ed. except iron. 2) More than 273 kinds of food consumed by the subjects were analyzed the content of Cu and database of Cu content were developed in the present study. The mean dietary intake of Cu per day in the RA was $0.99{\pm}0.07mg/d$ ($170.0{\pm}13.2%$ of the USA RDA) while it was $1.22{\pm}0.07mg/d$ ($203.4{\pm}13.1%$ of the RDA) in the UA. The percentage of dietary intakes of Cu less than 213 of the RDA was 8.6% in the RA in comparison to 0% in the UA. 3) The RA and the UA consumed more than 80% of total dietary intakes of Cu from plant foods. Thus, the RA and the UA consumed Cu from cooked rice, vegetables and fruits as a major source. However the RA had less Cu from meat and their products than did the UA (p<0.05) .4) Crab stew including crab and juice was the highest food source of Cu for the total subjects, followed by seasoned bud of aralia, cooked; beef rib meat, roasted; soybean paste soup w/mallow; and soybean paste soup w/mallow & beef. Major food source of Cu was similar for the RA and the UA such as cooked rice, vegetables and fruits. 5) Mean concentration of serum Cu in the RA was $18.1{\pm}0.7{\mu}M/L$ that was in the normal value, and all subjects in this group were in more than normal value. In the RA serum Cu concentration related positively with serum ceruloplasmin concentration, serum vitamin C concentration and EC SOD activity, respectively. However, serum Cu concentration did not relate with serum TBARS concentration in the RA. Above results showed that the RA had good status of Cu nutrition based upon dietary intake and serum concentration, however some of the RA had lower intake of Cu than the RDA. The overall children in the UA had good Cu nutrition. Therefore, the subgroup of the RA should be supported to improve their Cu nutrition, and this support could give them better antioxidant status based upon positive relationship between serum Cu concentration and serum antioxidant status in the RA.

Optimization of Peach Wine Fermentation by Response Surface Methodology (반응표면분석에 의한 복숭아주 발효 최적화)

  • To prepare peach wine using peach juice, fermentation properties were monitored, and fermentation conditions (initial sugar concentration, temperature, and time) were optimized by a response surface methodology. Alcohol content for peach wine fermentation ranged from 3.4~9.2% [$R^2=0.9229$ (P<0.01)] and 8.54% (maximum value) at $18.73^{\circ}Brix$, $16.81^{\circ}C$, and 12.99 day. Acidity ranged from 0.30~0.74%, and 0.25% (minimum value) at $15.11^{\circ}Brix$, $17.09^{\circ}C$, and 13.61 day. Residual sugar concentration was $6.67^{\circ}Brix$ (maximum residual sugar content) at $17.79^{\circ}Brix$, $20.63^{\circ}C$, and 3.37 day. Yellow color intensity was 18.92 (maximum Hunter's color b value) at $13.19^{\circ}Brix$, $20.81^{\circ}C$, and 12.81 day. Based on the above study results, optimization conditions for peach wine fermentation were 9 days, below $20^{\circ}C$, and $19^{\circ}Brix$ peach juice.

Study on the Method of Differentiating between Fresh and Frozen Chicken Meat by Using Mitochondrial Malate Dehydrogenase Activity (Mitochondrial Malate Dehydrogenase 활성을 이용한 냉장계육과 냉동계육의 판별법에 관한 연구)

  • This study was performed to develop the method of differentiation fresh and frozen meat by using the measurement of mitochondrial malate dehydrogenase. The principle of this experiment is based on the fact the enzyme proteins associated with mitochondria membrane could be released by freezing. The methods were studied by measurements of protein concentration of meat press juice, WHC (water-holding capacity), drip loss and mitochondrial malate dehydrogenase enzyme activity. Samples were stored at 4$^{\circ}C$ and -18$^{\circ}C$ during storage period, respectively. Protein concentration of meat press juice was ranged from 8.5 mg/mL to 12.7 mg/mL and increased by freezing below at -18$^{\circ}C$(p<0.05). The WHC was not significantly different between fresh meat and frozen chicken meat (p>0.05). The amount of drip loss of fresh and frozen chicken meat at 4$^{\circ}C$ and -18$^{\circ}C$ was not significantly different (p>0.05). Mitochondrial malate dehydrogenase activity of frozen meat (-18$^{\circ}C$) was significantly higher (p<0.05) than that of fresh meat. Also, enzyme activity of frozen meat was maintained at the same level after 3 minutes reaction. But fresh meat had not this reaction. From these results, it suggests that mitochondrial malate dehydrogenase can be used as a promising enzyme to differentiate between fresh and frozen meat.

Fermentation Properties and in vitro Anticancer Effect of Young Radish Kimchi and Young Radish Watery Kimchi (열무김치 및 열무물김치의 발효특성과 in vitro 항암효과)

  • Fermentation properties and in vitro anticancer effect of young radish (YR) kimchi and young radish watery (YRW) kimchi were investigated during fermentation at 5℃. The fermentation of YR kimchi during 2-3 weeks led to the decrease of pH down to pH 4.3, increased acidity, and the highest Leuconostoc sp. counts. YR kimchi showed the acidity of 1.04-1.27% at the pH 4.3, when the kimchi was ripened properly. The fermentation of YRW kimchi during 9 days led to the decrease of pH down to pH 4.3 and the acidity of 0.20%. Inhibitory effects of the juices of YR, YR kimchi, and YRW kimchi on the growth of AGS human gastric adenocarcinoma cells in MTT assay were increased with the added concentration. The juice of YR kimchi had a higher inhibitory effect on the growth of AGS human gastric adenocarcinoma cells than that of YRW kimchi at same concentration. The juice of YR kimchi showed similar inhibitory effects on the growth of AGS human gastric and HT-29 human colon adenocarcinoma cells in MTT assay to baechu kimchi, which the inhibition rates are more than 50%.

Studies on the Taste Sensitivity and Eating Habits of Koreans (한국인(韓國人)의 맛에 대한 감도(感度)와 식습관(食習慣)에 대한 연구(硏究))

  • In order to provide basic data for food processors and to improve eating habits and healthy diets, four primary tastes and hot taste threshold of Koreans, physio-chemical properties of foods and food preference were investigated. The results of this study were as follows; 1. The recognition threshold concentration of four primary tastes was 0.016 % of salt, 0.245${\sim}$0.249 % of sweet, 0.004${\sim}$0.008 % of sour and 0.008${\sim}$0.012 % of bitter. Threshold concentration of hot taste was 170,000${\sim}$600,000 Scoville Heat Unit(S.H.U.) 2. The most acceptable tastes were 0.3 % saltiness in 0.375 % broth(at $60^{\circ}C$), 6 % sweetness in 2 % instant coffee(at $60^{\circ}C$) and 19.3 sugar-acid ratio(16.8 brix/0.8736 % citric acid) in 100 % valencia orange juice(at $20^{\circ}C$). 3. The salt concentrations of soup were 1.127 % in average. S. H. U. (scale of hot taste) of soybean sprout soup and spinach bean paste soup ranged between 12,500 and 47,500 equivalent to oleoresin capsicum content of 1.990${\sim}$5.911 (mg %). 4. 'Jigae' (Korean style stew) was the most favorable food and it was revealed that the father influences the formation of his children's eating habits.
