Effect of Cell Density on Xylitol Fermentation by Candida parapsilosis (Candida parapsilosis에 의한 Xylitol 생산시 균체농도가 미치는 영향)

  • Effect of cell density on the xylitol production from xylose by Candida parapsilosis KFCC 10875 was investigated. The concentrated cells were obtained by centrifugation of culture broth. The xylitol production rate was maximum at the cell concentration of 20 g/l and the specific xylitol production rate decreased when the cell concentration was increased due to oxygen limitation. Effect of the initial concentration of xylose on the xylitol production was also examined using the concentrated cells of 20 g/l. The xylitol production rate, specific xylitol production rate, and xylitol yield from xylose were maximum at 170 g/l xylose. Above 170 g/l xylose, the xylitol production rate was remarkably decreased. The concentrated cells could also be obtained by adjusting the dissolved oxygen (DO) during fermentation. The rapid accumulation of cells up to 20 g/l was achieved by maintaining an increased level of DO during the exponential growth phase and then, for the efficient xylitol production, the DO was changed to a low level in the range of 0.7-1.5%. A fed-batch fermentation of xylose by adjusting the DO level was carried out in a fermentor and the final xylitol concentration of 140 g/l from xylose of 200 g/l could be obtained for 56 h fermentation.

  • The feasibility of replacing the tope cell of pn GaInP homojunction with our GaInP/AlGaInP heterojunction structure in III-V semiconductor multijunction photovoltaic (MJPV) cells having the highest current conversion efficiency was investigated. The performance of photovoltaic (PV) cells grown on $2^{\circ}$ and $10^{\circ}$ off-oriented GaAs substrates were compared to each other. The PV cells on the $10^{\circ}$ off-cut substrate showed higher short-circuit current density ($J_{sc}$) and conversion efficiency values than that of using the $2^{\circ}$ one. For $2{\times}2mm^2$ area PV cell on $10^{\circ}$ off substrate, the $J_{sc}$ of $9.21mA/cm^2$ and the open-circuit voltage of 1.38 V were measured under 1 sun illumination. For $5{\times}5mm^2$ cell on $10^{\circ}$ off substrate, the conversion efficiency was decreased from 6.03% (1 sun) to 5.28% (20 sun) due to a decrease in fiill factor (FF).

  • An optimal condition for the continuous production of ${\delta}-aminolevulinate$(ALA) was investigated using high concentrated resting cells of Rhodocyclus gelatinosus KUP-74. The increase of the amount of extracellular ALA versus the concentration of resting cells showed rectangular hyperbolic pattern until 20 mg cells/ml, but no further increase in the ALA amount by increasing its concentration was occurred. The highest yield of the extracellular ALA was observed after 3 hr of incubation of 1 ml reaction system containing 20 mg cells, 4 mM levulinate and 5 mM L-glutamate. On the other hand, the immobilized cells prepared by Ca-alginate inclusive method needed to incubate for 6 hr with 6 mM levulinate and 10 mM L-glutamate to give maximal yield of the extracellular ALA. In addition, under these conditions the resulted continuous productivities of the ALA by immobilized cells and highly concentrated resting cells were appeared 50 percent decreases after incubations for 185 hr and 100 hr, respectively, and the method of the cells to be immobilized was more efficient to recover the extracellular ALA produced.

Inhibitory Effects of Extracts of Houttuynia cordata Thumb (Saururaceae) on Cytokine Release of Human Monocytic THP-1 Cells and Human Eosinophilic EoL-1 Cells

  • In the present study, we investigated whether Houttuynia cordata Thumb (Saururaceae; HC) extracts have an anti-inflammatory effect in human monocytic THP-1 cells and human eosinophilic EoL-1 cells. The dried and powdered whole plants of HC were extracted with 80% EtOH. The combined extract (HC-1) was concentrated under reduced pressure. The residue was diluted with water, and then successively partitioned with n-hexane, EtOAc, and BuOH to produce the n-hexane (HC-2), EtOAc (HC-3), BuOH (HC-4), and the water-soluble fractions (HC-5), respectively. HC extracts have no cytotoxicity on THP-1 cells and EoL-1 cells at a high concentration of $10\;{\mu}g/ml$ for 24 h, except HC-2 extract ($10\;{\mu}g/ml$). Interleukin-6, Interleukin-8 and Monocyte chemoattractant protein-1 in THP-1 cells were increased after the treatment with the extract from house dust mite or LPS. The increase of cytokine production was strongly suppressed by HC-3 extract, in comparision with other extracts. HC-3 also had inhibitory effect on Interleukin-6 production increased by mite extract and LPS in EoL-1 cells. However, HC-3 extract increased Interleukin-8 production induced by mite extract and LPS in EoL-1 cells. These results suggest that HC extracts may be used as useful agents for treating allergic disorders such as asthma and atopic dermatitis.

High Cell Density Culture of Micro-algal Dunaliella bardawil (미세조류 Dunaliella bardawil의 고농도 세포배양)

  • High cell density cultivation of microalga Dunaliella bardawil using nitrogen fed-batch cultures was studied in batch flask. Optimum environmental conditions include concentrated nutrients except NaCl and carbon sources, carbon sources, pH, light, agitation, nitrate and phosphate ions. Cell growth, consumption rates of nitrate and phosphate ions were monitored. Optimal conditions for higher cell density were found to be(in the range tested): 5 times concentrated media(1 times-10 times concentrated media) pH 8.0 (7.0-9.0) white light(blue and red light) 15mM of nitrate (0.94-15mM) 250mM $NaHCO_3$ and $CO_2$ gas. However, the addition of phosphate ions did not enhance the algal maximum cell density and specific growth rate. Nitrate was found to be effective for the cell growth. The maximum cell density of fed-batch culture using nitrate ions in $8.955{\times}106$cells/ml after 189hr incubation.

  • The artificial increase in the solar intensity incident on solar cells using lenses or mirrors can allow solar cells to generate equivalent power with a lower cost. In application areas of Fresnel lenses as solar concentrators, several variations of design were devised and tested. Some PV systems still use commercially available flat Fresnel lenses as concentrators. In this study, we designed and optimized flat Fresnel lens and the 'light pipe' to develop 500X concentrated solar PV system. We performed rigorous ray tracing simulation of the flat Fresnel lens and light-pipe. The light-pipe can play imporatant roles of redistributing solar energy at the solar cell and increase the mechanical tolerance so that it can increase the lifetime of the high-concentration solar PV system and decrease the cost of manufacturing. To investigate the sensitivity of the solar power generated by the concentrated solar PV according to the performance of lens and light pipe, we performed raytracing and executed a simulation of electrical performance of the solar cell when it is exposed to the non-uniform illumination. We could conclude that we can generate 95 % or more energy compared with the energy that can be generated by perfectly uniform illumination once the total energy is given the same.

  • Some microorganisms, including actinomycetes, cyanobacteria, and other bacteria, algae, fungi, and yeast, can accumulate and retain relatively high quantities of heavy metals and radionuclides from their external environments (1-4). Both living and dead cells can be used for biosorptive metal/radionuclide removal from solution. Thus microorganisms and products excreted by or derived from microbial cells (2) may provide an alternative or adjunct to conventional techniuqes of metal removal and recovery. Recent approaches have separated the microbial growth and metal removal process to manipulate production of metal-adsorptive capacity of bacteria and metal removal process. If pre-grown cells are immobilized and used for metal removal, mathematical modeling can be applied to predict immobilized cell reactor behavior under specific process conditions. Waste and microbial adsorbent could be separated from the treated flow in one step. Once treated, the metal waste is concentrated in a small volume of sorbed form for easy metal disposal or recovery.

  • The incorporation of ³H-proline by epithelial and connective tissue elements of rat palatal mucosae was studied in order to investigate the relative levels of protein synthesis by the epithelium and underlying connective tissue cells. Following a sixty minutes incorporation of the radioactive tracer in vitro, it was found that the suprabasal cells had most grains per unit area. Furthermore, the grains were more concentrated over the cytoplasm than the nucleus. This was in contrast with the labeling of basal cells which had twice as many grains over the nucleoplasm than that over the cytoplasm. In intermediate cells; i.e., the spinous layer, the number of silver grains per unit area was decreased from that of the suprabasal cells. In areas where desmosomes were more prominent, many grains were in touch with such desmosomes. However, the labeling appeared to be reduced as soon as the cells became flattened. Moreover, the epidermal keratohyalin granules were relatively free of grains. Except for certain intercellular surfaces the keratinized cells were generally free of grains. On the connective tissue side, silver grains were primarily localized over the fibroblasts with occasional grains being found over palatal muscle cells, neural elements and so on. Most grains over collagenous fibers were found in relation to mature collagen fibrils. Thus, protein synthesis in isolated mucosae of the rat palate appeared to take place both in epithelial and connective elements. There were no apparent tissue alterations caused by the in vitro incorporation procedure utilized under conditions of this study.

  • Inhibition of the growth of human cancer cells by proteins secreted from 373-L1 cells was investigated in the present study. The growth of human cancer cells was inhibited by co-culture with 373-L1 cells under 10% FBS and DME, DME, GIT and serumless medium, respectively. The conditioned medium of cultured 373-L1 cells under serumless medium was concentrated 100-fold through an ultrafiltration cell with a 10,000 molecular weight cutoff at 4$^{\circ}C$ under positive pressure using nitrogen(373-L1 EM). 373-L1 EM inhibited the growth of HeLa, Hep G 2, KHOS-Np, A43l and MCF-7 cells. 3T3-L1 EM was purified with FPLC, DEAE-ion exchange chromatography and phenyl-sepharose chromatography. The major protein of 373-L1 EM has a molecular weight of 66,000-68,000 in SDS-PAGE analysis. The results suggest that the inhibitory activity of 373-L1 EM appears to be due to some protein(m.w.66,000-68,000) secreted by 373-L1 cells.

Physiological Function of Endothelial Cells Cultured on Polyurethsne Coated by ECM (인조혈관재료 표면에 도포된 혈관내피세포의 생리적 변화에 관한 연구)

  • Antithrombogenic surFace is one of the most important things to the artificial vascular prostheses. This problem will be solved if the surface of prosthesis is covered with endothelial cells. The attachment and the growth of endothelial cells onto vascular prosthesis are very difficult. So many studies have been concentrated on the attachement of endothelial cell. But no good performance of the in uiwo experiments has been shown until now. In this study, we used the whole extracellular matrix (ECM) excreted from fibroblasts as an underlying matrix, and the endothelial cells were seeded to obtain the long term patency of vascular graft(i.e., for the patent 8 week implanted wafts in the animal model of rat). In order to study the antithrombogenic functions of cultured endothelial cells, prostaglandin(PGF 1 a) synthesis and platelet adhesion were assayed. The concentration of PGF a of stimulated group was sisnificantly higher than that of control group(21.97 $\pm$ 3.45 vs 4.93 $\pm$0.71 pg/1000 cells). The platelet adhesion of the polyurethane sheet covered with endothelial cells was lower than that of polyurethane sheet or sheet covered with ECM(1.04$\pm$0.28, 2.87$\pm$0.77, 2.89$\pm$0.70, % radioactivities, respectively). Endothelial cells grew well on polyurethane coated with ECM, synthesized the prostacyclin and functioned well as antithrombogenic. Therefore the endothelialization onto the ECM excreted from fibroblasts may be a good method for the vfudig prosthesis.
