제주지역의 돼지 써코바이러스 2형 관련 장염에 대한 병리학적 특징 (Pathological characteristics on porcine enteritis associated with porcine circovirus type 2 in Jeju)

  • From 2006 to 2009, 50 pigs suspected of enteritis associated with porcine circovirus type 2 (PCV-2) (EAPC) were collected. Gross and histopathologic examinations and immunohistochemistry (IHC) were performed on the small intestine of 50 pigs. The pigs with EAPC were concentrated in grower pigs (68%), and diarrhea (84%) was the most common clinical sign. Grossly, the walls of the small intestine were thickened, and mesenteric lymph nodes were enlarged. The histopathologic features in the small intestine with EAPC were characterized by lymphoid depletion, histiocytic infiltration, and formation of basophilic grape-like inclusion bodies and multinucleated giant cells in gut-associated lymphatic tissue (GALT) and lamina propria. According to IHC, PCV-2 antigens were more intense and distributed widely in GALT (98%) than lamina propria (82%) of the small intestine. Co-infection with other enteric pathogens was detected in 26 pigs (52%). Twenty-four pigs (48%) were confirmed to be a PCV-2 single infection. Collectively, infected PCV-2 in the small intestine may induce an immunosuppressive status of individuals and then allow secondary co-infections in the digestive system. This study shows that PCV-2 can induce diarrhea and enteric lesions in pigs without a co-infection of other enteric pathogens.

박테리아 셀룰로오스의 생산 및 개질 (Production of Bacterial Cellulose and Its Modification)

  • The bacterial celluloses are very different in its physical, chemical and morphological structures compared to wood cellulose. These fibers have many unique properties that are potentially and commercially beneficial. This study was aimed to elucidate the production of bacterial celluloses and to improve their physical properties by chemical pretreatment. Bacterial celluloses produced by static culture had gel-like pellicle structure. The pellicle thickness was increased with the increasing time, and its layer was about 1.8cm after one-month incubation. The pellicles extruded from the cells of Acetobacter had a non-crystalline structure during initial growing stages, gradually getting crystaliyzed with the incubation time elapse, and eventually fumed to the cellulose I crystals. Young's modulus of bacterial cellulose sheet was increased with increasing NaOH concentration, and resulted in the highest at 5% NaOH concentration. Similar results with NaClO3 pretreatment can be observed. Too concentrated alkali solutions induced the destruction of cellulose fibrils and changed the mechanical properties of the sheets. These alkaline pretreatment have removed non-cellulosic components(NCC) from the bacterial cellulose, and enhanced inter-abrillar bonding by direct close contact among cellulosic fibrils.

Stability of Retroviral Vectors Against Ultracentrifugation Is Determined by the Viral Internal Core and Envelope Proteins Used for Pseudotyping

  • Retroviral and lentiviral vectors are mostly pseudotyped and often purified and concentrated via ultracentrifugation. In this study, we quantified and compared the stabilities of retroviral [murine leukemia virus (MLV)-based] and lentiviral [human immunodeficiency virus (HIV)-1-based] vectors pseudotyped with relatively mechanically stable envelope proteins, vesicular stomatitis virus glycoproteins (VSVGs), and the influenza virus WSN strain envelope proteins against ultracentrifugation. Lentiviral genomic and functional particles were more stable than the corresponding retroviral particles against ultracentrifugation when pseudotyped with VSVGs. However, both retroviral and lentiviral particles were unstable when pseudotyped with the influenza virus WSN strain envelope proteins. Therefore, the stabilities of pseudotyped retroviral and lentiviral vectors against ultracentrifugation process are a function of not only the type of envelope proteins, but also the type of viral internal core (MLV or HIV-1 core). In addition, the fraction of functional viral particles among genomic viral particles greatly varied at times during packaging, depending on the type of envelope proteins used for pseudotyping and the viral internal core.

The Effect of Milk Protein on the Biological and Rheological Properties of Probiotic Capsules

  • Probiotics are often infused into functional foods or encapsulated in a supplement form to maintain a healthy balance between the gut microbiota and their host. Because there are milk-based functional foods such as yogurt and cheese on the market, it has been suggested that milk-based probiotics could be incorporated into skim milk proteins in a liquid capsule. Skim milk is mainly composed of casein and whey protein, which create a strong natural barrier and can be used to encapsulate probiotics. In this study, we compared the encapsulated probiotics prepared with milk-based concentrated cell mixtures using commercial probiotics. Probiotic capsules were emulsified with skim milk proteins using vegetable oil to form a double coating layer. The product was heat-stable when tested using a rheometer. The survival rate of the milk-based probiotic cells in the lower gastric environment with bile was significantly higher than commercial probiotics. Thus, milk-encapsulated probiotics exhibited greater efficacy in the host than other types of probiotics, suggesting that the former could be more viable with a longer shelf life under harsh conditions than other form of probiotics. Our findings suggested that, compared with other types of probiotics, milk-based probiotics may be a better choice for producers and consumers.

1,2-dimethylhydrazine을 투여한 rat 장관의 증식세포 분포와 종양발생에 대한 면역조직화학적 연구 (Immunohistochemical study on distribution of proliferating cells and tumor formations in intestinal tracts of 1,2-dimethylhydrazine-treated rats)

  • The present investigation was focussed mainly on the development of the tumors and proliferating cells on the intestinal tracts of 1, 2-dimethyl-hydrazine(DMH)-treated young or adult rats. 26 rats(Wistar, 14 young rats weighting approximately 130~180gm and 12 adult rats weighting approximately 500~550gm) were given subcutaneously once weekly with 20mg of DMH/kg body weight(BW)/week for 8~22 weeks. Individual body weight were recorded weekly at the same day and time. The rats were killed at 8, 13, 15. 17, 19, 21 and 22 weeks. The intestinal tracts were opened longitudinally and carefully examined for tumors. The localization, number, and size of tumors were noted. Tumor-bearing areas were dissected out and fixed on neutral buffered 10% formalin and normal-looking mucosa from 8~22 weeks rats were also taken for fixation. Paraffin sections were stained by H-E for histopathological examination or with immunohistochemical stain for bromodeoxyuridine(Brdur) positive cells. 1. The growth proportion of body weight appeared to be decreased in the DMH-treated young rats than in control young rats and body weight of DMH-treated adult rats appeared to be 13.4% or less lower than weighted on 0 week. 2. Macroscopically, the developed tumors in the intestinal tracts were not observed as early as the 13 weeks after DMH treatment. The number of developed tumors per rat was found to be 14.3, 18.8, 22.3 in 15, 17 and 22 weeks. The numbers of tumors in intestinal regions per rat were 2.1, 4.3, 5.4, 2.5 in duodenum, jejunum, ilium and colon on 15 weeks, 2.3, 6.4, 7.8, 2.3, on 17 weeks, and 2.7, 9.3, 9.0, 1.3 on 22 weeks, respectively and the ileum and jejunum were higher in appearance rate of tumors and tumor types are dome shapes and diameter of largest tumor were 6.3mm. 3. Histopathologically, intestinal mucosa were thickened by the irregular distorted and distended crypts following hyperplasia. The tumors developed on the mucosa and submucosa and were recognized to be adenocarcinoma. 4. Immunohistochemically, the labeling index(LI) was calculated as the ratio of the number of Brdur-labeled cells to the total number of column cells of the crypts with longitudinal axis. LI of Brdur positive cells per crypt were 5.6%, 8.0% on small intestine of control and 22 week group, respectively and 3.7%, 12.7% on large intestine of control and 22 week group, respectively and were appeared to be increase in 22 week group than in control group and to be more number of proliferating cells in 22 week group than in control group. 5. LI of Brdur positive cells in 1, 2, 3, 4, 5 segments of crypt column were 11.7%, 10.7%, 3.8%, 0.6%, 0% in small intestine of control group and 23.5%, 11.8%, 2.3%, 2.4%, 0.8% in small intestine of 22 week group, and 5.4%, 7.4%, 3.8%, 1.0%, 0.4% in large intestine of control group and 29.5%, 20.3%, 5.9%, 6.3%, 1.3% in large intestine of 22 week group respectively. So results indicate that the number of proliferating cells by DMH treatment increase and were concentrated on the 1, 2 segments of crypt columns.

NIH3T3와 NIH3T3(ras) 세포에서 Dynamin II 발현 및 형태적 비교 (Dynamin II Expression and Morphological Comparison of NIH3T3 and NIH3T3 (ras) Cells)

  • Ras 신호전달체계는 세포내 다양한 결합 분자들과 더불어 세포의 분열과 세포의 이동에 관여한다. Dynamin 단백질은 endocytosis와 분비과정에서 vesicle를 분리하는데 관여하는 것으로 알려져 있으며, 3가지 아형으로 구분된다. Dynamin I은 신경조직에서 만 발현되고, dynamin II는 모든 조직에서 발현되지만 dynamin III는 정소를 포함한 생식기계에서만 발현된다. 선행된 연구에서 NIH3T3 세포를 이용하여 ras과발현 세포주를 만들었으며, dynamin II와 ras의 신호전달체계에 있는 Grb2가 결합한다는 것을 보고하였다. 따라서, 본 연구는 ras 단백질이 과발현되는 세포 (NIH3T3 (ras))와 대조세포인 NIH3T3의 형태학적인 차이점을 분석하고, 이 두 세포들에서 dynamin II 단백질의 발현의 차이를 비교하고자 하였다. Dynamin II의 발현차이를 분석하기 위해 형광염색을 하여 공초점 레이저현미경으로 세포내 분석을 하였으며, western blot을 시행하여 생화학적인 발현차이를 보았다. 또한, 두 세포의 미세구조적인 분석을 위하여 SEM과 TEM을 사용하였다. Dynamin II는 NIH3T3 (ras) 세포에서 발현이 증가 하였으며, NIH3T3 세포에 비하여 좀더 방추형이 었으며, 작은 세포질 돌기가 세포막을 따라 다수 신장되어있음이 관찰되었다. 또한, NIH3T3 (ras) 세포의 endocytotic vesicle이 형성되는 부위에서 dynamin II의 발현이 증가하였다. 이러한 결과로 dynamin II는 ras신호전달체계의한 신호전달분자로서 작용을 할 것으로 사료된다.

횡성호의 육수학적 조사와 인부하 (The Limnological Survey and Phosphorus Loading of Lake Hoengsung)

  • 횡성호에서는 수중폭기 기간에도 수온약층이 형성되었으며, 수중폭기로 인한 영향이 크지 않은 것으로 판단된다. 모든 정점의 표충(0-5m)평균 탁도는 0.1-8.5NTU로 우기 이후에 중층 및 심층에서 높았다. 투명도는 0.9-3.5m의 범위로 댐 하류(St. 1)정점에서 크게 나타났으며, 상류로 갈수록 낮았다. 용존산소는 2000년 7월과 8월에 전 지점의 0m에서 $8.4-14.1\;mgO_2\;{\cdot}\;L^{-1}$로 높았으나 심층에서는 고갈 상태를 보였다. 또한 2001년 7월과 9월에도 정점 1의 심층에서 $1\;mgO_2\;{\cdot}\;L^{-1}$ 내외로 매우 낮았다. 전 정점의 표층 (0-5 m)평균 총인 농도는 5-46 mgP ${\cdot}$ $m^{-3}$로 초봄의 turnover 직후 및 우기이후에 높았으며 용존 무기인 농도는 0.2-13.2 mgP ${\cdot}$ $m^{-3}$로 2001년 9월을 제외하고는 대부분10mgp ${\cdot}$ $m^3$이하 이였다. 횡성호의 연간 인부하량은 약 4.45ton 정도로 유역내에서 발생되는 총인 발생량 71.34ton중 약 6.2%만이 호수내로 유입되는 것으로 나타났다. 수면적당 인부하량은0.77 gp ${\cdot}\;m^2\;{\cdot}\;yr^{-1}$으로 횡성호의 환경용량이라 할 수 있는 과잉임계부하량 0.72 gp ${\cdot}\;m^2\;.\;yr^{-1}$을 0.05 gP ${\cdot}\;m^2\;.\;yr^{-1}$ 초과한 수준이다. 전 정점 표층 (0-5 m)평균 총질소 농도는 0.83-3.55 mgN ${\cdot}\;L^{-1}$로 우기 이후에 중충 이하 지역에서 높았다. 질산성 질소는 전 정점의 표층(0-5m)평균이 0.09-3.33 mgN ${\cdot}\;L^{-1}$로 2001년 6월에 가장 낮았다. 암모니아성 질소는 전 정점의 표층(0-5m)평균이 0.04-0.23 mgN ${\cdot}\;L^{-1}$이였다. 전 정점 표층 (0-5 m)평균 규소농도는 0.5-9.6 mgSi ${\cdot}\;L^{-1}$로 봄철에 낮았으며, 우기이후에 다시 증가하였다. 평균 TSI는 2000년과 2001년에 각각 59와 57이었다. 전 정점 표층 (0-5 m)평균 엽록소 a농도는 0.3-32.4 mg ${\cdot}\;m^{-3}$로 여름철에 높은 경향을 보였으며, 정점별로는 상류지역에서 높았다. 식물플랑크톤은 2000년 7월에 녹조류인 Scenedesmus sp.가 10,480 cells ${\cdot}\;mL^{-1}$,8월과 9월에는 남조류인 Microcystis sp.가 각각 3,492와 296 cells ${\cdot}\;mL^{-1}$로 우점하였다. 10월에는 녹조류인 Coelastrum microporum이 133 cells ${\cdot}\;mL^{-1}$, 11월에는 규조류인 Asterionella formosa가 2,654 cells ${\cdot}\;mL^{-1}$, 12월에는 규조류인 .Aulacoseira granulata가 29 cells ${\cdot}\;mL^{-1}$우점종이였다. 특히 2000년 여름철에 우점종이였던 남조류가 2001년에는 출현하지 않았으며 종 다양성 (diversity)지수는 2000년 7월에 2.22로 가장 높았으며, 생물량도 조사기간중 36,640 cells ${\cdot}\;mL^{-1}$로 가장 많았다.

암컷 마우스 생식기관 내 수은 화합물의 위치와 시간에 따른 축적된 수은 농도 변화 (Localization and Accumulated Concentration Changes of Mercury Compound in Reproductive Organs of Female Mice with Time)

  • 본 연구에서는 암컷 생식기관 내 축적된 수은 화합물의 위치와 시간에 따른 수은의 농도 변화를 확인하기 위하여 수행하였다. Methylmercuric chloride를 일주일에 한 번씩 총 3주간 사춘기 암컷 마우스에 피하 주사하였다. 시간에 따른 수은의 농도 변화를 확인하기 위해 투여 종료 후 10일, 150일, 300일째에 희생하였다. 투여 종료 후 10일에 희생한 경우 체중에 유의한 차이가 있었지만, 그 외 경우에는 체중과 난소의 무게에 있어서 대조군과 수은 투여군 사이에 유의한 차이가 없었다. 축적된 수은의 위치를 오토메탈로그라피 방법으로 확인하였고, 자궁, 난소, 난자에 축적된 수은 화합물의 위치를 광학현미경으로 분석하였다. 자궁의 경우, 투여 종료 후 10일째에 수은이 지질 세포와 자궁바깥막의 중피에 위치하였다. 150일째에는 수은 농도가 감소하였으며 300일째에는 나타나지 않았다. 투여 종료 후 10일째 난소의 경우, 수은이 피질 부위의 지질 세포와, 난포를 둘러싸는 난포막 세포, 황체에 분포하였다. 150일째에는 수은이 난소의 수질 부위에 축적되었으며, 300일째에는 분포하지 않았다. 투여 종료 후 10과 150일째 난자의 경우, 수은이 난자의 주변부에 주로 분포하였으며, 300일째에는 수은 농도가 감소되고 난자 전체에 고르게 분포하였다. 이런 결과는 암컷 마우스에서 수은에 의해 호르몬 생성, 착상, 그리고 발생 중인 배아가 영향을 받을 수 있다는 사실을 제시해 준다.

착생식물 기근의 형태 및 세포학적 특성 (Morphological and Cellular Characteristics of Aerial Roots in the Epiphytic American Ivy (Parthenocissus sp.))

  • 착생식물인 American Ivy의 기근이 흡착 표면에 부착할 때 착생에 따라 일어나는 형태 및 세포수준에서의 분화현상을 전자현미경적으로 연구하였다. 발달 중의 어린 기근은 향축면과 배축면이 구별되지 않는 곤봉상의 형태로 세포소기관들이 비교적 발달하며 색소체가 분포하는 유세포로 구성되어 있다. 이때 유세포 내 여러 액포에는 전자밀도가 높은 물질(EDS)의 축적이 두드러지게 나타나기 시작하였고, 이러한 현상은 기근 표피의 최외층 및 인접한 세포층에서 가장 신속하게 일어났다. 착생하는 기근의 세포는 훨씬 확장되었으나, EDS의 축적으로 인한 액포의 융합으로 세포질은 세포벽 주위로 밀려 극히 일부만 분포하였다. 착생과 거의 동시에 이들 세포층에는 급격한 변화가 일어나 세포질 내 미세구조들이 분해 소실되었고 액포 내 다량의 EDS도 극히 소량만 잔류하거나 거의 사라져 세포는 공동화되었다. 이와 함께, 세포벽 또한 변형되어 점차 불규칙적인 형태의 세포들로 변하였다. 착생이 더욱 진행되면 양면의 표피조직 및 세포들은 모두 불규칙적인 형태로 변화되고, 표면 부위는 일련의 분비물질들이 축적되었다. 흡착이 진행되어 대상 물체표면에 강력히 착생하면서 조직은 더욱 밀착되고 구성 세포들은 찌그러져 기근 조직의 대부분 부위에서 세포사멸이 일어났다. 이와 같이 기근 조직의 기능적 단계에 따라 EDS의 급격한 변화가 나타났는데, 이는 아마 기근이 흡착면에 부착할 때 EDS가 착생과 연관된 기능을 수행하는 것과 관련이 있을 것으로 추정되고 있다.

면역세포의 cytokine 유리에 미치는 substance P의 영향 (Effects of Substance P on the Release of Cytokines from Immune Cell Lines)

  • The neuropeptide substance P(SP) has been implicated in the mediation of inflammation and immune-mediated disease such as arthritis. Recently, it was reported that SP was markedly increased around the blood vessels in inflamed gingiva as well as in close association with the inflammatory cell infiltrate. These results support that SP may contribute to the pathophysiology of neuronal inflammation in human periodontal tissues. SP may regulate inflammatory/immune responses by stimulating the proliferation of human T cells, differentiation and antibody-secreting potential of B cells, macrophage respiratory burst, connective tissue proliferation, and the secretion of cytokines from monocytes and T cells. Here, I studied potential role of SP as a costimulatory chemical signal in inflammatory/immune responses, by determining the released proinflammatory cytokines such as $MIP-1{\alpha}$, $IL-1{\beta}$, and IL-6 from culture supernatants of homogeneous immune cell lines. Serum free cell supernatants were concentrated with TCA precipitation, fractionated with SDS-PAGE, and subjected into western blot analysis. Among 15 cell lines tested, macrophage/monocyte cell line RAW264.7 and WRl9m.1 showed the highest level of induction of $MIP-1{\alpha}$ when stimulated with LPS. Discrete IL-6 bands with multiple forms of molecular mass were detected from supernatants of B cell lines A20(32kDa), Daudi(32, 35kDa), and SKW6.4(29kDa), which were expressed constitutively. $IL-1{\beta}$ could not be detected by the method of western blot analysis from supernatants of all cell lines tested except RAW264.7, WRl9m.1, and erythroid cell line K562 which showed the least amount of $IL-{\beta}$ secretion. SP $10^{-9}M$ with suboptimal dose of LPS treatment showed synergistic induction of $MIP-1{\alpha}$ release from RAW264.7 or WR19m.1, and also IL-6 release from A20, but this synergism is not the case in costimulation of RAW264.7 or WRl9m.1 with SP $10^{-9}M$ and TPA. Although treatment of T cell line CTLL-R8 with SP $10^{-7}M$ or PHA+TPA induced modest level of $MIP-1{\alpha}$ secretion, synergism was not observed when they are applied together. These findings all together suggest the possibility of a regulatory role of SP in inflammatory/immune reaction through differential modulation of bioactivities of other chemical cosignals.
