This work aims to develop a dry lubricant for oilless bush, especially a solid lubricant, thereby creating a coating method with improved properties of anti-friction and load-carrying capacity without oil lubrication. In this work, spherical-shaped powders of thermosetting resin such as polyimide (PI) are mixed with a binder matrix obtained by mixing a fluorocarbon compound resin such as Polytetrafluoroethylene (PTFE) or Ethylene tetra fluoro ethylene (ETFE) with itself or with a non-fluorocarbon thermoplastic resin such as Polyether ether ketone (PEEK). And these dry lubricant mixtures are thickly coated (200-300 mm in the thickness) on the inner surface of the bush by using a wet-typed air-spray deposition method. It was found that the load-carrying capacity of the solid lubricant for excavator bush (60 mm in diameter) that operates under a high load condition (at 40 MPa) is greatly improved owing to the spherical-shaped powders of thermosetting resin. In addition, the coefficient of friction at the sliding surface is also reduced less than 0.1. Thick coating also lowers the contact stress at the edge of a bush that results in better tribological performances. The result suggests that the lubrication performance and durability life of the bush can be remarkably improved even without lubrication (oil or grease).
The effects of heat treatments (T6 and T73) on the microstructure, mechanical properties, and high cycle fatigue behavior of modified AA7075 alloys were investigated. A modified 7075 alloy was manufactured using modified-Mg (Mg-Al2Ca) instead of the conventional element Mg. Based on the microstructure, the average grain size was 4.5 ㎛ (T6) and 5.2 ㎛ (T73). Regardless of heat treatment, the modified AA7075 alloys consisted of Al matrix containing homogeneously distributed Al2CuMg and MgZn2 phases with reduced Fe-intermetallic compound. Room temperature tensile tests showed that the properties of modified 7075-T6 (Y.S.: 622MPa, T.S: 675MPa, elongation: 15.4%) were superior to those of T73 alloy (Y.S.: 492MPa, T.S: 548MPa, elongation: 12.8%). Experimental data show that the fatigue life of T6 was 400 MPa, about 64% of its yield strength. However, the fatigue life of T73 alloy was 330 MPa and 67%. Irrespective of the stress level, all crack initiation points were located on the specimen surface, and no inclusions acting as stress concentrators were seen. Superior mechanical properties and high cycle fatigue behavior of modified AA7075-T6 alloy are attributed to the fine grains and homogeneous distribution of small second phases such as MgZn2 and Al2CuMg, in addition to reduced Fe-intermetallic compounds.

We aimed to develop and validate a sensitive analytical method of nannozinone A, active metabolite of Nannochelins A extracted from the Myxobacterium Nannocytis pusilla, in mouse plasma using a liquid chromatography-tandem mass spectrometry (LC-MS/MS). Mouse plasma samples containing nannozinone A and 13C-caffeine (internal standard) were extracted using a liquid-liquid extraction (LLE) method with methyl tert-butyl ether. Standard calibration curves were linear in the concentration range of 1 - 1000 ng/mL (r2 > 0.998) with the inter- and intra-day accuracy and precision results less than 15%. LLE method gave results in the high and reproducible extraction recovery in the range of 78.00-81.08% with limited matrix effect in the range of 70.56-96.49%. The pharmacokinetics of nannozinone A after intravenous injection (5 mg/kg) and oral administration (30 mg/kg) of nannozinone A were investigated using the validated LC-MS/MS analysis of nannozinone A. The absolute oral bioavailability of nannozinone A was 8.82%. Plasma concentration of nannozinone A after the intravenous injection sharply decreased for 4 h but plasma concentration of orally administered nannozinone A showed fast distribution and slow elimination for 24 h. In conclusion, we successfully applied this newly developed sensitive LC-MS/MS analytical method of nannozinone A to the pharmacokinetic evaluation of this compound. This method can be useful for further studies on the pharmacokinetic optimization and evaluating the druggability of nannozinone A including its efficacy and toxicity.

The objective of this study was to establish a multi-residue quantitative method for the analysis of anthelmintic and antiprotozoal drugs in various livestock products (beef, pork, and chicken) using ultra-high-performance liquid chromatography-tandem mass spectrometry. Each compound performed validation at three different levels i.e., 0.5, 1, and 2× the maximum residue limit according to the CODEX guidelines (CAC/GL 71-2009). This study was conducted according to the modified quick, easy, cheap, effective, rugged, and safe procedure. The matrix-matched calibrations gave correlation coefficients >0.98, and the obtained recoveries were in the range of 60.2%-119.9%, with coefficients of variation ≤32.0%. Furthermore, the detection and quantification limits of the method were in the ranges of 0.03-3.2 and 0.1-9.7 ㎍/kg, respectively. Moreover, a survey of residual anthelmintic and antiprotozoal drugs was also carried out in 30 samples of beef, pork, and chicken collected in Korea. Toltrazuril sulfone was detected in all three samples. Thus, our results indicated that the developed method is suitable for determining the anthelmintic and antiprotozoal drug contents in livestock products.

BACKGROUND: The objective of this study was to develop a comprehensive and simple method for the simultaneous determination of 59 veterinary drug residues in livestock products for safety management. METHODS AND RESULTS: For sample preparation, we used a modified liquid extraction method, according to which the sample was extracted with 80% acetonitrile followed by incubation at -20℃ for 30 min. After centrifugation, an aliquot of the extract was evaporated to dryness at 40℃ and analyzed using liquid chromatography combined with tandem mass spectrometry. The method was validated at three concentration levels for beef, pork, chicken, egg, and milk in accordance with the Codex Alimentarius Commission/Guidelines 71-2009. Quantitative analysis was performed using a matrix-matched calibration. As a results, at least 52 (77.6%) out of 66 compounds showed the proper method validation results in terms of both recovery of the target compound and coefficient of variation required by Codex guidelines in livestock products. The limit of quantitation of the method ranged from 0.2 to 1119.6 ng g-1 for all matrices. CONCLUSION(S): This method was accurate, effective, and comprehensive for 59 veterinary drugs determination in livestock products, and can be used to investigate veterinary drugs from different chemical families for safety management in livestock products.

Because particulate matter has emerged as a major contributor to air pollution, the tire industry has conducted studies to reduce particulate matters from tires by improving tire performance. In this study, we compared the conventional blending method, in which rubber, filler, and additives are mixed simultaneously, to the Y-blending method, in which masterbatches are blended. We manufactured carbon black (CB)-filled natural rubber (NR)/butadiene rubber (BR) blend and silica-filled epoxidized NR/BR blend compounds to compare the effects of the two blending methods on the physical properties of the compounds and the amount of particulate matter generated. The Y-blending method provided uniform filler distribution in the heterogeneous rubber matrix, improved processability, and exhibited low rolling resistance. This method also improved physical properties owing to the excellent filler-rubber interaction. The results obtained from measuring the generation of particulate matter indicated that, the Y-blending method reduced PM2.5 particulate matter generation from the CB-filled and silica-filled compounds by 38% and 60%, and that of PM10 by 29% and 67%, respectively. This confirmed the excellence of the Y-blending method regarding the physical properties of truck bus radial tire tread compounds and reduced particulate matter generated.

Periodontal disease is a chronic inflammatory disease that leads to the gradual destruction of the supporting structures of the teeth including gums, periodontal ligaments, alveolar bone, and root cementum. Recently, interests in alleviating symptoms of periodontitis (PD) using natural compounds is increasing. Avenanthramide-C (Avn-C) is a polyphenol found only in oats. It is known to exhibit various biological properties. To date, the effect of Avn-C on PD pathogenesis has not been confirmed. Therefore, this study aimed to verify the protective effects of Avn-C on periodontal inflammation and subsequent alveolar bone erosion in vitro and in vivo. Upregulated expression of catabolic factors, such as matrix metalloproteinase 1 (MMP1), MMP3, interleukin (IL)-6, IL-8, and COX2 induced by lipopolysaccharide and proinflammatory cytokines, IL-1β, and tumor necrosis factor α (TNF-α), was dramatically decreased by Avn-C treatment in human gingival fibroblasts and periodontal ligament cells. Moreover, alveolar bone erosion in the ligature-induced PD mouse model was ameliorated by intra-gingival injection of Avn-C. Molecular mechanism studies revealed that the inhibitory effects of Avn-C on the upregulation of catabolic factors were mediated via ERK (extracellular signal-regulated kinase) and NF-κB pathway that was activated by IL-1β or p38 MAPK and JNK signaling that was activated by TNF-α, respectively. Based on this study, we recommend that Avn-C may be a new natural compound that can be applied to PD treatment.
NUC1 (Nutraceutical compound 1) is an ethanol extract composed of a formulation based on medicinal herbs traditionally used for the treatment of arthritis in Korea and China. This study investigated the therapeutic effects of NUC1 on osteoarthritis (OA). The protective effect of NUC1 on OA was tested in a rabbit model of collagenase-induced arthritis (CIA) for 4 weeks. Results were compared among four groups (n = 9 per group): the normal group (untreated), the CIA group (vehicle control), the NUC1 group (CIA rabbits treated with 200 mg/kg NUC1), and the JOINS group (positive control, CIA rabbits treated with 200 mg/kg JOINS tablet). NUC1 significantly inhibited NO production (p < 0.05 at 125 ㎍/ml, p < 0.01 at 250 ㎍/ml, and p < 0.001 at 500 ㎍/ml) and iNOS expression in macrophages, in a concentration-dependent manner. NUC1 also inhibited the release and protein expression of MMP-1, 3, and 13, in TNF-α-induced chondrosarcoma cells in a concentration-dependent manner. In vivo, the MMP-1 and MMP-3 levels in synovial fluids were significantly (p < 0.05) lower in NUC1 group (77.50 ± 20.56 and 22.50 ± 7.39 pg/ml, respectively) than in the CIA group (148.33 ± 68.58 and 77.50 ± 20.46 pg/ml, respectively). Also, in histopathological, NUC1 ameliorated articular cartilage damage in OA by increasing the abundance of chondrocytes and proteoglycan in the articular cartilage. Thus, NUC1 showed promise as a potential therapeutic agent, and it can be generalized to a broader study population in different OA animal models.
본 연구는 농산물 중 2017년 기준신설예정농약 spiroxamine에 대한 공정시험법을 개발하고자 수행하였다. Spiroxamine은 모화합물만이 잔류물로 정의되므로 이를 분석하기 위한 시험법을 개발하였고, 확립된 시험법은 실험실 내 및 실험실 간 검증을 통해 공정시험법으로의 유효성을 확인하였다. Spiroxamine의 물리화학적 특성을 고려하여 분석을 위해 LC-MS/MS를 분석기기로 사용하였으며, acetonitrile으로 추출 후 유기용매를 이용하여 액액분배조건 및 florisil cartridge를 이용한 SPE 정제조건을 확립하여 시료의 불순물을 효과적으로 정제하는 시험법을 개발하였다. 개발된 Spiroxamine 시험법의 직선성은 결정계수($r^2$)가 0.99 이상으로 우수하였으며, 검출한계 및 정량한계는 각각 0.0005, 0.001 mg/kg으로 높은 감도를 나타내었다. 대표 농산물 5종(현미, 감자, 대두, 감귤, 고추)에 대한 시험법 검증결과 평균 회수율(5 반복)은 71.9~98.8%였고, 분석오차는 10.0% 이하로 정확성 및 재현성이 우수함을 확인할 수 있었다. 또한 광주지방식약청과의 실험실 간 검증 결과에서 평균 회수율은 70.6~104.6%를 나타내어 모두 Codex 가이드라인 및 식약처 가이드라인의 기준에 부합하는 것으로 확인되었다. 개발된 시험법은 낮은 검출한계 및 정량한계, 우수한 직선성, 회수율 시험을 통한 양호한 정밀성 및 재현성 등이 입증되어 농산물 중 Spiroxamine의 분석을 위한 식품공전 공정시험법으로 활용되기에 적합한 것으로 판단된다.
수분 보유력이 우수한 hyaluronan (HA)은 피부 보습에 관여하는 여러 물질들 중 하나로 피부의 extracellular matrix를 구성하는 주요 성분 중 하나이다. Glycosaminoglycans (GAGs)의 구성 성분의 하나로 과거에는 진피에서 유래하는 것으로 알려져 왔으나 최근 연구들을 통해 표피에서 합성되는 것이 확인되었다. Polyphenolic compound의 일종인 kaempferol과 quercetin은 채소류 같은 식물성 음식에 많이 존재하는 것으로 알려져 있으며, kaempferol은 인체 표피세포에서 glutathione 합성을 증가시키고 quercetin은 lipoxygenase inhibitor로 PPAR (peroxisome proliferator activated receptor) - mediated 표피세포 분화를 억제하는 것으로 알려져 있다. 본 연구에서 우리는 표피 세포주에서 이들 flavonoids -kaempferol, quercetin -의 HA 합성에 미치는 영향을 알아보고자 하였다. 물질 처리에 따른 HA 합성 효소인 hyaluronan synthase 1, 2, 3 (HAS1, 2, 3) 유전자 발현의 변화를 semi-quantitative RT-PCR을 통해 살펴보았다. 이들 flavonoid들에 의해 24 h 후 HAS2, 3 mRNA 발현이 증가되는 것을 발견하였다. 또한 HA 합성량의 변화를 알아보기 위해 ELISA를 수행하였다. 24 h 물질 처리 후 배지를 수거하여 HA 합성량을 살펴본 결과 이들 물질에 의해 합성이 유의하게 증가함을 알 수 있었다. 비록 합성 촉진에서의 효과가 retinoic acid에는 못 미치지만 kaempferol과 quercetin은 표피 세포주에서 농도 의존적으로 HA 합성을 증가시켰다. 위의 결과를 통해 flavonoid류인 kaempferol과 quercetin이 피부에서 HA 생산을 촉진시킴을 알 수 있었고 이를 통해 피부 보습과 잔주름 개선에 효과를 볼 수 있을 것으로 생각된다.
