Enterobacter agglomerans U-1의 다당류 생산을 위한 배양조건 (Cultural Condition of Enterobacter agglomerans U-1 for Polysaccharide Production)

  • 점질성 물질을 생산하는 세균인 Enterobacter agglomerans의 다당류 생산조건을 검토하였다. 최적 조건은 배지로서 $sucrose(23.75g/L),\;peptone(2.06g/L),\;yeast\;extract(0.5g/L),\;KH_2PO_4(1.0g/L)$$MgSO_4{\cdot}7H_2O(1.0g/L)$를 함유하며 배양 pH와 온도는 6.5와 $30^{\circ}C$이었다. 이 조건에서 3일 배양후 8.5g/L의 다당류가 생산되며 점도는 240 mPa.s.를 나타내었다. 이때의 다당류 생산수율 및 다당류 생산 속도는 각각 36%와 142.07 mg/g/h이었다.

  • All of us have no time to predict and prepare for a rapidly-approaching future change in the 21st century, thereby being reality that we feel uneasy about future society. Accordingly, there is necessity for re-consolidating environment and daily culture in preparation for future society, namely, aging society. Given well preparing for super-aging society that will approach in the future, by which this effort becomes what bad luck often brings good luck, our country's role is thought to get probably bigger even in the globally silver industrial market in near future. We need to synthetically recognize a human being's aging psychological and physical conditions through the aging experience exhibition available for publicizing properly this historical phenomenon, and to pay more attention to importance of interaction between a human being and product environment. The experience exhibition is a concept for the exhibition form and the spatial composition, which emphasized interaction, with having escaped from the simply exhibition space. Given trying to communicate information more scientifically with escaping from the conventional exhibition of seeing and listening, it is also important to research Into the basis for exhibition as the experience space based on interaction. After all, a human being's aging is the unavoidably future phenomenon. And, importance of environment, which influences a human being, is very mutual as well. Our country is desired to develop a research on interaction of the exhibition experience space available for broadening recognition of senior-friendly and for properly experiencing this, thereby being an opportunity that the business entity, which plans silver industry, and the academic circles, which research and develop this, do mutually cooperate and can make silver industry grow further.

액정 고분자/에틸렌-아크릴산 이오노머 블렌드의 반응상용화에 관한 연구 (Reactive compatibilization of liquid crystalline polymer/ethylene-acrylic acid ionomer blends)

  • 방향족 열방성 액정 고분자와 에틸렌-아크릴 산 공중합체 및 그 금속염 고분자간의 반응 상용화에 관하여 연구하였다. 블렌드는 배치 믹서에서 제조하였으며 액정 고분자의 에스터 그룹과 이오노머의 아크릴산 사이에 발생하는 acidolysis 반응으로 형성되는 그라프트 공중합체의 발생 유무를 평가하였다. 반응의 정도는 배치 믹서에서 시간에 따른 토크의 증가와 열분석 및 형태학 상 구조를 통해 평가하였다. poly(ethylene-co-acrylic acid) (EAA) 이오노머의 Na 염이 반응을 증진시키는데 특히 효과적임을 알 수 있었다. 반응의 진행 정도는 금속염의 종류 뿐 아니라 이오노머의 조성과 반응 시간에 따라 변하였다. 액정 고분자와 Na로 중화된 이오노머 간의 반응으로 생성된 그라프트 공중합체는 액정 고분자와 EAA 이오노머 블렌드의 반응상용화제로 효과가 높음을 미세 구조 분석을 통하여 알 수 있었다.

산도변화와 침지시간이 광중합 복합레진의 잔류단량체 유출에 미치는 영향 (EFFECT OF PH AND STORAGE TIME ON THE ELUTION OF RESIDUAL MONOMERS FROM POLYMERIZED COMPOSITE RESINS)

  • The purpose of this study was to determine whether pH and time has any influence on the degradation behavior of composite restoration by analyzing the leached monomers of dental composites qualitatively and quantitatively after storage in acetate buffer solution as a function of time using high performance liquid chromatography (HPLC) / mass spectrometer. Materials and Methods:Three commercial composite restorative resin materials (Z-250, Heliomolar and Aeliteflo) with different matrix structure and filler composition were studied. Thirty specimens (7mm $diameter{\times}2mm$ thick) of each material were prepared. The cured materials were stored in acetate buffer solution at different pH (4, 7) for 1, 7 and 45days. As a reference, samples of unpolymerized composite materials of each product were treated with methanol (10mg/ml). Identification of the various compounds was achieved by comparison of their mass spectra with those of reference compound, with literature data. and by their fragmentation patterns. Data were analysed statistically using ANOVA and Duncan's test. Results:1. Amounts of leached TEGDMA in Aeliteflo were significantly larger than those of UDMA in Z-250 and Heliomolar at experimental conditions of different storage time and pH variation (p<0.001). 2. As to comparison of the amounts of leached monomers per sorage time, amounts of leached TEGDMA in Aeliteflo and UDMA in Z-250 and Heliomolar were increased in the pH 4 solution more significantly than in the pH 7 solution after 1day, 7days and 45days, respectively (p<0.001). 3. In total amounts of all the leached monomers with storage times, the overall amounts of pH 4 extracts were larger than those of pH 7 extracts for all resin groups, but there was no significant difference (p>0.05).

응용아연(응용亞鉛) 도금공정(鍍金工程)에서 아연부착량(亞鉛附着量)에 관한 연구 (A Study of Zn-coating Weight in Hotdip Galvanizing Process)

  • 本 실험에서는 熔融亞鍍金工程에서 다른 諸要因을 一定하게 하고 鋼金溫度와 浸積時間에 따른 亞鉛附着量과 銑損失을 組織학的인 면에서 考察하였다. 銑損失에 要하는 活性化에너지를 구하였으며 試料範위內에서 附着量에 미치는 최적조건을 찾으므로서 不당한 銑損失을 막고 亞鉛原단位를 줄이는데 目的이 있다. 銑損失量과 드로스(Dross)발생량이 상호 密接한 關係에 있으며 도로스 발생량이 감소되면 亞鉛原단位가 감소되었다. 실驗結論은 다음과 같다. 1. 同一時間內에서 詩片-I은 470$^{\circ}$까지 도금층 두께 형성이 온도에 비례하며 浸積時間 8分정도에서 最大값을 나타낸다. 2. 詩片-5는 浸積時間에 比例해서 附着量이 급증함을 나타내었는데 이는 Si의 영향이 지배적이며 그 이외의 試料에서는 20分이후 附着量增加率이 극히 적었다. 浸積時間은 20分이 가장 적당하며 더 이상 오해 浸積시키면, 銑損失量만, 증가할 것이다. 3. 銑損失에 要하는 活性化에너지는 다음과 같으며 이와같은 값은 ${\gamma}$相의 成長에 미치는 活性化에너지 값과 거의 비슷하리라 생각됨. 詩片 - I = 18,500 Cal/mole 詩片 - III = 22,000 Cal/mole 詩片 - V = 42,300 Cal/mole

Mineral, Nutritional, and Phytochemical Profile, Total Phenolic Content, and Radical Scavenging Activity of Philippine Bamboo "Bolo" Gigantochloa levis (Blanco) Merr. Leaves

  • The study is a pioneering effort to determine the mineral, nutritional, and phytochemical composition and phenolic content and to determine the free radical scavenging activity of Gigantochloa levis (Blanco) Merr, a native bamboo species (locally known as "bolo") in the Philippines. Proximate analysis showed that air-dried G. levis leaves contain 15.8% ash, 22.6% crude protein, 1.2% crude fat, 29.3% crude fiber, and 19.7% total sugar. Phytochemical tests indicated the presence of diterpenes, triterpenes, saponins, phenols, tannins, and flavonoids in both the ethanolic and aqueous leaf extracts, while phytosterols were only detected in the ethanolic extract. Folin-Ciocalteu assay determined the total phenolic content in gallic acid equivalents (GAE) to be $85.86{\pm}3.71$ and $32.32{\pm}1.01mg\;GAE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The total phenolic content in quercetin equivalents (QE) was $74.44{\pm}3.11$ and $29.43{\pm}0.85mg\;QE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The radical scavenging activity of the different solvent fractions containing varying concentrations of the extract was determined using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay. The ethyl acetate and 1-butanol fractions were found to have the highest radical scavenging activity. Mineral analysis via Energy Dispersive X-Ray Spectrometry (EDS) of the ash of G. levis leaves showed that Si is the major component, followed by K and Mg. These results point to the potential of G. levis leaves as a source of minerals and bioactive compounds with medicinal value.

Compositional Analysis of Major Saponins and Anti-inflammatory Activitiy of Steam-Processed Platycodi Radix under Pressure

  • Platycosides are the saponins in Platycodi Radix and they have several beneficial effects such as antiinflammatory and anti-obesity activities. This study was designed to determine the changes in the saponin composition in Platycodi Radix (platycosides) after being processed under steam and pressure and to investigate the anti-inflammatory effects of their extracts. The change of the platycoside compositions was investigated after 1, 2, 3, 6 and 9h heat processing of Platycodi Radices by using HPLC coupled with an evaporative light scattering detection (ELSD) system. After heat treatment ($125^{\circ}C$, 1, 2, 3, 6 and 9 h), the contents of several platycosides such as platycoside E, platycodin $D_3$, platycodin D, polygalacin D, and platycodin A decreased as the processing time was longer. While the total contents of the saponins decreased, the contents of deapi-forms of deapiplatycoside E, deapi-platycodin $D_3$, and deapi-platycodin D increased relatively. These results indicate that the linkage between apiose and xylose located at C-28 is labile to heat and pressure. The LPS-induced iNOS inhibitory activities of the samples treated for 1 and 2 hours were enhanced and after then, the activities were reduced. These results suggested that heat treatment of the samples affect the content of the total saponins and the saponin content may be the important criteria representing the anti-inflammatory activity.

닭고기 너겟의 닭고기 스킨 첨가수준에 따른 품질 특성 (Quality Properties of Chicken Nugget with Various Levels of Chicken Skin)

  • 본 연구는 닭고기 스킨의 첨가가 닭고기 너겟의 품질 특성에 미치는 영향을 조사하였다. 처리구별로 닭고기 스킨을 0%(control), 5%, 10%, 15%를 첨가하여 제조하였다. 수분함량은 닭고기 스킨의 첨가량이 증가할수록 유의적으로 감소하였으나(P<0.05), 지방함량은 닭고기 스킨의 첨가량이 증가할수록 유의적으로 증가하였다(P<0.05). 단백질함량은 닭고기 스킨의 첨가량이 증가할수록 감소하는 경향을 보였다. 회분함량은 닭고기 스킨의 첨가량이 증가할수록 증가하였으나, 대조구와 처리구 간에 유의적 차이는 발견하지 못하였다. 명도는 닭고기 스킨을 첨가함에 따라 유의적으로 증가하는 경향을 보였으나(P<0.05), 적색도와 황색도는 닭고기 스킨의 첨가량이 증가할수록 감소하는 경향을 보였다. 튀김수율은 닭고기 스킨의 첨가량이 증가할수록 감소하는 경향을 보이며, 경도(hardness), 검성(gumminess)과 씹힘성(chewiness)은 닭고기 스킨의 첨가량이 증가함에 따라 감소하는 경향을 보였다. 관능평가에서 다즙성(juiciness)과 전체적인 맛(overall acceptability)은 닭고기 스킨의 첨가량이 증가할수록 증가하는 추세를 보였다. 이러한 결과로 보아 닭고기 너겟에 닭고기 스킨을 첨가 시 품질 특성을 증진시키는 것으로 나타났으며, 관능평가를 기초로 하여 물성 및 튀김수율을 고려하였을 때 약 10%의 닭고기 스킨 첨가가 닭고기 너겟의 적정 첨가비율이라고 판단된다.

고분자 연료전지용 MEA 연속 코팅공정 개발 (Continuous Coating Process Development for PEFC Membrane Electrode Assembly)

  • Membrane electrode assembly (MEA) for polymer electrolyte fuel cell (PEFC) are commonly prepared in the research laboratory by spraying, screen-printing and brushing catalyst slurry onto membrane or other support material like carbon paper or polyimide film in a batch style. These hand applications of the catalyst slurry are painstaking process with respect to precision of catalyst loading and reproducibility. It has been generally mentioned that the adoption of continuous process is very helpful to develop the reliable product. In the present work, we report the results of using continuous type coater with doctor-blade to coat catalyst slurry for preparing the MEA catalyst layers In a faster and highly reproducible fashion. We show that while expectedly faster than batch style, the machine coater requires the use of slurry of appropriate composition and a properly selected transfer decal material in order to achieve superior MEA plat lnw loading reproducibility. To make highly viscous catalyst slurry that is imperative for using coater, we use 40wt.% Nafion solution and minimize the content of organic solvent. And the choice of proper high surface area catalyst is important in the viewpoint of making well-dispersed slurry. After catalyst coating onto the support material, we transferred the catalyst layer to both sides of Nafion membrane by hot-pressing In this case, the degree of transfer was Influenced by hot-pressing condition including temperature, pressure, and time. To compare the transferring ability, we compared so many films and detaching papers. And among the support, polyethylene terephthalate(PET) film shows the prominent result.

고추를 이용한 생선용 복합 분말 조미료 개발 및 평가 (Development of Mixed Seasoning Products for Fish Dishes using Korean Chili Peppers(Capsicum annuum L.))

  • 미국산 복합 분말 조미료에 기초하여 수출용 고부가가치 고추 가공 제품으로 생선용 복합 분말 조미료를 개발하였다. 한국산 고춧가루와 한국 고유의 재료인 유자, 버섯, 그리고 마늘을 첨가하여 짠맛과 단맛, 매운맛을 조화롭게 하여 맛의 차별화를 유도하였고, 생선의 비린내를 제거하였다. 수출용 생선 복합 분말 조미료 AI는 한국산 고춧가루(20%), 오레가노, 파슬리, 타임, 흑후추 등의 총 허브류(31.7%), 소금(11.5%), 유자(2.8%), 버섯(8.6%), 마늘(5.8%)의 식품 재료로 구성되었고, 그 외 파프리카, 설탕, 올리브 오일 등을 첨가하여 제조하였다. 미국인을 대상으로 한 기호도 평가에서 개발된 샘플(AI)은 미국산 복합 분말 조미료 제품보다 선호도가 높게 평가되었다.
