The changed isotropic absolute Raman intensities of the phosphate residue in the complexes of positive charge oligopeptides, lys-lys, arg-arg, lys-aromat-lys, negative charge diethyl phosphoric acid (DEP) and polyriboadenylic acid{poly(rA)} were reported and discussed. Our measurements showed that the absolute intensities of phosphate stretch vibration in complexes were different according to the reaction partners. Due to the partial electrical charge and molecular structure of oligopeptides for the complex formation lysine can interact more strongly than arginine when the reaction partners have short chain and no steric hindrance. Owing to these reasons the intensity of phosphate stretching vibration is very sensitive according to the circumstance of reaction. From our results we could suggest that we can discriminate any one of the the lysine and arginine in the complicated biological molecule during interaction between nucleotides and proteins. The activity of reaction of two basical oligopeptides is not quite similar for complex formation in aqueous solution. The activity of dipeptides depends upon the structure of molecule and environment for complex formation. Aromatic ring contributes to electrostatic interaction in complexes. The amount of the absolute intensity for pure stacking interaction is smaller than electrostatic interaction in macromolecular complexes.
Using the Chlorella cells which had been uniformly labeled with $^{32}P$, the distribution of phosphorus in various fractions of cell material was investigated. Uniformly $^{32}P$-labeled Chlorella cells were further grown in a P-free medium, and some protions of the cells were taken out at intervals during the culture, and subjected to analyze the contents of $^{32}P$ in various fractins of the cell constituents. 2. Analysis of the $^{32}P$-labeled Chlorella cells showed that the highest in P-content was the fraction of RNA followed by those of lipid, RNA-polyphosphate complex, acid-insoluble polyphosphate, acid-soluble polyphosphate, DNA and protein. 3. During the culture of $^{32}P$-labeled Chlorella cells in a P-free medium, amounts of phosphate in DNA, protein and lipid fractions increased, while the P-contents in the fraction of RNA-polyphosphate complex decreased as well as those of acid-insoluble polyphosphate and acid-soluble polyphosphate fractions. 4. It was inferred that phosphorus used in the syntheses of DNA and protein was taken from polyphosphates of the cells, and RNA-polyphosphate complex would play an important role as a phosphate pool.
The drinking water industry faces a growing number of difficultiesin the treatment of groundwater for drinking water production. Groundwater sources are frequently contaminated with nitrates and phosphates due to usage of chemical fertilizer In this study, feasibility of micellar enhanced ultrafiltation (MEUF) was investigated to remediate groundwater contaminated with nitrate and phosphate. Ultrafiltration membrane was cellulose acetate with molecular weight cut off (MWCO) 10,000 and celtyl pyridinium chloride (CPC) was used to form pollutant-micelle complex with nitrate and phosphate. The results show that nitrate and phosphate rejections are satisfactory. The removal efficiency of nitrate and phosphate show 80% and 84% in single pollutant system, respectively with 3 molar ratio of CPC to pollutants. In the multi-pollutant systems, the removalefficiency increased to 90 % and 89 % for nitrate and phosphate, respectively, The presence of nitrate in the solutions did not affect the removal of phosphate and that of phosphate did not affect the removal of nitrate. The concentration of CPC in the permeate and removal efficiency of CPC was a function of the concentration of CPC in the feed solutions.
Dicalcium phosphate dihydrate (DCPD)를 2% HPMC 및 35% PAA와 각각 1:1의 분액비로 30초간 섞은 후 cone and plate geometry를 가지는 rheometer를 사용하여 frequency sweep test와 time sweep test를 통해 shear storage modulus (G'), shear loss modulus (G''), 그리고 complex viscosity (${\eta}^*$)를 측정하였다. 2% HPMC군과 35% PAA군의 complex viscostiy의 차이를 Mann-whitney test with Bonferroni's collection을 사용하여 분석하였다. 실험결과 2% HPMC 및 35% PAA 군 모두에서 shear thinning과 yield behavior등 pseudoplastic property를 보였으며 complex viscosity는 HPMC 군에서 PAA 군보다 통계적으로 유의성 있게 높았다. Chlorella ellipsoidea were grown in a Mg-free medium. Aliquots of the algal cell were taken out at the beginning and predetermined time intervals during the culture and were analyzed the contents of phosphate in various fractions of the cell constituents. The results obtained were compared with those of the control. When Chlorella cells were grown in a Mg-free medium, the contents of phosphate in the DNA protein, RNA-olyphosphate complex, nucleotidic-lbileP, and PCA-soluble, fractions decreased compared with those of the control, while the content of acid insoluble polyphosphate increased significantly. On the otherhand, RNA-P and lipid-P showed the tendency of decrease at the early stage of the culture, but they were increased more than those in the control as culture proceeds. It is showed that phosphate turnover from acid-insoluble polyphosphate into DNA, protein, and RNA-polyphosphate complex was inhibited by magnesium-deficiency of the cells.
Adsorption on goethite of individual component from a solution containing phosphate, sulfate, or copper ion was investigated. Competitive adsorption in the binary and ternary solution systems composed of phosphate, sulfate, and copper ions was also investigated. In competitive adsorption systems with phosphate and sulfate ions, the presence of phosphate ion reduced the adsorption of sulfate ion largely. On the other hand, the presence of sulfate ion caused only a small decrease in phosphate adsorption. This result suggests that phosphate ion is a stronger competitor for adsorption on goethite than sulfate ion, which is consistent with the higher affinity of phosphate for the surface compared to sulfate ion. Compared to the results from single-sorbate systems, adsorption of copper ion in the binary system of sulfate ion and copper ion was found to be enhanced in the presence of sulfate ion. Addition of sulfate ion to the binary system of copper ion and phosphate ion resulted in a small enhancement in copper sorption. This result implies that the presence of sulfate ion promotes adsorption of the ternary complex FeOHCuSO$_4$. The adsorption isotherms could be well described by the Langmuir adsorption equation.
Escherichia coli has a PhoR-PhoB two-component regulatory system to detect and respond to the changes of environmental phosphate concentration. For the E. coli W3110 strain growing under phosphate-limiting condition, the changes of global gene expression levels were investigated by using DNA microarray analysis. The expression levels of some genes that are involved in phosphate metabolism were increased as phosphate became limited, whereas those of the genes involved in ribosomal protein or amino acid metabolism were decreased, owing to the stationary phase response. The upregulated genes could be divided into temporarily and permanently inducible genes by phosphate starvation. At the peak point showing the highest expression levels of the phoB and phoR genes under phosphate-limiting condition, the phoB- and/or phoR-dependent regulatory mechanisms were investigated in detail by comparing the gene expression levels among the wild-type and phoB and/or phoR mutant strains. Overall, the phoB mutation was epistatic over the phoR mutation. It was found that PhoBR and PhoB were responsible for the upregulation of the phosphonate or glycerol phosphate metabolism and high-affinity phosphate transport system, respectively. These results show the complex regulation by the PhoR-PhoB two-component regulatory system in E. coli.
DNA 모델 화합물인 bis(p-nitrophenyl)phosphate(BNPP)에 대한 2핵 닉켈(II) 착 화합물, ${\mu}-aquapentaaqua[{\mu}-3,6-bis(6'-methyl-2'-pyridyl)pyridazine]chlorodinickel(II)$ trichloride trihydrate(APNT)의 촉매 가수분해 반응성을 검토하였다.
