Distribution of Phlorotannins in the Brown Alga Ecklonia cava and Comparison of Pretreatments for Extraction

  • The brown seaweed Ecklonia cava is known to be a rich source of phlorotannins that have diverse biological activities. Among the phlorotannins in E. cava, concentrations of dieckol and phlorofucofuroeckol-A, which were identified as major active components, were determined in different parts of the tissue. We compared the efficacy of different pretreatments for their extraction. A high-performance liquid chromatography (HPLC) method to determine phlorotannin concentrations showed good accuracy (92.64 and 94.02%, respectively), precision (3.92 and 3.94%, respectively), and linearity (r>0.996). Mature thalli contained 1.5-fold more dieckol (1.82 mg/g-dry tissue) than young thalli. In the tissues of E. cava, blade tissue contained more phlorotannins than the stipe or holdfast. Among differently dried thalli, approximately 90% or more dieckol and phlorofucofuroeckol-A were extracted from shadow-dried tissue as compared with lyophilized tissue. In sun-dried and oven-dried thalli, approximately 60% of the phlorotannins were extracted. Thalli washed with fresh water, boiled thalli, and steamed thalli showed reduced extraction of the compounds.

Comparison on Efficacy of Administration Time of Levothyroxine in Patients with Hypothyroidism: A Meta-analysis (갑상선 기능 저하 환자에서 levothyroxine 투여 시간에 따른 효능 비교: 메타분석)

  • Background: Levothyroxine is an essential drug for the treatment of hypothyroidism or related diseases. Several studies have reported an association between the effects of levothyroxine treatment and time of administration, which can be inconsistent. Objective: This study was conducted to compare the levels of thyroid-stimulating hormone or free thyroxine between morning and nighttime dosing of levothyroxine. Methods: We reviewed previously reported relevant articles and conducted a meta-analysis. Results: In total, five studies were included in this meta-analysis. Results showed that thyroid-stimulating hormone (standard difference in means [SE]=0.321; 95% confidence interval [CI], -0.016 to 0.657) and free thyroxine (SE= -1.367; 95% CI, -2.943 to 0.210) levels did not differ significantly between morning (before breakfast) and nighttime (before bedtime) administration. Conclusion: This is the first meta-analysis to evaluate the effects of time of administration on levothyroxine levels in patients with hypothyroidism. Based on our results, we suggest considering patients' lifestyles or daily routines when counselling them on the optimal time of administration for levothyroxine.

  • Breast cancer screening makes extensive utilization of mammography. Even so, there has been a lot of debate with regards to this application's starting age as well as screening interval. The deep learning technique of transfer learning is employed for transferring the knowledge learnt from the source tasks to the target tasks. For the resolution of real-world problems, deep neural networks have demonstrated superior performance in comparison with the standard machine learning algorithms. The architecture of the deep neural networks has to be defined by taking into account the problem domain knowledge. Normally, this technique will consume a lot of time as well as computational resources. This work evaluated the efficacy of the deep learning neural network like Visual Geometry Group Network (VGG Net) Residual Network (Res Net), as well as inception network for classifying the mammograms. This work proposed optimization of ResNet with Teaching Learning Based Optimization (TLBO) algorithm's in order to predict breast cancers by means of mammogram images. The proposed TLBO-ResNet, an optimized ResNet with faster convergence ability when compared with other evolutionary methods for mammogram classification.

Usefulness of Brushing Cytology in the Diagnosis of the Patients with the Stricture of Biliary Tree (담도계 협착 환자의 진단에 솔질 세포검사의 유용성)

  • Pancreaticobiliary tract strictures are frequent Indications for endoscopic retrograde cholangiopancreatography(ERCP). We have investigated the brushing cytology in order to determine its efficacy for diagnosis of pancreaticobiliary malisnancies. Brushing cytology during ERCP was evaluated in 56 patients with biliary tract stricture presenting to the Catholic Hospital of Taegu-Hyosung from April 1997 to August 1999. A comparison was made between the cytologic and histologic diagnoses on 32 cases from 30 patients. A diagnosis of malignancy was establishied in 78.1%, benign in 15.6%, and inadequate in 6.3% of the cases. Statistical data on cytologic diagnoses in strictures of the bile duct were as follows; specificity and sensitivity of blushing procedure was 100% & 83.3%, respectively: sensitivity of interpretation was 89.3%: with no false positive cases and 3 false negative cases: predictive value for malignancy was 100% & 100%, respectively: predictive value for benign was 28.6% & 40%,, respectively: overall diagnostic efficiency was 84.4%. It is concluded that brush cytology is a diagnostically reliable, highly specific technique for malignant lesions encounted at ERCP, although a negative result does not rule out the diagnosis of malignancy.

Comparison of Differential Analgesic and Adverse Effects of 0.125% and 0.15% Ropivacaine Coadministrated Epidurally with Fentanyl (경막외강으로 Fentanyl을 첨가한 0.125% 및 0.15% Ropivacaine 투여시 술후 진통 효과 및 부작용의 차이 비교)

  • Background: This purpose of this study is to compare the analgesic efficacy, occurrence of motor block and other side effects of 48 hr continuous postoperative epidural infusions with 0.125% ropivacaine with fentanyl or 0.15% ropivacaine with fentanyl. Methods: Forty patients undergoing Cesarean section were randomly allocated into two groups. Both groups received epidural injection of 2.0% lidocaine 19 ml and 0.75% ropivacaine 2-5 ml with fentanyl $50{\mu}g$ for 20 minutes before surgery. Following surgery, a continuous epidural infusion using a two- day infusor was given for 40 minutes for post-operative pain control. Group 1 (n = 20) then received 0.125% ropivacaine and 6 ug/ml of fentanyl at a rate of 2 ml/h. Group 2 (n = 20) received 0.15% ropivacaine and 6 ug/ml of fentanyl at the same rate. Visual analog scales (VAS) for pain during rest and movement, sensory change and motor blockade were assessed for 48 hr after surgery. Results: There were no significant differences in VAS for pain during rest or movement. The incidence of side effects was similar in both groups. Conclusions: Using 0.125% ropivacaine with fentanyl via an epidural route provided similar pain relief and side effects as 0.15% ropivacaine with fentanyl.

Identification of an Entomopathogenic Fungus, Beauveria bassiana SFB-205 toxic to the Green Peach Aphid, Myzus persicae

  • To select entomopathogenic fungi controlling aphids effectively, several isolates were screened against second instars of Myzus persicae nymphs in the glasshouse using conidia suspension at $1.0{\times}10^5$ conidia/ml. Among these isolates, SFB-205 conidia showed the highest insecticidal activity about 32.7% efficacy to M. persicae at 4 days after application in the glasshouse. The attachment of SFB-205 conidia on the surface of M. persicae nymphs, and germination and penetration were observed using scanning electron microscopy. SFB-205 was identified as Beauveria bassiana species through the comparison of 5.8 s rRNA genes. There were 24 polymorphisms between SFB-205 and the previously reported isolate, B. bassiana ATCC74040 using six kinds of primer combinations in amplified fragment length polymorphism (AFLP) analysis. The B. bassiana SFB-205 might be used as a practical biological control agent for the green peach aphid, M. persicae in the field.

Effect of warming local anesthesia solutions before intraoral administration in dentistry: a systematic review

  • Background: The aim of the present systematic review was to evaluate and compare the efficacy of warmed and unwarmed local anesthesia solutions in reduction of pain during intraoral injection administration. Methods: PubMed, Ovid SP, and Cochrane Central Register of Controlled Trials were searched from publication years 1990 to 2020 with relevant MeSH terms. Studies were screened by titles and abstracts, followed by full-texts evaluation of the included studies. Results: A total of four studies were included in the systematic review. Outcomes evaluated were subjective and objective pain during administration of the warmed local anesthesia solution in comparison with the unwarmed local anesthesia solution. Among the four studies that evaluated the self-reported pain score, three studies showed significantly lower pain scores associated with warmed local anesthesia. Only two studies evaluated the observed pain score, and both of them reported a significantly lower pain reaction with the warmed local anesthesia solution. Conclusion: Within the limits of this systematic review, warming the local anesthesia solution to body temperature (37℃) before administration seemed to reduce the discomfort during intraoral local anaesthesia administration, and more high-quality studies should be carried out to validate the same.

Comparison of the Genetic Alterations between Primary Colorectal Cancers and Their Corresponding Patient-Derived Xenograft Tissues

  • Patient-derived xenograft (PDX) models are useful tools for tumor biology research and testing the efficacy of candidate anticancer drugs targeting the druggable mutations identified in tumor tissue. However, it is still unknown how much of the genetic alterations identified in primary tumors are consistently detected in tumor tissues in the PDX model. In this study, we analyzed the genetic alterations of three primary colorectal cancers (CRCs) and matched xenograft tissues in PDX models using a next-generation sequencing cancer panel. Of the 17 somatic mutations identified from the three CRCs, 14 (82.4%) were consistently identified in both primary and xenograft tumors. The other three mutations identified in the primary tumor were not detected in the xenograft tumor tissue. There was no newly identified mutation in the xenograft tumor tissues. In addition to the somatic mutations, the copy number alteration profiles were also largely consistent between the primary tumor and xenograft tissue. All of these data suggest that the PDX tumor model preserves the majority of the key mutations detected in the primary tumor site. This study provides evidence that the PDX model is useful for testing targeted therapies in the clinical field and research on precision medicine.

A Preliminary Comparison of the Efficacy of Auricular Acupuncture, Transdermal Nicotine Patch and Combination Therapy for Smoking Cessation (금연 이침과 경피적 니코틴 패치 및 병용 요법 간의 금연 효과에 대한 예비적 비교 연구)

  • Objectives : The purpose of this study was to compare three therapies - auricular acupuncture, transdermal nicotine patch and therapy - in combination with auricular acupuncture and nicotine patch. Methods : The subjects of this study included 188 male smokers in their 20s, 30s, 40s and 50s, who visited Wonmi Public Health Center. They were evaluated with the Fagerstrom test for nicotine dependence, amounts of daily smoking and success rate of smoking cessation. The subjects decided on a therapy method strictly of their own choice. They were divided into three groups according to the therapy; auricular acupuncture group (62 persons), nicotine patch group (69 persons) and combination therapy group (57 persons). Results : All three groups resulted in statistically significant reducing effects of nicotine dependence and amounts of daily smoking. In the combination therapy group, nicotine dependence was significantly decreased by more than those of the other groups. There were no statistically significant differences in decrements of daily smoking and success rate of smoking cessation among the three groups. Conclusions : The above results suggest that auricular acupuncture and transdermal nicotine patch have significant effects of smoking cessation. If they are combined, it is expected that therapies for smoking cessation would be developed.

Effects of Brief Character Strength Workshop for Enhancement of Positivity (긍정성 증진을 위한 단기 성격 강점 워크샵의 효과)

  • Objective : The purpose of this study is to develop the one-session positive psychology based strength enhancing workshop and verify effect of this program. This program is composed of lecture based on positive psychology and activities for identifying and utilizing personal character strengths. Methods : Total 81 participants were participated in this study. Positive Resources Test was assessed the change of personal positive resources at pre-, post program and subjective satisfaction for the workshop was measured at the post program. Repeated-t test was performed to identify the effect of this program. Results : Among the positive resources, satisfaction with life, positive affect, gratitude, acceptance, personal growth, efficacy, flow, resilience, hope, spirituality are significantly increased after program. Conclusion : Although this study is a single-session program without control group for comparison, it is meaningful to try a short-term intervention concise the program consisting of more than 8 sessions. This study will be based on the future development a variety of positive interventions.
