Molecular Interaction Between Interleukin-8 Receptor and G$_\alpha$16 subunit G protein (Interleukin-8 수용체와 G$_\alpha$ 16 subunit G protein 간의 분자상호 작용에 관한 연구)

  • In order to identify the domains of the G$_{\alpha}$16 subunit G protein that are responsible for its activation by the Interleukin-8 receptor, a serious of chimeras between G$_{\alpha}$16 and G$_{\alpha}$11 were assessed for their abilities to be activated by these receptors. Co-expression of IL-8 receptor and chimeras in which the carboxyl-terminal regions of G$_{\alpha}$11 were replaced from 30 up to 156 amino acid residues with the corresponding regions of G$_{\alpha}$16 demonstrated that C-terminal 156 amino acid residues of the G$_{\alpha}$16 were not sufficient to confer IL-8 receptor interaction specificity. Testing of a reciprocal serious of chimeras composed of G$_{\alpha}$16 sequences at the amino terminus and G$_{\alpha}$11 sequences at the carboxyl terminals revealed that sequences extending from the amino tar- minus to amino acid 209 of G$_{\alpha}$16 were sufficient to 7ndow the chimera with 75-80% of interaction specificity for 7-8-induced activation. Comparison of Extractive Nitrogenous Constituents in the Three Species of Raw Bloody Clams, Scaphrca broughtonii, S. subcrenata, and Tegillarca granosa Extracts (피조개, 새고막 및 고막의 함질소 엑스성분 조성 비교)

  • Extracts of raw bloody clams, broughton's ribbed ark (Scaphrca broughtonii), subcrenated ark (S. subcrenata), and granulated ark (Tegillarca granosa), were analyzed and compared foe extractive nitrogen, free amino acids, combined amino acids, ATP and related compounds, quaternary ammonium bases, and guanidino compounds. The contents of extractive nitrogen in broughton's ribbed ark, subcrenated ark, and granulated ark were 479, 506, and 432 mg/100 g, respectively. Twenty-eight or twenty-nine types of free amino acids were detected in all three extracts, among which taurine, ${\beta}-alanine$, glutamic acid, and alanine were the major ones. The composition of the major extractive components such as free amino acids, combined amino acids, ATP and related compounds, TMAO, TMA, and creatine in the extracts were similar among the extracts, but their contents were different.

Extractive Nitrogenous Constituents of Dried Layer, Porphyra dentata (잇바디돌김 건제품의 함질소 엑스성분 조성)

  • In order to investigate the composition of dried layer, Porphyra dentata cultured at the south coast of Korea, the dried layer was analyzed for extractive nitrogen, free amino acids, combined amino acids, ATP and its related compounds and quaternary ammonium basis, The extractive nitrogen contents of dried laver extracts were 670-1,304 mg/100 g (on dry basis). From twenty-eight to twenty-nine kinds of free amino acids were found in the dried laver extracts and their total amounts were $2,796\~6,277\;mg/100\;g$ (on dry basis). The extracts were rich in free amino acids such as alanine, taurine, glutamic acid, glutamine and phosphoserine, From eighteen to twenty-one kinds of combined amino acids were found in the extracts and their total amounts were $1,406\~2,142\;mg/100\;g$ (on dry basis). The amounts of ATP and its related compounds were $65.7\~124,7\;mg/100\;g(2.13\sim3.68{\mu}mol/g$ on dry basis), Homarine was detected in all samples but $\beta$-alaninebetaine, $\gamma$-bufobetaine and trigonelline disappeared during processing. TMAO and TMA were detected in all samples. During processing of dried layer, free amino acids, TMAO and TMA were increased but the other constituents such as combined amino acids, ATP and its related compounds and betaines were decreased in all specimens.

  • Subcellular localization is a key functional char acteristic of proteins. With the number of sequences entering databanks rapidly increasing, the importance of developing a powerful tool to identify protein subcellular location has become self-evident. In this paper, we introduce a novel method for predic ting protein subcellular locations from protein sequences. The main idea was motivated from the observation that amino acid pair composition data is redundant. By classifying from multiple feature subsets and using many kinds of amino acid pair composition s, we forced the classifiers to make uncorrelated errors. Therefore when we combined the predictors using a voting scheme, the prediction accuracy c ould be improved. Experiment was conducted on several data sets and significant improvement has been achieve d in a jackknife test.

Preparation of Branched-chain Amino Acid (BCAA)-enriched Hydrolysates from Corn Gluten (고 분지아미노산 함유한 옥수수 단백가수물의 제조조건 탐색)

  • The process of the preparation of branched-chain amino acid (BCAA)-enriched hydrolysates from corn gluten was optimized through the parameters of pre-treatment (heating and cellulosic hydrolysis), hydrolysis method (acid, protease, and microbe plus protease), concentration, and spray drying condition. The protein yield of corn gluten was increased by heating and cellulase treatments. Among three different hydrolysis methods, the combined use of microbes and protease was the most effective in terms of free amino acid (FAA) and BCAA content of the corn gluten hydrolysates. In addition, the FAA and BCAA content in the hydrolysates prepared by microbial and enzymatic combined treatment were improved by a concentration process. Spray drying conditions for the preparation of the powder from the hydrolyzed reactant were an inlet temperature of $185^{\circ}C$, outlet temperature of $80^{\circ}C$, and the use of maltodextrin as an anticaking agent. Thus, this study established an economical process for preparation of value-added hydrolysates of excellent productivity and quality, in terms of high BCAA content and product stability.

  • Vegetables contain more or less calcium. By the traditional method the total calcium in the vegetable was analyzed. A part or calcium in the vegetable is fixed as calcium oxalate and the calcium does not utilize for nutrition. In this investigation the free calcium was determined by extraction in boiling water, the combined calcium with oxalic acid was determined by extraction in 0.1 M hydrochloric aci and the other calcium combined with protein, amino acid etc. was estimated. The amount of calcium obtained from the difference between the total calcium and calcium exmtaining in calcium oxalate was estimated to be nutritious.

Dietary Factors for Secretary Digestive Enzyme from the Pancreas in the Chicken (계의 췌장소화효소 분비에 미치는 사료성분에 관한 연구)

  • The present study was done to investigate the mechanism of Pancreatic digestive enzyme secretion in response to dietary components in chicks. A simplefied pancreatic juice collection method, useful for a short-term experiment, was developed. By wing vein injection, it was shown that the increased trypsinogen and chymotrypsinogen, while neither other single amino acids nor glucose affected the secretion of enzymes, amylase, trypsinogen and chymotrypsinogen. Cholecystokinin (CCK) had an immediate effect on pancreatic enzyme secretion and this response was in a dose dependent fashion. The injection of CCK seemed to have selective stimulation favoring the secretion of chymotrypsinosen followed by amylase and trypsinogen. Simultaneous injection of single amino acid with CCK increased digestive enzyme secretion to various extents depending on the kind of amino acids whereas the injection of glucose with CCK did not affect when compared with that of CCK'alone. By varying doses, synergetic action of CCK plus amino acid on the secretion of pancreatic digestive enzymes was observed at 0.5mM for Val and 5mM for Arg. A further attempt was made to examine the effect of combined administration of amino acids with CCK on pancreatic enzyme secretion. The injected substances were an AAs mixture and combination of selected amino acids, i.e. Thr+Phe+Ile, Thr+Phe. Thr+Ile or Phe+Ile. When increases in enzyme outputs for the first 30 min were compared , it was shown that the responses of three enzymes, amylase, trypsinogen and chymotrypsinogen, brought about by the administration of the AAs mixture was almost entirely accounted for by the combined injection of Thr+Phe. Thus, it was well demonstrated that CCK and amino acids had a synergetic action on the secretion of a specific pancreatic digestive enzyme depending on a kind of amino acid injected.

A Study on the Amino-Carbonyl Reaction (아미노-카르보닐 반응(反應)에 관한 연구)

  • Reaction conditions in the amino-carbonyl reaction, and the effect of amino acids on the reactivity of amino-carbonyl reaction were investigated. Results obtained are as follows : 1. When the pH of the reaction mixture was increased above the isoelectric point of an amino acid, a significant increase in the color intensity was observed. 2. The color intensity increased gradually up to 1 : 1 of the molar ratio of reactants. This result was interpreted to show that sugar and free amino group combined in 1 : 1 ratio. 3. Amino-carbonyl reaction showed a significant time and temperature-dependences. The activation energy at 0.2 M glucose and 0.2 M glycine system was 37.5 Kcal/mole. 4. Among amino acids tested, glycine, lysine and $\beta$-alanine caused a significant increase in the color intensity, but acidic amino acids showed the least color intensity. The latter was interpreted to show that one of carboxyl groups of acidic amino acid has an inhibiting effect on the reactivity of the amino group. 5. The color intensity of sugars tested was in the order of xylose>arabibose>fructose>glucose>maltose>lactose.

THE EFFECTS OF DIETARY ENERGY ON THE TOTAL SULPHUR AMINO ACID REQUIREMENTS OF BROILERS DURING TWO GROWTH PERIODS

  • There levels of dietary ME (3,000, 3,200 and 3,400 kcal/kg) and four levels of Total Sulphur Amino Acid (TSAA) (0.73, 0.83, 0.93 and 1.03%) were studied in the starter period (0-3 wks) of the broilers. Three levels of dietary ME (3,000, 3,200 and 3,400 kcal/kg) combined with four levels of TSAA (0.65, 0.72, 0.79 and 0.86%) were studied in the grower period (3-6 wks). The crude protein content of the diet of the starter period was 23% while the diet of the grower period was 20%. The performance data of the starter broilers indicated that the dietary energy levels had no significant effects on body weight gain, feed intake and feed:gain ratio. However, TSAA levels had a significant influence on the growth and feed parameters. The response pattern for the grower period was similar to the starter period. The present experiment showed that in the tropics the TSAA requirement for the starter period was between 0.83 to 0.93% which is similar to the values recommended by NRC while for the grower period the TSAA requirement was between 0.79 to 0.86% at all the three energy levels which is higher than the values recommended by NRC.

Transfer RNA Acceptor Stem Determinants for Specific Aminoacylation by Class II Aminoacyl-tRNA Synthetases

  • A critical step in the faithful translation of genetic information is specific tRNA recognition by aminoacyl-tRNA synthetases. These enzymes catalyze the covalent attachment of particular amino acids to the terminal adenosine of cognate tRNA substrates. In general, there is one synthetase for each of the twenty amino acids and each enzyme must discriminate against all of the cellular tRNAs that are specific for the nineteen noncognate amino acids. Primary sequence information combined with structural data have resulted in the division of the twenty synthetases into two classes. In recent years, several high-resolution co-crystal structures along with biochemical data have led to an increased understanding of tRNA recognition by synthetases of both classes. The anticodon sequence and the amino acid acceptor stem are the most common locations for critical recognition elements. This review will focus on acceptor stem discrimination by class II synthetases. In particular, the results of in vitro aminoacylation assays and site-directed and atomic group mutagenesis studies will be discussed. These studies have revealed that even subtle atomic determinants can provide signals for specific tRNA aminoacylation.
