Development and Validation of HPLC Method for Pharmacokinetic Study of Promethazine in Human (염산프로메타진 체내동태 연구를 위한 혈청 중 프로메타진의 HPLC 정량법 개발 및 검증)

  • A rapid, selective and sensitive reversed-phase HPLC method for the determination of promethazine in human serum was developed, validated, and applied to the pharmacokinetic study of promethazine. Promethazine and internal standard, chlorpromazine, were extracted from human serum by liquid-liquid extraction with n-hexane containing 0.8% isopropanol and analyzed on a Capcell Pak CN column with the mobile phase of acetonitrile-0.2 M potassium dihydrogen phosphate (42:58, v/v, adjusted to pH 6.0 with 1 M NaOH). Detection wavelength of 251 nm and flow rate of 0.9 mL/min were fixed for the study. The assay robustness for the changes of mobile phase pH, organic solvent content, and flow rate was confirmed by $3^{3}$ factorial design using a fixed promethazine concentration (10 ng/mL) with respect to its peak area and retention time. In addition, the ruggedness of this method was investigated at three different laboratories using same quality control (QC) samples. This method showed linear response over the concentration range of 1-40 ng/mL with correlation coefficients greater than 0.999. The lower limit of quantification using 1 mL of serum was 1 ng/mL, which was sensitive enough for pharmacokinetic studies. The overall accuracy of the quality control samples ranged from 96.15 to 105.40% for promethazine with overall precision (% C.V.) being 6.70-11.22%. The relative mean recovery of promethazine for human serum was 63.54%. Stability (freeze-thaw and short-term) studies showed that promethazine was stable during storage, or during the assay procedure in human serum. However, the storage at $-80^{\circ}C$ for 4 weeks showed that promethazine was not stable. Extracted serum sample and stock solution were not allowed to stand at ambient temperature for 12 hr prior to injection. The peak area and retention time of promethazine were not significantly affected by the changes of mobile phase pH, organic solvent content, and flow rate under the conditions studied. This method showed good ruggedness (within 15% C.V.) and was successfully used for the analysis of promethazine in human serum samples for the pharmacokinetic studies of orally administered Himazin tablet (25 mg as promethazine hydrochloride) at three different laboratories, demonstrating the suitability of the method.

  • The first neutral beam injector (NBI-1) has been developed for the Korea Superconducting Tokamak Advanced Research (KSTAR) tokamak. A first long pulse ion source (LPIS-1) has been installed on the NBI-1 for an auxiliary heating and current drive of KSTAR core plasmas. Performance of ion and neutral beam extractions in the LPIS-1 was investigated initially on the KSTAR NBI-1 system, prior to the neutral beam injection into the main plasmas. The ion source consists of a JAEA magnetic bucket plasma generator with multi-pole cusp fields and a set of KAERI prototype-III tetrode accelerators with circular apertures. The inner volume of plasma generator and accelerator column in the LPIS-1 is approximately 123 liters. Final design requirements for the ion source were a 120 kV/ 65 A deuterium beam and a 300 s pulse length. The extraction of ion beams was initiated by the formation of arc plasmas in the LPIS-1, called as an arc-beam extraction method. A stable ion beam extraction of LPIS-1 has been achieved up to an 100 kV/42 A for a 4 s pulse length and an 80 kV/25 A for a 14 s pulse length. Optimum beam perveance of 1.21 microperv has been found at an accelerating voltage of 80 kV. Neutralization efficiency has been measured by using a water flow calorimetry (WFC) method of calorimeter and an operation of bending magnet. The full-energy species of ion beams have been detected by using the diagnostic method of optical multichannel analyzer (OMA). An arc efficiency of the LPIS was 0.6~1.1 A/kW depending on the operating conditions of arc discharge.

In a ratio of material quantity framed-tube structural system, The level of sensitivity affecting the horizontal displacement is greater then the beam's column, In case of braced tube structural system, Braced appeared to be most sensitive in comparison of material quantity ratio in columns and beams.

Analysis of the Structural Safety of a Wind-Protecting Wall Using ANSYS/CFX (ANSYS와 CFX를 이용한 방풍벽의 구조 안전성 분석)

  • This study was carried out to evaluate the structural safety fur both the attached wind-protecting wall in greenhouse and the detached one installed outside. Regarding the attached wind-protecting wall in greenhouse, the analysis was conducted by doing a fluid-structure coupled field analysis using both CFX-5.7 and ANSYS 8.1 and also under the design condition of an instantaneous maximum wind velocity of $30.9m{\cdot}s^{-1}$. Three kinds of the width ranged from 30 to 90cm were considered in this study. With regard to the detached wind-protecting wall, the structural saffty was analyzed under the pressure difference of 1,117 Pa which corresponded to a wind velocity of $50m{\cdot}s^{-1}$ and the analytical results were also compared with theoretical ones. The result showed that there was little difference in the distribution of velocity overall and total pressure on the lateral side according to the width of the attached wind-protecting wall, but greenhouse with wind-protecting widths of 30 to 60cm has been reinforced to the extent of about 11% when compared with the case of being without the wall. The result also showed that the detached wind-protecting wall with a main-column interval of 3m was not stable so that it was necessary for the detached wind-protecting wall to be adequately reinforced to secure structural stability. Finally, there was great difference between analytical results and theoretical studies. The difference meant that there was some possibility of including errors when a theoretical study was done in three dimensional structure.

Study of the Separation and Elution Behavior of Phenols as Priority Pollutants in Reversed Phase Liquid Chromatography (역상 액체 크로마토그래피에서 유기오염물질로서의 페놀류들의 분리 및 용리거동에 관한 연구)

  • The optimum condition for the separation of priority pollutant phenols using isocratic elution has been determined. The elution behavior of eleven phenols has been also studied to interpret the retention. The reversed phase liquid chromatographic methods were performed on a ${\mu}$-Bondapak $C_{18}$ column with methanol-water, acetonitrile-water, and THF water mixtures as mobile phases. The COF method, where Snyder's solvent triangle concept was combined with a mixture-design statistical technique, was used to optimize the strength and selectivity of solvents for the separation of phenols. The optimum solvent composition, which gives a complete separation of eleven phenols, was found to be $MeOH:ACN:H_2O$ = 7:40:53. The plots of ln k' vs. -${\Delta}H^{\circ}$ and ${\Sigma}{\pi}$ of phenols showed relatively good linearities. Effect of van der Waals volume, pi-energy and hydrogen bonding on the retention of phenols were investigated. The following equation with the correlation coefficient of 0.9927 for ACN-water solvent system was obtained; $log^{k'}=2.515{\times}10^{-2}VWV-1.301{\times}10^{-1}E-3.674{\times}10^{-1}$

  • The Direct Contact heat Exchanger(DCHX) has been widely studied in the chemical industry for many years due to its inherent simplicity as a counter-current divice for heat and mass transfer. In many solar systems, the DCHX unit can be combined with the thermal storage unit, or alternatively, it can be used separately from the storage unit, much like an external(to storage) closed heat exchanger system. In the present work, the spray column type of direct contact heat exchangers are studied extensively to harness the solar energy for hot water and spaced heating. Some of the major considerations that are involved in the design of heat exchangers in this study are that : working fluid is a hydrocaabon(such as Texaterm) or water which is either lighter or heavier than storage medium. The experimental data have revealed some interesting characteristics concerning the application of DCHXs for solar energy utilization. These experiments are carried out in the line of solar heating system, major results are as follows : 1) the flow and aspect of working fluid drop for maxium heat transfer 2) efficiency and volumetric heat transfer coefficient of D.C.H.X with a heavier working fluid are higher than those of D.C.H.X with a lighter working fluid.

Pharmacokinetic and Bioequivalence Study of Zolpidem Tartate in Healthy Volunteers

  • In this study simple and sensitive high performance liquid chromatographic method using a commercially available column, was developed and validated for the determination of zolpidem tartrate in human plasma. The developed method with suitable validation was applied to a bioequivalence study of two different kinds of zolpidem tartrate. Two different formulations containing 10 mg of zolpidem tartate (CAS : 99294-93-6) were compared in 24 healthy male volunteers in order to compare the bioavailability and prove the bioequivalence. The study was performed in an open, single dose randomized, 2-sequence, cross-over design in 24 healthy male volunteers with a one-week washout period. Blood samples for pharmacokinetic profiling were drawn at selected times during 12 h. The mean $AUC_{0-12h}$, $C_{max}$, $T_{max}$ and $T_{1/2}$ were $676.6{\pm}223.4$ $ng{\cdot}h{\cdot}mL^{-1}$, $177.4{\pm}34.2$ $ng{\cdot}mL^{-1}$, and $0.8{\pm}0.4$ and $3.5{\pm}2.1$, respectively, for the test formulations, and $640.7{\pm}186.6$ $ng{\cdot}h{\cdot}mL^{-1}$, $193.0{\pm}64.5$ $ng{\cdot}mL^{-1}$, and $0.9{\pm}0.4$ and $2.7{\pm}0.9$, respectively, for the reference formulation. Both primary target parameters $AUC_{0-12h}$ and $C_{max}$ were log-transformed and tested parametrically by analysis of variance (ANOVA). 90% confidence intervals of $AUC_{0-12h}$ and $C_{max}$ were in the range of acceptable limits of bioequivalence (80-125%). Based on these results, the two formulations of zolpidem tartate are considered to be bioequivalent.

Non-linear Time History Analysis of Piloti-Type High-rise RC Buildings (필로티형 고층 RC건물의 비선형시간이력해석)

  • Two types of piloti-type high-rise RC building structures having irregularity in the lower two stories were selected as prototypes, and nonlinear time history analysis was performed using OpenSees to verify the analysis technique and to investigate the seismic capacity of those buildings. One of the buildings studied had a symmetrical moment-resisting frame (BF), while the other had an infilled shear wall in only one of the exterior frames (ESW). A fiber model, consisting of concrete and reinforcing bar represented from the stress-strain relationship, was adapted and used to simulate the nonlinearity of members, and MVLEM (Multi Vertical Linear Element Model) was used to simulate the behavior of the wall. The analytical results simulate the behavior of piloti-type high-rise RC building structures well, including the stiffness and yield force of piloti stories, the rocking behavior of the upper structure and the variation of the axial stiffness of the column due to variation in loading condition. However, MVLEM has a limitation in simulating the abrupt increasing lateral stiffness of a wall, due to the torsional mode behavior of the building. The design force obtained from a nonlinear time history analysis was shown to be about $20{\sim}30%$ smaller than that obtained in the experiment. For this reason, further research is required to match the analytical results with real structures, in order to use nonlinear time history analysis in designing a piloti-type high-rise RC building.

Design and Implement of 50MHz 10 bits DAC based on double step Thermometer Code (50MHz 2단 온도계 디코더 방식을 사용한 10 bit DAC 설계)

  • This paper reports the test results of a 50MHz/s 10 bits DAC developed with $0.18{\mu}m$ CMOS process for the wireless sensor network application. The 10bits DAC, not likely a typical segmented type, has been designed as a current driving type with double step thermometer decoding architecture in which 10bits are divided into 6bits of MSB and 4bits of LSB. MSB 6bits are converted into 3 bits row thermal codes and 3 bits column thermal codes to control high current cells, and LSB 4 bits are also converted into thermal codes to control the lower current cells. The high and the lower current cells use the same cell size while a bias circuit has been designed to make the amount of lower unit current become 1/16 of high unit current. All thermal codes are synchronized with output latches to prevent glitches on the output signals. The test results show that the DAC consumes 4.3mA DC current with 3.3V DC supply for 2.2Vpp output at 50MHz clock. The linearity characteristics of DAC are the maximum SFDR of 62.02dB, maximum DNL of 0.37 LSB, and maximum INL of 0.67 LSB.

Pullout Test of Reinforcement with End Mechanical Anchoring Device (단부 기계적 정착장치를 갖는 철근의 뽑힘강도)

  • The development of reinforcing steel is required in reinforced concrete structures. The standard hooks that have been widely used for the tensile development in the beam-column joints tend to create difficulties of construction such as steel congestion as the member cross sections are becoming smaller due to the use of higher strength concrete and higher grade steel. Using the reinforcing bars with end mechanical anchoring device (headed reinforcement) provides potential economies in construction such as reduction in development lengths, simplified details, and improved responses to cyclic loadings. In this paper, the pullout strengths and behaviors of the headed reinforcement were experimentally studied. In 33 pullout tests performed using D25 deformed reinforcing bars, the test parameters were embedment depth, edge distance, head size, and the use of transverse reinforcement. The pullout strengths determined from tests closely agreed with the pullout strengths predicted using the CCD method. The pullout strengths increased with increasing embedment depths nd edge distances. The strengths tend to increase with the use of larger heads. From the experimental program where the effect of the transverse reinforcement was examined, a modification factor to the CCD was suggested to represent the effect of such reinforcement that is installed across the concrete failure plane on the pullout strengths.
