Antimicrobial Effect of Viscum album var. coloratum Extracts on Food-Borne Pathogens (식중독 유발 세균의 증식에 미치는 겨우살이 추출물의 영향)

  • This study was performed to investigate the antimicrobial effect of the Viscum album var. coloratum extracts against food-borne pathogens. First, the Viscum album var. coloratum was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol, respectively. The antimicrobial activity of the Viscum album var. coloratum extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Viscum album var. coloratum showed the highest antimicrobial activity against Bacillus cereus and Shigella dysenteriae. Synergistic effect in inhibition was observed when Viscum album var. coloratum extract was mixed with Perillae folium extract as compared with each extract alone. Finally, the growth inhibition curves were determined by using petroleum ether extracts of Viscum album var. coloratum against Bacillus cereus and Shigella dysenteriae. The petroleum ether extract of Viscum album var. coloratum had strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. At this concentration, the growth of Bacillus cereus was retarded more than 24 hours and up to 12 hours for Shigella dysenteriae. In conclusion, the petroleum ether extracts of Viscum album var. coloratum inhibit efficiently Bacillus cereus and Shigella dysenteriae.

  • Antioxidant, α-glucosidase inhibition, and anticoagulant effects of 80% ethanolic extract from mistletoe (Viscum album var. coloratum) were investigated. The yield and polyphenol content of the mistletoe extract were 30.9±0.4% and 57.6±1.5 mg gallic acid equivalents/g, respectively. The antioxidant effects of the mistletoe extract such as free and cationic radical scavenging ability, nitrite scavenging ability, and reducing power increased in proportion to its concentration. Also the mistletoe extract inhibited the activity of α-glucosidase, and delayed plasma coagulation mainly by inhibiting the extrinsic and common pathways in blood coagulation system.

  • Lectin from mistletoe(Viscum coloratum) fermented by Lactobacillus plantarum for 1,2,3 days were obtained by salt fractionation, gel filtration, anion exchange chromatography and SDS-PAGE, and compared with the lectin from unfermented mistletoe. The new lectin of molecular weight of about 18,500D from fermented mistletoe was identified.

  • An antioxidant was isolated from Korean mistletoe (Viscum album var. coloratum) by consecutive purification using silica gel, Sephadex LH-20, and RP-HPLC. The active principle was identified as homo-flavoyadorinin-B (3',7-dimethoxyluteolin-4'-O-[apiosyl $(1{\rightarrow}2)$ glucoside]) by spectral analyses. It inhibited 74.6% of hydroxyl radical and 30.6% of superoxide anion radical at 0.01 mM; however, th~compound did not show any scavenging activity against hydrogen peroxide radical. At 0.1 mM, above compound scavenged superoxide anion radical about twice as effective as positive controls, BHT and ${\alpha}-tocopherol$. Radical scavenging activities of homo-flavoyadorinin-B on DPPH, hydroxyl, and hydrogen peroxide radicals were almost same with those of positive controls.

  • The mistletoe lectins are major active components in the extract of European mistletoe (Viscum album L) that have been widely used in adjuvant chemotherapy of cancer. This study was performed to investigate the antitumor activity of extract of Korean mistletoe (Viscum album var. coloratum) modified with Korean mistletoe lectin (Viscum album var. coloratum agglutinin, VCA). Compared with the results of VCA, survival rate was increased and experimental lung metastasis was reduced by treatment of modified extract (VCM). In addition, the treatment of VCM reduced angiogenesis and VCA-induced toxicity measured by a CAM assay. And VCM inhibited proliferation and induced apoptosis in vitro in tumor cells originated from tissues which are possible to apply topically without surgery. Taken together, the antitumor activities of VCM-treated group outperformed the activities of the VCA-treated group.

  • References in the modern Korean literature in this area to the Korean medicinal plant name written in Chinese script and expressed as Sanggisaeng have been extremely confusing. This name has been considered as six species, specifically Loranthus parasiticus, Loranthus tanakae, Taxillus yadoriki, Taxillus chinensis, Korthalsella japonica, and Viscum album var. coloratum. Recently, it was suggested that sanggisaeng is not distributed in Korea; however, the plant name Sanggisaeng recorded in old Korean literature sources, such as in the Hyang-yak-chae-chwi-wol-lyeong, Hyang-yak-jib-seong-bang, and Dong-ui-bo-gam sources, among others, was identified using the name V. album var. coloratum, whereas in China, Sanggisaeng was referred to as T. sutchuenensis. The Hangeul name of Sanggisaeng had been Ppongnamugyeousali, but now the name Gyeousali is widely used.

Antifungal Activities of meOH Extracts from Three Korean Mistletose against Tyromyces palustris, Endothia nitschkei and Trichophyton rubrum (3종의 한국산 겨우살이 메탄올 추출물의 Tyromyces palustris, Endothia nitschkei 그리고 Trichophyton rubrum에 대한 항균활성)

  • The traditional Korea medical book already recorded various biological activities of the Korean mistletoes. The objective of this study was examine antifungal activities of MeOH extract from the Korean mistletoe through column chromatography on three fungi, such as Tyromyces palustris Endothia nitschkei and Trichophyton rubrum. No mistletoes had anti-fungal activity against T. palusties. Extracts of V. album var. coloratum showed the highest hyphal growth-inhibitory activity against E. nitschkei and leaf extract of this species had higher activity than twig extract. Further fractionation of most active fraction and following antifungal assay showed that its anti-fungal activity might be caused by synergism if its components. It was suggested that Viscum album var. coloratum shows significantly antifungal activities against E. nitschkei and T. rubrum. Further examination is needed to find out more exact active compounds.

  • In this study, we investigated whether Korean mistletoe (Viscum album L. var. coloratum) has anti-inflammatory effects that play key roles in the regulation of pathological mechanism of atopic dermatitis (AD). Four kinds of fractions, hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC), were used and RAW264.7 mouse macrophages and RBL-2H3 rat basophils were used to measure various inflammatory markers. EA significantly decreased mRNA expression and protein secretion levels of tumor necrosis factor alpha (TNF-α), interleukin (IL)-6, and IL-4 but HX did not affect these markers. In addition, BU decreased mRNA expressions of IL-4 and IL-6 whereas MC decreased IL-6 and TNF-α mRNA expressions. As a result, Korean mistletoe can show anti-inflammatory effects by inhibiting the secretion of cytokines related to AD, so it is thought that it will be possible to develop functional cosmetics related to this.

Isolation and Characterization of Lectin from Viscum coloratum (겨우사리 중의 렉틴성분 분리 및 특성)

  • Lectin from mistletoe (Viscum coloratum) were obtained by salt fractionation, gel filtration and anion exchange chromatography. A molecular weight of about 60,000D has been determined by SDS-PAGE and two basic subunits which have molecular weights of 33,000D and 28,000D are linked by a disulfide bond. The partially purified mistletoe lectins agglutinated human B erythrocytes. Agglutinating activity was relatively stable at varied pH $(3.77{\sim}8.71)$ and at temperature range of $0{\sim}40^{\circ}C$ and not affected by 9 metal ions. Galactose, lactose and N-acetyl-D- galactosamine inhibited agglutinating activity of lectin. Lectin from mistletoe was more mitogenic to murine lymphocytes than concanavalin A.

  • The study was carried out to evaluate the preliminary toxicity and general pharmacology of KML-IIU, a purified pectin from Korean Mistletoe (Viscum album coloratum). KML-IIU was administered intravenously to ICR mice and Spargue-Dawley rats to investigate the acute toxicity. LD50 values in mice and rats were above 30 $\mu$g/kg. KML-IIU had no effects on the general behaviors, acetic acid induced writhing syndrome, pentobarbial induced sleeping time, pentylenetetrazole induced convulsion and the change of body temperature. In addition, KML-IIU did not show any effects on digestive system and blood coagulation system.
