Acacia ferruginea Inhibits Tumor Progression by Regulating Inflammatory Mediators-(TNF-α, iNOS, COX-2, IL-1β, IL-6, IFN-γ, IL-2, GM-CSF) and Pro-Angiogenic Growth Factor-VEGF

  • The aim of the present investigation was to evaluate the effect of A ferruginea extract on Dalton's lymphoma ascites (DLA) induced tumours in BALB/c mice. Experimental animals received A ferruginea extract (10 mg/kg.b.wt) intraperitoneally for 14 consecutive days after DLA tumor challenge. Treatment with extract significantly increased the life span, total white blood cell (WBC) count and haemoglobin (Hb) content and decreased the level of serum aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), gamma glutamyl transferase (${\gamma}$-GT) and nitric oxide (NO) in DLA bearing ascites tumor models. In addition, administration of extract significantly decreased the tumour volume and body weight in a DLA bearing solid tumor model. The levels of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin-1 beta (IL-$1{\beta}$), interleukin-6 (IL-6) and granulocyte monocyte-colony stimulating factor (GM-CSF), as well as pro-angiogenic growth factors such as vascular endothelial growth factor (VEGF) and inducible nitric oxide synthase (iNOS) were elevated in solid tumour controls, but significantly reduced by A ferruginea administration. On the other hand, the extract stimulated the production of interleukin-2 (IL-2) and interferon-gamma (IFN-${\gamma}$) in animals with DLA induced solid tumours. Increase in $CD4^+$ T-cell population suggested strong immunostimulant activity for this extract. GC/MS and LC/MS analysis showed quinone, quinoline, imidazolidine, pyrrolidine, cyclopentenone, thiazole, pyrazole, catechin and coumarin derivatives as major compounds present in the A ferruginea methanolic extract. Thus, the outcome of the present study suggests that A ferruginea extract has immunomodulatory and tumor inhibitory activities and has the potential to be developed as a natural anticancer agent.

  • The protoplast formation of Rhizoctonia solani in the fast growing anastomosis groups (AGs) 1 and 4, the intermediate AG-2 and AG-5, and the slow AG-3 yielded the most, moderate and the least in that order, respectively. Sclerotia formation varied with AGs. A high yield of protoplasts from AGs was obtained with a combined lytic enzyme system containing cellulase 'Onozuka' R-10, macerozyme R-10 and ${\beta}-glucuronidase$. When 3g (fresh weight) of 30 hr old mycelia was incubated for 3 hr at $32^{\circ}C$ with the enzyme mixture in 0.6 M mannitol, maximum protoplasts were obtained in the five AGs. A protoplast fusion between sclerotia forming AG-1 inactivated with heat and non-forming AG-5 was induced by polyethylene glycol and ${Ca}^{2+}$. Seven fusants obtained were based on characteristics of colony and sclerotium formation on culture plates. The fusants were confirmed by isozyme patterns of esterase and killing reaction between AG-1 and a fusant F1501.

  • The sprig of Sojekbojungwhan(消積保中丸) has been used for curing as a traditional medicine without any experimental evidence to support the rational basis for their clinical use. This experiment was carried out to evaluate the possible therapeutic or antitumoral effects of Sojekbojungwhan extract against cancer, and to study some mechanisms responsible for its effect. The cytotoxic and antitumor effects were evaluated on human cell lines(A549, hep3B, Caki-1, Sarcoma 180) after esposure to Sojekbojungwhan extract using in ILS, colony forming efficiency and SRB assay which were regarded as a valuable method for cytotoxic and antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows. 1. As a result of exposure to Sojekbojungwhan extract, the proliferation of A549, hep3B, Caki-1, good correlations were shown from the results of SRB assay and those of clogenetic assay. 2. The oral administration of Sojekbojungwhan extact showed significant effects of increase of MST(mean survival time) and ILS(increased life span) depending on the increasing concentration. 3. Against squamous cell carcinoma induced by MCA, Sojekbojungwhan decreased not only the frequency of tumor production but also the number and weight of tumors per tumor bearing mice(TBM). Sojekbojungwhan also significantly suppressed the development of 3LL cell-implanted tumors by frequency and their size, and some developed tumors were regressed by the continuous treatment of Sojekbojungwhan extract into TBM. 4. Sojekbojungwhan extract also increased NK cell activities. According to the above results, it could be suggested that Sojekbojungwhan extract has some antitumor effects.

  • Bioenzyme was made up of two predominant strain, which were nomenclated PN-1 and PN-2, respectively. PN-1 possessed spore forming ability, motility, gram positive, catalase activity, but didn't have oxidase activity, urease activity, nitrate reduction and indole forming ability. Taxonomical characteristics of the PN-2 were similar to PN-1 strain, but colony color of PN-2 was yellow-white and nitrate reduction ability was positive. Predominant strains were ascertained by physiologically and morphologically. PN-1 and PN-2 were identified Bacillus lichenifonnis and Bacillus thuringinensis, respectively. Yield of lettuce significantly increased in first and second bioenzyme treatment. Fresh weight, length of shoot, width of leaves significantly increased in N,P,K and N,P,K followed by Bioenzyme application.

  • One major constraint in the development and implementation of a successful and cost-effective area-wide integrated pest management (AW-IPM) programme with a SIT component for Bactrocera oleae (Diptera: Tephritidae) is the ability to produce a large number of high quality mass-reared individuals. The aim of this study was to develop a more efficient and practical egg collection system in an attempt to improve the mass-rearing of this species. The following basic parameters were examined: egg production per female, egg hatch, pupal recovery, pupal weight, adult emergence and percentage of fliers. Three different strains (Israel wild-type, France wild-type, and Greece laboratory) were tested and each strain was evaluated for six generations. Female flies of the Israel strain produced significantly more eggs per female than the other two strains, but egg hatch was significantly lower. Egg hatch of the France wild type and the Greece laboratory strain was similar. For all other parameters, there was no significant difference between strains; however, there was a significant generational effect for all parameters observed. As a result of this study, a protocol was developed for the mass-rearing of this species that included the use of large adult holding cages that could house up to 96,000 flies per cage. The newly developed method of egg collection using a flat wax panel as one of the sides of an adult holding cage proved to be cost-effective, efficient, making colony growth easier for industrial mass-rearing.

Quality Characteristics of Korean Wheat Bread prepared with Substitutions of Naturally Fermented Blueberry Starters (블루베리 천연 발효액종을 첨가한 우리밀 식빵의 품질 특성)

  • In this study, a natural fermentation starter formulation was developed for manufacturing Korean bread products by substituting baker's yeast with naturally fermented blueberry starters. As the incubation time of the blueberry extracts increased, the pH and total titratable acidity increased. The sweetness (brix%) of blueberry extracts containing various amounts of sugar were higher than the other sample. The result of alcoholicity for naturally fermented blueberry extracts, the fermented blueberry extract containing 20% sugar was highest. Lactic acid bacteria counts increased until the 4th day; however, it decreased from the 5th day, and viable yeast counts increased consistently until the 5th day. The volume for naturally fermented blueberry extracts increased as the incubation time increased. As the fermentation time of blueberry starters increased, the pH of bread dough decreased. The RVA analysis conveyed that wheat flour retrogradation was retarded by increasing the blueberry starter content. The weight of pan breads containing blueberry starters were higher than that of the control, while the volume, specific volume and baking loss rate were lower than those of the control. The moisture content of pan breads containing blueberry starter decreased as storage time increased. In analyzing the visible mold colony during 7 days of storage at $28^{\circ}C$, mold growth in pan breads containing the blueberry starter was retarded. The hardness of breads containing blueberry starters were significantly increased as storage time increased. The breads containing 50% naturally fermented blueberry starter have acceptable sensory properties. In conclusion, these results indicated that 50% of natural fermentation blueberry starter could be very useful as a substitute for yeast when making naturally fermented bread.

  • The carbon dioxide, nitrogen, and phosphate removals from wastewater using microalgae have extensively been studied. Especially, the green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. These hydrocarbons suggest that the possibility of renewable biofuels to be converted into useful fuels such as gasoline by simple catalytic cracking. The poor recovery (18 - 32%) of hydrocarbon from B. braunii culture in two-phase bubble column seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. In order to overcome low recovery efficiency, two-stage extraction culture system including culture vessel and two-phase separator is now under development, resulted improving contact between solvent phase and cell suspension. Hydrocarbon recovery using this process was more than two times as that using two-phase extraction culture.

Biocontrol Potential of Fungal Endophytes against Fusarium oxysporum f. sp. cucumerinum Causing Wilt in Cucumber

  • Endophytic fungi have received much attention as plant growth promoters as well as biological control agents against many plant pathogens. In this study, 30 endophytic fungal species, isolated from various plants in China, were evaluated using in vitro dual culture assay against Fusarium oxysporum f. sp. cucumerinum, causing wilt in cucumber. The results of the present study clearly showed that all the 30 endophytic fungal isolates were highly capable of inhibiting the mycelial colony growth of Fusarium oxysporum f. sp. cucumerinum with inhibition % over 66% as compared to control treatments. Among all of them, 5 isolates were highly effective such as, Penicillium sp., Guignardia mangiferae, Hypocrea sp., Neurospora sp., Eupenicillium javanicum, and Lasiodiplodia theobromae, respectively. The Penicillium sp. and Hypocrea sp. were highly effective as compared to other isolates. From in vitro results 10 best isolates were selected for greenhouse studies. The results of the greenhouse studies showed that among all of them 3 endophytic fungal isolates successfully suppressed wilt severity when co-inoculation with pathogen Fusarium. oxysporum f. sp. cucumerinum. The endophytic fungi also enhanced plant growth parameters of the host plants, the antagonistic fungal isolates increased over all plant height, aerial fresh, and dry weight as compared to control.

Effects of dietary supplementation with Pediococcus acidilactici ZPA017 on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation

  • Objective: This study was conducted to determine the effects of dietary supplementation with Pediococcus acidilactici (P. acidilactici) ZPA017 as a probiotic on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation. Methods: A total of 94 sows (Large White×Yorkshire, average 4.50 parities) were randomly allotted to two dietary treatments: control diet and the diet supplemented with P. acidilactici ZPA017 (2.40×109 colony-forming unit/kg of diets). The study started at d 90 of gestation and conducted until d 28 of lactation. Results: Compared to sows fed the control diet, supplementation of P. acidilactici ZPA017 increased the number of weaning piglets, weaning weight of litter and piglets, survival rate of piglets at weaning (p<0.05), and decreased diarrhea rate of piglets in lactation (p<0.05). Dietary P. acidilactici ZPA017 increased fecal Lactobacillus populations (p = 0.030) and reduced fecal Escherichia coli and Staphylococcus aureus populations (p<0.05) of sows at weaning. Moreover, the supplementation of P. acidilactici ZPA017 increased serum concentrations of immunoglobulin G, immunoglobulin A and total protein (p<0.05), while decreased serum haptoglobin concentration and alanine aminotransferase activity (p<0.05) of sows at weaning. Conclusion: Administration of P. acidilactici ZPA017 in diets during late gestation and lactation had positive effects on the reproductive performance, intestinal microflora balance and immunity of sows.

In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. To improve recovery efficiency, mixed-solvent of extractive solvent (octane) and biocompatible solvent (octane) was tested in two-phase column for in situ extraction. In two-phase extraction culture using mixed-solvent, the algal growth was intensely inhibited even at low concentration of polar octanol solvent. the hydrocarbon recovery in two-stage cell-recycle extraction showed a 2.9 fold increase (57%) over that in two-phase extraction. Up to 60 % of hydrocarbon could be recovered without serious cell-damage in the case of downstream separation for 6 h at the high recycle flow rate using this process after batch culture.
