Biodistribution of a Promising Probiotic, Bifidobacterium longum subsp. longum Strain BBMN68, in the Rat Gut

  • Bifidobacterium longum subsp. longum BBMN68, isolated from centenarians in Guangxi, China, has been proved to be a promising probiotic strain for its health benefits. In this study, the biodistribution of this strain in the rat gut was first investigated using the quantitative realtime PCR assay and propidium monoazide. Strain-specific primers were originally designed based on the BBMN68 genome sequence. Healthy rats were orally inoculated with either a single dose of BBMN68 (1010 colony-forming units/kg), or with one dose per day for 7 days and bacterial concentrations were analyzed in detail from the intestinal contents and feces of four different gut locations, including stomach, small intestine, colon, and rectum. Results indicated that strain BBMN68 could overcome the rigors of passage through the upper gastrointestinal tract and transiently accumulate in the colon, even though survival in the stomach and small intestine was not high. A good level of BBMN8 could stay in vivo for 72 h following a 7-day oral administration, and a daily administration is suggested for a considerable and continuous population of BBMN68 to be maintained in the host intestine.

  • 본 연구의 목적은 어린이들이 섭취하는 비타민 제제들의 우식원성을 치아우식 유발지수와 Streptococcus mutans의 활성정도 및 산생성능을 통해 분석하는 것이다. 4가지 비타민, 노마(NM), 세노비스 키즈(CK), 애니멀 퍼레이드(AP), 캐릭터 비타민(CV)를 대상으로 진행하였다. 치아우식 유발지수를 산출한 결과 AP, CV, CK, NM 순으로 작아졌다. pH 측정 시 모든 실험군들의 초기 값은 산성을 나타내었다. S. mutans의 군집 형성 단위를 분석한 결과 NM, CV은 대조군에 비해 더 높은 증식률을 보였고(p < 0.05), CK와 AP (p < 0.05)는 대조군에 비해 더 낮은 증식률을 보였다. 공초점 현미경으로 관찰하였을 때 실험군들은 대조군에 비해 높은 세균 활성도를 보였다. 비타민 제제들의 세균 활성도와 산도를 고려해 볼 때, 어린이의 구강 건강을 위해서는 세심하게 고려하여 섭취해야 할 것이다.

Candidacidal Activity of Xylitol and Sorbitol

  • Purpose: It has been reported that xylitol and sorbitol affect antifungal activities by enhancing antimicrobial activities of other substances. The purpose of this study was to investigate the direct candidacidal activities of xylitol and sorbitol at a wide range of concentration. Methods: Xylitol and sorbitol solubilized with simulated salivary buffer at a range of $0.8{\mu}M$ to 1.05 M were used. Candida albicans strains, ATCC strains 10231, 11006, and 18804 were used for the candidacidal assay. The candidacidal activities of xylitol and sorbitol were determined by comparing the numbers of colony forming units between in the presence and absence of xylitol or sorbitol and calculating the percent loss of cell viability. Results: There were some differences in the candidacidal activities according to the types of sugar alcohols and C. albicans strains. The candidacidal activity of more than 10% was observed when a final concentration of 32.9 mM in xylitol or sorbitol was maintained and that of about 20% was observed when a final concentration of 131 mM was maintained. Even at a high concentration of 1.05 M, the candidacidal activity of xylitol or sorbitol was about 20%. Conclusions: Xylitol and sorbitol at the concentrations used in commercial oral health care products had some levels of candidacidal activities.

  • Frost injury of crops is closely related to the epiphytic population dynamics of ice nucleation-active (INA) bacteria, and the injury can be reduced by decreasing the INA bacterial population. In order to predict the epiphytic population of INA bacteria on crops, a rapid and accurate detection method has to be developed. In the previous report, we produced some antibodies against INA proteins purified from the outer membrane of INA bacteria. However it was difficult to produce the antibodies because the purification procedures of the INA proteins were complicated, and the final yield was too low. We designed a specific peptide from the N-terminal region of INA protein by computer analysis and synthesized the peptide in vitro in this experiment. The peptide sequence was Asp-Ser-Por-Leu-Ser-Leu-His-Ala-Asp, that is corresponding to the highly conserved region in several INA proteins, with predicted beta turn, coiling, and hydrophilic region. A polyclonal anti-INA peptide antiserum produced specifically recognized INA bacteria as few as 10 colony-forming units (CFU) in the ELISA reactions and did not respond to other non-INA bacteria. Serological specificity of the anti-INA peptide antiserum will facilitate the forecasting of the INA bacterial population dynamics on crops.

Control of Crisphead Lettuce Damping-off and Bottom Rot by Seed Coating with Alginate and Pseudomonas aeruginosa LY-11

  • Seedling damping-off and bottom rot caused by Rhizoctonia solani are yield limiting diseases of crisphead lettuce. To provide biocontrol measure in the management of the diseases, biocontrol strain Pseudomonas aeruginosa LY-11 was isolated from lettuce rhizosphere and introduced into crisphead lettuce rhizosphere by the seed coating delivery method. Alginate was used as a coating material to generate beads containing $10^6-10^{6.5}$ colony-forming units (CFUs) of viable bacterial cells of LY-11. When seeds germinated from the alginate beads containing the strain LY-11, the bacteria established mostly in plant rhizosphere to maintain at least $10^4$ CFU per gram of plant tissues. Crisphead lettuce seedlings germinated from the entrapped seeds were less affected from damping-off and bottom rot with disease control values of 70.4% and 85.4% respectively. Although P. aeruginosa LY-11 colonized plant rhizosphere and not phyllosphere, the result indicated that bottom rot caused by the foliar inoculation of R. solani was effectively reduced by the rhizobacteria. All data suggested that immobilized rhizobacterial application in seeds by alginate coating could control damping-off and induce induced systemic resistance of crisphead lettuce to reduce bottom rot.

Sensitization of Periodontitis Disease Causing Bacteria by Low Power He-Ne Laser Radiation

  • The present investigation is an attempt to create an optimized protocol for a bactericidal modality of different powers of He-Ne laser radiation to eliminate periodontitis disease causing bacteria from dental plaques. Periodontitis is most prevalent infectious disease of men and caused by a limited number of Gram negative oral bacteria. Porphyromonas gingivalis and Streptococcus sanguis are the important bacteria responsible for periodontitis diseases. Effect on periodontitis disease causing bacteria were produced by the exposure of different powers of He-Ne laser light i.e. 9 mW, 17 mW and 26 mW of red colour of wavelength 632.8 nm in two different periods of time i.e. 10 min. and 20 min. in the presence of dye Methylene blue (MB) used as a photosensitizer. The results have been shown in terms of percentage inhibition of colony forming units (cfu.) of bacteria. This study has shown that maximum inhibition of cfu. were observed in Laser+MB-20 min. exposure time. This inhibition was followed by Laser+MB-10 min., but minimum inhibition was seen in Laser only at 10 min. exposure. In case of effect of methylene alone on the cfu. of bacteria, it was seen that MB have not shown more inhibition of cfu. and it had shown that the no. of cfu. are very similar to that of control. The above observation of the present study was seen in case of every 3 different type of used powers of laser for both the bacteria. Maximum percentage inhibition of cfu. were seen in case of 26mW powers of He-Ne laser, which was 67. 28% to 61.42% for Porphyromonas gingivalis and Streptococcus sanguis respectively. So, increasing the power of laser (safe range for dentistry is 3-30 mW) under conditions shows an increased percentage inhibition of cfu. Thus the present investigation may be a useful adjunct with mechanical debridement in the prevention of recolonization of subgingival lesions by pathogenic microorganisms which are harmful and drug resistant.

식품제조공장 내 공중부유미생물 오염도와 오염진균동정 (Identification and Concentration of Airborne Microbes in Food Manufacturing Plants)

  • To evaluate the indoor air quality of food manufacturing plants, the presence of viable bacteria and fungi was assessed in the indoor air of the facilities at which 9 food items were manufactured. Air samples were collected from the general zone, low clean zone and clean zone of each factory with an air sampler, in combination with plate counts agar using for bacteria, and dichloran-glycerol agar for fungi. The samples were incubated at $25^{\circ}C$ for 4 to 7 days. After culture, the colony forming units (CFU) on each plate were counted and corrected with a positive hole conversion table. The average concentration of bacteria was $2.2{\times}10^3\;CFU/m^3$ in the general zone, $1.2{\times}10^3\;CFU/m^3$ in the low clean zone and $7.3{\times}10^2\;CFU/m^3$ in the clean zone. The average concentration of fungal microbes was $2.5{\times}10^3\;CFU/m^3$ in the general zone, $2.6{\times}10^3\;CFU/m^3$ in the low clean zone, and $2.0{\times}10^2\;CFU/m^3$ in the clean zone. No meaningful differences were detected between the general zone and the low clean zone, but the clean zone had significantly lower concentrations than the other zones. Additionally, the identification of the fungi was performed according to morphological method using a giant culture and slide culture. The fungi were identified as belonging to 18 genera, and the genera Cladosporium(33%), Penicillium(29%) and Aspergillus(26%), predominated. Aspergillus isolates were identified to species level, and A. ochraceus, a mycotoxigenic species, was identified. As part of the effort to control the quality of the indoor air of food manufacturing plants, our results show that continued studies are clearly warranted.

Production of Ready-to-Reconstitute Functional Beverages by Utilizing Whey Protein Hydrolysates and Probiotics

  • This investigation was aimed at developing a ready-to-reconstitute beverage by utilizing probiotics and whey protein hydrolysates carrying bioactive peptides. Cheddar cheese whey was ultrafiltered. The 18% protein retentate was subjected to protein hydrolysis using Neutrase. The hydrolyzed retentate was further condensed to 35% total solids and spray-dried at $75^{\circ}C$ outlet air temperature. Different levels of sugar, citric acid and stabilizer were blended for spray-dried hydrolysates. Spray-dried hydrolysate was further inoculated with different levels of probiotics grown in a whey medium and dried in fluidized-bed drier at $40^{\circ}C$ to obtain a ready-to-reconstitute beverage. Hydrolysis was greatest at an enzyme:substrate ratio of 1:25 for 3 h. Spray-dried hydrolysate reconstituted to 1% protein and blended with 15% sugar, 0.2% citric acid and 0.15% xantham gum resulted in a superior product with no sedimentation. Accordingly, sugar, citric acid and xanthum gum were dry-blended with spray-dried hydrolysates. Bifidobacterium bifidum and Lactobacillus acidophilus that was grown separately in a whey medium, blended to produce 2% spray-dried hydrolysate and dried as described above resulted in a readyto-reconstitute beverage mix. The fluidized dried product typically exhibited a probiotic count of $10^8$colony forming units (CFU)/g. However, blending of probiotic to the retentate and direct spray-drying precipitously reduced the probiotic count to $10^4$ CFU/g of powder.

Assessment of indoor air micro-flora in selected schools

  • Quantification of viable forms of microbial community (bacteria and fungi) using culture-dependent methods was done in order to characterize the indoor air quality (IAQ). Role of those factors, which may influence the concentration of viable counts of bacteria and fungi, like ventilation, occupancy, outdoor concentration and environmental parameters (temperature and relative humidity) were also determined. Volumetric-infiltration sampling technique was employed to collect air samples both inside and outside the schools. As regard of measurements of airborne viable culturable microflora of schools during one academic year, the level of TVMCs in school buildings was ranged between 803-5368 cfu/$m^3$. Viable counts of bacteria (VBCs) were constituted 63.7% of the mean total viable microbial counts where as viable counts of fungi (VFCs) formed 36.3% of the total. Mean a total viable microbial count (TVMCs) in three schools was 2491 cfu/$m^3$. Outdoor level of TVMCs was varied from 736-5855 cfu/$m^3$. Maximum and minimum VBCs were 3678-286 cfu/m3 respectively. Culturable fungal counts were ranged from 268-2089 cfu/$m^3$ in three schools. Significant positive correlation (p < 0.01) was indicated that indoor concentration of viable community reliant upon outdoor concentration. Temperature seemed to have a large effect (p < 0.05, p < 0.01) on the concentration of viable culturable microbial community rather than relative humidity. Consistent with the analysis and findings, the concentration of viable cultural counts of bacteria and fungi found indoors, were of several orders of magnitude, depending upon the potential of local, spatial and temporal factors, IO ratio appeared as a crucial indicator to identify the source of microbial contaminants.

Rapid Screening of Salmonella spp. Using PBM BioSignTM Salmonella Test and Evaluation of the PBMS Test

  • The PBM ${BioSign}^{TM}$ Salmonella (PBMS) test kit based on an mmunochromatographic method was evaluated for the screening of Salmonella spp. in pure cultures, and 80, 15, and 10 artificially and naturally contaminated, and negative controlled food samples, respectively. The PBMS test involves presumptive qualitative procedures, detecting the presence of Salmonella spp. in foods within 26 h total testing period and allowing the user to release negative products 70 h earlier than the conventional methods. The PBMS test using Buffered Peptone Water and Rappaport-Vassiliadis broth was evaluated for 10 different food types for various Salmonella spp. It showed detection limits of 1 to 25 colony forming units (CFU)/25 g. No cross-reaction was observed, particularly to other gramnegative bacteria. These results indicate the PBMS test is a rapid and inexpensive procedure for the screening of Salmonella spp. present at low concentrations (1 to 25 CFU/25 g) in foods.
