향나무 추출물의 Galectin-3 저해에 의한 암 전이 억제 효과 (Anti-Metastatic Activity of Juniperus chinensis Extract by Galectin-3 Inhibition)

  • 향나무 에탄올 추출물에 대한 암 전이 억제 효과를 측정하기 위하여 세포 수준에서 암 전이에 밀접히 관련되어 있는 galectin-3에 대한 억제 효과를 colon 26-M3.1 lung carcinoma에서 측정하였다. 향나무 추출물 $5{\mu}g/m{\ell}$, $50{\mu}g/m{\ell}$$500{\mu}g/m{\ell}$ 처리 시, 농도 의존적으로 galectin-3에 대한 저해 효과를 나타내었다. 한편, 암 전이 관련 있는 또 다른 단백질로 MMP 효소에 대한 직접적인 억제효과를 측정한 결과, 향나무 추출물 $5{\mu}g/m{\ell}$, $50{\mu}g/m{\ell}$$500{\mu}g/m{\ell}$ 처리 시 직접적으로 MMP-1, MMP-2 및 MMP-9 효소에 대해서 농도의존적인 저해 효과는 나타났으나, 강한 활성으로 평가되지는 못하였다. Colon 26-M3.1 lung carcinoma을 이용한 in vivo mouse 모델에서 향나무 추출은 농도 의존적으로 전이 억제 효과를 나타내었고, MMP-2 활성화 물질인 relaxin과 병용투여 시, relaxin의 기능에 영향을 받지 않는 것으로 나타났다. 따라서 in vivo mouse 모델에서 나타난 암 전이 억제 효과는 galectin-3 저해로 인한 효과로 사료된다.

보중익기합대칠기탕과 Doxorubicin의 병용이 Colon-26의 항암효과에 미치는 영향 (The Anti-tumor Effect of Bojungikkeehapdaechilkitang with Doxorubicin in Colon-26)

  • Objectives : To evaluate the anti-tumor and synergic effect of Bojungikkeehapdaechilki-tang (BJDC) with doxorubicin. Methods : The inhibitory concentration (IC), $IC_{50}{\;}and{\;}IC_{90}$ of single use of doxorubicin and BIDC with their concomitant treatment against Colon-26 (murine rectum carcinoma) cell line were observed using MTT (microculture tetrazolium test) assay. In addition, their anti-tumor effects were also observed in xenograft nude mice models against Colon-26 cell line. Results : BJDC had only minimal direct anti-tumor effect against Colon-26 cell line but it reduced general depressed signs induced by implantation of the tumor cell lines and increased the total WBC and lymphocyte numbers. Conclusions : It is considered or expected that BJDC extract is reducing the critical toxicity of doxorubicin and has favorable synergic anti-tumor effect when administered conconitently with doxorubicin.

대장암에서 17, 18번 염색체의 이형접합성 소실 (Loss of Heterozygosity (LOH) on 17th and 18th Chromosome from Colorectal Carcinoma)

  • Colorectal carcinoma is occurred frequently to Korean and so ranked the fourth from various cancers. Due to western dietary life, this cancer has been increased continually. Therefore, the study will be needed to find a candidate gene involved in the development and progression of colorectal carcinoma and to diagnose and treatment helpfully. The striking feature from cancer suppressor genes is known for LOH (loss of heterozygosity), which is the method to find allele genetic loss or mutation of cancer cell. The purpose of this study was designed to find a carcinogenic gene from colon cancer using microsatellite marker on 17th and 18th chromosome from 30 subjects. The LOH was investigated in order of D18S59 57% (17/30), TP53CA 50% (15/30), D18S68 47% (14/30), D18S69 43% (13/30). The genetic mutation depends on loci of colorectal carcinoma was shown higher with 2.44 from colon cancer than with 1.25 from right colorectal carcinoma (p<0.032). The genetic mutation with lymph nodes was investigated higher with 2.69 at mutated group than with 1.14 at non-mutated group (p<0.003). At genetic mutated pattern depends on disease stage, there was higher significant difference at III-IV stage 2.50 than that of I-II stage 1.17, respectively (p=0.015). There was no difference at comparison between histological classification and serological CEA increase. The loss on 18q21 found in this study is highly recurrence loci and was observed 43% for Korean with high recurrence. Therefore, LOH is a very useful tool to detect 18q21 loci in clinical application, prior to the treatment of colorectal carcinoma. After the operation of colorectol carcinoma, the efficient application using LOH at operated part tissue which is designed to protect the recurrence as well as its cure will be needed.

Zinc and Zinc Related Enzymes in Precancerous and Cancerous Tissue in the Colon of Dimethyl Hydrazine Treated Rats

  • Trace element zinc deficiency or excess is implicated in the development or progression of some cancers. The exact role of zinc in the etiology of colon cancer is unclear. To cast light on this question, an experimental model of colon carcinogenesis was applied here. Six week old rats were given sub cutaneous injections of DMH (30 mg/kg body weight) twice a week for three months and sacrificed after 4 months (precancer model) and 6 months (cancer model). Plasma zinc levels showed a significant decrease (p<0.05) at 4 months and a greater significant decrease at 6 months (p<0.01) as compared with controls. In the large intestine there was a significant decrease in tissue zinc levels (p<0.005) and in CuZnSOD, and alkaline phosphatase activity (p<0.05) in the pre-cancerous model and a greater significant decrease in tissue zinc (p<0.0001), and in CuZnSOD and alkaline phosphatase activity (p<0.001), in the carcinoma model. The tissue zinc levels showed a significant decrease in the small intestine and stomach (p<0.005) and in liver (p<0.05) in the cancer model. 87% of the rats in the precancer group and 92% rats in the cancer group showed histological evidence of precancerous lesions and carcinomas respectively in the colon mucosa. This study suggests that the decrease in plasma zinc, tissue zinc and activity of zinc related enzymes are associated with the development of preneoplastic lesions and these biochemical parameters further decrease with progression to carcinoma in the colon.

Effects of Parafibromin Expression on the Phenotypes and Relevant Mechanisms in the DLD-1 Colon Carcinoma Cell Line

  • Background: Parafibromin is a protein encoded by the HRPT2 (hyperparathyroidism 2) oncosuppressor gene and its down-regulated expression is involved in pathogenesis of parathyroid, breast, gastric and colorectal carcinomas. This study aimed to clarify the effects of parafibromin expression on the phenotypes and relevant mechanisms of DLD-1 colon carcinoma cells. Methods: DLD-1 cells transfected with a parafibromin-expressing plasmid were subjected to examination of phenotype, including proliferation, differentiation, apoptosis, migration and invasion. Phenotype-related proteins were measured by Western blot. Parafibromin and ki-67 expression was detected by immunohistochemistry on tissue microarrays. Results: The transfectants showed higher proliferation by CCK-8, better differentiation by electron microscopy and ALP activity and more apoptotic resistance to cisplatin by DNA fragmentation than controls. There was no difference in early apoptosis by annexin V, capase-3 activity, migration and invasion between DLD-1 cells and their transfectants. Ectopic parafibromin expression resulted in down-regulated expression of smad4, MEKK, GRP94, GRP78, $GSK3{\beta}$-ser9, and Caspase-9. However, no difference was detectable in caspase-12 and -8 expression. A positive relationship was noted between parafibromin and ki-67 expression in colorectal carcinoma. Conclusions: Parafibromin overexpression could promote cell proliferation, apoptotic resistance, and differentiation of DLD-1 cells.

Enhanced proliferation of SNU-407 human colon cancer cells by muscarinic acetylcholine receptors

  • We investigated the expression of muscarinic acetylcholine receptors (mAChRs) and their possible involvement in the regulation of cell proliferation in four colon cancer cell lines (SNU-61, SNU-81, SNU-407, and SNU-1033) derived from Korean colon carcinoma patients. A ligand binding assay showed that all four cell lines expressed mAChRs. Treatment of the four cell lines with the cholinergic agonist carbachol led to the activation of extracellular signal-regulated kinases 1 and 2 (ERK1/2). In SNU-407 cells, carbachol significantly stimulated cell proliferation, which could be abolished by the muscarinic antagonist atropine and the ERK1/2 kinase inhibitor PD98059. These results indicate that mAChRs specifically mediate the proliferation of SNU-407 colon cancer cells via the ERK1/2 pathway.

대장암에서 7, 20번 염색체의 Amplification (Amplification on 7th and 20th Chromosome from Colorectal Carcinoma)

  • Colorectal carcinoma from various cancers is fourth ranked occurred to Korean. Due to western dietary life, this cancer has been increased continuously. Therefore, the further study will be needed to find a candidate gene involved in the development and progression of colorectal carcinoma as well as to diagnose and treatment helpfully. The purpose of this study was designed to find a carcinogenesis gene using microsatellite marker on chromosomes 7th and 20th from 30 colon cancer patients. The amplification was investigated in order of D20S97 57% (17/30), D20S101 57% (17/30), D20S119 53% (16/30), D7S483 50% (15/30), D7S495 47% (14/30), D7S498 47% (14/30). The genetic mutation pattern depends on loci of colorectal carcinoma was shown highly amplified with 3.77 from colon cancer than with 2.08 from right colorectal carcinoma (P<0.018). The genetic mutation with lymph nodes was investigated higher with 4.13 at metastasized group than with 1.93 at non-metastasized group (P<0.001). There was no difference at comparison between histological classfication and serological CEA increase as well as on genetic mutated pattern depends on disease stage. It is suggested that the amplification on chromosomes 7q and 20q determines a pivotal role from first stage to metastasis cancer and also functions as an useful marker on diagnosis and treatment of colorectal carcinoma patients as well as follow-up checkup. Recently, the diagnosis and study using genetic analyzer are necessary for efficient application. Fortunately, several university hospitals run this genetic analyzer currently so it is expected that this method makes full use of clinical application.

Colon26-L5 대장암 세포를 이용한 간전이 모델에 산삼약침 처치가 혈중 cytokine에 미치는 영향 (Effects of Cultivated Wild Ginseng Herbal Acupuncture to the serum cytokine on Hepatic Metastatic Model using Colon26-L5 Carcinoma Cells)

  • Objective : This experiment was conducted to evaluate inhibitory effects against hepatic metastasis by cultivated wild ginseng Herbal Acupuncture. Methods : Colon26-L5 carcinoma cells were injected through hepatic portal vein to induce hepatic metastatic cancer. After treated cultivated wild ginseng Herbal Acupuncture and investigated various kinds of cytokine level using cytokine chip. Results : 1. Mice treated with cultivated wild ginseng Herbal Acupuncture reduced the level of $IL-l{\alpha}$, $IL-{\beta}$, and $TNF-{\alpha}$ compared to the control group. 2. Mice treated with cultivated wild ginseng Herbal Acupuncture was not showed significant change in the level of IL-4, IL-l0, IL-12 and $INF-{\gamma}$ compared to the control group. 3. Observing the level of various kinds of cytokine, cultivated wild ginseng Herbal Acupuncture was suppressed pro-inflammatory cytokine. These findings indicate cultivated wild ginseng Herbal Acupuncture is possible to use the inflammatory disease and futher studies carry out for the explanation of anticancer mechanism.

In Vitro Cytotoxic Effect of N-(Phosphonacetyl)-L-Aspartic Acid in Liposome Against C-26 Murine Colon Carcinoma

  • We have investigated the in vitro cytotoxic effect of liposome-encapsulated N-(phospho-nacetyl)-L-aspartic acid (PALA) against C-26 murine colon cancer cells, and have compared it in this regard to free PALA. Three different PALA-containing liposomal formulations using distearoylphosphatidylcholine (DSPC), distearoylphosphatidylglycerol (DSPG), and polyethyle-neglycol-derivatized distearoylphosphatidylethanolamine (PEG-DSPE) were made and their cytotoxicity was measured. In 72 hr continuous exposure experiment with C-26 cells, the 50% growth inhibitory concentration ($IC_50$) of DSPG-PALA liposome formulation was $0.09\mu\$, which showed about 65-fold more potent than unencapsulated free PALA ($5.1\mu\$). Similar degree of increase in cytotoxicity was also observed in 1 hr exposure experiment. However the $IC_50$ of PEG-DSPE-PALA liposome and DSPC-PALA liposome were $10.7\mu\$and $11.8\mu\$respectively, which showed slightly less potent than unencapsulated free PALA. Physical characteristics of PALA-liposomes, such as the size and drug:lipid ratio were also determined. In conclusion, negatively-charged DSPG-PALA liposome showed the highest cytotoxic effect among tested on the C-26 cells in vitro.

Importance of Sulfonylimidazolidinone Motif of 4-Phenyl-1-arylsulfonylimidazolidinones for Their Cytotoxicity: Synthesis of 2-Benzoyl-4-phenyl[1,2,5]thiazolidine-1,1-dioxides and Their Cytotoxcity

  • For probing the importance of planarity of imidazolidinone motif of 4-phenyl-1-(benzenesulfonyl)imidazolidinones 1 for their cytotoxicity, 4-phenyl-2-(benzoyl)[1,2,5]thiadiazolidine-1,1-dioxide (2a), 4-phenyl-2-(p-toluoyl)[1,2,5]thiadiazolidine-1,1-dioxide (2b), 4-phenyl-2-(phenylcarbamoyl)[1,2,5]thiadiazolidine-1,1-dioxide (3a), and 4-phenyl-2-(p-tolylcarbamoyl)[1,2,5]thiadiazolidine-1,1-dioxide (3b) were prepared along with their regioisomers (5a, 5b, 9a, 9b) and their cytotoxicity were measured against human lung carcinoma (A549), human colon carcinoma (COLO205), human ovarian cancer (SK-OV-3), human leukemic cancer (K562), and murine colon adenocarcinoma (Colon26) cell lines in vitro. All compounds prepared do not show any activity against all five cancer cell lines unlike 1. Compounds 1 possess planarity of imidazolidinone, especially in sulfonylurea moiety ($-SO_2$NHCONH-). However compounds 2 and 3 have nonplanar 5-membered ring, [1,2,5]thiadiazolidine-1,1-dioxides. Such structural differentiation might result in the loss of activity. Therefore the inactivity of 2 and 3 could also be an indication for the necessity of planarity of imidazolidinone ring of 1 for their cytotoxic activity.
