Investigation of atrophic piglets diseases in northern area of the Gyeongnam province, Korea (경남 북부지역 위축자돈에 대한 질병조사)

  • This study was investigated to diagnose pathogenic organisms of atrophic piglets in northern area of the Gyeongnam province, Korea. Samples such as feces, blood and necropsy specimens of 42 atrophic piglets (${\leq}10$ weeks old) were taken from May to December 2013 for this survey. Samples were examined by reverse transcriptase-polymerase chain reaction (RT-PCR) assay and bacteria isolation for detection of pathogenic agents. 93 pathogens were isolated from 42 samples can be classified into ll groups. We identified bacterial agents in 56 cases (60.2%) and viral agents in 31 cases (33.3%). However, 6 cases (6.5%) were undetected. Among these pathogens, the most prevalent disease were porcine reproductive and respiratory syndrome (PRRS) in 22 cases (23.7%). The major diseases were Colibacillosis in 15 cases (16.1%), Glasser's disease in 12 cases (12.9%), and porcine epidemic disease (PED) in 9 cases (9.7%). Mixed infections were accounted for 77.8% of atrophic piglets. In particular, the rate of mixed infections with PRRS virus showed the highest frequency (71.4%). In addition, there is a seasonal variation. Viral pathogens were dominantly detected in winter, but in the rest of the season bacterial agents were mainly detected. Gastrointestinal diseases occurred mainly in the pre-weaning piglets, the respiratory diseases and wasting diseases occurred mainly in the post-weaning piglets.

Studies on enterotoxigenic Escherchia coli isolated from piglets with diarrhea (설사 자돈에서 분리된 병원성 대장균에 관한 연구)

  • This study was carried out to Investigate the biochemical characteristics, antibiotic susceptibility, serogroups and pili producibility test of enterotoxigenic Escherichia coli(ETEC) isolated from piglets with diarrhea in Kangwon province from March to October 1996. 1. Sixty eight E coli strains were isolated from 72 piglets with diarrhea and the biochemical and cultural reaction were compared with the classification criteria of Edwards and Ewing. 2. The serogroups of 26 isolates were classified as 08 : K87 6(8.8%), O20 : K1O1 4(5.9%), O141 : K85 4(5.9%), 09 : K103 : P987 3(4.4%), O45 : K 2(2.9%) 0139 : K82 2(2.9%), O64 : $K^{-}$2(2.9%), O149 : K91 1(1.5%), O157 : K88ac 1(1.5%) and O115 : $K^{-}$1(1.5%), respectively. 3. In antibiotic susceptibility test, the isolates showed high susceptible to Ak, Eno, Na, Gm, Am and Km, whereas resistance to Tc, Sm and Cf. 4. Sixty one strains(89.7%) of 68 I coli Isolates were resistant to one or more drugs. The isolates resistant to 2 and 3 or more drugs were 60.3% and 19.1%, respectively. Amog the 16 multiple resistant patterns, Sm Tc(11.5% ), Cf Sm Tc(11.5% ), Cf Cp Sm Su Tc(9.8% ) and Cf Cp Sm Su Tc(8.2%) patterns were frequently observed. 5. MRHA of guinea pig erythrocytes was detected in 9 out of 26 OK serotype and 9 out of 42 unidentified serotypes. MRHA titers of serotypes showed from 16 to 32 in O141 : K85 and no titers in O139 : K82. 6. By the GM1 ganglioside ELISA, $\beta$-, $\alpha$-, and $\gamma$-hemolysin producing strains was detected as 36, 6, and 5 from heat labile enterotoxin(LT) of 47 ETEC, respectively. The distribution of LT toxin from 112 isolates was showed $\beta$- hemolysin, 2 isolates $\alpha$-hemolysin and 3 isolates $\gamma$-hemol-ysin from 26 OK serotypes.

Characteristics of Escherichia coli isolated from piglets with diarrhea ; antimicrobial susceptibility, genotypes of enterotoxins and pili and plasmid profiles (설사 자돈으로부터 분리한 Escherichia coli의 특성에 관한 연구 ; 항균제 감수성, 장독소 및 섬모의 유전형의 분포 및 plasmid profiles)

  • The antimicrobial susceptibility, genotypes of enterotoxins(LT, STa) and pili(K88, 987P), and plasmid profiles were investigated with 102 Escherichia coli isolated from piglets showing diarrhea in Korea. Almost of them were susceptible to ceftiofur(99%), cefquinone(97.1%). However they showed resistance to bacitracin(100%), streptomycin(98%), vancomycin(97%), trimethoprim/sulfamethoxazole(87.2%), tetracycline(84.3%) in antimicrobial susceptibility test. Moreover, all of the isolates demonstrated resistance to more than 2, and 78% of them were resistant to more than 8 of total 17 drugs. Multiplex PCRs for genotyping of enterotoxins(LT, STa) and pili(K88, 987P) were established with primers designed based on sequences from Genebank. Seventeen strains(16.6%) of the isolates had STa gene, 11 strains(10.8%) of them had both STa and LT genes, and 18 strains(17.8%) had K88 gene. But none of the isolates harbored a gene exclusively encoding LT. The gene encoding 987P pili was not found in all isolates. Fifty-four strains of 102 isolates(52.9%) had plasmid with various sizes ranging from 125kb to 1.1kb. Numbers of plasmid per isolates were also various, from 1 to 9. Distinctive relationship between plasmid profiles and genotypes of enterotoxins and pili in the isolates was not found. These results might provide the basic knowledge to establish strategies for the treatment and prevention of colibacillosis in piglets.

Effect of chitosan-oligosaccharides on hydrophobicity of pathogenic Escherichia coli (Chitosan-oligosaccharides가 병원성 대장균의 소수성(疎水性)에 미치는 영향)

  • The purpose of this study was to evaluate effect of chitosan-oligosaccharides (CHIOL) on hydrophobicity of pathogenic E coli including a field isolate from suckling piglet with diarrhea, E coli-O157 : H7, and E coli-O149 : K88ac. E coli field isolate appeared adhesion of 100% to n-hexadecane between 0.00125% and 0.05% CHIOL. E coli-O157 : H7 occurred adhesion of 69% and 64% under the level of 0.00125% and 0.025% CHIOL, respectively. E coli-O149 : K88ac showed adhesion of 100% in higher than 0.025% CHIOL. For cationic action, the adhesion of E coli isolate and E coli-O149 : K88ac to n-hexadecane were inhibited at level of higher than 10mM $Ca^{2+}$ but did not induce any difference among the concentrations used(p < 0.01). However, the adhesion of E coli-O157 : H7 to n-hexadecane was inhibited at level of higher than 50mM $Ca^{2+}$. In a field trial, control piglets showed average mortality of up to 58% during 3 days after the onset of diarrhea. In contrast, the prevalence of E coli-induced diarrhea in CHIOL-treated groups without mortality was dropped down to average 34% on the 1st day after the treatment of CHIOL, and average 2% on the 4th day. After then, piglets with diarrhea was not present. In conclusion, the low concentrations of CHIOL were most likely to associate with the enhancement of hydrophobicity to pathogenic E coli. Calcium inhibited the hydrophobicity of E coli by CHIOL. These results suggested that CHIOL could be played an efficient and reliable role in treating enteric colibacillosis of piglets.

Isolation of the Pathogenic Bacteria from Chicken and Antimicrobial Drug Sensitivity of the Strain Isolated (가금유래 주요병원성세균의 분리와 분리균주에 대한 약제감수성조사)

  • A total of 1503 specimens were submitted to the Poultry Disease Diagnostic Service Laboratory during the year 1966 and 1978. The most frequently diagnosed diseases in order of prevalence were avian mycoplasmosis, staphylococcosis, colibacillosis, salmonellosis and pullorum disease, the percentages of the conditions being 24.6%, 20.0%, 18.0%, 12.6% and 6.4%, respectively, The drug resistance of pathogenic mirnoorganisms isolated during the year 1978 from chicken with colicabacillosis, staphylococcosis or salmonellosis were investigated by the use of disc diffusion technique, the results being as follow. 1) Drug resistance of 63 strains of Escherichia coli More than 95% of the strains tested were sensitive to colistin and gentamicin. The percentages of strains sensitive to kanamycin, chloramphenicol, ampicillin and nitrofurantoin were 66.7%, 60.3%, 60.3% and 47.6%, respectively. Majority of the strains were highly resistant to streptomycin and tetracyline. All the strains were resisistant to bacitracin lincomycin, oleandomycin, penicillin and erythromycin. All the strains tested were resistant to more than two among 10 drugs in common use such as penicillin, erythromycin, streptomycin, tetracycline, neomycin, chloramphenicol, kanamycin, ampicillin and gentamicin, and 27 different resistance patterns were noted. The most frequent multiple resistance pattern was PC, EM, SM and TC (11.1%). 2) Drug resistance of 48 strains of Salmonella More than 95% of the strains tested were sensitive to colistin, gentamicin ana ampicillin. The percentages of st rains sensitive to kanamycin, tetracycline, neomycin and nitrofurantoin were 81,3%, 79%, 72.9%, and 68.0% respectively. None of them was sensitive to streptomycin, oleandomycin, erythromycin, lincomycin and bacitracin. All the strains were resistant to more than one among 7 drugs in common use such as streptomycin, erythromycin, neomycin, tetracycline, kanamycin, ampicillin and gentamicin. The most frequent resistance pattern was SM and EM(66.7%). 3) Drug resistance of 54 strains of Staphylococci All the strains tested were sensitive to gentmaicin, kanamycin and cephalothin. Majority of them were highly sensitive to bacitracin, methicillin, nitrofurantoin and chloramphenicol. The Percentages of strains sensitive to streptomycin, ampicillin, lincomycin and tetracycline were 66.7%, 55.6%, 44.4% and 27.8%, respectively. Among them, 51 strains were resistant to more than one among 11 drugs in common use such as tetracycline, lincomycin, ampicillin, penicillin, streptomycin, erythromycin, neomycin, oleandomycin, chloramphenicol, methicillin and bacitracin, and thirty one different resistance patterns were noted.

Virulence factors and antimicrobial resistance patterns of pathogenic Escherichia coli isolated from fecal samples of piglets (자돈 분변 유래 병원성 대장균의 병원성 인자 및 항생제 내성 양상)

  • Pathogenic Escherichia coli is the cause of a wide range of diseases in pigs, including diarrhea, edema disease, and septicemia. Diarrhea caused E. coli may result in significant economic losses, making pathogenic E. coli an important pathogen for the swine industry. This study investigated the prevalence of virulence factor genes, antimicrobial resistance phenotypes, and resistance genes in E. coli isolated from feces of piglets in Korea between 2017 and 2020. As a result, 119 pathogenic E. coli isolates were obtained from 601 fecal samples. The F4 adhesin gene and the STb enterotoxin gene were commonly present in E. coli isolated from diarrhea samples. The dominant virulotypes of isolates from diarrhea samples were STb, Stx2e, and F4:LT:STb. More than 80% of the screened isolates were resistant to ampicillin, sulfisoxazole, chloramphenicol, or tetracycline. To confirm the resistance mechanisms for β-lactam or quinolone, we investigated the genotypic factors of resistance. Each of the ceftiofur-resistant E. coli produced an extended-spectrum β-lactamase encoded by blaCTX-M-14, blaCTX-M-27, and blaCTX-M-55. And all ciprofloxacin-resistant E. coli harbored mutations in quinoloneresistance-determining-regions. In addition, some of the ciprofloxacin-resistant E. coli contained the plasmid-mediated-quinolone-resistance genes such as qepA, qnrB1, or qnrD. This study has confirmed that the F4 fimbria and the STb enterotoxin are the most predominant in pathogenic E. coli isolated from piglets with diarrhea in Korea and there is a great need for responsible and prudent use of antimicrobials to treat colibacillosis.

Survey on Korean-native Calves Diseases and Mortality (한우 송아지의 질병발생과 폐사율 조사)

  • From September 1996 to September 1999, 419 Korean-native calves with diseases under 6-month old collected from Kyonggi, Chungcheong, Chonlla and Kyongsang were examined by clinical, microbiological, parasitic, hematologic and histopathological mean. Among them, 124 cases were tested about the neutralization antibodies against infectious bovine rhinotracheitis virus(IBRV), Parainfluenza-3 virus(PI-3V), bovine uiral diarrhea virus(BVDV), bovine ephemeral fever virus(BEFV). In calf diseases in the survey, enteric diseases(72.8%) were most frequently involved and the following orders were taken by respiratory(17.4%) and reproductive (5.0%) disorders. In the causative pathogens associated with calf diseases and motality, 48.4% was induced by bacteria origin and also 35.6% by viral agents. Calf mortality was up to 76.3% in the cae of bacterial diseases and 55.7% in viral diseases. In bacterial diseases, frequent disorders were composed of colibacillosis(52.7%), salmonellosis(13.8%), pasteurellosis(12.8%) and campylobacteriosis(3.9%) and their mortalities showed 73.8% in colibacillosis, 73.0% in pasteurellosis, 67.9% in salmonellosis and 50.0% in campylobacteriosis (50.0%). Among the outbreaks of viral diseases, there were BVD(22.8%), bovine rotavirus infection(20.8%), bovine coronavirus infection(16.8%), bovine respiratory syncytial virus infection(15.4%), IBR(15.4%). Akabane disease(4.7%) and Chuzan diseases(3.4%). Interesting results through this studies were obtained the first isolate to Chuzan virus and Ainovirus in Korea which could be promised the development for diagnostic method and vaccines soon. Calf mortality to Akabane and Chuzan diseases was 100%. Main parasitic diseases were occupied by coccidiosis and babesiosis and their mortality of babesiosis was 20.0%. Other diseases were abomasal impaction(6.7%) and toxicosis(4.5%). The mortality of abomasal impaction was 89.3%. In some causes with malformations(1.9%) were confirmed as anasarca, derodidymus, polymelia, humerus hypoplasia, and tracheal collapse. Calf diseases had mostly been occurred in one month old grout (52.5%) and its prevalence was 25.1% in two to three month old group and 22.4% in four to six month old group. In calf mortality by age, there were 37.9% in one month old group, 18.1% in two and three month old group, and 13.8% in four to six month old group, respectively. The older the age of calf, the less the prevalence of calf enteric diseases. Respiratory diseases in calves to be tested frequently occurred in one to two month old group (41.4%). In one month old calves, the prevalence of enteric disease was 80.0%(p<0.05) and that of reproductive and respiratory disease was 9.5% and 8.2%, respectively. In two month old and four to six month old, enteric disease was 65.7% and 63.8% and respiratory disease was 28.6% and 26.6%. Seasonal prevalence and mortality of Korean-native calf diseases were not a significant difference. Prevalence of calf diseases in summer(31.5%) frequently occurred to compare that in winter(20.3%). Abortion and malformation in calves frequently occurred in spring. Hematological values in 84 calves with clinical signs showed mild to marked leukocytosis. Also, there was slight increase in hematocrit, platelet, mean corpuscular volume and mean plasma volume, but all of those were included the higher level to normal ranges. Calves with respiratory signs showed slightly erythrocytosis. One hundred seventy three calves without clinical signs were not significant different to ill cases in hematological values, but number of platelets was in higher normal range. In 125 calves, 84.8% was taken the antibody to IBRV, but 72% with the antibody had recorded the titer level lower than log$_2$5. The neutralizing antibody levels of higher than $log_{2}5$ to PI-3V and BVD virus were 60.8% and 67.2% cases, respectively. There were the cases of 57.6% had the neutralizing antibody level lower than log$_2$5 to BEFV.

Studies on Epidemiological Survey of Infectious Disease of Chicken in Korea (국내 닭 전염성 질병에 관한 역학적 조사 연구)

  • A total of 9, 012 cases was submitted for diagnosis of chicken diseases to Veterinary Research Institute, Rural Development Administration from domestic chicken farms during 18 years from 1971 to 1988. Of them, 6, 181 cases diagnosed as the infectious disease were investigated for the detection rate of infections on basis of you, season , and chicken age. The results obtained were summarized as followings:1. Detection rate or the infections was lowest as 49.3% in the year 1973, and highest as 78.6% in 1985 (average 68.6%). 2. Of infections detected, bacterial diseases were most frequent (32.6%), and followed in order by viral (26.3%), parasitic (7.7%), and fungal diseases (2.1%) in geneal. 3. The most frequently detected bacterial diseases in order of prevalence were mycoplasmosis (8.8%), colibacillosis (8.5%), and staphylococcosis (5.8%), and followed by salmonellosis pullorum disease , yolk sac disease, and salpingitis (0.8-1.5%). 4. In viral diseases, 7.5% of infections detected was lymphoid leukosis and 7.2%-Marek's disease, 4.4%-Newcastle disease, 2.0%-infectious laryngotracheitis, 1.7%-infectious bursal disease, and 1.0%-avian encephalomyelitis, while detection rate of infectious bronchitis, egg drop syndrome '76, and inclusion body hepatitis was less than 1.0%, respectively. 5. The most prevalent parasitic disease was coccidiosis (4.5%), followed by ascariasis (1.4%). The detection rate of other parasitic diseases including leucocytozoonosis, black head , heterakiasis, and ectoparasitosis was very as 0.2-0.7%, respectively: In fungal diseases, 2.0% of infections was detected as aspergillosis, and followed by candidiasis (0.1%). 6. Detection rate of the infections on basis of season was somewhat higher in summer. (27.7%), and autumn (27.7%) than in winter (23.5%), and spring (21.5%) in general. In bacterial, viral, and fungal diseases, there were the similar tendencies of detection rate as in infections, while parasitic diseases were much highly detected in summer (34.3%), and autumn (39.5%) than in any other season. 7. Among bacterial diseases colibacillosis was most frequently detected in summer, and staphylococcosis in autumn. In detection rate of viral diseases, Marek's disease, infectious laryngotracheitis, and infectious bursal disease was highest in summer, lymphold leukosis, fowl pox and egg drop syndrome '76 in autumn, and infectious trachitis in winter, repectively. The majority of important parasitic diseases including coccidiosis were highly detected in summer and autumn. 8. On basis of chicken age, detection rate of infections were highest in chicken of growing period between 30 and 150 days of age (41.4%), and followed by 35.3% in laying chicken over 150 days of age, and 17.3% in chicken of brooding age under 30 days of age. Bacterial, and parasitic diseases were most frequently detected in chicken of growing period, viral diseases in chicken of growing, and laying period as nearly equal rate of detection, and fungal diseases in chicken of brooding age.

Study on disease prevalence to Holstein calves reared in Chonnam area (전남 지방의 홀스타인 송아지의 질병 발생율 조사)

  • The prevalence of major calf disease was investigated in 117 Holstein dairy calves in Chonnam area. All of them were moved in the College experimental farm which is operated in intensive units. clinical signs were daily examined throughout two months after the introduction of the College farm. Among calves, 92 cases(78.6%) died in the two months after the introduction in it. Outbreaks of respiratory and alimentary diseases were their main causes of their fatality. The incidence of respiratory disorders during the full period of the experiment was up to 42.8%, and the alimentary diseases were occurred 35.9% of the herd. Most of the mortality was related with respiratory(59.9%) and alimentary(52.1%) pathogens. Also calf mortality by combined infection claimed 6.6% among 100 morbidity cases. Principle pathogens to cause mortality were Pasteurella spp(44.4%), E coli(29.9%), bovine viral diarrhea virus(16.2%), IBRV(12.0%), respectively. Viruses also played as an important role in increasing calf morbidity to secondary respiratory bacterial pathogens. Pasteurella infection combined with infectious bovine rhinotracheitis virus(11 cases), parainfluenza virus type-3(9 cases), or bovine respiratory syncytial virus(7 cases) was appeared as major pattern to mortality. colibacillosis in causing enteritis was concurrently infected with BVD(19 cases), bovine coronavirus infection(14 cases), salmonellosis(5 cases), coccidiosis(5 cases) and clostridial infection(4 cases). Ninty-two cases to death were appeared to have 100% neutralizing antibodies to BCV; Among them, 73.8% had the neutralizing antibody level higher than 64. Calves with neutralizing antibodies higher than 16 to BVDV were 50%. The cases with neutralizing antibody level lower than 8 to BEFV were 89.4% that means the necessity of appropriate vaccination.

Production of a Specific Yolk Antibody against Enterotoxigenic E. coli F41 Fimbrial Antigen (장독성대장균 F41 섬모항원에 대한 특이난황항체 생산)

  • Enteric colibacillosis has economically become an important disease of young animals as a result of increasing intensification of farrowing management. The objective of this experiment is to isolate fimbrial antigen from enterotoxigenic E. coli F41, to develop specific polyclonal IgY which can effectively neutralize or reduce the proliferation of pathogens in feed or living animal system, and to apply IgY technologies to animal industry. The results obtained were as follows: The molecular weight of the purified F41 antigen was 29,500 dalton on sodium dodecyl sulfate-polyacrylamide gels. Fimbrial antigen was confirmed by the western blot method. It was observed that after immunization the level of serum antibody titer of laying hen was shown in two weeks and gradually increased. The antibody titer in egg yolk appeared two weeks after it was shown in serum antibody. The titers of egg yolk antibody were gradually increased to the maximum level of 320,000 (antigen 50${\mu}g$/$m\ell$), 450,000 (antigen 200${\mu}g$/$m\ell$), and 320,000 (antigen 600${\mu}g$/$m\ell$). According to the results of specificity test by ELISA, the anti-F41 antibodies from chicken serum and egg yolk reacted only with ETEC F41 antigen. There was no cross reaction with other ETEC strains (K88, K99, and 987P). In vitro condition, as a result of antigen binding ability of yolk antibodies, bacterial concentration was rapidly decreased to $10^5$ CFU/$m\ell$ from $10^9$ CFU/$m\ell$ when 2${\sim}$4 mg/$m\ell$ of freeze dried WSF (water soluble fraction) was added.
