Antibiotic resistance pattern of avian pathogenic Escherichia coli isolated from chickens (닭에서 분리된 조류 병원성 대장균의 항생제 내성 양상)

  • Avian pathogenic Escherichia coli (APEC) is an important bacterial pathogen of chickens and causes colibacillosis such as airsacculitis, perihepatitis, omphalitis, peritonitis, salpingitis, and pericarditis. As the transfer of antibiotic resistance from animal to humans can be possible, surveillance on antibiotic resistance of APEC is very important. A total 34 APEC isolates from diseased chickens during the period from 2007 to 2009 were obtained. The susceptibility of the isolates to 13 antibiotics was determined by disc diffusion assay. Resistance to erythromycin was found in 97.1% of APEC isolated, followed by resistance to tetracycline (85.3%), doxycycline (82.3%), ampicillin (73.5%), sulfisoxazole (67.6%), enrofloxacin (67.6%), ciprofloxacin (64.7%), norfloxacin (61.7%) trimethoprim/sulfamethoxazole (52.9%), gentamycin (26.5%), amoxicillin (8.8%), colistin (5.9%), and amikacin (2.9%). The blaTEM genes were detected in 25 (100%) of the 25 ampicillin-resistant APEC isolates. Among the 29 tetracycline-resistant APEC isolates, tetA and tetB genes were detected in 18 (62.1%) and 9 (31%) isolates, respectively. Twenty six (76.5%) isolates were multiresistant to at least 6 antibiotics and seven (20.1%) isolates were multiresistant to at least 10 antibiotics. This results indicated that multiple antibiotic-resistant APEC is widespread in chicken flocks in Korea.

  • Poultry production in korea is a very complex situation. Large modernized farms and old styles of small farming coexist with one another. This gives rise to a tangled epidemiological situation in terms of infectious diseases. The main poultry diseases of economic importance are colibacillosis, pullorum diseases, Mycoplasma gallisepticum infection, infectious coryza, infectious synovitis, Newcasyle disease, fowl pox, Marek's disease, avian encephalomyelitis, infectious bursal disease, infectious laryngotracheitis, infectious bronchitis and coccidiosis. Avian influenza, fowl cholera and fowl typhoid have not been reported for a few decades, and these are rated as exotic diseases.

Isolation of chicken anemia agent (virus) from naturally infected chickens (자연감염된 닭으로부터 chicken anemia agent (virus)의 분리)

  • Attempts to isolate chicken anemia agent (CAA) were made by inoculating tissue homogenates into MDCC-MSBl or LSCC-1104B1 cell lines and passaging the cells serially. CAA was isolated from the liver and thymus of 11 weeks old layer chickens and from the liver of 10 weeks old broiler breeder chickens. The layer flock experienced approximately 45% mortality during 9 to 14 week of age from gangrenous dermatitis and lymphoid organs of affected chickens were severely atrophied. The broiler breeder flock experienced approximately 7% mortality during 7 to 9 weeks of age and affected birds showed lesions of colibacillosis, staphylococcal arthritis, and coccidiosis together with atrophied lymphoid organs. The isolated viruses were identified as CAA by the indirect fluorescent antibody test and virus neutralization test using CAA immune sera including one to Gifu-1 strain of CAA. The CAA isolate 89-69, when inoculated into susceptible 1 day old SPF chicks, induced anemia 14 to 16 days after inoculation. It did not induce any cytopathic effects in chicken embryo liver and chicken embryo fibroblast cell cultures. Infectivity of the isolate was not affected by the treatment of chloroform or heat ($70^{\circ}C$ for 15 minutes).

A Study on Bioserotype and Drug Resistance of Salmonella and Escherichia coli Isolated from Feces in Zoological Animals (동물원의 야생동물 분변에서 분리한 살모넬라균과 대장균의 생물형, 혈청형 및 약제내성에 관한 연구)

  • This study was undertaken the bioserotype and drug resistance of Salmonella and Escherichia coli isolated from feces for the prevention and treatment of salmonellosis and colibacillosis in zoological animals. The results obtained from the research were as follows 1. Salmonella were isolated 19, or 4.7% from 408 samples and E. coli were isolated 12, or 40.0% from 30 diarrheal samples. 2. The biotypes in 19 Salmonella were Subspecies 1. 3. The serogroups of Salmonella isolated were 47.4% in B group, 31.6% in C, 5.3% in D and 15.8% in other, and serotype of E. coli was 100% in 0127a. 4. The antibiotic resistance of Salmonella and E. coli isolated were 13, or 68.4% and 7, or 58.3% strains, respectively 5. The multiple resistant patterns of antibiotics in Salmonella were 2drugs- and 3 drugs-resistance 30.8%, respectively, and those in E. coli were mono drug-, 2 drugs- and 7 drugs-resistance 28.6%, respectively. 6. The transferred rate of resistance to recipients (E. coli ML 1410 NA$^{r}$ ) in Salmonella was 38.5%, but that in E. coli was 71.4%.

Survey on enteric diseases in chicken (닭의 장염발생 조사)

  • The purpose of the survey was to understand the field situation on the occurrence of enteric diseases in poultry farms in Korea. During the period from January of 1991 to December, 1992, 279(35.8%) out of 779 farms surveyed experienced the cases of enteritis in their chickens. The farms with diarrheal problems consisted of 157(45.1%) of 348 broiler flock, 28(32.6%) of 86 breeder flock, and 94(27.2%) of 345 layer flock. The main diseases causing enteritis were frequently appeared as infectious bursal disease(27.3%), colibacillosis(22.6%), and coccidiosis(8.6%). Necrotic enteritis(NE) in chicken occurred in 21 farms(2.7%) among 779 farms examined. The outbreaks of NE were divided 4.7% in breeders, 4.0% in broilers, and 0.9% in layers, respectively. NE occurred mainly in the age(71.4%) of 20~30 days and the mortality was varying from 1.4% to 10.0% for broilers, 3.0% for layers and 0.8% for breeders. Seasonal prevalence of NE showed the highest as 52.4% in summer and followed by 33.3% in autumn, 9.5% in spring and 4.5% in winter.

  • Reproductive performance of gilts and sows in ordinary piggeries was investigated with special reference to litter size, piglets weaned per litter, the cause of preweaning mortality and the incidence of diarrhea. Biochemical properties of Escherichia coli isolated from piglets with diarrhea were also determined. The results obtained are summarised as follows: 1. Of the 2,226 gilts and sows farrowed, average number of piglets born alived and weaned piglets per litter were 8.9 and 7.1, respectively, indicating that both gilts and sows lost more than 20% of all their piglets prior to weaning. 2. The causes of preweaning mortality in order of prevalence were diarrhea (39.3%), pneumonia (20.0%), crushing (13.8%), starvation (11.0%) and born weak (10.3%). 3. Incidence of 3 diarrheal syndromes of piglets, i.e. 1 week diarrhea, 3 week diarrhea and post-weaning diarrhea were 18.4%, 66.1% and 15.5%, respectively, showing that most farms were suffering from so called 'white scours' in piglets 14~28 days old. 4. Biochemical properties of 268 cultures of E. coli isolated from diarrheal piglets were tested and compared with those of standard strains of porcine origin. All those properties of isolates were matched to standard E. coli teated while variable results were obtained with haemolytic capabilities of cultures tested. 5. Of 147 isolates 16 cultures (10.9%) were identified as colicin producers.

Prevalence of pili and enterotoxins of Escherichia coli associated with diarrhea in preweaning piglets (포유자돈 소장에서 분리된 대장균의 섬모항원과 장내독소 분포양상)

  • A comprehensive study of 132 Escherichia coli isolates from 150 piglets with colibacillosis included detection of heat-labile enterotoxin, heat-stable enterotoxin, and identification of K88 (F4), K99 (F5), 987P (F6), and F41. Four pili were examined by haemagglutination and slide agglutination test. Heat-labile(LT) and heat-stable(ST) enterotoxin was determined by reverse passive latex agglutination and precipitation test, respectively. Among 132 E coli isolates, 26 had K88 (19.7%), 16 had K99 (12.1%), 3 had 987P (2.3%), and 2 had F41 (1.5%). Three had K88 and K99 (2.3%), 3 had K88 and 987P (2.3%), 2 had K99 and 987P (1.5%), 5 had K99 and F41 (3.8%), and 8 E coli strains had K88, K99 and F41 (6.1%) simultaneously. Among 132 E coli isolates, 5 produced LT only (3.8%), 55 produced heat-stable toxin ST only (41.7%), and 4 produced both LT and ST (3.0%). Three major pathotypes accounted for 27.9% of E coli isolates: $K99^+$ (8.3%), $K88^+ST^+$ (9%) and $K88^+$ (10.6%). Results of this study indicated that piliated enterotoxin-producing E coli was prevalent and was associated with diarrhea in preweaning piglets.

  • The prevalence of diarrhea caused by enterotoxigenic Escherichia coli was surveyed on 445 calves in 6 farms which were located in the central part of Korea. The incidence of enterotoxigenic Escherichia coli in calves with diarrhea was investigated by detecting the K99 and F41 antigens from the isolated strains of Escherichia coli The incidence of colibacillosis in calves was 23.3%. Of 238 strains of Escherichia coli isolated from calves with diarrhea, 73 strains(30.6%) were proved possessing the K99 antigen by mannose-resistant hemagglutination(MRHA) using horse red blood cells and 79(33.1%) possessing F41 antigens by MRHA using guinea-pig red blood cells. The minca medium, nutrient broth, tryptic soy broth and brain heart infusion were tested for yield of K99 and F41 pili. The production of pili was greatest in minea medium. The best detachment method of the K99 and F41 pili from the cells was heat treatment for 20 minutes at $60^{\circ}C$ and concentration by precipitation with ammonium sulfate. The purified antigens of K99 and F41 were polypeptides with molecular weights of 18,500 and 29,500, respectively by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

Prevention of Scours in Neonatal Kids after Oral Administration of an Organic Acid Solution or Antibiotics

  • The efficacy of an organic acid solution in the prevention of neonatal kid diarrhoea, was examined in this study and was compared with that of different antibiotics. In a goat farm, kids and their mothers were divided in 4 experimental groups. The kids of Group I were not given any treatment serving as negative controls. The kids of Group II were treated orally for 3 consecutive days with a solution of organic acids starting approximately 12 h after their birth. The kids of Groups III and IV were treated once orally with ampicillin and oxytetracycline respectively, approximately 12 h after birth. All groups were compared as regards the occurrence of diarrhoea, its duration, and the rate of mortality. The results have shown that the number of cases and the duration of diarrhoea were significantly reduced in all treated groups, when compared to the control group (p<0.05). However, no or little difference was noted with respect to morbidity, mortality and diarrhoea characteristics when treated groups were compared with each other (p>0.05). Samples of diarrhoeic faeces from kids in the control group resulted in the isolation of K88 and K99 strains of Escherichia coli. It was concluded that, early oral administration of organic acids can be effective in the prevention of scours in neonatal kids, possibly caused by enterotoxigenic E. coli strains, and to a degree of protection similar to that seen with the used antibiotic schemes.

Molecular epidemiologic analysis of pathogenic Escherichia coli isolated from poultry in Korea (국내 가금 유래 병원성 대장균의 분자역학적 분석)

  • Among 203 avian pathogenic Escherichia coli (APEC) isolated from poultry with colibacillosis in korea, 14 isolates were selected from total 68 isolates transferred R plasmid and classified into 5 groups on the basis of antimicrobial minimal inhibitory concentration (MIC) pattern, farm source and O serotype. An association between clonal origin and R plasmid of them was investigated by R plasmid profile, restriction endonuclease analysis and pulsed-field gel electrophoresis (PFGE). The strains that showed the same or very similar antimicrobial MIC pattern, but different farm source and O serotype, revealed different PFGE pattern, which seemed to be different clonal origin. And the strains that showed the same MIC pattern and O serotype, revealed different PFGE pattern, seemed to be originated from different clone. Also the strains showing the same MIC pattern and farm source, but different O serotype, revealed to be different clonal origin. The strains that showed the same or similar MIC pattern, farm source, and O serotype, revealed identical or similar PFGE pattern, which seemed to belong to be one clone. Meanwhile, horizontal transfer of R plasmid seems to be common in APEC with regardless of O serotype and clone of the strains. These results indicate that rapid and accurate epidemiological survey of APEC can be possible by the combination of O serotyping, plasmid profiling and PFGE analysis following the classification of them into groups of antimicrobial drug resistance pattern.
