식품 보존 스트레스에서의 식중독세균의 생체막 생성 (Biofilm Formation of Food-borne Pathogens under Stresses of Food Preservation)

  • 세균이 외부stress에 대한 자가저항으로 biofilm형성을 하는 것은 식품 뿐만아니라 식품기기등의 세척, 소독등의 식품안전 확보에 많은 어려움을 주게 된다. 본 연구에서는 glass wool과 mlcrotiter plate assay를 이용하여 주요 식중독 세균인 Salmonella, E. coli, B. cereus, S. aureus를 여러가지 식품보존하에서 상해와 biofilm형성 정도를 비교하였다. 이들 세균은 외부의 stress없는 조건하에서도 상해를 받지 않았고 모두 biofilm이 형성되어 glass wool에 부착되었다. Microtiter plate assay에서의 상해별 biofilm형성은 acid stress에서 10%이내의 상해를 받은 E. coli와 약 40%의 상해를 받은 S. aureus에서 높게 나타났다. $4^{\circ}C$의 cold temperature에서는 30-50% 상해를 나타낸 B. cereus와 E. coli가 높은 biofilm 형성을 보였고 cold starvation에서는 다른 stress에 비해 전체적으로 biofilm형성도가 낮은 값으로 측정되었다. 그리고 6% sodium chlorine solution에서 30-55%의 상해를 입은 Salmonella가 높은 biofilm 형성도를 보였다. 그러나 같은 종의 식중독 세균이라도 외부의 stress 대하여 다양한 정도의 biofilm을 생성하는 것으로 보인다. 따라서 식품으로부터 이들 식중독 세균을 제어하기 위해서는 대상식품의 보존환경에 따른 biofilm 형성특성을 고려해야 할 것으로 사료된다.

이동원(李東垣) 내외상변(內外傷辨) 관련 논의들에 나타난 내외(內外) 관계성에 대한 고찰 (A Study on the Relationship between the Internal and External from Discussions on Li Dongyuan's Differentiation of Internal and External Damage)

  • Objectives : This study aims to examine the multi-layered meanings of Li Dongyuan's concept of 'internal damage' through examining the origins of the 'internal damage' concept together with discussions derived from his 'Differentiation of Internal and External Damage'. Methods : Rather than Li's original works, those done by later doctors who were influenced by Li were thoroughly examined in this study to determine the relationships among internal and external damage, and external contraction. Results : Based on discussions related to the 'differentiation of internal and external damage' following Li, his concept of 'internal damage' could be understood as multi-layered, as it contains both spatial as well as pathogenic meaning. Therefore the meaning behind categorizations of internal and external damage, and internal damage and external contraction could be different. Li's 'internal damage' concept includes not only internal pathogen but elements of external contraction of wind and cold, which seems to have been integrated into the 'Cold Damage Theory' after Zhu Danxi, seemingly having changed the concept of cold damage. Therefore, discussions on the crossing between internal damage and external contraction have on one hand expanded the boundaries of each concept, while on the other hand, have caused confusion between the two at times. Conclusions : The concept of 'internal damage' has been widely accepted due to Li Dongyuan, on which related discussions have been centered. However, the concept was originally multi-layered, naturally leading to various discussions. Future studies should focus on the relationship of 'internal damage' with 'external damage' or 'external contraction', beyond the theories of Li.

Block Copolymer Thin Films: Nanotemplates for New Functional Nanomaterials

  • In this study, the applications of the block copolymer thin films are introduced. For this purpose, we first obtained cylindrical nanodomains in polystyrene-block-poly(methyl methacrylate) copolymer perpendicularly oriented to a substrate. Then, nanoporous templates were prepared after removing the PMMA nanodomains by UV treatment. By using electropolymerization, high density nanowire arrays of conducting polymer of poly(pyrrole) and poly( 3-hexyl thiopene) were obtained and their electric properties were measured. Also, these nanoporous thin films were found to be very useful for the separation of human Rhinovirus type 14 (HRV 14), major pathogen of a common cold in humans, from the buffer solution.

First Report of Postharvest Gray Mold Rot on Carrot Caused by Botrytis cinerea in Korea

  • In February 2014, gray mold rotting symptoms were observed in carrots in cold storage at Gangneung, Gangwon province, Korea. The typical symptom of gray mold rot showed abundant blackish gray mycelia and conidia was observed on the infected root. The pathogen was isolated from infected root and cultured on PDA for further fungal morphological observation and confirming its pathogenicity according to Koch's postulates. Results of morphological data, pathogenicity test and rDNA internal transcribed spacer (ITS 1 and 4) sequence showed that the postharvest gray mold rot of carrot was caused by Botyrtis cinerea. This is the first report of postharvest gray mold rot on carrot in Korea.

  • Current food sanitation systems for meat and livestock in Australia ar underpinned by HACCP-based quality assurance. HACCP-based quality assurance programs have been implemented by all australian red meat industry sectors, from 'farm-to-fork'. These quality assurance programs are (along with other quality and food safety considerations) the collection of a series of sanitation steps and strategies that aim to ensure that only healthy, clean livestock are presented for slaughter; those healthy, clean animals enter a clean processing plant; contact of carcase surfaces with potential sources of contamination is avoided; carcase surfaces are decontaminated before chilling; and subsequent growth of potential contaminants is avoided. Prospects for food sanitation systems for meat and livestock in Australia lie largely in enhancing current and applying new sanitation strategies and procedures within our HACCP-based quality assurance framework. Prospects include increased focus on actual (Versus perceived) risks; on-farm/feedlot pathogen elimination; increased implementation of existing (or new) decontamination technologies; and an improved cold chain.

The First Report of Postharvest Stem Rot of Kohlrabi Caused by Sclerotinia sclerotiorum in Korea

  • In March 2014, a kohlrabi stem rot sample was collected from the cold storage room of Daegwallyong Horticultural Cooperative, Korea. White and fuzzy mycelial growth was observed on the stem, symptomatic of stem rot disease. The pathogen was isolated from the infected stem and cultured on potato dextrose agar for further fungal morphological observation and to confirm its pathogenicity, according to Koch's postulates. Morphological data, pathogenicity test results, and rDNA sequences of internal transcribed spacer regions (ITS 1 and 4) showed that the postharvest stem rot of kohlrabi was caused by Sclerotinia sclerotiorum. This is the first report of postharvest stem rot of kohlrabi in Korea.

The Ozone Stress Transcriptome of Pepper (Capsicum annuum L.)

  • We used cDNA microarrays to monitor the transcriptome of ozone stress-regulated genes (ORGs) in two pepper cultivars [Capsicum annuum cv. Dabotop (ozone-sensitive) and Capsicum annuum cv. Buchon (ozone-tolerant)]. Ozone stress up- or down-regulated 180 genes more than three-fold. Transcripts of 84 of these ORGs increased, transcripts of 88 others diminished, and those of eight either accumulated or diminished at different time points in the two cultivars or changed in only one of the cultivars. 67% (120) of the ORGs were regulated differently in ozone-sensitive and ozone-tolerant peppers, most being specifically up-regulated in the ozone-sensitive cultivar. Many were also represented in the plant defense transcriptome against non-host pathogen infection, and some in the transcriptomes for cold, drought, and salinity stresses.

A pathogen-induced osmotin-like protein gene, CAOSMl, from pepper: Differential expression and in situ localization in pepper tissues during pathogen infection and abiotic stresses

  • An osmotin-like protein (CAOSMl) gene was isolated from pepper leaves infected with the avirulent strain Bv5-4a of Xmthomonas campestris pv. vesicatoria. The cDNA encodes a polypeptide of 250 amino acids with a molecular mass of 27, 361 Da. Its amino acid sequence is highly homologous to various osmotin-like proteins from other plant species. The CAOSMl gene expression was organ- and tissue-specifically regulated In pepper plants. The CAOSMl mRNA was intensely localized in the endodermis area of root tissue and in the phloem cells of vascular bundles of red fruit tissue, but not in leaf, stem, and green fruit tissues of healthy pepper plants. Infection by X. c. pv vesintoria, Colletotrichum coccodes, or Phytopkhora capsici iinduced CAOSMl transcription in the leaf or stem tissues. Expression of the CAOSMl gene was somewhat higher in the incompatible than the compatible interactions of pathogens with pepper. The CAOSMl mRNA was prevalently localized in the phloem cells of the vascular bundle of leaf tissues infected by C. coccodes. The CAOSMl gene was activated in leaf tissues by treatment with ethylene, methyl jasmonate, high salinity, cold acclimation and mechanical wounding, but not by abscisic acid (ABA) and drought. These results indicate that the pepper CAOSMl protein functions in response to Pathogens and some abiotic stresses in pepper plants

Cloning And Characterization of Pathogen-Inducible EREBP-Like Transcription Factor(CaNR19) From Hot Pepper (Capsicum annuum L.)

  • An EREBP/AP2-type transcription factor (CaPFl) was isolated by DDRT-PCR following inoculation of soybean pustule pathogen Xanthomonas axonopodis pv. glycines Bra which induces HR on pepper leaves. Genomic Southern blot analysis revealed that the CaPFl gene is present as a single copy within the hot pepper genome. The deduced amino acid sequence of CaPFl has two potential nuclear localization signals, a possible acidic activation domain, and an EREBP/AP2 motif that could bind to a conserved cis- element present in promoter region of many stress-induced genes. The mRNA level of CaPFl was induced by both biotic and abiotic stresses. We observed higher-level transcripts in resistance-induced pepper tissues than diseased tissues. Expression of CaPFl is also induced upon various abiotic stresses including ethephon, MeJA, cold stress, drought stress and salt stress treatments. To study the role of CPFI in plant, transgenic Arabidopsis and tobacco plants which express higher level of pepper CaPFl were generated. Global gene expression analysis of transgenic Arabidopsis by cDNA microarray indicated that expression of CaPFl in transgenic plants affect the expression of quite a few GCC box and DRE/CRT box-containing genes. Furthermore, the transgenic Arabidopsis and tobacco plant, expressing CaPFl showed tolerance against freezing temperature and enhanced resistance to Pseudomonas syrnigae pv. tabaci. Taken together, these results indicated that CaPFl is a novel EREBP/AP2 transcription factor in hot pepper plant and it may has a significant role(s) in regulation of biotic and abiotic stresses in plant.

농산폐기물(農産廢棄物)의 메탄발효(醱酵)와 그 이점(利點) (Anaerobic Digestion of Agricultural Wastes and 1ts Benefits)

  • Anaerobic digestion has recently attracted all over the world and Korea also shows no exception. The major benefits of anaerobic digestion are energy production, water pollution control, pathogen reduction and effective manure production. In Korea it was recognized in late sixties that there was a positive need to find alternative energy for farmers household. The main traditional energy sources in rural area were crop residues and forestry products. Therefore Office of Rural Development through its Rural Guidance Bureau disseminated about 29,000 household biogas units from 1969 to 1975 to provide cooking fuel for farmers household and to improve the mode of farmers living standards. The units were welcomed by farmers at that time. Now, however, most of them are not using due to a number of reasons associated with cold winter and some techno-economical problems (in those day, fossil fuel was quite expensive to compare with other prices and since then farmers income was quickly increased). The author studied on bag type household biogas plant to solve some technical problems of existing household biogas plants, but this also has little appeal for the farmers. From 1977 author studied on village scale biogas plant with two pilot plants. From the viewpoint of energy production, COD removal, kill rate of pathogen and fertilizer value, the results obtained from the experiments were quite promising, but the construction cost of the village scale biogas plant was too high for the farmers in Korea. To find most suitable biogas plant for farmers in Korea through the simplifying the biogas digester, the author developed batch-load biogas plant. By feeding coarse crop residues and manures, total solids concentrations of the batch-load biogas plant are about 28 percent which is much higher than continous digester of 5-8 percent. The batch-load biogas plant was welcomed by many farmers in Korea when it was reported on TV and newspapers. The plant was disseminated 154 units in 1982, 766 units in 1983 and 812 units in 1984 as a promissing project. Besides these biogas plant experiments, studies were also conducted 1) to determine gas production rate with agricultural wastes, 2) to evaluate the effect of loading rate, dilution, retention time on biogas production, 3) to project the amount of potencial energy from agricultural wastes.
