  • We have obtained the following results, at the production of cellulase of Chaetomium globosum and its properties of crude enzyme. 1. At the production of enzyme, wheat bran solid culture was more active than surface or shaking culture. 2. The production of enzyme was maximum between the eighth and the tenth days, but slightly decreased thereafter. 3. The optimum condition of the reactions in saccharification with CMC were obtained the following results. 1) The optimum pH was within the range of from 4.0 to 5.0 and stable pH range was within 3.5 to 6.5. 2) The optimum temperature was $40^{\circ}C$ and thermal stability was below $50^{\circ}C$ and completely inactivated at $70^{\circ}C$ 4 Dialyzed crude enzyme was activated by $Mn^{++}\;Mg^{++}\;Fe^{++}\;and\;Mo^{++}\;respectively\;but\;Hg^{++}$ was inhibited its enzyme action.

Regeneration Effects of Lespedeza cuneata Ethanol Extract on Experimental Open Wound in Rat (흰쥐의 외과적 창상에 대한 야관문 에탄올 추출물의 피부재생 효과)

  • The purpose of this study was to examine the skin regeneration ability of Lespedeza cuneata extract (LC) as well as verify its wound healing effects on an open wound in rats. Rats were divided into six groups (NO, saline treated group; CO, 1% carboxy methyl cellulose (CMC) treated group; PC, fucidin treated group; LCL, 1% LC treated group; LCM, 3% LC treated group; LCH, 5% LC treated group), and the experimental material was applied for 5 weeks. Elastase inhibition rate of the LCM group was 2.7% lower than that of butylated hydroxy anisole (BHA), which is an antioxidant. Futher, the collagenase inhibition rate of the LCM showed 7% higher activity than that of BHA. The left wound areas in the LCL group, LCM group, and LCH group after the 21st day were noticeably reduced in wound area by 54.2%, 53.5%, and 48.7%, respectively, compared to the CO group. This suggests that Lespedeza cuneata extract has healing effects on surgical wounds by promoting regeneration of skin epithelial tissue and synthesis of collagen.

  • This study was conducted to develop a robust navigator which could be in positioning for precision farming through developing a plural GPS receiver with 4 sets of GPS antenna. In order to improve positioning accuracy by integrating GPS signals received simultaneously, the algorithm for processing plural GPS signal effectively was designed. Performance of the algorithm was tested using a simulation program and a fixed point on WGS 84 coordinates. Results of this study are aummarized as followings. 1. 4 sets of lower grade GPS receiver and signals were integrated by kalman filter algorithm and geometric algorithm to increase positioning accuracy of the data. 2. Prototype was composed of 4 sets of GPS receiver and INS components. All Star which manufactured by CMC, gyro compass made by KVH, ground speed sensor and integration S/W based on RTOS(Real Time Operating System)were used. 3. Integration algorithm was simulated by developed program which could generate random position error less then 10 m and tested with the prototype at a fixed position. 4. When navigation data was integrated by geometrical correction and kalman filter algorithm, estimated positioning erros were less then 0.6 m and 1.0 m respectively in simulation and fixed position tests.

  • Transpiration cooling is the most effective cooling technique for the high-performance liquid rockets and air-breathing engines operating in aggressive environments with higher pressures and temperatures. When applying transpiration cooling, combustor liners and turbine blades/vanes are cooled by the coolant(air or fuel) passing through their porous walls and also the exit coolant acting as an insulating film. Practical implementation of the cooling technique has been hampered by the limitations of available porous materials. But advances in metal-joining techniques have led to the development of multi-laminate porous structures such as Lamilloy$^{(R)}$ fabricated from several diffusion-bonded, etched metal thin sheets. And also with the availability of lightweight, ceramic matrix composites(CMC), transpiration cooling now seems to be a promising technique for high-performance engine cooling. This paper reviews recent research activities of transpiration cooling and its applications to gas turbines, liquid rockets, and the engines for hypersonic vehicles.

Evaluation of in vivo Genotoxicity of Plant Flavonoids, Quercetin and Isoquercetin (식물유래 플라보노이드 Quercetin과 Isoquercetin의 생체 내 유전독성평가)

  • In vivo genotoxic potential of isoquercetin, a plant common flavonoid, in comparison with quercetin was investigated for the DNA breakage and the clastogenicity endpoints. Male ICR mice were administered by oral gavage for 3 days with $3{\times}0.5%$ carboxymethyl cellulose (CMC), 3 ${\times}$ isoquercetin (250, 500 mg/kg/day), 3 ${\times}$ quercetin (250, 500 mg/kg/day) and 2 ${\times}$ ethyl methanesulfonate (EMS, 200 mg/kg/day). Tissues were collected 48 hours after the first treatment and within 3 hours after the last treatment. The DNA damages were evaluated using Comet assay in liver and stomach, while the clastogenicities were determined using micronucleus test in bone marrow of same animals. The treatment of isoquercetin as well as quercetin did not cause the DNA damages in liver and stomach, and not induce the frequencies of micronucleus polychromatic erythrocytes in bone marrow. In conclusion, isoquercetin as well as quercetin did not cause the DNA breakages and the chromosomal damages in vivo system in these study conditions.

Isolation and Characteristics of Composting-promoting-bacteria (부숙촉진 미생물 분리 및 분리균의 특성)

  • This experiment was conducted to determine the safety and feasibility of using compost-promoting-bacteria. Compost-promoting-bacteria was isolated from livestock compost containing sawdust. The isolated bacteria was identified as Bacillus subtilis LYH201 by the method of the composition of the fatty acid with MIDI system and Bergey's manual. This Bacillus subtilis LYH201 had the following characteristics : Gram-positive, straight rod ($0.5{\sim}0.7{\mu}m$ width, $2.5{\sim}3.0{\mu}m$ length), facultatively aerobic and product of xylanase, CMCase, catalase, oxidase, protease and $0.5{\sim}0.7{\mu}m$-amylase. Growth of Bacillus subtilis LYH201 at saccharose as carbon source(0.5%) was faster than other carbon source. Activity of cellulase. $0.5{\sim}0.7{\mu}m$-amylase and protease from Bacillus subtilis LYH201 after 24 hours at $50^{\circ}C$ by agar diffusion method was higher than that of low temperature. Optimum growth condition of Bacillus subtilis LYH201 was $50^{\circ}C$ and pH 6.

Effects of Hydrophobic Chain Structure of Nonionic Surfactanets on Surfactant Adsorption and Diesel Removal from Kaolin Soil (비이온계 계면활성제의 소수성 구조가 카올린 토양에서 흡착 및 경유 제거에 미치는 영향)

  • The effects of hydrophobic chain length and its structure of nonionic surfactants on surfactant adsorption and contaminated diesel removal were studied in kaolin soil. Hydrocarbon chain length and double bond in hydrophobic tail group of nonionic surfactants affected surfactant adsorption and diesel removal efficiency from kaolin soil. The degrees of surfactant adsorption and diesel removal were closely related each other. Among nonionic surfactants we studied, surfactants with shorter hydrophobic chain length and higher HLB value showed lower degree of adsorption and higher efficiency of diesel removal. The existence of unsaturated carbons in the structure of hydrophobic chain enhanced diesel removal by reducing surfactant adsorption to kaolin soil. The best diesel removal was obtained after adsorption saturation was reached. If surfactant concentration was higher than a critical value, diesel removal was reduced probably because of precipitation. liquid crystal formation, or coacervation of surfactants at high concentration.

An influence on EDC/PPCPs adsorption onto single-walled carbon nanotubes with cationic surfactant (단일벽 탄소나노튜브의 미량유해물질 흡착거동에서 양이온 계면활성제의 영향에 관한 연구)

  • Recent studies have been reported the presence of Endocrine Disrupting Compounds, Pharmaceuticals and Personal Care Products (EDC/PPCPs) in surface and wastewater, which could potentially affect to the complicate behavior in coupled presence of nano-colloid particles and surfactants (adsorption, dispersion, and partitioning). In this study, the adsorption of EDC/PPCPs by Single Walled Carbon Nanotubes (SWNTs) as a representative of nano-particles in cationic surfactant solutions were investigated. Hydrophobic interactions (${\pi}-{\pi}$ Electron Donor-Acceptor) have been reported as a potential adsorption mechanisms for EDC/PPCPs onto SWNTs. Generally, the adsorptive capacity of the relatively hydrophobic EDC/PPCPs onto SWNTs decreased in the presence of cationic surfactant (Cetyltrimethyl Ammonium Bromide, CTAB). This study revealed that the competitive adsorption occurred between CTAB cations and EDC/PPCPs by occupying the available SWNT surface (CTAB adsorption onto SWNTs shows five-regime and maximum adsorption capacity of 370.4 mg/g by applying the BET isotherm). The adsorption capacity of $17{\alpha}$-ethinyl estradiol (EE2) on SWNT showed the decrease of 48% in the presence of CTAB. However, the adsorbed naproxen (NAP) surely increased by forming hemimicelles and resulted in a favorable media formation for NAP partition to increase SWNTs adsorption capacity. The adsorbed NAP increased from 24 to 82.9 mg/g after the interaction of CTAB with NAP. The competitive adsorption for EDC/PPCPs onto SWNTs is likely to be a key factor in the presence of cationic surfactant, however, NAP adsorption showed a slight competition through $CH_3-CH_3$ interaction by forming hemimicelles on SWNT surface.

  • In this study, we evaluated optimum condition of enzyme with pH and temperature for preparation of microfibillated cellulose(MFC). Well-known endo-glucanase, three enzymes were used and CMC was used for substrate. Enzyme activity was evaluated using DNS method and absorbance with UV/VIS spectrophotometer. The enzyme shown the greatest activity was reacted with pulps at optimum condition for 1 hour and treated pulps beated until 100 mL CSF. Enzyme B and Enzyme L was the higher enzyme activity below 0.1% concentration and Enzyme N was the lowest enzyme activity. At various pH and temperature conditions, enzyme activity of Enzyme B was higher than the others at the same concentration. Especially enzyme activity at $50^{\circ}C$ of Enzyme B was almost not changed over pH 6.0. Optimum condition of three enzyme was pH 6 or pH 7 and $50^{\circ}C$ or $60^{\circ}C$. Also beating efficiency of enzyme treated pulps with Enzyme B is 55.6%.

  • 급성 간질환모델에 대한 인진호 추출분획의 간장 약효검색, 방법 1. $CCl_4$ 간장해 : SD계 수컷 흰쥐에 $CCl_4$와 olive oil 혼합액(1:4v/v%)을 체중100g당 0.2$m\ell$씩 복강내 투여하였으며, 시험약물은 $CCl_4$ 혼합액 투여 4시간전 및 6시간후에 경구로 2회 투여하였다. 48시간후에 부검하여 혈청을 얻어 간기능검사 항목인 ALT 및 AST 활성을 측정하였다. 2. D-Galactosamine 간염 : SD계 수컷 흰쥐에 D-GalactosamineㆍHCl을 650mg/kg씩 복강내 투여하였으며, 시험약물은 $CCl_4$ 간장해시험과 동일하게 2회 경구투여하였다. D-Galactosamine 투여 24시간 후에 부검하여 혈청을 얻어 간기능검사 항목인 ALT 및 AST 활성을 측정하였다. 3. 담즙울체모델 : SD계 수컷 흰쥐에 ANIT 100mg을 olive oil 1$m\ell$에 현탁시켜 80$m\ell$/kg b.wt. 용량으로 1회 경구투여하였으며, 시험약물은 ANIT 투여전 2시간, 투여 후 6, 22, 28시간 간격으로 4회 경구투여하였다. ANIT 투여 47시간 후에 1시간 동안 담즙을 채취하였고, ANIT 투여 48시간째 채혈하여 혈청내 총빌리루빈치를 측정하였으며 담즙배출량, 담즙중 담즙산량도 측정하였다. 4. 약물투여 음성대조 : 1% CMC-Na 용액(10$m\ell$/kg b.wt.) 양성대조 : Silymarin(25mg/kg), UDCA(25mg/kg), DDB(37.5mg/kg) 인진호추출분획 : 인진호 원료의 수침액인 BE분획의 수득률을 기준으로 하여, KP(180mg/kg), PS-1 및 PS-2(300mg/kg), EE(500mg/kg), HH(640mg/kg), BE(1500mg/kg)
