천연염료를 이용한 한지염색에 관한 연구(IV) - 정향나무를 중심으로 - (Studies on the Dyeing of Hanji by Natural Dye-stuffs(IV) - With a focus on the clove tree -)

  • The objective of this study was to find in what color Hanji(Korean hand-made paper) is dyed when it is dyed with a pigment extracted from clove tree using different kinds of mordant, and how the paper is discolored and variety of strength under the condition of accelerated aging test. The results of this experiment are as follows. The Hanji dyed with aluminum acetate mordant was colored yellowish brown at pH 4.82, discoloration after aging was as much as a slight difference of color was recognized, and the decrease of strength after aging test was small. Thus, the method using aluminum acetate mordant was usable in coloring Hanji. The second most useful Hanji dyeing moth of was using distilled water and ferrous sulfate mordant, which dyed Hanji light brown at pH 6.03. However, when pigment was extracted using distilled water and copper sulfate mordant was used, discoloration was satisfactory but strength decreased too much and pale brown was obtained. Thus, this method was not usable.

Function of Blending Essential Oil in the Development of Anti-Dandruff Products

  • Purpose: In this paper, we show our blending ratio of 10 types of Essential Oils that survives beneficial bacteria and kills harmful bacteria in the scalp, and we investigate the possibility of application of our blending ratio to the development of anti-dandruff products and the possibility of being used as a raw material for clinical beauty and customized cosmetics. Methods: The scalp microorganisms used in our study were M. furfur, S. epidermidis, E. coli, and P. nitroreducens. There are a total of 10 Essential Oils such as True Lavender, Lime, Roman chamomile, Rosemary camphor, Cedarwood, Geranium, Clove, Tea tree, Palmalosa, and Peppermint. The antibacterial test of the blended Essential Oil was carried out according to the test method of the standardized evaluation methodology of "Food and Food Additives Code". Since M. furfur is related to the growth of sebum in the scalp, in this study we used the fnLNB and the fnLNA with 20 ㎖ of whole fat cow milk added. Results: The blending ratio of EO, which inhibits dandruff-causing bacteria such as M. furfur, S. epidermidis, E. coli, and does not inhibit P. nitroreducens showing dominant growth in a healthy scalp, was B8(Clove 0.2%, Roman chamomile 0.5%, Tea tree 0.3%), B9(Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.3%), B10(Clove 0.1%, Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.2%). Conclusion: It is thought that the blending ratio of BEO obtained as a result of this study can provide a basis for use as an alternative to antibiotics in developing anti-dandruff drugs and emerge as a new alternative to solve scalp microbial imbalance. In order for EO to be used as a useful raw material for anti-dandruff preparation, researches on 1) Standardization (the effects of products differ according to the types, regions, climate, extraction methods, etc.), 2) Antimicrobial effects, 3) Safety, etc., must be established.

수목 방향유 15종의 산화방지, 항세균, 항진균 및 항염증 효과 (Antioxidant, antibacterial, antifungal, and anti-inflammatory effects of 15 tree essential oils)

  • 본 연구는 수목 방향유 15종의 산화방지 활성, 항세균, 항진균 활성에 대해 연구하였다. 먼저 2가지 다른 실험방법을 통하여 산화방지 효과를 확인한 결과 15가지 수목 방향유 중 clove bud, 붓순나무, 왕초피나무, 곰취는 농도의존적으로 상당한 산화방지 활성을 나타내었다. 다음으로 S. aureus, L. monocytogenes, E. coli O157:H7, S. typhimurium, 그리고 S. mutans에 대한 항세균 활성은 액체배양법으로 측정한 결과 clove bud에서 현저하게 억제 효과가 나타났다. 또한, 페이퍼디스크 확산법을 통해 연구한 결과 앞에서 언급한 5가지 균주의 5, $2.5{\mu}L/disc$에서 농도의존적으로 균의 성장을 억제하였고 그 중 식중독 균주인 S. aureus의 성장에 대해 clove bud ($0.625-5{\mu}L/disc$)는 $1.25{\mu}L/disc$ 농도까지 눈에 띄게 억제 효과가 나타났다. 추가적으로 A. fumigatus, C. albicans, T. rubrum, A. ochraceus의 4가지 균주에서 clove bud의 항진균 효과가 다른 수목 방향유보다 좋았으며, 이를 바탕으로 페이퍼디스크 확산법을 통해 억제환의 크기를 측정한 결과 $1-2{\mu}L/disc$의 농도에서 양성 대조 약물인 ketoconazole ($20{\mu}g/disc$) 보다 더 우수한 효능을 나타내었다. 이 같은 결과를 종합하여 산화방지 활성과 항균 활성이 뛰어난 clove bud의 항염증 효과를 평가하였고, clove bud는 0.01% 농도에서 염증 관련 인자인 COX-2와 iNOS 단백질 발현을 LPS 처리군과 비교하여 COX-2 억제제 celecoxib와 유사하게 감소시켰다. 이와 같은 결과를 종합하면 수목 방향유는 산화방지, 항세균, 항진균, 항염증과 같이 여러 활성을 지니고 있으며, 방향유는 산화방지 및 항균성 화장품 소재로서 응용할 수 있으며, 염증반응 완화를 포함한 다양한 산업화에 응용될 수 있을 것으로 사료된다.

Grapefruit seed extract와 7종의 Essential oil 및 혼합 Essential oil의 항균 활성 (Antibacterial activity of grapefruit seed extract and seven kinds of essential and blended essential oils)

  • 본 배경 : 질염은 항생제를 복용하는 방식으로 치료를 하고 있으며, 이러한 항생제의 지속적인 치료는 내성을 유발할 수 있다. 연구 방법 : Lactic acid bacteria 2종에 에센셜 오일을 이용한 항균 활성을 보고자 한다. SE(Grapefruit Seed Extract), eucalyptus, tea tree, clove bud oil, cinnamon oil, lemongrass oil, thyme oil, ginger oil을 일정 비율로 넣어 배양 후, 병원성 미생물- E. coli, C. albicans와 Lactic acid bacteria은 균주에 맞는 배지를 사용하여 균 수를 측정하였다. 결과 : Essential oil 7종과 GSE가 병원성 미생물에 억제 효과가 있으며, 병원성균(E. coli, C. albicans)에 대한 Grapefruit seed extract(GSE)의 억제농도를 확인하였다, 병원성균은 억제하고 Lactic acid bacteria는 억제하지 않는 배합비도 확인하였다(GSE 농도가 200ppm에서 Eucalyptus globulus(EG) oil 50㎕와 Melaleuca alternifolia oil(Tea tree oil, TTO) 50㎕(pH 5.0, 5.5, 6.0)). 결론 : 본 실험에서 Essential oil은 다양한 항균 활성 가지고 있어 항생제 대안으로도 생각할 수 있으며, 장기 항생제 치료환자에 대한 보조 항균제로서도 유용할 것으로 생각된다.

식물정유의 굼벵이에 대한 살충활성 (Insecticidal Activity of Essential Oils against Whitegrub)

  • 굼벵이는 각종 작물과 나무에 피해를 줄 뿐만 아니라 특히 골프장 잔디에 피해를 주는 주요해충의 하나이다. 본 연구는 18종의 식물정유(anise oil, camphor, castor, cinnamon, clove oil, citronella, fennel, geranium, lavender, lemongrass, linseed, neem, peppermint, pine, rosemary, tea-tree, thyme, turpentine)를 이용하여 굼벵이의 환경친화적 방제 가능성을 알아보고자 실내와 온실, 야외에서 실험을 수행하였다. Cap vial실험에서 홈줄풍뎅이 3령충에 효과가 높은 정유는 anise (88.9%)와 linseed oil (100%) 및 tea-tree oil (88.9%)이었다. 온실 pot에서 왜콩풍뎅이 3령충에 대한 anise와 linseed oil의 효과는 낮게 나타났다. 야외에서 anise와 linseed 및 tea-tree oil의 효과도 대상 굼벵이의 종류에 따라 차이를 보였다. 주둥무늬차색풍뎅이 번데기에 대해서는 tea-tree oil 처리에서만 32.6%의 보정사충율을 보였으나 등얼룩풍뎅이 3령충에 대해서는 anise와 linseed, tea-tree oil이 각각 54.8%, 51.6%, 56.5%의 보정사충율을 보였다.

Antifungal Activities of the Essential Oils in Syzygium aromaticum (L.) Merr. Et Perry and Leptospermum petersonii Bailey and their Constituents against Various Dermatophytes

  • This study was carried out in order to investigate the potential of using plant oils derived from Leptospermum petersonii Bailey and Syzygium aromaticum L. Merr. Et Perry as natural antifungal agents. The antifungal effects of essential oils at concentrations of 0.05, 0.1, 0.15, and 0.2 mg/ml on the dermatophytes Microsporum canis (KCTC 6591), Trichophyton mentagrophytes (KCTC 6077), Trichophyton rubrum (KCCM 60443), Epidermophyton floccosum (KCCM 11667), and Microsporum gypseum were evaluated using the agar diffusion method. The major constituents of the active fraction against the dermatophytes were identified by gas chromatography-mass spectrometry and high-performance liquid chromatography analysis. The antifungal activities of S. aromaticum oil (clove oil) against the dermatophytes tested were highest at a concentration of 0.2mg/ml, with an effectiveness of more than 60%. Hyphal growth was completely inhibited in T. mentagrophytes, T. rubrum, and M. gypseum by treatment with clove oil at a concentration of 0.2 mg/ml. Eugenol was the most effective antifungal constituent of clove oil against the dermatophytes T. mentagrophytes and M. canis. Morphological changes in the hyphae of T. mentagrophytes, such as damage to the cell wall and cell membrane and the expansion of the endoplasmic reticulum, after treatment with 0.11 mg/ml eugenol were observed by transmission electron microscopy (TEM). At a concentration of 0.2 mg/ml, L. petersonii oil (LPO) was more than 90% effective against all of the dermatophytes tested, with the exception of T. rubrum. Geranial was determined to be the most active antifungal constituent of L. petersonii oil. Taken together, the results of this study demonstrate that clove and tea tree oils exhibited significant antifungal activities against the dermatophytes tested in this study.

김치의 선도유지를 위한 천연보존제의 탐색 (Screening of Natural Preservatives to Inhibit Kimchi Fermentation)

  • As a primary step to develop natural preservative for extending the shelf-life of kimchi, the effect of 102 edible plants, 21 antimicrobial agents and related compounds on kimchi fermentation was studied. Among 42 oriental medicinal plants tested, Baical skullcap and Assam indigo were found to be highly effective for maintaining the fresh state of kimchi. Although Bugbane, Red mangolia, Bushy sophora, Szechuan pepper, Chinese quince and Scisandre significantly inhibit the growth of Lactobacilli, their effect was not high enough to be used as raw materials for kimchi preservative. When the effect of 32 herbs and spices was tested, peppermint, cinnamon, lemon balm, clove, hop, rosemary, sage, horseradish and thyme showed high antimicrobial activity against kimchi microorganisms. Among them, the effect of clove ranked top. When it was added to fresh kimchi, initial cfu value ($2.4{\times}10^{6}cfu/g$) changed little even after 2 day's fermentation ($2.6{\times}10^{6}cfu/g$). Sensory test was not a good criteria to evaluate the effect of herbs and spices, since their highly specific flavors affected the taste of kimchies. Twenty eight fruits, vegetables and related plants were tested, but only leaves of pine tree, persimmon and oak leaves showed a significant bactericidal effect, finally contributing to the storage of kimchi. In addition, when 21 natural preservatives and other compounds were added individually to fresh kimchi, nisin and caffeic acid could inhibit fermentation.

Dyeing and Color Fastness Properties of Natural Dyed Actual Size Hanji

  • After manufacturing the natural dyed actual size Hanji using 11 kinds of plant natural dyeing materials and 2 kinds of animal natural dyeing materials, the color characteristics and color change and color fastness after post-mordanting were compared and discussed. The hues of 13 types of natural dyed Hanji were black, PB, and RP, each with 1 type, YP with 3 types, and Y with 7 types. Among the natural dyeing materials, Chinese ink, indigo and Lac showed high color yield and color difference, and violet-root cromwell and gardenia seeds showed low color yield. The color fastness of Hanji dyed with turpentine diluted Ottchil, Pagoda tree seeds, Chinese ink and indigo was excellent, while that of gardenia seeds and violet-root cromwell were very poor. After post-mordanting of natural dyed Hanji with Al, Cu, and Fe mordants, the hue changes were show up the Alnus firma, clove, lac and cochineal. In addition, the color difference was very diverse and was overall the most biggest due to Fe mordant. After 72 hr. of UV irradiation on post-mordanting natural dyed Hanji, hue change was observed in 3 types and color fastness was improved in 8 types by post-mordanting. The Hue and color fastness are significantly different depending on the type of natural dying materials and post-mordants. Therefore, when dyeing Hanji with natural dyes, it is necessary to dye with sufficient knowledges and informations about the desired color and fastness.

Investigation of bacteremia after toothbrushing in orthodontic patients

  • Objective: The aim of this study was to investigate the occurrence rate of bacteremia following toothbrushing with toothpastes composed of several antibacterial agents and compare the results with the conventional oral hygiene maintaining methods in orthodontic patients. Methods: This clinical study included 100 adult orthodontic patients who were divided into 4 groups. Each group comprised of 25 patients, wearing fixed orthodontic appliances. In the first group, bacteremia was assessed after toothbrushing without using any toothpaste. In the second group, a 0.2% chlorhexidine gluconate mouthrinse was used before brushing with no toothpaste. In the third group, subjects brushed with a commonly used toothpaste which did not include an additional antimicrobial agent. The fourth experimental group used toothpaste which included tea tree oil, clove oil, peppermint oil and bisabolol as antimicrobial elements. Pre- and post-brushing blood samples were obtained using a strict aseptic technique. All samples were microbiologically evaluated using blood culture bottles. Results: Toothbrushing in orthodontic patients yielded to an increase in the occurrence rate of bacteremia when using normal toothpaste or no toothpaste at all. Conclusions: The use of chlorhexidine mouthwash before toothbrushing, and brushing with antimicrobial toothpaste did not show a statistically significant difference in preventing the occurrence of bacteremia (p > 0.05).

Neem과 mustard oil이 곤충병원성 선충과 누에에 미치는 영향 (Effect of Neem and Mustard oils on Entomopathogenic Nematodes and Silkworm)

  • 13종의 식물 정유(anise oil, clove oil, marigold, mustard oil, neem oil, quassia, quilaja, rosemary oil, rotenone, tea tree extract, thyme oil, wintergreen oil, and yucca)와 caffeine이 산업곤충인 누에(Bombyx mori)와 생물적 방제 인자의 하나인 곤충병원성 선충 Steinernema carpocapsae GSN-1 계통(Sc)과 Heterorhabditis sp. Gyeongsan 계통(Hg)에 미치는 영향을 실내 검정하였다. 1,000 ppm 농도의 식물체 추출물들 중 neem oil이 누에에 대한 살충활성이 가장 높았다. Neem oil을 처리한 뽕잎을 공급하였을 때, 급상 5일과 10일 후 누에의 치사율은 각각 55.3%와 100%였다. 그리고 neem oil이 처리된 뽕잎을 섭식한 누에는 번데기와 고치를 형성하지 못하였다. Rotenone을 처리한 뽕잎을 공급받은 누에의 고치와 번데기 무게는 각각 0.27 g과 1.01 g으로 가장 적었다. Mustard oil은 곤충병원성선충에 대하여 살선충 활성이 높았다. X-plate에서는 mustard oil 20 ppm 처리 시 곤충병원성선충 Sc와 Hg의 치사율이 처리 3일 후 각각 69.0%와 100%였으며 5 ppm 농도에서도 4%와 36%의 치사율을 보였다. Sand barrier에서 Sc를 100 ppm 농도의 mustard oil과 혼합 처리 시 꿀벌부채명나방(Galleria mellonella) 노숙 유충의 치사율은 무처리구와 차이가 없었으나 Hg의 경우는 무처리구에 비하여 30% 낮은 꿀벌부채명나방 치사율을 보였다. 꿀벌부채명나방 유충의 체내에 정착한 선충 수는 Hg가 Sc에 비하여 적었다. Sand barrier내 생존 Sc 선충 수는 200 ppm 이하 농도에서는 무처리구와 차이가 없었다.
