Effects of Antibiotics on Suppression of Agrobacterium tumefaciens and Plant Regeneration from Wheat Embryo

  • Antibiotics used for suppressing Agrobacterium in plant transformation procedure might have negligible effects on plant tissues and regeneration. The effects of antibiotics on growth suppression of Agrobacterium and plant regeneration were investigated for enhancing Agrobacterium-mediated transformation using wheat mature embryos. Antibiotics tested, except carbenicillin, were able to suppress that embryos were coated with a layer of Agrobacterium cells in callus induction medium. Agrobacterium growth was suppressed minimally at 50 mg/l of timentin, while cefotaxime and clavamox were completely suppressed at relative high concentration of 250 mg/l. In the treatment of carbenicillin, initiation of growth suppression of Agrobacterium occurred at 750 mg/l of concentration because Agrobacterium KYRT1 contains the carbenicillin resistant gene. In Agrobacterium inoculation, effects of antibiotics were significantly different on the rate of callus induction and shoot formation. Almost embryos were induced calli at 50 mg/l of timentin whereas callus induction rate was achieved above 90% at 100 mg/l and 250 mg/l of cefotaxime and clavamox, respectively. Shoot formation rate was higher in the treatment of timentin than that of cefotaxime and clavamox at 500 mg/l of concentration, respectively. Timentin can be used as a good antibiotics in Agrobacterium-mediated wheat transformation.

개에서 피부개선충(Canine Scabies)과 귀진드기(Ear Mite) 혼합감염에 대한 주 2회 selamectin 적용 일례 (Efficacy of Twice a Week Selamectin Application as a Treatment for Mixed Canine Scabies and Ear Mite Infestation in a Dog)

  • 6개월령 수컷 아메리칸 코커스파니엘이 심한 소양감, 탈모 그리고 머리를 심하게 흔드는 증상으로 내원하였다 신체검사에서 전신적으로 가피. 비듬, 구진. 발적의 병변이 관찰되었다. 전반적인 피부검사결과 개선충, 귀진드기, 구균과 Malasseria의 혼합 감염으로 진단하였다. Selamectin 6mg/kg을 주 2회로 총 4회 경부 피부에 적용하였다 마지막 적용 후 14일에 피부를 재검사 하였을 때 개선충과 귀진드기는 관찰되지 않았으며 소양증과 비듬의 증상이 현저하게 개선되었다. 개선충과 귀진드기에 대한 치료가 끝난 후 구균과 Malassezia 치료를 위해 clavamox 와 ketoconazole을 3주간 투여 하였으며 동시에 sebolytic 샴푸와 chlorohexidine 샴푸를 3주동안 주 2회 실시하였다. 2개월 후 피부 증상은 현저히 감소하였고 피부 병변은 회복되었다. 이 치료법에 따른 selamectin 적용은 고가이지만 개선충과 귀진드기가 감염된 개에서 매우 효과적이며 임상적으로 scabies 감염증의 치료적 진단법으로 유용한 제제로 판단된다

Optimization of Agrobacterium-mediated transformation procedure for grapevine 'Kyoho' with carrot antifreeze protein gene

  • We report an Agrobacterium-mediated transformation procedure optimized for 'Kyoho' that is a major table grapevine cultivar in Korea, and its transgenic plants with antifreeze protein gene of carrot (DcAFP). The full length of DcAFP coding region in accordance with the previous report was isolated from young leaves of carrot and recombined into a plant transformation vector. Ethylene inhibitors such as silver nitrate and aminoethoxyvinylglycin (AVG) supplemented in a co-cultivation medium distinctly increased frequency of shoot regeneration when explants were sub-cultured in a selection medium: particularly ten-fold higher in treatment with 0.1 mg/L AVG than one without ethylene inhibitor. Among various antibiotics and their concentrations, the combination of 150 mg/L cefotaxime plus 150 mg/L $Clavamox^{TM}$ was selected for elimination of Agrobacterium cells in addition to minimization of adverse effect on shoot regeneration, while 50 mg/L kanamycin monosulfate effectively suppressed regeneration of non-transgenic shoots. Applying the elucidated culture condition, we finally obtained a total of 5 transgenic 'Kyoho' plantlets with DcAFP, of which integration with the grapevine genome and transcription was confirmed by nucleic acid analyses.
