Taxonomic Revision of the Genus Chirosia Rondani (Diptera: Anthomyiidae) in Korea (한국산 양치꽃파리속의 분류학적 재검토)

  • In this paper, a total of 11 species of the genus Chirosia are treated. Among them, a species, Chirosia flavipennis (Fallen, 1823), is newly added to Korean fauna. The keys to Korean species, host plants and domestic localities are given. Some illustrations of male genitalic features are included. The cladistic analysis of the Korean species is performed using Musca domestica (Muscidae), Scathophaga stercoraria (Scathophagidae), Delia platura (Anthomyiidae) and Anthomyia angulata (Anthomyiidae) as outgroups.

  • 혹무늬 톡토기과의 56개 아과들에서 20개 형질복합을 추추랗여 형질평가방법과 Wagner algorithm의 형질 비평가방법을 사용하여 계통수를 작성, 비교하였다. 전자의 방법을 이해 진화적 형질순서와 극성을 결정하고 고등급의 파생공유 형질을 토대로한 단계통군과 자매군을 확인하여 계통분화를 추적하였다. 이 두가지 방법에 의한 계통수에서 분지 방식은 매우 다르게 나타났으나 나열순서는 유사했고, 단지 마지막의 2개 아과가 후자에서는 반대로 나타났다. 그 원인은 후자의 방법으로는 파생혈질수가 많은 분류군에서의 형질상태가 Wagner algorithm에서 누적효과를 일으킨 때문인 것으로 생각되며, 아울러 1, 10 및 20 번 형질의 적응이동(adaptive shift) 상 나타낼 중요한 의의를 적절히 평가 반영할 수 없는 것으로 추정된다.

A Sustematic Study of Korean Entomobryidae(Collembola, Insecta) Based on Cladistic Analysis of Phenotypic nd Allozyme Data (한국산 털보톡토기과(곤충강)의 표현형과 효소유전자형질에 대한 분지분석)

  • 한국산 털보톡토기과(Collembola, Insecta)의 계통유연관계를 알아보기 위해 이 과에 속하는 3개속(Sinella, Entomobrya, Homidia)의 6종과 가시톡토기과의 1종(Tomocerus kinoshitai)에 대하여 모서식과 형태적 조사 외에 전기영동을 이용한 효소분석을 시리시하였다. 이들 모든 조사에서 32개의 형태형질과 33개의 모서식형질 및 80개의 효소유전자 형질을 얻어 분지분석하였는데, 이중 효소유전자에 대해서는 UPGMA방법도 적용하였다. 이러한 분석 결과 다양한 분지도를 얻었으나, Homidia종들은 언제나 두 개의 무리로 분리되었으며, Homidia munda의 두 아종이 가장 유연관계가 높았다. 반면에 Homidia koreana의 두 개체군은 다소 분리되어 같은 종으로 보기 의심스럽다는 소견을 얻었다. 또한 형태형질분석으로 얻은 계통수에서 가시톡토기과의 Tomocerus종이 털보톡토기과의 속들 사이에 끼어들어 나타나는데 이것은 형질에 따라 비중을 달리두는 평가방법이 아니기 때문에 나타난 것으로 추측된다. 본 연구 결과 대체로 외부형질과 모서식, 그리고 효소유전자의 세가지 유형의 형질들을 종합 분석할 때 도출되는 계통수가 자연분류에 가장 가까운 근연성을 보이는 것으로 판단되었다.

Phylogenetic Study of Two Problematic Subgenera of Tomoceridae (Insecta : Collembola) from Korea (한국산 가시톡토기 과 (곤충 강: 톡토기 목)의 문제 2아속의 계통분화)

  • Two problematic subgenera of Tomoceridae (Collembola, Insecta) were investigated for their phylogenetic relationships based on morphological characters and allozyme study from three relevant species. Different dendrograms were obtained obtained between morphological and allozyme studies. The morphological data did not give rise to any result distinctive enough to separate the two subgenera whereas the allozyme analysis produced a clear separation by the high genetic distance value. They were consistent, however, whether given rise to by using distance or cladistic methods and also whether character weighting or unweighting approaches employed in the morphological character analysis. As a consequence, it is strongly suggested that any prominent morphological trait might work as a good taxonomical character when supported by a strong genetic divergence as evidenced by allozyme analysis for instance.

  • Polymerase chain reaction(PCR)-based inter-simple sequence repeats(I-SSR) markers were analyzed in 48 megagametophytes of a single tree of Abies koreana $W_{ILS}$. Nineteen of the 35 primers, screened with 6 megagametophyte DNA and produced the clearest amplification products in the preliminary experiment, were used for PCR with 48 megagametophyte DNAs sampled from a single tree. On the basis of the chi-square test, a total of 51 amplicons, amplified by the 19 primers, were revealed to be segregated according to the Mendelian ratio(i.e., 1 : 1 segregation ratio) in the 48 megagametophytes at 5% significance level. Based on the linkage analysis, the observed 51 Mendelian loci turned out to be unlinked each other, which suggested that they are evenly distributed in the genome. However, majority of RAPD markers are known to belong to the independent linkage blocks, which frequently results in the amplification of RAPD markers from the restricted regions of the genome. Owing to the nature of even distribution of the 51 loci observed in this study, the I-SSR markers could give better resolution of estimating genetic diversity from the whole genome than RAPD markers. And I-SSR markers are also more suitable than RAPD markers for reconstructing phylogenetic relationship by a cladistic method which requires to fulfil the assumption of independent evolution of the different characters.

The morphological study of Galium L. (Rubiaceae) in Korea (분계분석을 이용한 한국산 갈퀴덩굴속(Galium L.) 식물의 외부형태학적 연구)

  • To clarify the morphological variations of Galium, its morphological characteristics were examined using maximum parsimony analysis. The key characteristics as well as the range of variation in each characteristic were investigated at the species level and were employed to elucidate the relationships between the taxa. This study confirmed that species belonging to genus Galium formed a monophyletic group and comprised two main clades. Sect. Cymogaliae and sect. Leptogalium are polyphyly groups, and other sections are monophyletic group. This study also suggests that the fruit hairs, the types of petal apexes, and the colors of the petals are the most valuable taxonomic characteristics for differentiating different sections. The numbers of the leaves and leaf shape provide useful taxonomic characteristics for the identification of different species.

Molecular Diversity of pagA Gene from Baciilus anthracis (탄저균 pagA 유전자의 분자적 다양성)

  • Bacillus anthracis is a gram-positive spore-forming bacterium that causes the disease anthrax. The anthrax toxin contains three components, including the protective antigen (PA), which binds to eucaryotic cell surface receptors and mediates the transport of toxins into the cell. In this study, the entire 2,294-nucleotide protective antigen gene (pag) was sequenced from 4 of B. anthracis strains to identify potential variation in the toxin and to further our understanding of B. anthracis evolution in Korea. Sequence alignment of the entire PA gene from 30 strains representative of the four B. anthracis diversity groups revealed mutations. The mutation of B. anthracis BAK are located adjacent to a highly antigenic region crossing the junction between PA domains 3 and 4 shown to be critical to LF binding. The different mutational combinations observed in this study give rise to 11 PA genotypes and 4PA phenotypes. Three-dimensional analysis of all the amino acid changes (Ala to Val) observed in BAK indicated that these changes are not only close sequentially but also very close in three-dimensional space to the antigenic region importan tfor LF binding. Phylogenetic (cladistic) analysis of the pag corresponded with previous strain grouping based on chromosomal variation, suggesting that plasmid evolution in B. anthracis has occurred with little or no horizontal transfer between the different strains.

Taxonomic study of the genus Diplazium (Woodsiaceae) in Korea (한국산 참빗고사리속(우드풀과)의 분류)

  • Cladistic analysis was performed to reveal the interspecific relationship of nine taxa of Korean Diplazium (Woodsiaceae). A total of 29 taxa with 20 taxa of eastern asiatic species as ingroup and three taxa of Athyrium as outgroup, were analyzed with 17 morphological characters. Diplazium nipponicum, recently found from Jeju Island, was most closely related to D. chinense by having the acuminate apex of pinnule segment and fascicled fronds. Diplazium nipponicum was distinguished from D. chinense by having scales and pinnule segments of serrated margin, and non-overlapped pinnules. Diplazium nipponicum and D. chinense were closely related to D. mesosorum and D. hajizoense in sequence, by having long acute apex in pinnae. These species formed same clade with D. subtripinnatum, which is endemic to Japan and D. kawakamii, which is distributed in Japan and Taiwan.

Sceptridium atrovirens and S. microphyllum (Ophioglossaceae): First report in Korea and its phylogenetic position based on morphology (숲고사리삼과 애기고사리삼(고사리삼과): 한반도 미기록 식물의 형태로 본 계통적 위치)

  • Two unrecorded species (Sceptridium atrovirens and S. microphyllum), previously known only in Japan, were found in a forest in Dosun-dong, Seogwipo-si, Jeju-do for the first time in Korea. S. atrovirens was distinguished from other related species of the genus Sceptridium in Korea by having the thickest rhizome and root, irregularly crenulate or roughly serrate, and a shorter sporophore blade. The local name was newly given as 'Sup-go-sa-ri-sam' for the species, denoting the habitat. S. microphyllum was distinguished by having the smallest plant height, compact sterile blades, and partly lateritious tropophyll in winter. The local name was newly given as 'Aegi-go-sa-ri-sam' for species, meaning of a small size. To reveal the interspecific relationships within the genus Sceptridium in Korea, cladistic analysis was performed for 27 morphological characteristics. The morphological characteristics and illustrations of the two taxa, as well as photographs at the habitat, are provided with a taxonomic key to the species of Sceptridium in Korea.

Systematic Relationships Among Ten Species of Subgenus Drosophila (Diptera: Drosophilidae) in Korea (한국 초파리아속(쌍시목: 초파리과) 10종에 대한 계통적 유연관계)

  • This study sought to observe systematic relationships through taximetrical analyses by morphological characters among the 10 species, on three species in the virilis section (D. virilis, D. tsigana and D. lacertosa) and seven species in the quinaria section (D. angularis, D. brachvnephros, D. curvispina, D. kuntzei, D. nigromaculata, D. takadai and D. unispina) of the subgenus Drosophila. In the cluster and the cladistic analysis among the members of subgenus Drosophila, 10 species was divided into the 1st group of D. virilis, D. tsigana, and D. lacertosa and the 2nd group of D. angularis, D. brachynephros, D. curvispina, D. kuntzei, D. nigromaculata, D. takadai, and D. unispina. In cluster analysis, the 2nd group had three sister groups; one sister group that clustered D. angularis and D. brachynephros then D. unispina was clustered to them, another sister group clustered D, curvispina and D. takadai then D. kuntzei was clustered to them and the other sister group of D. nigromaculata. In the 10 species, D. virilis and D. lacertosa were the first to be divided and then D. tsigana. Although 1st group which D. virilis was belonged can be determined as more primitive than the 2nd group, it seemed that this group was not the direct ancestor of the 2nd group, rather there should be another ancestor. Among the quinaria species group, D. nigromaculata was the first to be divided and D. kuntzei was the most recent species to be divided.
