팔당호 섬모충 플랑크톤의 분류 및 생태학적 연구 (Taxonomic and Ecologic Studies on Ciliate Plankton in Pal'tang Reservoir, Korea)

  • 국내산 담수 섬모충 플랑크톤의 분류 및 생태학적 특성을 파악하기 위해 2000년 12월 15일부터 2001년 12월6일까지 팔당호에서 표층수를 채집하였다. 조사결과 총 6목 7과 10속 12종의 우점종이 분류, 동정 되었으며 이들은 모두 국내 미기록 종으로서 생태학적 특성과 함께 기재하였다. 우점적으로 동정된 섬모충 플랑크톤의 대부분은 부영양수역에서 보고된 종들이었으며, 저수온기인 동계와 춘계에는 nanoplankton을 먹이로 하는 비교적 대형종인 Codonella cratera, Rimostrombidium lacustris, Strobilidium caudatum, Limnostrombidium viride, Pelagostrombidium fallax, Stylonychia sp., Pseudostrombidium planktonticum, Phascolodon vorticella등이 출현하는 반면, 고수온기인 하계와 추계에는 박테리아를 먹이로 하는 Rimostrombidium hyalinum, Rimostrombidium brachykinetum, Halteria gradinella, Vorticella convallaria와 같은 소형종들이 우점하였다.

  • Eight Holstein cows weighing 419-673kg with 2 to 10 years of age were allocated into two groups, four animals each, to study the effect of feeding urea and urea plus Zeolite on rumen pH and the population of rumen ciliates. Rumen fluid samples were taken after morning feed for 7 times at one hour intervals. Experimental results obtained were as follows; 1. Rumen pH was gradually dropped from 7.272 to 7.163 by 3.01 hour and from 7.18 to 7.07 by 2.87 hour and reached 7.352 and 7.29 at 7 hour after feeding, respectively on urea and urea plus Zeolite feeding group. 2. Total ciliate numbers decreased from 209,540 to 113,755 by 4.311 hour and from 381,430 to 203.125 by 4.406 hour and gradually increased to 151,030 and 265,230 by 7 hour after feeding, respectively on urea and urea plus Zeolite feeding group. 3. Population of ruminal ciliates was not changed and Entodinium simplex was the major ciliate population for both treatments. It was from 81.21 to 89.12% on urea feeding group and from 84.6 to 88.3% of total number of ciliates on urea plus Zeolite feeding group.

Growth and ingestion rates of heterotrophic dinoflagellates and a ciliate on the mixotrophic dinoflagellate Biecheleria cincta

  • To explore the interactions between the mixotrophic dinoflagellate Biecheleria cincta (previously Woloszynskia cincta) and heterotrophic protists, we investigated whether the common heterotrophic dinoflagellates Gyrodinium dominans, Gyrodinium moestrupii, Gyrodinium spirale, Oxyrrhis marina, and Polykrikos kofoidii, and the ciliate Strobilidium sp. were able to feed on B. cincta. We also measured growth and ingestion rates of O. marina and Strobilidium sp. on B. cincta as a function of prey concentration. In addition, these rates were measured for other predators at single prey concentrations at which the growth and ingestion rates of O. marina and Strobilidium sp. were saturated. All grazers tested in the present study were able to feed on B. cincta. B. cincta clearly supported positive growth of O. marina, G. dominans, and Strobilidium sp., but it did not support that of G. moestrupii, G. spirale, and P. kofoidii. The maximum growth rates of Strobilidium sp. and O. marina on B. cincta (0.91 and 0.49 $d^{-1}$, respectively) were much higher than that of G. dominans (0.07 $d^{-1}$). With increasing the mean prey concentration, the specific growth rates of O. marina and Strobilidium sp. on B. cincta increased, but either became saturated or slowly increased. The maximum ingestion rate of Strobilidium sp. (1.60 ng C $predator^{-1}\;d^{-1}$) was much higher than that of P. kofoidii and O. marina (0.55 and 0.34 ng C $predator^{-1}\;d^{-1}$) on B. cincta. The results of the present study suggest that O. marina and Strobilidium sp. are effective protistan grazers of B. cincta.

Feeding by common heterotrophic protist predators on seven Prorocentrum species

  • Species belonging to the dinoflagellate genus Prorocentrum are known to cause red tides or harmful algal blooms. To understand the dynamics of a Prorocentrum sp., its growth and mortality due to predation need to be assessed. However, there are only a few Prorocentrum spp. for which heterotrophic protist predators have been reported. We explored feeding by the common heterotrophic dinoflagellates Gyrodinium dominans, Oxyrrhis marina, Pfiesteria piscicida, Oblea rotunda, and Polykrikos kofoidii and the naked ciliate Strombidinopsis sp. (approx. 90 ㎛ cell length) on the planktonic species Prorocentrum triestinum, P. cordatum, P. donghaiense, P. rhathymum, and P. micans as well as the benthic species P. lima and P. hoffmannianum. All heterotrophic protists tested were able to feed on the planktonic prey species. However, O. marina and O. rotunda did not feed on P. lima and P. hoffmannianum, while G. dominans, P. kofoidii, and Strombidinopsis sp. did. The growth and ingestion rates of G. dominans and P. kofoidii on one of the seven Prorocentrum spp. were significantly different from those on other prey species. G. dominans showed the top three highest growth rates when it fed on P. triestinum, P. cordatum, and P. donghaiense, however, P. kofoidii had negative growth rates when fed on these three prey species. In contrast, P. kofoidii had a positive growth rate only when fed on P. hoffmannianum. This differential feeding on Prorocentrum spp. between G. dominans and P. kofoidii may provide different ecological niches and reduce competition between these two common heterotrophic protist predators.

The First Case of Diarrhoea in Tibetan Sheep, Ovis aries, Caused by Balantidium coli in the Qinghai Tibetan Plateau Area, China

  • This study was carried out to determine the pathogen-causing diarrhoea in sheep Ovis aries in the Qinghai Tibetan Plateau Area, China. A trophozoite was identified as species of ciliate alveolates infecting the sheep based on morphological characteristics examined by microscope. It was mostly spherical, colourless and transparent, with many vesicles. Macronucleus and contractile vacuoles could not be distinguished. Size of the trophozoite was $80-180{\times}70-150{\mu}m$ and its surface was covered with cilia. Molecular analysis based on sequences of 18S rRNA and ITS genes confirmed the ciliate species as Balantidium coli. According to the literature, there have been many epidemiological investigations of B. coli infection in pigs, monkeys and humans. To our knowledge, this was the first report of B. coli infections in sheep in the Qinghai Tibetan Plateau Area of China, or eleswhere around the world. Importantly, the sheep case was rare but raised our concern that B. coli may spread across species and expand its host range.

Non-specific activation of mouse peritoneal macrophages by a freshwater ciliate, Tetrahymena pyriformis

  • Toxoplasma-killing activities of mouse peritoneal macrophages activated by the extracts of Tetrahymena pyriformis (Korean and Chinese strains) were evaluated, and the active protein fractions from both strains were partially characterized by a method including chromatographies and SDS-PAGE. The first peak in Korean strain and the second peak in Chinese strain of T. pyriformis obtained by DEAE-Sephadex A-50 chromatography were most effective in the activation of macrophages to kill Toxoplasma gondii tachyzoites in vitro. Subsequent fractionations of obtained peak fractions were performed on a Sephadex G-200 gel. The first peaks fractionated from both strains of T. pyrtyomis had the highest toxoplasmacidal activities, and when subjected to the SDS-PAGE, one prominent band was visualized for each of the strains showing the same molecular weight of ca. 52.6 kDa. This active protein is suggested to be related to non-specific activation of mouse peritoneal macrophages.

전기방전식 잡초방제기 개발을 위한 기초연구 (Basic Studies on Development of Electrical Weed Control System)

  • Motivated by the need fir developing the new method of weed control in place of chemical weedicide, this paper aims at making good use of electricity against environmental pollution. Compared with chemical weedicide, the electric method of weed control is functionally more versatile, effectively more rapid, and lower in terms of cost. In particular, this method will contribute to environmental protection. In detail the electrical weed control system which is the simple circuit for generating the electric current of high voltage is comprised of step-up transformer which rectifies the current and the capacitor which stores the energy. The effectiveness of electrical weed control system is evaluated by germination rate and control of Dgitaria Sangvinalis(Galinsoga ciliate). As a result, the electrical weed control system(high voltage spark discharge) can kill weeds effectively

In vitro efficacy of formalin, hydrogen peroxide and copper sulfate on the scuticocilliate Uronema marinum at low salinity

  • The scuticocilliate, Uronema marinum is a histophagous ciliate and the causative agent of 'scuticociliatosis'in cultured olive flounder Paralichthys olivaceus. In the present study, in vitro efficacy of hydrogen peroxide, formalin and copper sulfate on the scuticocilliate at low salinity was investigated. Each chemical showed synergistic parasiticidal effects with low salinity (salinity in 5 ppt) compared to each chemical alone (salinity in 33 ppt). At low salinity (5‰), ciliates were killed completely within 1.5h by exposure to 50ppm formalin (37% formaldehyde), at 100ppm hydrogen peroxide (30% solution) and at 100ppm copper sulfate (20% solution). The formalin was the most effective chemical against the parasites at low salinity.

Morphology of one Frontonia and two Spathidium ciliates (Ciliophora: Intramacronucleata) from Korea

  • Three ciliate species, Frontonia schaefferi Bullington, 1939, Spathidium curiosum Foissner, 2016, and S. muscicola Kahl, 1930, were collected from freshwater and terrestrial habitats in Korea. Their morphology was investigated based on live observations and protargol impregnation. Frontonia schaefferi was characterized by a cell size of $89-112{\times}45-53{\mu}m$ (after protargol impregnation) and three vestibular kineties. Spathidium curiosum was characterized by a cell size of ca. $125{\times}25{\mu}m$ in vivo and the unusual shape of the extrusomes. Spathidium muscicola was characterized by a cell size of ca. $135{\times}40{\mu}m$ in vivo and four to six macronuclear nodules. These three species are new records for Korea.

Subcellular Localization of GTP Binding Protein in Stentor coeruleus

  • The heterotrichous ciliate Stentor coeruieus shows a step-up photophobic response to visible light In the previous paper, the existence of GTP-binding proteins was confirmed by using the antisera against the carboxy terminal decapeptide of transducin $\alpha$ subunit. The photoreceptor, stentorin, is localized in the pigment granule. If the immunoreactive G-protein directly interacts with the photoreceptor stentorin, the G-protein expected to be located in the pigment granule rather than plasma membrane. To elucidate the function of the immunoreactive G-protein, the localization of the G-protein in Stentor coeruleus was studied. The results suggest that this G-protein is located in the myoneme involved in the contraction and extension of the cell rather than in the pigment granule.
