Effect of Fibrinogen Genotype and Other Characteristics on Plasma Fibrinogen Levels

  • This study was carried out to investigate the effect of fibrinogen genotype and other characteristics on the plasma fibrinogen levels. Many studies have conformed that high plasma fibrinogen levels are associated with the increased risk of ischaemic heart disease, stroke and arterial disease. And fibrinogen levels are related with age, obesity, cholesterol and alcohol consumption, genotypes. For this study the blood samples were collected from 93 healthy Koreans (66 males and 27 females). The blood samples were individually analyzed by smoking status, cholesterol levels, genotype, age, and gender. The plasma fibrinogen was assayed by clotting method (modified Clauss assay) and cholesterol was assayed by cholesterol oxidase method. Subjects were classified by current smokers, ex-smokers (<6 month), or nonsmokers. The $\beta$-fibrinogen genotype was detected by PCR of relevant region and digestion with HaeIII, with the H$_1$H$_1$ allele allowing cleavage by this restriction enzyme and H$_2$H$_2$ allele being refractory. In conclusion, the study shows that the factor of the increasement in the fibrinogen level was closely related with the cholesterol level, smoking status and genotype (H$_1$H$_2$); but there was no significant difference by gender, Especially, among the people over 50 years of age, fibrinogen level was higher with the increasement of cholesterol level (<200 mg/dl), current smoker, and genotype H$_1$H$_2$.

The Effect of Medium Chain Triglyceride Diet on the Serum Lipids and Lipoprotein Levels of Rats Fed with Cholesterol (Medium Chain Triglyceride첨가(添加) 식이(食餌)가 Cholesterol투여(投與) 흰쥐의 혈중(血中) 지질(脂質) 및 Lipoporotein에 미치는 영향(影響))

  • The purpose of the study was to find an effect of medium chain triglyceride (MCT)diet on the serum lipids and lipoprotein of Sprague-Dawley rats when the were fed with cholesterol. All experiment-groups were fed with different diet such as soybean oil, shortening,MCT and MCT mixed with oil respectively. After fed to each group, the levels of serum lipids and lipoprotein among fed rats were examined and the data from the experiment were compared with a control group. The results obtained from the study are as follows : (1) The average boby weight gain in MCT diet and soybean oil diet groups were lighter than that of the control group, while the food efficiency ratio in each experimental diet group was almost same as than in the control group. (2) Total cholesterol levels in serum of all experimental diet groups were lower than that of the control group except the level in shortening diet group. Especially, the levels of total cholesterol in MCT diet and soybean oil diet groups were significantly lower than that of the control group. (3) VLDL, LDL-cholesterol levels of all experimental groups were almost same except the case of the MCT diet and soybean oil diet group which was significantly low. (4) The ratio of VLDL, LDL-cholesterol to HDL-cholesterol, so called, atherogenic index in serum of MCT and soybean oil diet groups were significantly lower than that of the control group. (5) The ratio of total cholesterol to phospholipids in serum of MCT diet group was decreased, because total cholesterol levels were decreased more than phospolipid levels. (6) The ratio of the serum VLDL, LDL to HDL in MCT diet group. in MCT and soybean oil mixed diet group, and in soybean diet group was decreased. In other wards, it means that the decrease of the serum VLDL, LDL in each group and the increase of the serum HDL.

Effect of Nicotine on Plasma Cholesterol Level in Spontaneously Hypertensive Rats (Nicotine에 의한 자발성고혈압백서(自發性高血壓白鼠)의 혈장 Cholesterol 농도의 변동)

  • The present study was undertaken to find out fasting total plasma cholesterol levels of SHR at different phases(labile, established and malignant) in the course of hypertension development and also to investigate effect of nicotine on plasma cholesterol levels of these animals. In nicotine administered group, 2.18 mg/kg nicotine was given daily for six weeks. The results obtained are as follows; 1) Mean total plasma cholesterol levels of SHR at labile, established and malignant phases were $104.6{\pm}3.1\;mg/dl$, $120.8{\pm}2.6\;mg/dl$ and $136.6{\pm}2.5\;mg/dl$ respectively. The total plasma cholesterol level generally increased with age of experimental animals. 2) After administration of nicotine (2.18 mg/kg/day) for six weeks, mean total plasma cholesterol levels of SHR at labile, established and malignant phases were $130.2{\pm}3.1\;mg/dl$, $150.4{\pm}3.3\;mg/dl$ and $166.6{\pm}3.2\;mg/dl$, respectively. The result of present study strongly suggests that nicotine has a positive relationship with plasma cholesterol levels in SHR.

Effects of Dietary Calcium and Sodium Levels on Lipid Metabolism in Hyperlipidemic/Hypercholesterolemic Rats (고지혈증 모델 흰쥐에서 칼슘과 소디움 섭취수준이 체내 지질대사에 미치는 영향)

  • The effects of dietary Ca and Na levels on lipid metabolism in hyper lipidemic/hypercholesterolemic rats were examined. In Expt. 1, normal rats were divided into six groups and fed high fat(15%, w/w)/cholesterol(1%, w/w) diet containing two levels of Na, low (0.05) or high(1.5%) and three levels of Ca, low(0.1%), normal (0.5%), or high(1.5%) for 8 weeks. In Expt. 2, hyperlipidemia / hypercholesterolemia rats were induced by feeding high fat / cholesterol diet for 4 weeks. They were divided into four groups and fed the high fat / cholesterol diet, containing two levels of Na, low or high and two levels of Ca, low or high for 4 weeks. In Expt. 1, total lipid and total cholesterol contents in serum and liver were significantly lower in rats fed high Ca diet than in rats fed normal or low Ca diet regardless of dietary Na levels. Serum TG was the highest in rats fed low Ca and low Na diet. In Expt. 2, Serum total lipid, TG, and total cholesterol levels decreased by 24, 35, 26% respectively in rats fed high Ca diet regardless of dietary Na levels. Serum total lipid level tended to increased in rats fed low Na diet. The total lipid and TG contents in liver slightly decreased in rats fed high Ca diet. Another observation was that high Ca intake significantly faciliated the fecal lipid and cholesterol excretion regardless of dietary Na levels. There results suggest that the hypolipidemidc/hypocholesterolemic effects of high Ca diet could be partly due to increase in lipid and cholesterol excretion and these effects may be independent of dietary Na levels.

Effects of Dietary Xylooligosaccharide on Lipid Levels of Serum in Rats Fed High Cholesterol Diet (고콜레스테롤 식이를 급여한 흰쥐에서 Xylooligo당의 혈중 지질개선 효과)

  • The purpose of this study was to investigate the effects of dietary xylooligosaccharide on lipid levels of serum in rats fed high cholesterol diet. Male Sprage-Dawley rats weiging 100$\pm$10g were randomly assigned to groups of two normal(N, N+10X) and four high cholesterol diets which contained 1%(w/w) cholesterol. High cholesterol diet groups were classified to xylooligosaccharide free diet (C group), 5% xylooligosaccharide diet(C+5X group), 10% xylooligosaccharide diet(C+10X group) and 15% xylooligosaccharide diet(C+15X group) according to the level of dietary xylooligosaccharide supplementation. These experimental diets were fed ad libidum for 4 weeks. The weight gain of high cholesterol diet group were significantly increased more than that of normal group, but those of 10% and 15% dietary xylooligosaccharide groups were significantly decreased more than that of high cholesterol diet(C)group. The higher content of xylooligosaccharide, the more food intake was increased. The food efficiencies of 10%, 15% cholesterol diet groups were lower than that of high cholesterol diet(C)group. The levels of serum triglyceride(TG) and total cholestoral were significantly high in cholesterol diet groups compared with normal diet group but were decreased in groups fed 5% and 10% dietary xylooligosaccharide. Especially, the lowest level showed in group fed high cholesterol diet containing 10% xylooligosacchride. High cholesterol diet group containing 10% xylooligosaccharide increased HDL-cholesterol level and then decreased LDL-cholesterol level and atherogenic index compared with other groups. The level of TBARS(thiobarbituric acid reactive substances) in serum was decreased in rat group fed xylooligosaccharide in high cholesterol diet. The higher content of xylooligosacchride, the more gastrointestinal transit time was decreased. The results indicate that dietary xylooligosaccharide can improve status of TG and total cholesterol and repress lipid peroxidation in serum lipid at hypercholesterolemia induced by high cholesterol diet.

Egg phospholipids exert an inhibitory effect on intestinal cholesterol absorption in mice

  • BACKGROUND/OBJECTIVES: Consumption of cholesterol-rich foods, such as eggs, has a minimal effect on circulating cholesterol levels in healthy humans. To gain insight, we investigated whether phospholipids rich in eggs (EPL) interfere with intestinal cholesterol absorption in vivo. MATERIALS/METHODS: To investigate the acute effect of EPL on intestinal cholesterol absorption, male C57BL/6J mice were orally administered with 6, 11, or 19 mg of EPL for three days. We also tested the effect of chronic EPL consumption on cholesterol metabolism in the small intestine and the liver in mice with diet-induced hypercholesterolemia. Male C57BL/6J mice were fed a high fat/high cholesterol (HF/HC; 35% fat, 0.25% cholesterol, w/w) diet for 4 weeks to induce hypercholesterolemia, and subsequently the mice were either fed 0, 0.4 or 0.8% (w/w) of EPL for 6 weeks. RESULTS: Intestinal cholesterol absorption was significantly decreased by the highest dose of acute EPL administration compared to control. Chronic EPL supplementation did not significantly alter intestinal cholesterol absorption nor plasma levels of total cholesterol and low-density lipoprotein cholesterol. In the small intestine and the liver, EPL supplementation minimally altered the expression of genes which regulate cellular cholesterol levels. CONCLUSION: Although chronic EPL consumption was not able to counteract hypercholesterolemia in HF/HC-fed mice, acute EPL administration decreased intestinal cholesterol absorption. This study provides in vivo evidence that acute administration of PLs in eggs prevent cholesterol absorption in the intestine, suggesting a mechanism for a minimal effect of egg consumption on circulating cholesterol levels.

Effect of Ginkgo biloba Extract (EGb 761) on Serum Cholesterol Levels in Wild-type C57Bl/6 Mice

  • Ginkgo biloba extract (EGb 761) is a standardized extract of Ginkgo biloba leaves and has anti- atherosclerosis properties. Many patients with atherosclerosis disorders take Ginkgo biloba extracts to supplement current therapy. In addition, normal healthy individuals also take Ginkgo biloba extracts for prophylactic purposes. However, it is unknown whether supplementation of Gingko biloba extracts in healthy individuals offer a benefit. In this study, we assessed whether EGb 761 could provide beneficial effects on serum cholesterol levels in normal mice. Wild-type C56Bl/6 mice were orally administered EGb 761 at 25 mg/kg (Group 3) or 50 mg/kg (Group 4) every other day for 40 days. We found that the serum levels of HDL-cholesterol (HDL-C) were significantly increased in EGb 761 and lovastatin treated groups. Treatment with EGb 761 and lovastatin resulted in reduced serum total cholesterol and LDL-cholesterol (LDL-C) compared to control group. Serum lecithin cholesterol acyltransferase (LCAT) levels were higher in EGb 761 and lovastatin treated group compared to the control group. However, no difference was observed in serum APO A-I levels between the control group and treatment group. These results suggest that EGb 761 can increase HDL-C resulting in increased serum LCAT levels.

The Relationship between Serum Cholesterol Levels and Dietary Fatty Acid Patterns, Plasma Fatty Acids, and Other Lipid Profile among Korean Adults (성인 남녀의 혈청 콜레스테롤의 수준과 지방산 섭취 양상, 혈중 지질 및 지방산 조성의 상관 관계 연구)

  • The purposes of this research were to assess dietary fatty acid patterns and to elucidate the relationship between the serum cholesterol levels and dietary fatty acid patterns, plasma fatty acid compositions, BMI (body mass index), and other lipid profile. The subjects were 151 adults aged 23 to 80 years, selected from the Outpatient Clinic and Cardiova-scular Department of the Seoul Municipal Hospital. Dietary data were obtained using three day food records. Sixteen dietary fatty acids were analyzed using Korean and US nutrient databases. The subjects were divided into three serum cholesterol levels: desirable (< 200 mg/dl, N = 44), borderline-risk ($\geq$ 200 - < 240 mg/dl, N = 35), and high-risk ($\geq$ 240 mg/dl, N = 72) groups. The high-risk group had higher BMI, waist, and waist to hip ratio (WHR) than the desirable and borderline-risk groups. Serum concentrations of triglyceride, LDL cholesterol and LDL/HDL cholesterol ratio were significantly higher in the high-risk group as compared to those in the other two groups. The serum cholesterol levels were highly correlated with BMI (r = 0.435), triglyceride (r = 0.425) and LDL/HDL cholesterol (r = 0.870) ratio. The highest fatty acid intake was from oleic acid (33 - 34% of total fatty acid intakes), which was followed by linoleic acid (27%), palmitic acid (19%), and stearic acid (7%). There was no correlation between the serum cholesterol levels and the dietary fatty acid intakes, polyunsaturateumonounsaturateusaturated fatty acids (P/M/S) and $\omega$6/$\omega$3 ratios. The correlation between plasma fatty acids such as myristic acid, oleic acid, linoleic acid, and docosahexaenoic acid and serum cholesterol levels was also weak. Elevation Factors of Fibrinogen in the Elderly Koreans

  • Plasma fibrinogen is risk factor of vascular disease including stroke, ischemic heart disease, atherosclerosis and thrombosis. Many studies have confirmed that high plasma fibrinogen levels are related with age, obesity, cholesterol, alcohol consumption, and genotype. This study was carried out to investigate the effect of fibrinogen genotype and other characteristics on the plasma fibrinogen levels in the elderly Koreans. For this study the blood samples were collected from 178 healthy elderly Koreans (102 males and 76 females, $55{\sim}80$ year olds). The blood samples were analyzed by smoking status, cholesterol levels, genotype, age, exercise, drinking, and gender. The plasma fibrinogen was assayed by clotting method, cholesterol being assayed by cholesterol oxidase method. The $\beta$-fibrinogen genotype was detected by PCR of relevant region and digestion with Alu I. The alleres with the restriction site and the non cleavable alleres were designated $A_1$ and $A_2$. In conclusion, genotype $A_1A_2$ and exercise are increased and associated with plasma fibrinogen levels. But, there were no significant differences by smoking, gender, age, drinking and cholesterol.

Effects of Sodium Alginate and Cellulose on Postprandial Plasma Lipoprotein and Cholesterol Metabolism in Rats(II) (Sodium Alginate와 Cellulose 가 식후 혈장 Lipoprotein 조성과 콜레스테롤 대사에 미치는 영향 (II))

  • This study was undertaken to dtermine whether dietary fibers had an effect on postprandial plasma lipoprotein and liver lipid composition in rats. Each experimental diet ocntained 10 % dietary fiber by weight. All animals were sacrificed in postprandial state. Sodium alginate-fed animals has significantly lower plasma cholesterol, VLDL-, LDL-cholesterol, TG, and protein levels than did the fiber-free control group. In addition, liver cholesterol and TG concentrations were the lowest in sodium alginate -fed animals. There was no significant change in HDL-cholesterol levels among experimental groups. Cellulose-fed animals also have lower plasma cholesterol and TG levels than fiber-free controls, but liver TG concentrations were not different from those in fiber-free controls. This study demonstrates that dietary fiber included in the diet of rats is able to alter postprandial lipoportein cholesterol and TG, and that sodium alginate, a soluble fiber, was the most effective in lowering plasma and liver cholesterol and TG.
