The effect of sulfur-containing amino acids on lipid metabolism was studied in rats fed casein as a protein source. Plasma cholesterol, HDL-cholesterol and atherosclerotic index decreased in the cysteine group compared to the methionine group. Plasma triglyceride and phospholipid level were not affected by the supplementation of the sulfur-containing amino acids. The levels of cholesterol and triglyceride in liver decreased by both methionine and cysteine. Cysteine increased the fecal excretion of coprostanol, total neutral steroid and bile acid. The results suggest that plasma cholesterol level is affected by dietary ratio of cysteine/methionine and that the hypocholesterolemic effects of cysteine is, at least in part, through reducing cholesterol absorption from small intestine and through enhancing fecal excretion of bile acids.
The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. The effect of feeding various diets on plasma lipids, liver lipids, fecal excretion of lipids and triglyceride secretion rate was studied in Sprague-Dawley rats fed for 1 month diets containing high fat(40% of energy ; HF) high cholesterol(1% ; HC), low fat/high sucrose(70% ; HS) or control diet(CT). Weight gain in HS group but there were no differences in TG contents of HF or HC groups compared to CT group. Fecal excretion of lipid was higher in HC than in HF, HS and CT groups. Hepatic TG secretion was not significantly different in four dietary groups. Accordingly, hypertriglyceridemia shown in HS group does not seem to be resulted from the increased TG secretion rate.
This study was undertaken to determine the effect of yellow and black soybeans on plasma and hepatic lipid composition and fecal lipid excretion in Sprague Dawley rats. Animals were fed with diets containing 52% yellow soybean, 50% black soybean, or 20% casein for 7 weeks. Feeding efficiency was significantly increased in the animals fed soybeans(p<0.05). Plasma cholesterol concentration was significantly lowered in the group fed yellow soybean compared with black soybean and casein-fed control (p<0.05). Hepatic triglyceride concentration was significantly lowered in soybean groups compared with casein-fed control(p<0.05), whereas hepatic cholesterol concentration was not affected by diet treatments. Soybean feeding significantly increased fecal weight, triglyceride and bile acid contents compared with casein feeding(p<0.05). It is concluded that soybean feeding in rats affects plasma and hepatic lipid levels by increasing the excretion of triglyceride and bile acids.
This study was carried out to determine the effects of sodium alginate and cellulose on the cholesterol metabolism in cholesterol-fed rats. Thirty male Sprague-Dawley rats were randomly assigned to three dietary treatments. Rats were fed, ad libitum, diets containing 10% dietary fibers as sodium alginate, cellulose or fiber-free with 1% cholesterol for 4 weeks. The results obtained were as follows: the feeding of sodium alginate with 1% cholesterol induced a significant decrease in plasma cholesterol and TG. The feeding of sodium alginate significantly decreased hepatic total lipids and TG levels, whereas the feeding of sodium alginate significantly increased hepatic HMG-CoA reductase activity. The feeding of sodium alginate and cellulose significantly increased fecal cholesterol and bile acid excretion. The excretion of TG in sodium alginate group, however, was two-fold and four-fold increased compared to cellulose and fiber-free group, respectively. As a result of this study, the ingestion of sodium alginate decreased plasma cholesterol and TG concentrations and liver TG concentration. This may be explained by the facts that fecal cholesterol, bile acid and TG level were increased significantly in sodium alginate group. The increased hepatic HMG-CoA reductase activity by sodium alginate feeding appears to be corresponded to whole-body cholesterol homeostasis.
This study was carried out to explore the hypocholesterolemic effect of dehyulled defatty soy flour and its possible mechanisms including endocrine status, cholesterol biosynthesis, and fecal excretion in rats. Animals fed casein were used as control and each phospholipid compared with casein feeding. Cholesterol concentrations in all lipoprotein fraction were significantly lower in defatted soy flour group compared with casein-fed control. Defatted soy flour feeding also significantly lowered hepatic total lipid, cholesteol and TG, and increaed fecal bile acid excretion by 270% compared with casein feeding. Defattd soy flour feeding had no significant effect on plasma thyroid hormone levels and hepatic 3-hydroxy-3-methyl glutary coenzyme A(HMG-CoA) reductase activity. However, plasma T4 concentration was slightly elevated and HMG-CoA reductase activity was suppressed in defatted soy flour group. These metabolic alterations partially explain the reduced plasma and hepatic cholesterol levels of rats fed defatted soy flour.
This study was conducted to investigate the hypocholesterolemic effects of porphyran and insoluble dietary fiber isolated from laver in rats fed high fat diet containing 1% cholesterol, 0.25% sodium cholate and 12% lard. Rats were fed, ad libitum, diets containing 5% diet fiber as cellulose(normal control or high fat control), porphyran or insoluble dietary fiber for 4 weeks. Among the groups fed high fat diet, liver weight was significantly lower in high fat porphyran group than high fat control. Plasma GOT, GPT, total cholesterol, cholesteryl ester, LDL-cholesterol and liver total cholesterol concentration were significantly lower in high fat porphyran group than high fat control. The feeding of porphyran significantly increased fecal cholesterol and bile acid excretion. The feeding of insoluble dietary fiber had no significant effect on either plasma or liver cholesterol levels, although fecal cholesterol level in the insoluble dietary fiber group was significantly higher than that in the high fat control. The results indicate that porphyran isolated from the laver may exert their hypocholesterolemic effect by increasing excretion of fecal bile acid and cholesterol.
The effect of milk in low and high cholesterol diet was invesigated on serum cholesterol metabolism and lipid contents of serum, aorta, liver of rats. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high(1.01% w/w) cholesterol-diets groups. Low cholesterol groups subdivided into four groups ; control group was given water and three milk groups were given low heat milk(LM), ultra-high heat milk(HM), and powder milk(PM), respectivily, instead of water. High cholesterol groups were consisted of three groups ; control, LM, and HM groups. After feeding these experimental diets for six weeks, lipid levels were measured in serum and tissure and dried feces were analyzed for neutral and acidic sterols. Results obtained from this study are as follows : 1) Nutrient intakes, body weight gains and aorta weights did not differ among groups, but liver weights were higher in high cholesterol fed rats than low cholesterol fed rats. 2) Serum protein contents were increased independently by intakes of high cholesterol and milk. 3) Serum total cholesterol and triglyceride levels were increased but phospholipid levels and HDL-cholesterol/total-cholesterol ratios were decreased by high cholesterol in diet. And milk supplementation decreased serum total cholesterol and triglyceride levels but increased phospholipid levels and HDL-cholesterol/total-cholesterol ratios. 4) Contents of cholesterol and triglyceride in aorta and liver were elevated by dietary high cholesterol and lowered by consumption of all three types of milk. 5) Levels of cholesterol and triglyceride among serum, aorta and liver were highly correlated (r=0.7-0.9, p<0.001). 6) Fecal excretion of total sterols was three times high in high cholesterol group, compared with low cholesterol groups and were increased about 20% by milk consumption. 7) The effects of milk were more pronouncely shown in low cholesterol groups and mostly confined to LM and HM groups, rarely shown in PM group. It is concluded from the present study that milk had the hypolipidemic as well as hypocholes terolemic effect, which appears to be mediated through increased fecal bile acid excretion. But the effect is likely to be shaded by excess consumption of dietary cholesterol and was almost absent in powder milk.
This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on lipid metabolism. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various chitosan and NOCC sources were given at the level of 0%(w/w) of diet. Total serum cholesterol and serum triglyceride concentrations were little affected by chitosan and NOCC supplements. However, HDL-cholesterol concentration and HDL : total cholesterol ratio were increased , and liver lipid, cholesterol , and triglyceride concentration were decreased by chitosan and NOCC supplements. The cholesterol and lipid lowering activity depends on fiber sources(crab and shrimp); type (chitosan and NOCC); and molecular weight(low , medium , and high). Among cadmium-free groups, chitosan-fed groups showed greater activity than NOCC -fed groups in lowering cholesterol and lipid levels, and greater fecal excretion of lipids and bile acids. Crab chitosans were more effective in cholesterol and lipid lowering activity than shrimp chitosans. The group fed high molecular weight crab chitosan showed the highest fecal excretion of lipids and bile acids. This indicated that high molecular weight crab chitosan was most effective in interfering with cholesterol and lipid absorption.
Demonstration of the highly positive correlation between blood cholestrol levels and heart disease has made consumers wary of the fats in meat, milk and eggs. The egg, as perhaps the single largest common source of cholesterol, has been cited by many members of the medical and scientific world as a food contributing to heart disease. In light of decreasing per capita egg consumption and continuing dietary egg cholesterol controversy, many researchers have focused their efforts on egg nutrition. The results reported, however, are often contradictory. In spite of the disputable scientific evidence, the egg has been labelled (erroneously) as a highly cholesterogenic food. The objective of this presentation is to present a general picture of the problem and discuss our laboratory findings relevant to the problem. An isotope technique was utilized to incorporate $^{14}C$-cholesterol into egg yolk lipoproteins and study the metabolic fate of dietary ovo-cholesterol in rats. Two hundred and fifty micro-curies of 4-$^{14}C$-cholesterol, emulsified in corn oil, were orally administered to five Single Comb White Leghorn laying hens. Eggs were collected, hard-boiled, and the hot dried egg yolk powder (HEY) was prepared. Total radioactivity excreted via feces was determined. The rat groups fed egg yolk powder excreted more than 95% of the ingested ovo-cholesterol, whereas the rat chow group excreted only 47%. No difference was observed between HEY and CEY treatments. Therefore, an unknown lipid factor present in egg folk accelerates cholesterol turnover rate and excretion via feces.
