This study was conducted to determine the morphological responses of barley seedlings to NaCl stress and to investigate histological changes of cells with transmission electron microscope(TEM) after NaCl stress. Plant height and root length of 10-day old barley seedlings with NaCl stress were reduced and inhibition level was found to be more severe in the plant height than in the root length. The leaf length, leaf width and leaf area were shorter as well with NaCl stress than without NaCl stress. However, there was no difference in the number of roots between NaCl treatments. The weight of dry matter decreased at higher NaCl concentrations, especially at 100mM NaCl. The water content of shoots tend to decrease at higher NaCl concentrations, but there was no difference in the water content of roots, The reduced sugar content was greatly increase than starch. Cellulose content was higher in NaCl stressed-plant than control, and tended to decreased at higher NaCl concentrations. Lignin content also decreased NaCl stressed-plant but there was no tendency at NaCl stress concentrations. Electric conductivity of cell sap with seedlings was high with NaCl stressed-plant. Amount of cell sap gradually increased with time in the roots than in the shoots, The grana of chloroplasts was changed by 150mM NaCl concentration. The christe of mitochondria in root meristematic sells ruined in structure and cell wall of leaf and root was also ruined by NaCl stress.
Korean ginseng has been widely used as medicine from ancient times in Asia. Current breeding efforts in Korea include the individual plant selection and the subsequent pure - line isolation, and considerable number of lines with desirable traits have thus been isolated. However, there were rare data on genetic maker and its analysis for selection of superior varieties. For taxonomic characterization and development of genetic markers for ginseng breeding, molecular biological methods including the RFLP and RAPD methods were applied. Cytoplasmic DNA of ginseng was analyzed for RFLP analysis. However. there is no different pattern among the chloroplast DNA or mitochondrial DNA of variants. In the case of RAPD analysis, the band patterns using 4 of 10 RAPD primers show the distinctive polymorphism among 9 ginseng variants, and lines, and Similarity Index(SI) on polymorphism was calculated for the extent and nature of these variabilities in ginseng. The sequences of 4 selected primers were TGCCGAGCTG, AATCGGGCTG. GAAACGGGTG, and GTGACGTAGG. By SI based on the polymorphic band patterns, Chungkyung - Chong and Hwangskoog - Chong, and JakyungChong 81783 and Jinjakyung of Russia showed the most close SI. The data of KG10l coincided with the fact that it was released from Hwangskoog - Chong. and Jakyung - Chong 81783 and Jinjakyung of Russia showed the most close SI. The data of KG101 coincided with the fact that it was released from Hwangskoog - Chong by breeding process. The data of Jakyung strains indicated the significant variation among the strains. From these results, RAPD analysis method could be succesively applied to the classification and genetic analysis for breeding of Korean ginseng.

Background: Both Panax ginseng Meyer and Panax quinquefolius are obligate shade-loving plants whose natural habitats are broadleaved forests of Eastern Asia and North America. Panax species are easily damaged by photoinhibition when they are exposed to high temperatures or insufficient shade. In this study, a cytohistological study of the leaf structures of two of the most well-known Panax species was performed to better understand the physiological processes that limit photosynthesis. Methods: Leaves of ginseng plants grown in soil and hydroponic culture were sectioned for analysis. Leaf structures of both Panax species were observed using a light microscope, scanning electron microscope, and transmission electron microscope. Results: The mesostructure of both P. ginseng and P. quinquefolius frequently had one layer of non-cylindrical palisade cells and three or four layers of spongy parenchymal cells. P. quinquefolius contained a similar number of stomata in the abaxial leaf surface but more tightly appressed enlarged grana stacks than P. ginseng contained. The adaxial surface of the epidermis in P. quinquefolius showed cuticle ridges with a pattern similar to that of P. ginseng. Conclusion: The anatomical leaf structure of both P. ginseng and P. quinquefolius shows that they are typical shade-loving sciophytes. Slight differences in chloroplast structure suggests that the two different species can be authenticated using transmission electron microscopy images, and light-resistant cultivar breeding can be performed via controlling photosynthesis efficiency.
Nuclear SSR (nSSR) and chloroplast SSR (cpSSR) markers were analyzed to assess the parameters of mating system in seed orchard, such as outcrossing rates, the number of potential pollen contributors, paternal contribution rates, degree of pollen contamination, and biparental inbreeding ($t_m-t_s$). In 2006, 2007, seeds were collected from the seed orchard of Pinus densiflora, established in 1977 at Anmyeon island. Estimates of outcrossing rates ranged from 94.9 to 100% (mean 98.9%) in 2006 and from 91.2 to 100% (mean 97.7%) in 2007 on the basis of the analysis of cpSSR haplotypes and from 90.3 to 100% (mean 95.9%) in 2006 and from 81.6 to 100% (mean 95.3%) in 2007 on the basis of the analysis of nSSR genotypes. By cross checking of both DNA markers, mean cumulative outcrossing rates of 100% and 98.9% were estimated in each year. Mean contamination rates were estimated as 48.9% and 42.4%, respectively. On the basis of cpSSR haplotype observed in each seed, paternal contribution rates (the number of pollen contributors) were estimated as 0.458 (mean 16.2) in 2006 and 0.512 (mean 14.8) in 2007. In conclusion, considering pretty high level of outcrossing rates observed in a seed orchard, there might be little to be influenced by inbreeding depression for genetic potential of the seeds induced by selfing. Estimates of the mating system parameters obtained from the two reproductive years were not statistically different, which revealed stable genetic quality of seeds produced in different years. Observed results from this study may provide useful information for the management and establishment of the seed orchard of the progressive generation.
The effect of soil EC on tissue morphology of leaf and shoot tip in cucumber (Cucumis sativus L. cv. Euinchim-baekdadagi) seedlings was investigated. Number of trichomes on leaf upper epidermis increased with the increase in soil EC from 1.0 to $3.0dS{\cdot}m^{-1}$, but the shape and number of stomata on lower epidermis remained unchanged. Epidermal cells of cucumbers grown in EC $1.5dS{\cdot}m^{-1}$ soil was occupied mostly by large vacuole whereas those grown in EC $3.0dS{\cdot}m^{-1}$ soil were filled with a nucleus, mitochondria, chloroplast and other micro-organelles. Sponge parenchima cells were also larger and contained fewer chloroplasts at EC $1.5dS{\cdot}m^{-1}$ than those grown at EC $3.0dS{\cdot}m^{-1}$. Leaf thickness decreased at high EC and the color of epidermal cells became significantly darker on the photograph of optical microscope. Normal tissue differentiation was greatly suppressed in plants grown in soils with $3.5dS{\cdot}m^{-1}$ or higher EC.
To investigate molecular phylogenetic relationships and to test hypothesis of hybrid origin of Ranunculus cantoniensis (Ranunculaceae), the sequences of nrDNA and chloroplast DNA were analyzed for 8 taxa and 25 accessions including 5 accessions of outgroup. In the phylogenetic trees by analyses of maximum parsimony and maximum likelihood for ITS nrDNA sequences and combined data of psbA-trnH, rps16 and trnL sequences of cpDNA, R. cantoniensis was most closely related to R. chinensis, and then to R. taciroi and R. silerifolius. The molecular phylogenetic relationships were not congruent with the previous report that R. cantoniensis was most closely related to R. silerifolius. In the sequence analysis of ITS and psbA-trnH, rps16, trnL for R. cantoniensis and the related taxa, R. cantoniensis showed polymorphism. It supported that the polymorphism also was reported in chromosome number and karyotype of R. cantoniensis. Ranunculus cantoniensis shared the marker gene of R. chinensis and R. silerifolius in ITS, and one of R. silerifolius in cpDNA. These results supported the hypothesis that R. cantoniensis was caused by hybridization between R. chinensis and R. silerifolius based on chromosome number and karyotype, and also estimated that R. silerifolius might be of maternal origin and R. chinensis be paternal.
Duckweed family (Lemnaceae Martinov), including the genus Lemna L., is a typical floating aquatic perennial plant, and about five genera and 40 species in the family are in wide distribution around the world except the polar regions. The genus Lemna is the smallest and the simplest plant among the angiosperms. It has a characteristic of doubling every three days with fast vegetative propagation, which helps the organisms to increase in rapid growth. As such, the plant is ideal for environmental pollution assessment and toxicity test. Although taxonomists and scholars have used different scientific names for the species, many of them have agreed that there is only one member of species of the genus Lemna in Korea. Paying attention to the external morphological variation observed in the Korean genus Lemna, we conducted a molecular phylogenetic analysis to identify the entity of the Korean Lemna species and to investigate the possibility of two or more members of the species existing in Korea. We determined and aligned the DNA sequences of the atpF-H region of the chloroplast DNA in 37 populations of the nationally distributed Lemna species. The results showed that the sequence length of the cp DNA atpF-H region was 463-483 bp, the length of the aligned sequences was 488 bp, and the number of variation site in nucleotide sequences was 47. There were two types of aligned sequences of the cp DNA atpF-H region from 37 populations of Lemna species in Korea. The maximum parsimony analysis revealed that the Korean Lemna consists of two clades, and one of them had two subclades. The results suggest that, contrary to the general understanding, at least two taxa (L.aequinoctialis, L.minor) exist in Korea.
Background: Although recent metagenomic approaches have characterized the distinguished microbial compositions in airways of asthmatics, these results did not reach a consensus due to the small sample size, non-standardization of specimens and medication status. We conducted a metagenomics approach by using terminal restriction fragment length polymorphism (T-RFLP) analysis of the induced whole sputum representing both the cellular and fluid phases in a relative large number of steroid $na{\ddot{i}}ve$ asthmatics. Methods: Induced whole sputum samples obtained from 36 healthy subjects and 89 steroid-$na{\ddot{i}}ve$ asthma patients were analyzed through T-RFLP analysis. Results: In contrast to previous reports about microbiota in the asthmatic airways, the diversity of microbial composition was not significantly different between the controls and asthma patients (p=0.937). In an analysis of similarities, the global R-value showed a statistically significant difference but a very low separation (0.148, p=0.002). The dissimilarity in the bacterial communities between groups was 28.74%, and operational taxonomic units (OTUs) contributing to this difference were as follows: OTU 789 (Lachnospiraceae), 517 (Comamonadaceae, Acetobacteraceae, and Chloroplast), 633 (Prevotella), 645 (Actinobacteria and Propionibacterium acnes), 607 (Lactobacillus buchneri, Lactobacillus otakiensis, Lactobacillus sunkii, and Rhodobacteraceae), and 661 (Acinetobacter, Pseudomonas, and Leptotrichiaceae), and they were significantly more prevalent in the sputum of asthma patients than in the sputum of the controls. Conclusion: Before starting anti-asthmatic treatment, the microbiota in the whole sputum of patients with asthma showed a marginal difference from the microbiota in the whole sputum of the controls.

Lipid droplets called plastoglobules are present in all plastid types. In chloroplasts, they are surrounded by the outer lipid monolayer from and connected to thylakoid membrane. The plastoglobule core contains the neutral lipids, which includes prenylquinones, triacylglycerols, and carotenoids. During stress and various developmental stages such as senescence, the size and number of plastoglobules increase due to the accumulation of lipids. Plastoglobules proteome revealed the presence of metabolic enzymes as well as structural proteins, plastoglobulins/fibrillins. Among the metabolic enzymes, the tocopherol cyclase, VTE1 and the NADPH quinine dehydrogenase, NDC1 have demonstrated that these participate in isoprenoid lipid metabolic pathways at the plastoglobule, notably in the metabolism of prenylquinones (tocopherol, plastoquinol and phylloquinone).
The genetic variation and intraspecific relationships between 10 individuals of seven cultivars and one Ulleungdo native of Wasabia japonica were investigated using RAPD (Randomly Amplified Polymorphic DNA) analysis. The 21 primers out of 50 random primers were amplified for all tested plants. The 68 (47.2%) among 144 bands derived from 21 primers showed polymorphism, and 3.2 bands per primer were observed. Number of bands per primer was ranged from 2 to 13, and average numbers were 6.8. The phenograms for 11 analyzed individuals by RAPD markers were not matched well with those of the result by morphological characters since they were clustered monophyletic at the similarity coefficient value ranged from 0.81 to 0.96. The Ulleungdo native individual was clustered sister to Daruma, Simanesairal, Sawa, and Hujidaruma cultivars. The RAPD markers were not useful to evaluate the intraspecific variations in Wasabia japonica cultivars, therefore need to more specific molecular phylogenetic characters such as AFLP technology and gene sequence of nuclear and chloroplast DNA.
