재(회) 처리의 미역 색소 안정화 효과 (PIGMENT STABILIZATION OF FRESH UNDARIA PINNATIFIDA WHEN TREATED WITH ASHES)

  • In this study, changes in chlorophyll and carotenoid pigment in fresh and ash treated Undaria pinnetifida were determined by column chromatography. And pigment stabilizing effects of ashes were discussed. The results obtained are summarized as follows: 1. The main carotenoids of Undaria pinnetifida are fucoxanthin, violaxanthin, lutein and $\beta-carotene$. 2. In storage of fresh Undaria pinnetifida at $2-5^{\circ}C$ for 30 days, chlorophyll a decreased rapidly and mainly converted to pheophytin a. Among carotenoids, lutein was markedly lost while the others retained approximately $30\%$ after 30 days' storage. 3, By treating the sample with ashes, the loss of chlorophyll a was restrained whereas that of carotenoids seemed to be promoted. It is considered that pigment stabilization was attributed to both effect, the pH control effect by the alkalinity of ashes and the fixation of the pigment by heavy metals in ashes. The stabilizing effect of chlorophyll a differed from the kind of ashes used. Reed ashes showed better effect than others such as straw or pine ashes. 4. Since fucoxanthin is extremely sensitive to alkali the treatment of ashes with a high alkalinity resulted in rapid decrease of the pigment during storage. 5. In the analysis of ashes, reed ashes which had better effect on chlorophyll a retention than the other ashes showed the highest content of iron.

Analysis on the Pigment Composition of Phytoplankton Assemblages using HPLC (High Performance Liquid Chromatography) in the Adjacent Waters of Nuclear Power Plants in Spring

  • The pigment composition and concentration of phytoplankton assemblages using HPLC in the adjacent waters of four nuclear power plants (Yonggwang, Kori, Wolsong and Ulchin) were investigated during the spring blooming in 2004. The mean concentration of chlorophyll a ranged from 563.8 to 2,949.0ng $l^{-1}$, with the lowest concentration at Kori and the highest concentration at Wolsong. Among the carotenoids, the amounts of fucoxanthin and chlorophyll $C_2$ were relatively higher than those of other pigments in the study site. As minor pigments, zeaxanthin, chlorophyll b, 19'-butanoyloxyfucoxanthin, diadinoxanthin, 19'-hexanoyloxyfucoxanthin, chlorophyll $C_3$ and peridinin were detected. The results of pigment composition and concentration showed that diatoms had an important proportion of phytoplankton community when a spring bloom occurred. Cyanobacteria was present relatively low density at the Wolsong and the green alga such as chlorophytes and prasinophytes were abundant at the Yonggwang and Kori, while dinoflagellates characterized by peridinin were common at Ulchin and Kori. The pigment composition and concentration of phytoplankton after passing through the cooling-water system of nuclear power plant were highly variable. No distinct trend of the change of each pigment composition and amount was detected but the variation of fucoxanthin and chlorophyll $C_2$ highly coupled with that of chlorophyll a. We pointed out that the diatom controlled the overall variation of phytoplankton biomass during the spring season.

  • The effect of water activity on degradation of pigments in dried lavers, Porphyra tenera Kjellm. was examined when stored at room temperature for fifty days. Chlorophyll pigment was extracted with methanol-petroleum ether mixture solvent(2:1 v/v), partitioned in ether, and analysed spectrophotometrically at 662 nm as chlorophyll a. The degradation products of chlorophyll were isolated on sugar-starch column(85:15 w/w) with n-propanol-petroleum ether solution(1:200 v/v) as a developing solvent. The isolated green colored zones were analysed individually at the wavelengths of 650, 662, and 667 nm as allomerized product, chlorophyll a retained, and pheophytin formed respectively. Carotenoida were also extracted with the methanol mixture solvent, partitioned in ether, and finally redissolved in acetone after the evaporation of ether in a rotary vacuum evaporator. The total carotenoid content was measured as lutein at 450 nm. From the results, it is noted that the rate of chlorophyll degradation reached a minimum at 0.11 to 0.33 water activity while progressively increased at higher moisture levels resulting in rapid conversion of chlorophyll to pheophytin. At lower activity, autocatalysed oxidizing reaction like allomerization seemed prevailing the acid catalysed conversion reaction. The loss of carotenoid pigment was also greatly reduced at the range of 0.22 to 0.34 water activity with much faster oxidative degradation at both higher and extremely lower moisture levels. These two moisture levels indicated above at which the both pigments exhibited maximum stability are considerably higher than the BET monolayer moisture which appeared 7.91 percent on dry basis at Aw=0.10 calculated from the adsorption isothermal data of the sample at $20^{\circ}C$. The rate of pigment loss in heat treated samples at 60 and $100^{\circ}C$ for 2 hours prior to storage somewhat decreased, particularly at higher moisture levels although the final pigment retention was not much stabilized.

인삼 모상근의 색소 생성 및 엽록체 발달에 미치는 광의 효과 (Effects of Light on the Pigment Production and Chloroplast Development of Ginseng Hairy Roots)

  • The effects of light on the pigment production and chloroplast development were examined on ginseng hairy roots cultured in 1/2MS liquid medium. The chlorophyll and carotenoid production were increased from 1,000 to 3,500 lux condition, but decreased drastically in 7,000 lux condition. The anthocyanin production was significantly increased with increment light intensity(1,000∼7,000 lux). The thylakoid membrane of chloroplast was proplastid in dark condition and it began to develop into thylakoid membrane in 1,000 lux condition and then intact thylakoid membrane was developed in 3,500 lux condition. However, the development of thylakoid membrane in 7,000 lux condition was inhibited comparing to 3,500 lux condition. The total chlorophyll production in blue light condition were high comparing to other wavelength and same as 40% of total chlorophyll on white light(3,500 lux) condition. The chlorophyll and carotenoid production by sucrose concentration were high in 3% sucrose condition and anthocyanin production was high in 4% condition. The production of chlorophyll and carotenoid by light periods was high when explants were cultured in dark condition for 1 week and then transferred to light condition for 4 weeks. Our results suggest that pigment production and chloroplast development could be accelerated by light Intensity of specific wavelength in cultures of ginseng hairy root.

NaNO3, NaHCO3 농도가 Arthrospira platensis 생장에 미치는 영향 (Effect of NaNO3 and NaHCO3 Concentration on Microglae Arthrospira platensis Growth)

  • Arthrospira platensis (A. platensis) is one of the most explored cyanobacteria and has been studied for proteins, vitamins, pigment (chlorophyll and carotenoids) and fatty acid. In this study, we tested the effect of $NaHCO_3$ and $NaNO_3$ on the microalgae growth under photoautothrophic culture in A. platensis. As a result, cell growth and dry cell weight were increased in proportion to the $NaHCO_3$ and $NaNO_3$ concentration. Pigment (chlorophyll and carotenoids) contents of A. platensis were increased with proportion to $NaHCO_3$ concentration. But, the content of pigment (chlorophyll and carotenoids) in 100% $NaNO_3$ medium of A. platensis was the highest, 40%, 140% and 200% $NaNO_3$ medium with pigment content of A. platensis was reduced. In conditions of $NaHCO_3$ (50%) or $NaNO_3$ (40%) limitation, A. platensis could accumulate lipids to high as 1.7-fold and 1.3-fold.

  • 디노프라젤레이트(dinoflagellate)들의 광채취색소단백질들은 안데나색소복합체들 내에서 카로테노이드(페리디닌)로부터 크로필 a로 실질적으로 100%의 효율을 갖는 에너지전달현상을 보여준다. 이와같이 디노프라젤레이트에서 광합성을 위한 태양에너지의 채취가 (특히 청(靑)색광에서) 높은 효율로 일어나는 것은 단백질표면의 갈라진 틈안에 위치한 페리디닌과 프로로필 a의 독특한 분자배치에 기인하는 것이다. 고등식물에서 가로테노이드와 크로로필 a 사이에 일어나는 에너지 전달메카니즘에 관해서도 디노프라젤레이트 안데나 색소복합체들과 비교해서 고찰하였다. Algae에서 광합성을 위한 태양에너지, 특히 적(赤)색광의 채취를 다룬 하나의 예로서 Chroomonas Species의 보조광합성색소단백질인 크로오오 모나스 피코시아닌의 분자토포그라피와 에너지전달도 역시 고찰하였다.

토마토의 생육과정에 따른 성분 변화 II - Ascorbic acid, Chlorophyll, Carotene, Lycopene 분석을 중심으로 - (The Changes of Components by Maturity Stage of Tomato II)

  • This was aim to study the changes of components by different parts and maturity of tomato. Tomato include the green pigment chlorophyll, the yellow pigment-carotene. the red pigment lycopene and the other component. These colors of tomatoes are reported to have important functional roles to promote health in human. The main objective of this study was to define changes in the content of the four tomato maturation that is ascorbic acid, chlorophyll, carotene, lycopene. A ascorbic acid contents of tomato fruits was the highest at 50th day. Chlorophyll contents of tomatoes fruits was the highest at 10th days after flowering. ${\alpha}$-carotene contained low-quantity while all growth period. ${\beta}$-carotene contained the highest at 50th days after flowering, $1036.97{\mu}g$. Lycopene contained the highest at 50th days after flowering, $5800{\mu}g$.

SO2 노출된 4개 수종의 엽내 광색소 함량 및 SOD 활성 변화 (Changes of Photosynthetic Pigment Contents and SOD Activity in the Leaves of Four Tree Species Exposed to SO2)

  • 목본 식물을 대상으로 SO$_2$에 의해 나타나는 생리적인 반응을 구명하기 위하여 소나무, 오갈피나무, 현사시, 상수리나무를 대상으로 SO$_2$를 500 ppb, 800 ppb로 하루에 8시간씩 7일간 처리하여 잎의 광색소 함량과 SOD 활성을 비교 분석한 결과는 다음과 같았다. SO$_2$의 처리 농도가 증가할수록 4개 수목의 엽 내 엽록소 함량은 감소하였으며, 엽록소 a와 엽록소 b, 카로티노이드 함량의 변화는 수종별, 처리별로 다른 경향을 보였다. 오갈피나무와 상수리나무의 엽록소 b와 a의 비는 SO$_2$는 500 ppb 처리구에서는 증가하다가 800ppb 처리구에서는 감소하였다. 즉 500ppb에서는 엽록소 a가 파괴되며, 800ppb에서는 엽록소 b도 파괴될 수 있음을 보여 주었으며, SO$_2$에 대한 민감성은 엽록소 a가 엽록소 b보다 높은 것으로 나타났다. 4개 수종의 잎 SOD활성은 수종별, 처리별 큰 차이를 나타냈다. 오갈피나무와 상수리나무는 500 ppb 처리구에서는 SOD 활성이 증가하다가 더 높은 농도에서는 활성이 감소하였으며, 소나무와 현사시의 경우는 500ppb 처리와 800ppb 처리에서 높은 SOD활성을 유지하여 내성을 보인다. 그러나 광색소와 SOD 활성을 기준으로 판단해 볼 때, SO$_2$에 대한 저항성은 현사시가 가장 높은 것으로 판단된다.

HPLC를 이용한 광합성색소 분석과 CHEMTAX 프로그램을 이용한 식물플랑크톤 군집조성 연구 (Application of Photosynthetic Pigment Analysis Using a HPLC and CHEMTAX Program to Studies of Phytoplankton Community Composition)

  • High-Performance Liquid Chromatograph(HPLC)를 이용하여 식물플랑크톤의 광합성색소를 분석한 결과를 토대로 CHEMTAX 프로그램을 이용하여 식물플랑크톤 군집조성을 추정하는 방법이 많이 이용되고 있다. CHEMTAX 프로그램은 식물플랑크톤 군집별(class level)로 체내에 존재하는 주요색소의 chlorophyll a에 대한 상대적인 비 값을 기초로 분석을 한다 본 연구는 국내 해역에서 분리한 식물플랑크톤 군집별 주요 종들이 가지는 광합성색소의 상대적인 비 값을 파악하고 이 자료를 기초로 남해안에 위치한 여자만과 가막만에서 측정한 광합성색소 분석결과에 대해서 CHEMTAX 프로그램을 이용하여 식물플랑크톤의 군집조성을 파악하였다. 국내해역에 존재하는 같은 군집종들 사이에서도 주요색소의 상대적인 비 값은 차이를 보였으며, 국외의 다른 해역에서 제시된 상대적인 비 값과도 상당한 차이를 보였다. 본 연구에서 얻은 비 값을 기초로 식물플랑크톤 군집조성을 분석한 결과, 다른 해역에서 제시된 비 값을 이용하여 분석한 결과와 유의한 차이를 보였다. 우점하는 군집(bacillariophyceae와 dinophytes)에 대해서는 5% 이내에서 차이를 보였으며, 비록 chlorophyll a에 대해서 낮은 기여도를 보이지만 pico와 nano 크기의 군집들(cyanophytes와 prasinophytes)에서는 상당한 차이(2배 이상)를 보였다. 비록 HPLC-CHEMTAX 분석방법은 식물플랑크톤 군집조성 연구에 유용하게 이용되고 있지만, 보다 정확한 결과와 식물플랑크톤의 생리, 생태를 이해하기 위해서는 연구해역에 존재하는 식물플랑크톤 종들에 대한 주요색소의 상대적인 비 값에 대한 연구가 계속적으로 진행되어야 할 것으로 판단된다.

금속이온이 청각 색소의 안정화에 미치는 영향 (Effect of Metal Ions on Stabilization of Codium fragile's Pigments)

  • 해양 녹조류인 청각에 함유되어 있는 chlorophyll을 유기용매를 사용하여 추출한 후 chlorophyll의 안정성을 증가시키기 위한 조건을 확립하였다. 추출 용매로 사용된 acetone, ethanol 그리고 methanol 중 methanol이 가장 효과적이었으며, 추출 수율은 건조 중량으로 $25.0{\pm}2.10\;mg/g$ 이었다. TLC 및 HPLC 분석을 통하여 청각 색소에는 chlorophyll a 와 chlorophyll b가 주성분이며, 이들은 각각 0.40 mg/g and 1.94 mg/g의 함량으로 존재하였다. 청각 색소에 함유된 총 엽록소 함량은 빛이 있는 곳과 없는 곳에서 30주간 $40^{\circ}C$로 저장하였을 경우, 초기 양의 23.2 %와 58.4 %로 각각 잔존하였다. 황산구리, 황산아연, 황화철, 황산마그네슘으로부터 해리 된, 구리, 아연, 철, 마그네슘과 같은 금속이 이온이 색소의 저장 안정성에 미치는 영향을 분석한 결과, 구리가 가장 효과적이었으며, 아연이 그 다음으로 효과적이었다. 구리와 아연이 1 mM의 농도가 존재할 경우, 청각색소는 30주 후에, 빛이 있는 상태에서는 초기 양의 47.0%, 빛이 없는 상태에서는 88.8 %로 잔존하였다. 청각 색소를 안정화시키는 구리와 아연의 최적 농도는 빛이 없는 조건에서 구리는 0.1 mM, 아연은 0.5 mM이었다.
