Comparison of Incidence of PSE Pork by Lairage Time, Use of Electrical Probe, Stunning Voltage and Chilling Condition (돼지의 계류시간, 전살전압 및 도체냉각조건에 따른 PSE 발생율 비교)

  • The incidence of PSE pork by lairage time, use of electric probe, stunning voltage condition and carcass chilling rate was compared. The incidence of PSE pork was 22.2% when pigs were held in lairage overnight without electric probe, whereas it was 59.3% when held in lairage for < 1.5 hr with electric probe before slaughter. The incidence of PSE pork was 42.37% when pigs were treated with 230 V of stunning condition, and was 66.67% when treated with 500V of stunning condition. The incidence of PSE pork was 52.2% when the ultimate carcass internal temperature was controlled higher than 38$^{\circ}C$ with the slow chilling(-5$^{\circ}C$ for 70 min), and was 39.6% when controlled lower than 30$^{\circ}C$ with the fast chilling(-15$^{\circ}C$ for 70 min). Results indicated that the incidence of PSE pork was decreased by 37.1% with lairage overnight and without use of electric probe, 24.3% with low voltage stunning treatment and 12.6% with fast chilling. However, the incidence of PSE pork was not significantly correlated with carcass weight.

Free Amino Acids, Collagen Solubility, and Meat Quality in Pork (Longissimus Muscle of Yorkshire) as a Function of Chiller Temperature and Aging

  • This study was conducted to identify the effect of chilling temperature (-3 and $6^{\circ}C$) and aging (1- and 7-day) on objective meat quality, collagen solubility, and free amino acids in pork (longissimus muscle of Yorkshire). Warner-Bratzler (WB)-shear force indicated that variation in chilling temperature had no detectable effect on meat tenderness and tenderization during the 7-day aging period. Among the 13 detected free amino acids, only 3 amino acids (histidine, valine, leucine) were significantly affected by the temperature treatment (p<0.05). Collagen solubility was significantly increased at $6^{\circ}C$ treatment (p<0.05). There was a significant linear relationship (r=0.67, p<0.05) between changes in free amino acids and WB-shear force during the 7-day aging period. These results confirmed that chilling conditions had significantly affected collagen solubility, and meat tenderization occurred in direct proportion to an increase in free amino acids.

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury. Studies on the Promoting of Seed Germination of Adenophora triphylla var. Japanica $H_{ARA}$ (잔대종자 발아촉진에 관한 연구)

  • The objective of this study is to promote germination for mass cultivation of Adenophora triphylla. After the seed soaking for I day in BA and $GA_3$ solution, seed germination was effectively promoted in the treatment of 500mg/L $GA_3$ and the highest germination rate was 94%.The average germination days were 1.5days earlier than those of non-treated seeds. In a chemicals treatment, seed germination rate was 54% in 1% $KNO_3$ treatment but KOH treatment was no significantly effected. The durations of chilling treatment for breaking of dormancy were longer than 2weeks. The optimal temperature for germination was $25^{\circ}C$. The promotion of seed germination is presumed to be due to the breaking of dormancy by $GA_3$, chilling treatment rather than seed coat maceration by KOH or $KNO_3$ treatments.

  • Terpene synthase plays a key role in biosynthesis of triterpene saponins (ginsenosides) and is intermediate in the biosynthesis of a number of secondary metabolites. A terpene synthase (PgTPS) cDNA was isolated and characterized from the root of Panax ginseng c.A. Meyer. The deduced amino acid sequence of PgTPS showed a similarity with A. deliciosa (AAX16121) 61%, V. vinifera (AAS66357) 61%, L. hirsutum (AAG41891) 55%, M. truncatula (AAV36464) 52%. And the segment of a terpene synthase gene was amplified by reverse transcriptase-polymerase chain reaction (RTPCR). We studied expression of terpene synthase under stressful conditions like chilling, salt, UV, and heavy metal stress treatment. Expression of PgTPS was increased gradually after exposure to stresses such as chilling, salt, and UV illumination. But its transcription seems to be reduced by cadmium and copper treatment.

  • Acute diarrhea is fastly progressing and its most symptom are caused by virus. It usually continue within 3 weeks. And also it company fever, chilling sign, anorexia and vomitting. Mostly, it is getting better by basic treatment but must be cautious about progressing to dehydration, sepsis or other complications. In this study, it is experienced that acute diarrhea patient, got sick by food, became better by treating in the point of Sasang Medicine. 75 years old male patient having abdominal pain with aqueous diarrhea (about 10th a day before admission), chilling sign, general weakness was classed as Soyangin having Shinhanbocktongmangeumjeung(身寒腹痛亡陰證). I prescribed Hwalsuckgosamtang(滑石苦蔘湯) by Peueumganggy(表陰降氣) therapy and his symptom was getting better. So report it.

Effect of Selenium on Storability of Basil (Ocimum basilicum) in Hydroponics (양액재배 시 selenium의 첨가가 basil의 저장에 미치는 영향)

  • The aim of this study was to select the proper temperature for MA storage and to study the effect of sodium selenate ($Na_2SeO_4$) on storability of hydroponically grown sweet basil. Sweet basil was cultured with 1 fold using the nutrient solution developed by European Vegetable R & D Center in Belgium. Three-weeks before harvest, sodium selenate ($Na_2SeO_4$) was treated 2, 4, 6 and $8mg{\cdot}L^{-1}$ in the nutrient solution. Although electrolyte leakage of sweet basil was remarkably increased because of chilling injury at $5^{\circ}C$ MA storage, storage life was extended for three more days by selenium supplement. The weight loss was lower at $10^{\circ}C$ than at $5^{\circ}C$ and vitamin C content was higher at $10^{\circ}C$ than $5^{\circ}C$ storage treatment. Se treatment increased vitamin C content and decreased ion leakage in the plant. Accumulation of Se in the leaves of basil was increased with Se concentration of the nutrient solution. Se accumulation was $66{\mu}g{\cdot}g^{-1}$ dry mass at $2mg{\cdot}L^{-1}$ treatment and $422{\mu}g{\cdot}g^{-1}$ dry mass at $8mg{\cdot}L^{-1}$ treatment. The results suggest that storage at $10^{\circ}C$ is good for reduction of chilling injury and maintain marketability of basil. Less than $2mg{\cdot}L^{-1}$ sodium selenate treatment is acceptable for human health.

Optimization of Curing Treatment and Storage Temperature of Chinese Yam (마의 수확후 처리 및 저장 온도 최적화)

  • Effects of curing treatments and storage temperature on the quality of Chinese yams (Dioscorea polystachya Turcz.) were investigated stepwise in three consecutive years for the optimization of postharvest handling procedures. Tuberous roots were harvested in early to mid November and cured under ambient or $29^{\circ}C$ heated air conditions for various periods according to the treatment conditions. Storage temperatures in the range of 0.5 to $7.5^{\circ}C$ were phased in to avoid chilling injury while examining storage potential from 4 to 7 months. As poststorage technology, short-term $60^{\circ}C$ hot-air exposure or low shelf temperature treatments were additively imposed. Curing treatments, especially heated air curing for 3-5 days tended to reduce the respiration and weight loss during storage while maintaining flesh firmness. Storage at $0.5^{\circ}C$ brought out typical chilling injury symptoms on the shelf with increases in respiration and lower flesh firmness by tissue breakdown resulting in the rapid loss of marketability. Optimum storage temperature appeared to be the $3-4^{\circ}C$ range which suppresses quality deterioration while avoiding chilling injury. Low shelf temperature seemed to be a necessary part of postharvest handling system to keep marketability through control of poststorage disorders such as rooting and decay. Overall results suggested that optimized postharvest program consisting of heated-air curing, storage at $3-4^{\circ}C$, and low shelf temperature could extend storage potential of Chinese yam to longer than 7 months.

Water relations of plants under environmental stresses: role of aquaporins

  • Effects of low temperature ($8^{\circ}C$) on the hydraulic conductivity of young roots of a chilling-sensitive (cucumber; Cucumis sativus L.) and a chilling-resistant (figleaf gourd; Cucurbita ficifolia Bouche) crop have been measured at the levels of whole root systems (root hydraulic conductivity, $Lp_r$) and of individual cortical cells (cell hydraulic conductivity, Lp). In figleaf gourd, there was a reduction only in hydrostatic $Lp_r$ but not in osmotic $Lp_r$ suggesting that the activity of water channels was not much affected by low root temperature (LRT)treatment in this species. Changes in cell Lp in response to chilling and recovery were similar asroot level, although they were more intense at the root level. Roots of figleaf gourd recovered better from LRT treatment than those of cucumber. In figleaf gourd, recovery (both at the root and cell level) often resulted in Lp and $Lp_r$ values which were even bigger than the original, i.e. there was an overshoot in hydraulic conductivity. These effects were larger forosmotic (representing the cell-to-cell passage of water) than for hydrostatic $Lp_r$. After a short term (1 d) exposure to $8\;^{\circ}C$ followed by 1 d at $20\;^{\circ}C$, hydrostatic $Lp_r$ of cucumber nearly recovered and that of figleaf gourd still remained higher due to the overshoot. On the contrary, osmotic $Lp_r$ and cell Lp in both species remained high by a factor of 3 as compared to the control, possibly due to an increased activity of water channels. After pre-conditioning of roots at LRT, increased hydraulic conductivitywas completely inhibited by $HgCl_2$ at both the root and cell levels. Different from figleaf gourd, recovery from chilling was not complete in cucumber after longer exposure to LRT. It is concluded that at LRT, both changes in the activity of aquaporins and alterations of root anatomy determine the water uptake in both species. To better understand the aquaporin function in plants under various stress conditions, we examined the transgenic Arabidopsisand tobacco plants that constitutively overexpress ArabidopsisPIP1;4 or PIP2;5 under various abiotic stress conditions. No significant differences in growth rates were found between the transgenic and wild-type plants under favorable growth conditions. By contrast, overexpression of PIP1;4 or PIP2;5 had a negative effect on seed germination and seedling growth under drought stress, whereas it had a positive effect under cold stress and no effect under salt stress. Measurement of water transport by cell pressure probe revealed that these observed phenotypes under different stress conditions were closely correlated with the ability of water transport by each aquaporin in the transgenic plants. Together, our results demonstrate that PIP-type aquaporins play roles in seed germination, seedling growth, and stress response of Arabidopsis and tobacco plants under various stress conditions, and emphasize the importance of a single aquaporin-mediated water transport in these cellular processes.

Studies on the Germination Characters of Korean Ginseng (Panax ginseng C.A. Meyer) Seed (고려인삼종자(高麗人蔘種子)의 발아특성(發芽特性)에 관(關)한 연구(硏究))

  • This study was conducted to define the optimal conditions for embryo growth during seed stratification and for breaking dormancy as well as seed germination of stratified ginseng seeds. The experiments were also carried out to detect some materials which were expected to induce seed dormancy in the ginseng seeds. The results summarized as follows; 1. The growth of embryo during seed stratification was significantly inhibited by the existence of endocarp. The fastest embryo growth was resulted at $15^{\circ}C$ and an estimated optimal temperature for embryo growth was about $18^{\circ}C$. 2. There was no significant difference between the embryo growth and germination ratio of ginseng seeds which were sown in seed bed at Aug-5 without seed stratification and that of artificial seed stratification. 3. Embryo growth and germination ratio was significantly inhibited by high temperature treatment at $30^{\circ}C$ for 24 hours or respiration stress by immersing seeds in water for 10 days or more. 4. When the seed stratification was started at $10^{\circ}C$, growth of embryo in the ginseng seeds were almost stopped. But, when the seeds were stratified first at $20^{\circ}C$ for 50 days and next at $10^{\circ}C$ for 50 days, the embryo growth was significantly promoted compared with the embryo growth in the seeds which were stratified at $20^{\circ}C$ for 100 days. 5. The successive embryo growth after seed stratification was significantly accelerated at $10^{\circ}C$ but the seeds chilled at $5^{\circ}C$ for 100 days were resulted in the highest germination ratio as well as the shortest days for germination. 6. The successive embryo growth during chilling treatment and seed germination were significantly inhibited by immersing seeds in water just before chilling treatment or during chilling treatment and by interruption of chilling treatment with raising temperature to $20^{\circ}C$ for 20 days during chilling treatment. 7. The germination ratio of ginseng seeds which finished chilling treatment was highest at $10^{\circ}C$ and 62.5% was the estimated soil moisture for the best germination of ginseng seeds. The ginseng seeds were found to require high amount of oxygen for germination. 8. Only water soluble material in homogenized ginseng seeds showed a significant inhibiting effect on the seed germination of sesame, millet and soybean. Water soluble material dissolved from undehisced ginseng seeds showed stronger inhibiting effect on the seedling growth of sesame than material from dehisced ginseng seeds. Extraction temperature did not influence the inhibiting effect of the material dissolved from ginseng seeds on the seedling growth of sesame. 9. Water soluble materials dissolved from the berry pulps, leaves, fresh roots and dried roots also showed a significant inhibiting effect on the seedling growth of sesame. 10. Water soluble materials dissolved from the ginseng seeds, leaves and fresh roots showed a significant inhibiting effect on the germination of true fungi and the growth of spawn but the growth of phytopathogenic bacteria was not. 11. Among the water soluble materials dissolved from ginseng seeds, the materials of low molecular weight less than 3,000 were resulted a significant inhibiting effect on the seedling growth of sesame and the materials of high molecular weight also showed an inhibiting effect.
