Clinical Study on 1 Case of Patient with Cellulitis Treated by Oriental and Western Medicine Simultaneously (봉와직염 환자에 대한 한 ${\cdot}$ 양방적 치료 1례)

  • Cellulitis is a infectious disease characterized by acute purulent inflammation clinically manifested by erythema, pain or heating sensation caused by palpation, chilling sign and mild to moderate fever. in western medicine most part of the treatment is focused on injecting routine antibiotics hoping for the pathogen(in the case bacteria) to be treated but instead causing the bacteria to be resistant to antibiotics and consequently leads to longer admission. In oriental medicine Cellulitis belongs to the cathegory of Ong(癰), Bal(發) or Dandok(丹毒) on symptoms. The chief cause of acute Cellulitis is the evil of wet and heat blended and the postraumatic infection evil. We experienced a case of acute cellulitis defined as Bicheonbal(비천발) and Chokbaebal(足背發). We attempted to use both methods using acupuncture, herbal medicine(Gamidangkwuijeomtong-tang(加味當歸拈痛湯)) as well as routine antibiotic treatment and as a result we have achieved remarkable results in laboratory tests though there was no difference in shortening the curing process compared to the average time that it take to cure when admitted to western medical center.

  • Cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.95), catalyzes the reduction of hydroxycinnamaldehydes to give hydroxycinnamyl alcohols, or "monolignols," the monomeric precursors of lignin. Lignins are important components of cell walls and lignified secondary cell walls play crucial roles in long distance transport of water and nutrients during plant growth and development and in plant defense against biotic and abiotic stresses. Here a cDNA clone containing a CAD gene, named as PgCAD, was isolated from a commercial medicinal plant Panax ginseng. PgCAD is predicted to encode a precursor protein of 177 amino acid residues, and its sequence shares high homology with a number of other plant CADS. The expression of PgCAD in adventitious roots and hairy roots of P. ginseng was analyzed using reverse transcriptase (RT)-PCR under various abiotic stresses such as salt, salicylic acid, wounding and chilling treatment that triggered a significant induction of PgCAD at different time points within 2-48 h post-treatment. This study revealed that PgCAD may help the plants to survive against various abiotic stresses.

Ethylene Production and Accumulation in Leaf Sheath and Its Relation to Tillering Suppression of Deep-Irrigated Rice Plants

  • The deep irrigation of rice plants brings about some beneficial effects such as reduced tiller production which results in the formation of bigger panicles, prevention of chilling injury, reduced weed growth, etc. The present study was carried out to examine the involvement of ethylene in the suppression of tiller production due to deep water irrigation in rice (cv. Dongjinbyeo). The ethylene production was induced in leaf sheath within 24 hours after the deep water irrigation and has increased even until 30 days after the treatment, recording 4.5-fold increase as compared to the shallow-irrigated rice plants. In the deep water irrigated rice plants, ethylene was accumulated to a high concentration in the air space of submerged leaf sheath as the irrigated water deterred the diffusion of ethylene out of the leaf sheath and ethylene biosynthesis was accelerated by the deep irrigation as well. The ethylene concentration recorded 35-fold increase in the deep-irrigated rice plants for 35 days. The tiller production was reduced significantly by the deep irrigation with water, the tiller bud, especially tertiary tiller bud differentiation being suppressed by the deepwater irrigation treatment, whereas the rice plants deep-irrigated with solutions containing $10^{-5}$ M or $10^{-6}$ M silver thiosulfate (STS), an action inhibitor of ethylene, showed the same or even higher production of tillers than those irrigated shallowly with water. This implies that the ethylene is closely linked with the suppression of tiller production due to deep water irrigation. In conclusion, ethylene, which was induced by hypoxic stress and accumulated in the leaf sheath due to submergence, played a key role in suppressing the tiller production of the deepwater irrigated rice.

Photosynthetic Responses and Photoprotection in Korean Hot Pepper (Capsicum annuum L.) against High Light Stress (광 스트레스에 대한 고추 잎의 광합성 반응과 광 보호 메커니즘)

  • Photoinhibition and photoprotection of PSII in the leaves of hot pepper (Capsicum annuum L.) grown in Hoagland solution and Tap water were compared. Though changes in the rates of $O_2$ evolution as a function of photon fluence rate (PFR) were comparable, the rates of respiration in the dark was 3 times higher in the Hoagland solution grown leaves than in the Tap-water grown ones. Compared to Hoagland solution grown plane, PSIIs of Tap water grown pepper leaves were more susceptible to photoinhibitory light treatment. In order to inactivate functional PSII to the same extents, Hoagland solution grown plants required almost 2-fold high light $(1600{\mu}molm^{-2}s^-)$ treatment than those of Tap water $(900{\mu}molm^{-2}s^-)$. Interestingly, the remaining fraction of PSII in Hoagland grown pepper was able to survive under prolonged illumination in the presence of lincomycin, which probably means that the growth condition of plant seemed to have an effect on the recovery of PSII from light stress. When PSII was severly photoinactivated at a chilling temperature, recovery was observed only if the residual functional PSII were not inhibited with DCMU, Nigericin and MV during recovery. In conclusion, PSIIs grown in the Hoagland solution was more resistant to excess light than in the Tap water grown one and the recovery of PSII from photodamage was more efficient in Hoagland grown pepper leaves than Tap water grown one, which means that the increased dark respiration may play a important role in the protection of PSII from photoinhibition by helping repair photosynthetic proteins (in particular, the D1 protein of PSII) degraded by photoinhibition.

  • Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.

Effects of Submersion Aging in Chilled Water on Tenderness and Microbial Growth of Vacuum-Packed Hanwoo Meat (냉수침지 숙성법이 진공포장 한우육의 연도 및 미생물 증식에 미치는 효과)

  • To investigate the effects of submersion aging in chilled water on tenderness and microbial growth of vacuum-packed beef, the semimembranosus muscles of Hanwoo (Korean cattle) was sampled at a commercial meat plant at 24 hrs postmortem. The samples were cut to 2.5 cm thick steaks and randomly assigned to following two treatments. The samples were stored in conventional refrigerator at 4$^{\circ}C$ after vacuum packaging for control whereas the other vacuum-packed samples were submersed in chilled water at 1$^{\circ}C$ for treatment, and both samples were stored for 14 days to measure total plate counts (TPC), sarcomere length, free calcium concentration, shear farce value and myofibrillar fragmentation index (MFI). The sarcomere length of treatment was significantly (p<0.05) longer than that of control at 3 days aging. Result suggested that submersion in chilled water of vacuum-packed beef might be effective to improve tenderness of meat compared to storage in conventional refrigerator. There were no significant differences in the shear force and MFI between control and treatment during storage. However, the free calcium concentration of samples from treatment was significantly (p<0.05) higher than that of control at 7 days of ageing. This result indicated that the lower shear force value and the longer sarcomere length of samples from treatment might be due to increasing the free calcium ion concentration in sarcoplasm during storage. On the other hands, samples from control showed significantly (p<0.05) higher number of microbial (TPC) compared to treatment during storage. from results obtained, submersion in chilled water of vacuum-packed beef could be recommended as a desirable aging method to improve tenderness of Hanwoo compared to aging in conventional refrigerator.

Improvement of Tomato Seedling Quality under Low Temperature by Application of Silicate Fertilizer (저온 저장 시 규산 처리에 의한 토마토 묘소질 향상)

  • The object of this study was to improve tomato seedling quality in low temperature(below 7, $10^{\circ}C$ during night time or daily mean air temperature was $18^{\circ}C$) by application of silicate fertilizer. Six different silicate fertilizer concentrations (8, 16, 32, 64, 128, and 256mM) or water as the control were applied to tomato seedlings twice a week for 20 days. Positive effects were observed in the growth parameters of the seedlings treated with 16 and 32mM silicate fertilizer; the most effective concentration of silicate at which seedlings showed the best performance was 16mM. However, a high concentration of silicate (256mM) caused negative effects on the growth. The transpiration rate decreased alongside with the increase of silicate concentration up to 32mM, possibly due to the increased stomatal diffusive resistance. Silicate stimulated the growth and development of tomato seedlings, resulting in increased growth parameters and root morphology. However, no significant differences were observed among treatment numbers of soil-drenching wuth the silicate (6, 10, or 20 times with 16mM) for 20 days, suggesting that silicate treatment with 6 times may be sufficient to induce the silicate effects. The application of 16mM of silicate fertilizer reduced relative ion leakage and chilling injury during low temperature storage. In addition, the seedlings treated with silicate fertilizer recovered faster than those without silicate treatment after low temperature storage.

Effect of Storage Temperature and Packing Materials on Storability of Fresh Salicornia europaea L. (저장온도와 필름종류가 신선 퉁퉁마디(Salicornia europaea L.)의 저장성에 미치는 영향)

  • These studies were carried out to find the effect of packing materials and storage temperature to enhance the storability of Salicornia europaea. The fresh weight loss was less than 2% at below $10^{\circ}C$ and non-perforated package, but $25^{\circ}C$ and perforated package treatment showed more than 4% of fresh weight loss that result to deteriorate quality. The carbon dioxide and oxygen contents remained 1~2% and above 15% in non-perforated package, and these contents were in proportion to gas permeability of packing materials. All the temperature treatments except 25c showed rapidly increasing ethylene concent from 7days after storage, and the highest ethylene content was $2^{\circ}C$ treatment that should appear chilling injury. The off-odor and deterioration ratio were lowest in $5^{\circ}C$ among the temperature treatments and $50{\mu}m$ thickness ceramic film treatment at $5^{\circ}C$ storage, while packing materials did not show any trends among the temperature treatments. The shelf life based on visual quality showed highest in $50{\ss}|$ thickness ceramic film packing and $5^{\circ}C$ treatment, and that was 28days.

Effects of 20-hydroxyecdysoneon Adult Development of Diapausing Fall webworm (Hyphantria cunea Drury) Pupae (휴면중인 미국흰불나방 번데기의 성충발육에 미치는 20-hyderoxyecdysone의 영향)

  • Studies were carried out to elucidate the efficacy of exogeneous 20-hydroxyecdysone treatment on terminating pupal diapause in the fall webworm. Hyphantria cunea Drury. And the difference was also investigated between normal adult development and post-diapause development after 20-hydroxyecdysone treatment. In the diapause termination rate of pupae treated with 20-hydroxyecdysone after storage for various periods at 16L:8D and $25\pm$$1^{\circ}C$, at the highest rate was observed from the group stored for the longest period and the lowest rate from those stored for 1.5 months. The time needed for adult emergence was inversely proportional to the chilling (at $0\pm$$1^{\circ}C$) period, and the longer its exposure period at low temperature, the higher its sensitivity to 20-hydroxyecdysone treatment. Pupal diapause duration was almost the same, regardless of storage period in the total darkness or at the photoperiod of 16L:8D, and also they successfully emerged to adult even without any experience at low temperature. The oxygen consumption rate in normally developing pupae showed nearly a typical U-form. But, that of diapausing pupae treated with 20-hydroxyecdysone slowly increased and jumped 14 days after the treatment. Pupal diapause began before formation of adult tissues, an a timing of adult tissue formation coincided with ascending timing of the metabolic rate in both normally developing pupae and diapausing pupae treated with 20-hydroxyecdysone. The diapausing pupae treated with 20-hydroxyecdysone was similar to normally developing pupae in band patterns of proteins from haemolymph or fat body.

Hot Water Dipping Treatment to Improve Storage Quality of Green Red Pepper (풋고추의 저장성 향상을 위한 열처리 조건 구명 및 저장 중 품질 변화)

  • We investigated the efficacy of heat shock treatments to improve storage quality of green red pepper. Green red pepper were stored at 10$^{\circ}C$ for 4 weeks after no treatment (control) or hot water dips at 50-65$^{\circ}C$ for 10s. Treatments at 65$^{\circ}C$ generally caused high levels of heat injury on the surface of fruit. The hot water dips had no effect on firmness, Hunter ‘a’ and ‘b’ values and viable cell count by 3 weeks of storage. But color of fruit changed toward darker and redder direction and firmness was decreased by heat treatment at 4 weeks of storage. The contents of capsaicin and dihydrocapsaicin were not caused significant influence by different heat treatment, were decreased through the storage. The optimal hot water dip condition for maintaining fruit quality after prolonged storage was found to be 55$^{\circ}C$ for 10s, which can lead to reduced chilling sensitivity of green red pepper.
