Attracting effect of baits used the by-product for swimming crab Portunus trituberculatus pots (부산물을 이용한 꽃게 통발용 미끼의 유인 효과)

  • In order to develop the substitutive materials for natural baits of swimming crab pots, the attracting effects of swimming crab such as the preference of baits which were made of the by-products of marine and stock raising through the water tank experiments and fishing experiments. On the investigation of mean entrapped catch number to the pot by the baits after putting the 4 kinds of baits, mackerel(M), mackerel with grinded mackerel s internals($MM_I$), mackerel with tuna s internals$MM_I$) and makerel with grinded krill(MK) each in one pot by turns, $MM_I$ and MK were entrapped mean 3.9(13.0%) and they were a little more comparing to M, and $MT_I$ is least with mean 2.1(7.0%)(F=12.913, P < 0.05). Otherwise, on the preference investigation of swimming crabs by the baits after putting the 4 kinds of baits in the 4 pots each, M was entrapped mean 3.0(10%), but $MM_I$, $MT_I$ and MK were mean 1.2(4.0%), 1.0(3.3%) and 1.5(5.0%) each and they were only 30-50% of M(F=13.398, P < 0.05). On the preference investigation of swimming crabs by the 5 kinds of baits, mackerel(M), and krill(K), manila clam($M_C$), pig s fat($P_F$) and chicken s head($C_H$) which were used in substitutive baits, M was entrapped mean 3.2(10.7%), but K was about 50% of catch of M with mean 1.6(5.3%), and $M_C$, $P_F$ and $C_H$ were very few with mean 0.1-0.2(0.3-0.7%)(F=89.186, P < 0.05). On the preference investigation of swimming crabs by the pots which were put each the 3 kinds of baits, original krill(K), grinded krill with gluten and soybean oil cake($K_GGS$) and grinded krill with gluten, soybean oil cake and glycine($K_GGSG_L$) in the blue fluorescent hexahedral plastic bait cages(BF), and which were put the mackerel(M) in the non-fluorescent hexahedral red plastic bait cage($RF_N$), it was entrapped mean 3.0(10.0%) in the pot which was put the mackerel in the $RF_N$, and the same level in the pots which were put the K and $K_GGSG_L$ in the BF, but it was mean 2.0(6.7%) in the pots which was put the $K_GGS$ in BF and it was decreased by 30% of catch comparing to $RF_N$(F=3.750, P < 0.05). On the preference investigation of swimming crab by the pots which was put grinded tuna with gluten, soybean oil cake and glycine($T_IGSG_L$) in the blue fluorescent hexahedral plastic bait cage(BF), and which was put mackerel(M) in the nonfluorescent hexahedral red plastic bait cage($RF_N$), it was entrapped mean 3.3(11.0%) in the pot which was put mackerel in $RF_N$, and mean 2.7(9.0%) in the pot which was put $T_IGSG_L$ in BF and it was about 15% less comparing to use bait M(t=1.387, P < 0.05). As a results of fishing experiments, a plan for enhancing catching efficiency of $T_IGSG_L$ will be required because catching efficiency of $T_IGSG_L$, alternative bait, was half of fish catching efficiency of natural bait using mackerel. Fishing experiments were conducted 3 times using reinforced substitutive artificial bait that is reinforced attractive effect of $T_IGSG_L$ and composed of tuna intestine, grinded mackerel, gluten, soybean cake, glycine and alanine($T_IM_GGSG_LA$). Catching efficiency of $T_IM_GGSG_LA$ was about 80% of that of natural bait made of mackerel.

Application for Identification of Food Raw Materials by PCR using Universal Primer (일반 프라이머를 이용한 PCR의 식품원료 진위 판별에 적용)

  • In order to determine an authenticity of food ingredient, we used DNA barcode method by universal primers. For identification of animal food ingredients, LCO1490/HCO2198 and VF2/FISH R2 designed for amplifying cytochrome c oxidase subunit1 (CO1) region and L14724/H15915 for cytochrome b (cyt b) region on mitochondrial DNA were used. Livestock (cow, pig, goat, sheep, a horse and deer) was amplified by LCO1490/HCO 2198, VF2/FISH R2 and L14724/H15915 primers. Poultry (chicken, duck, turkey and ostrich) was amplified by LCO1490/HCO 2198 and VF2/FISH R2 primers. But, Fishes (walleye pollack, herring, codfish, blue codfish, trout, tuna and rockfish) were only amplified by VF2/FISH R2 primers. For plant food ingredients, 3 types of primers (trnH/psbA, rpoB 1F/4R and rbcL 1F/724R) have been used an intergenic spacer, a RNA polymerase beta subunit and a ribulose bisphosphate carboxylase region on plastid, respectively. Garlic, onion, radish, green tea and spinach were amplified by trnH/psbA, rpoB 1F/4R and rbcL 1F/724R. The PCR product sizes were same by rpoB 1F/4R and rbcL 1F/724R but, the PCR product size using trnH/psbA primer was different with others for plants each. We established PCR condition and universal primer selection for 17 item's raw materials for foods and determine base sequences aim to PCR products in this study. This study can apply to determine an authenticity of foods through making an comparison between databases and base sequences in gene bank. Therefore, DNA barcode method using universal primers can be a useful for species identification techniques not only raw materials but also processed foods that are difficult to analyze by chemical analysis.

  • In trying to predict the effect of genetics on the broiler in the year 2000, this is a relatively short period of time as far as broiler genetics in concerned. Modern broiler genetics started around 1945 and tremendous gains when made in past 35 years. Futher improvements on broiler will depend on the evolution and revolution: 1. Evolution: (1) Growth rate has been made 4-5% per year. (2) Feed conversion has improved approximately 1% per year. (3) Abdominal fat is becoming a major complaint in broiler. (4) Because of the changing life-style, broiler meat sales in the future will be more and more in cut-up form. (5) Breeding for stress resistance and selection for docile temperament can be important in order to funker improve fled efficiency. (6) In female parent stock, reproduction characteristics are in many can negatively correlated with the desired broiler traits. (7) Egg production and hatchability in moot commercial parent nod m at a fairly high level. (8) In male parent stock, the heavier and mon super-meat-type male lines are desired to Product better broilers. 2. Revolution: Trying to forecast revolutionary change in broiler genetics is highly speculative, as sudden change are aften unpredictable. (1) Species hybridization, such as a turkey-chicken cross (2) Biochemical tools, such as blood typing. (3) Mutation breeding by radiation or chemical mutagentia. (4) Broiler breeding would be to change the phenotypic appearance by single gene, such as naked, wingless.   • In this study, effective gene extraction methods were compared to identify raw materials of processed meat products through molecular biological methods. Species specific primers were used to identify ingredients of processed foods and, as a sample, 13 kinds of processed meat products including beef, pork and chicken. According to the type of sample, 13 kinds of samples were classified into liquid type, source type and powder type. The samples were pre-treated (centrifugation) and (or) performed Whole Gene Amplification (WGA) kit for amplification of the extracted DNA. As a result, it was possible to identify the raw material of products through the centrifugation of sample 1 ml for liquid type of processed meat products. For source type of products after gene extraction, it was required to perform WGA for the identification of ingredients. For powder type products did not required any further pre-treatment and WGA. In this study, it was an opportunity to confirm the possibility of identification of raw material from the gene extraction of processed meat products and this method could be used to examine the authenticity of raw material of products.

The Study on Consummer Behaviour of Poultry meat and Egg (닭고기와 계란의 소비에 대한 조사 연구)

  • The purpose of this research is to analyze the tendencies of poultry consumption in Korea. The information used is the data collected three times during the period from Sept. 1, 1985 to April 30, 1987 throughout the major cities. Those who participated in the survey are 2, 598 including housewives, nutritionists, cookers, group feeding institutions, woman's organizations, and the students of doing nutrition-related studies. Consumer preferences for poultry meat and eggs tend to move toward high quality and more strict sanitary standards. Following this line of consumer preference changes, the poultry product marketing supported by cold chain system is rapidly developing. Household consumption of poultry meat largely consists of hi-and semi-broilers but the household demand for these two broilers tends to decrease steadily over time. In general poultry meat consumption appears to be affected by consumer taste rather than market factors such as prices. In addition consumer choices are quite different depending on poultry meat parts which are preferred in order of drum sticks, wings, and breasts. In particular drum sticks are most preferred than any other parts. An important problem in poultry production is related to consumption seasonality since large part of poultry meat consumption is concentrated during the summer season. Another problem is associated with little development of cooking methods. At present there are two types of primary commercial cooking techniques, fries and samgaetang (boiled chicken with jinsang and rice). For promoting domestic poultry meat disposal and reducing the demand seasonality, new cooking methods should be developed and followed by more aggresive advertisements. In domestic egg trade, smaller packing units(i.e., 10 eggs per unit) tend to bi preferred to large ones (i.e., 30 eggs per unit). In consumers egg purchasing decisions nearness to the shops and convenience appear to be important factors. For egg shell colors consumers recognize that there is no difference in nutritional values. However, survey results show that consumers highest preference lies in eggs with brown color. Eggs are most popular among children and preferred in order of middle-and high-school students, 17-25 age people, and adults. Egg prices are concieved relatively cheap to its nutritional values. In house-holds eggs are consumed in the forms of fries, side dishes, and lunch basket dishes. However, high level of cholesterol content in eggs appears to be an important problem in promoting eggs consumption.

Temperature Changes during Freezing and Effect of Physicochemical Properties after Thawing on Meat by Air Blast and Magnetic Resonance Quick Freezing (식육의 송풍식 냉동과 전기자장 냉동 중 온도 변화 및 이화학적 품질에 미치는 영향)

  • This study was carried out to investigate the effects of various temperatures of electro-magnetic resonance and air blast freezing methods on the physicochemical quality of meat. Beef (loin and round), pork (belly and ham) and chicken (breast and leg) were purchased at a commercial market, and the meat was frozen using three methods: air blast freezing ($-20^{\circ}C$ and $-45^{\circ}C$) and electro-magnetic resonance quick freezing. Changes in the physicochemical properties of meat were analyzed by drip loss, cooking loss, water holding capacity (WHC) and proximate compositions. In comparison, regardless of the animal species and cuts of meat, electro-magnetic resonance quick freezing (2 h) resulted in a completely frozen product in a much shorter time than $-20^{\circ}C$ and $-45^{\circ}C$ air blast freezing (24 h and 8 h, respectively). Drip loss of loin which had underwent electro-magnetic resonance quick freezing were significantly (p<0.05) lower than those of the other two treatments, but cooking loss and water holding capacity were the highest at 43.7% and 60.7%, respectively (p<0.05). Characteristics such as crude protein, crude fat and moisture compositions showed significant differences, depending on the cuts and freezing methods (p<0.05). The fat composition of electro-magnetic resonance quick frozen loin and round were significantly low (p<0.05). However, moisture content was the highest compared to other freezing methods, as 67.1% and 71.9%, respectively (p<0.05). Electro-magnetic resonance quick freezing was an appropriate way to reduce the deterioration of meat quality due to freezing, and the drip loss was least for the part with low moisture, low protein, and high fat.

Analysis of Consumer Attitudes to Washed Fresh Ginseng (세척 및 시판 수삼 이용실태와 소비자 인식도 분석)

  • We focused on the condition of washed, fresh ginseng in the market and analyzed consumer attitudes to the product to improve the consumption of fresh ginseng. A preference for fresh ginseng was high among every age class, and was particularly notable in older individuals. Fresh ginseng was usually eaten as ginseng chicken soup, mostly in summer and winter, and as a valuable health food, rather than because of desirable qualities of taste or smell. A total of 66.5% of those surveyed reported that they had purchased fresh ginseng, mostly from big discount stores. Such stores were preferred as outlets owing to a reputation for quality and shopper convenience (the stores were located near interviewees' homes). More than 70% of purchasers emphasized that they chose to purchase in department stores and traditional markets owing to the reputation for quality of such sources. Most consumers considered quality to be a prime element in purchase decisions. A total of 32.9% of those surveyed had bought washed fresh ginseng to give as a gift to a friend. Of those surveyed, 67.1% had never bought washed fresh ginseng. The following reasons were proposed to explain the preference for purchase of ginseng with attached soil. Most consumers (40.2%) thought the soil was a reflection of quality. Those who preferred washed fresh ginseng considered that the material was hygienic and convenient to store. When questioned about their intention to buy washed fresh ginseng, 55.0% of consumers replied in the affirmative and 17.1% in the negative. When consumers were asked whether they would be willing to pay an additional charge for the washing and packing of fresh ginseng, 58.8% answered in the affirmative.
