Genetic Variability and Relationships of Native Japanese Chickens Assessed by Microsatellite DNA Profiling - Focusing on the Breeds Established in Kochi Prefecture, Japan -

  • Blood samples were collected from eight native Japanese breeds of chickens (Miyadi-dori, Ohiki, Onaga-dori, Shoukoku, Tosa-Jidori, Tosa-Kukin, Toutenkou and Uzurao) and two foreign breeds of chickens (White Leghorn and Rhode Island Red) to examine the genetic variability and relationships among the breeds by using a microsatellite DNA technique. Except for the Shoukoku breed, the other Japanese chicken breeds all originate from Kochi Prefecture. Ohiki, Onaga-dori, Tosa-Jidori, Toutenkou and Uzurao are fancy fowl, and Miyadi-dori and Tosa-Kukin are utility fowl. Among the fancy fowl, Ohiki, Onaga-dori, and Toutenkou males have thick and long feathers in the saddle and tail. Genetic variabilities of the 20 microsatellites examined, varied depending on the breed: the mean number of alleles per locus ranged from 2.05 (Miyadi-dori) to 3.90 (Rhode Island Red); proportion of polymorphic loci ranged from 0.75 (Miyadi-dori) to 1.00 (Rhode Island Red, Shoukoku and Uzurao); and mean expected heterozygosity ranged from 0.330 (Miyadi-dori) to 0.607 (Rhode Island Red). Unique microsatellite alleles were detected in each breed. Using the neighbour-joining method, phylogenetic trees were constructed based on the genetic distances of D$_{A}$ and D$_{ST}$. Among the breeds originating from Kochi Prefecture, fancy and utility breeds belonged to different clusters. Among the fancy breeds, those having thick and long feathers in the tail and saddle showed a close genetic relationship to the Shoukoku breed, which also has thick and long feathers in the tail and saddle.

Effect of Hybridization on Carcass Traits and Meat Quality of Erlang Mountainous Chickens

  • Native chickens hold a significant share of the market in China. In response to the huge demand from the market, the productivity of Chinese native chickens needs to be improved. Cross breeding is an effective method to increase productivity, although it might affect meat quality. In this study, two pure lines (SD02 and SD03) of Erlang mountainous chickens were hybridized with a yellow feather and faster growing line (SD01). The effect of hybridization on carcass and meat quality (physiochemical and textural traits) was measured in the $F_1$ population at d 91 of age. The hybrids exhibited higher body weight and dressed weight, and amount of semi-eviscerated, eviscerated, breast muscle and abdominal fat (p<0.05). Abdominal fat yield also increased (p<0.05) compared to the offspring of the two pure-lines. Meanwhile, there was no significant difference in meat quality traits except for the myofiber diameter and density and the shear force of the breast muscle. Overall, the offspring of cross-lines were similar to pure lines in meat color, pH value, inosinic acid, crude protein, crude fat, dry matter, moisture content and amino acid composition in the breast muscle. These results suggest that productivity can be improved via cross-breeding while maintaining meat quality of the Erlang mountainous chicken.

Effect of pre-miRNA-1658 gene polymorphism on chicken growth and carcass traits

  • Objective: Polymorphisms occurring in the precursor region of microRNAs (miRNAs) affect the target gene and alter the biogenesis of miRNAs, resulting in phenotypic variation. The purpose of the study was to investigate the genetic effects of rs16681031 (C>G) mutation in the precursor region of gga-miR-1658 on the economic traits of the Gushi-Anka chicken F2 resource population. Methods: To explore the effect of miR-1658 polymorphisms on chicken economic traits, the SNP was genotyped by MassArray matrix-assisted laser desorption/ionization-time of flight mass spectrometry. The association between the SNP and chicken body size, growth and carcass traits was determined by linear mixed models. Results: The SNP was not only significantly associated with body weight at the age of 6, 8, 10, 12 weeks, respectively, but also with the breadth of the chicken chest, body slanting length and pelvic breadth at 4 weeks, chest depth at 8 weeks of age, and body slanting length at 12 weeks (p<0.05), respectively. Conclusion: Our data serve as a useful resource for further analysis of miRNA function, and represent a molecular genetic basis for poultry breeding.

Analysis on Association of a SNP in the Chicken OBR Gene with Growth and Body Composition Traits

  • Leptin receptor (OBR) is a member of the class I cytokine receptor family. It signals mainly via the JAK/STAT pathway and plays an important role in regulating body energy storage and metabolism. This study was designed to investigate the effects of the OBR gene on chicken growth and body composition. Broiler lines selected divergently for or against abdominal fat were used. Primers for the exon9-region in the OBR gene were designed using chicken genomic sequences from the public genome domain. A C/A single nucleotide polymorphism (SNP) was found and its three genotypes (AA, AB and BB) were identified in this population. The results showed that the OBR polymorphism was associated with fatness traits, such as abdominal fat weight and abdominal fat percentage. This research suggests that OBR or a linked gene has effect on fat deposition in the chicken.

닭에서 품종 확인을 위한 DNA 마커에 관한 고찰 (DNA markers in chicken for breed discrimination)

  • There is an emerging interest in using DNA markers for breed identification in animals. This article reviews the breed identification markers in chicken, mainly developed in Chungnam National University, with particular emphasis on the mitochondrial DNA markers and the nuclear DNA markers including the SNPs in MHC region and the plumage color related MC1R markers. This information would be very useful for an appropriate conservation breeding program as well as for the establishment of molecular markers for chicken breed identifications.

  • 열교환기 환기장치는 실내로부터 배출되는 고온의 배기류를 이용하여 내부 환기를 위하여 투입되는 차가운 외기를 가온시킨다. 이를 통해 환기로 인해 발생되는 에너지 비용의 절약을 도모할 수 있고, 차가운 외기의 유입으로 인한 열적 균일성 감소 및 그에 기인한 호흡기성 질병 발생을 미연에 방지하는 효과가 있다. 본 연구에서는 열교환기 환기장치의 원격제어 및 원격계측을 위한 제어 및 계측 시스템을 개발하고 개발된 열교환기 환기장치의 제어 및 계측 시스템을 육계 사육장에 설치하여 내부의 온도와 습도의 원격제어 및 원격계측을 통해 성능검증을 수행하였다. 성능검증실험에서 육계 사육장 내부에서 2도 이내의 온도 제어가 가능함을 확인하였다.

Quantitative Trait Loci Affecting Rous Sarcoma Virus Induced Tumor Regression Trait in F2 Intercross Chickens

  • We performed a genome-wide linkage and quantitative trait locus (QTL) analysis to confirm the existence of QTL affecting Rous Sarcoma Virus (RSV) induced tumor regression, and to estimate their effects on phenotypic variance in an F2 resource population. The F2 population comprised 158 chickens obtained by crossing tumor regressive White Leghorn (WL) and tumor progressive Rhode Island Red (RIR) lines was measured for tumor formation after RSV inoculation. Forty-three tumor progressive and 28 tumor regressive chickens were then used for genome-wide linkage and QTL analysis using a total of 186 microsatellite markers. Microsatellite markers were mapped on 20 autosomal chromosomes. A significant QTL was detected with marker LEI0258 located within the MHC B region on chromosome 16. This QTL had the highest F ratio (9.8) and accounted for 20.1% of the phenotypic variation. Suggestive QTL were also detected on chromosomes 4, 7 and 10. The QTL on chromosome 4 were detected at the 1% chromosome-wide level explaining 17.5% of the phenotypic variation, and the QTLs on chromosome 7 and 10 were detected at the 5% chromosome-wide level and explained 11.1% and 10.5% of the phenotypic variation, respectively. These results indicate that the QTLs in the non-MHC regions play a significant role in RSV-induced tumor regression. The present study constitutes one of the first preliminary reports in domestic chickens for QTLs affecting RSV-induced tumor regression outside the MHC region.

육계사료에 항생제 첨가 효과와 경제성에 관한 조사연구 II (the additional effects of antibiotics for the chicken breeding feed and economical efficiency.)

  • The purpose of this experiment is to clarify the additional effect of antibiotics for the chicken breeding feed. This experiment was carried out with 6,560 individuals which were collected from four farms, Dugil, Hyubhoi, Sanggog and Kumsan. The duration of this experiment was 6 months from July to December in 1981. 1. Body weight and body gain. (1) The range of the body gain was 1.91-7.60% The most effective sections was T$_3$, and the effect was T$_2$, T$_4$, T$_1$, T$\sub$5/ in order. (2) The environmental sanitation of the Sanggog farm was not so good. In this farm the body gain was severely occurred in T$_3$ and T$_2$ sections. Mere gain obtained from T$\sub$5/ section. (3) The environmental sanitation was good in Dugil, Hyubhoi and Kumsan. A little body gain was occurred in these farms, but differences were shown according to the place or breeds. 2. Amount of feed intake feed conversion ratio. (1) Amount of feed intake was increased 0.13-3.19%, and feed conversion ratio was improved 1.68-4.62% compared with control. T$_3$ section showed high percent and T$_4$ T$_1$, T$\sub$5/, Is, C in order. (2) The keeping method of the chicken breed in Hyubhoi was floor brooding. From Hyubhoi and Sanggog farms, the efficiency was increased 9.4-9.8%. (3) From Dugil and Kumsan, the rate of improvement was low. 3. Mortality : Mortality was merely decreased in Sanggog and Kumsan farms which the sanitary and fowl caring conditions had been had, by additional antibiotics. But there was no worthy effect. 4. The ratio of carcass and the degree of skin coloration. The additional antibiotics in feed had no effect on the ratio of carcass but had the influence on the yellowish skin coloration. Skin coloring was good in T$\sub$7/ section and T$_1$, T$\sub$6/, T$_4$, C, T$_3$, T$\sub$5/, T$_2$ in order. 5. Economical efficiency (1) The addition of the antibiotics in feed increased 3.76-10.95% of gross earning. The highest earning was occurred in T$_4$ section and T$_3$, T$_1$, T$_2$, T$\sub$5/, C sections in order. (2) The earning rate was influenced by the antibiotic products. (3) In Dugil and Kumsan farms, there was no difference in earning, however, in Hyubhoi and Sanggog farms there was wide difference in earning. Conclusionally, the addition of antibiotics in chicken breeding feed is necessary. Especially in bad condition of environmental sanitation and floor brooding, the effect of additional antibiotics is clear. Selection of antibiotics from various kinds, maker's confidence and quality problems are important.

OCX-32 유전자 내 c.494A>C 및 c.267T>G SNP이 한국 재래닭 산란형질에 미치는 효과 분석 (Effects of c.494A>C and c.267T>G SNPs in OCX-32 Gene of Korean Native Chicken on Egg Production Traits)

  • 가금 사육 프로그램에서 경제적으로 중요한 형질에 잠재적인 후보 유전자의 식별 및 활용은 점점 더 중요해지고 있다. Ovocalyxin-32(OCX-32) 유전자는 닭의 9번 염색체에 위치하며, 난각을 형성하는데 중요한 역할을 한다. 본 연구의 목적은 한국 재래닭의 OCX-32 유전자 내 SNP의 유전자형 결정과 산란 형질과의 연관성을 분석하기 위해 수행하였다. PCR-RFLP 방법을 통해 한국 오골계 46수, 백색 46수, 회색 43수, 흑색 46수를 포함한 총 181수의 한국 재래닭 암컷 4종의 SNP을 분석하였다. 산란 형질은 시산일령, 시산난중, 산란율, 난중의 4가지 항목을 포함하여 측정하였다. OCX-32 유전자 내 c.494A>C SNP은 오골계의 산란율과 유의적인 차이를 나타냈으며(p<0.001), 백색 재래닭에서는 난중과 유의적인 상관관계가 있었다(p<0.05). c.267T>G SNP은 오골계의 난중과 유의적인 연관성이 나타났다(p<0.05). 하지만 회색과 흑색 재래닭에서는 유의적인 상관관계가 나타나지 않았다. 본 연구 결과, 한국 재래닭의 사육 프로그램에서 산란형질을 선발하는 마커로 사용되기까지 추후 더 많은 개체군을 통한 연구가 요구되나, OCX-32 유전자 내 c.494A>C와 c.267T>G의 단일염기변이가 한국 재래닭 중 오골계와 백색 재래닭에서 산란 특성에 따른 DNA 선발 마커로서 활용할 수 있을 것으로 사료된다.

전북지방 닭의 내부기생충 감염상황 (Prevalence of parasite infection of chickens in Jeonbuk province)

  • In order to detection of the intestinal parasites, fecal samples were taken from broiler (n=290), parent stock (n=168) and laying hen (n=114) in Jeonbuk province. The prevalence and identification of intestinal parasites were determined by the fecal examination using the floatation method and microscopical examination, respectively. The detection rate of parasite-eggs from 3 flocks (total=572) was 44.9%. In the breed and type of breeding, infection rate of parasite-eggs was detected 65.5% as broiler (floor breeding, 290 chickens), 20.2% as parent stock (floor breeding, 168 chickens) and 28.9% as laying hen (cage breeding, 114 chickens), in order. In the concern of mixed infection such as single and double, the rate were 40.7% and 4.2%, respectively. Six kinds of infective eggs were isolated 257 fecal sample from 3 flock. They were classified 74.7% as Eimeria spp, 18.1% as Ascaridia galli and 6.0% as Capillaria spp and 0.4% as Heteratkis gallinarum, Railleina spp or Trichostrongylus spp, single or in combination.
